During in door or outdoor mass culture, Porphyra have been easily contaminated with bacteria, protozoa and microalgal species. Several axenic treatments for Porphyra thalli have been published (Polne-Fuller and Gibo 1984; Chen and McCracken 1993), but axenic techniques for neutral spores and conchocelis we not developed. Microalgae research is gaining momentum because of their potential biotechnological applications, including the generation of biofuels. Genome sequencing analysis of two model microalgal species, polar free-living Coccomyxa sp. C-169 and symbiotic Chlorella sp. NC64A, revealed insights into the factors responsible for their lifestyle and unravelled biotechnologically valuable proteins. However, genome sequence analysis under-explored cytochrome P450 monooxygenases (P450s), heme-thiolate proteins ubiquitously present in species belonging to different biological kingdoms. In this study we performed genome data-mining, annotation and comparative analysis of P450s in these two model algal species. Sixty-nine P450s were found in two algal species. Coccomyxa sp. showed 40 P450s and Chlorella sp. showed 29 P450s in their genome. Sixty-eight P450s (>100 amino acid in length) were grouped into 32 P450 families and 46 P450 subfamilies. Among the P450 families, 27 P450 families were novel and not found in other biological kingdoms. The new P450 families are CYP745-CYP747, CYP845-CYP863, and CYP904-CYP908. Five P450 families, CYP51, CYP97, CYP710, CYP745, and CYP746, were commonly found between two algal species and 16 and 11 P450 families were unique to Coccomyxa sp. and Chlorella sp. Synteny analysis and gene-structure analysis revealed P450 duplications in both species. Functional analysis based on homolog P450s suggested that CYP51 and CYP710 family members are involved in membrane ergosterol biosynthesis. CYP55 and CYP97 family members are involved in nitric oxide reduction and biosynthesis of carotenoids. This is the first report on comparative analysis of P450s in the microalgal species Coccomyxa sp. C-169 and Chlorella sp. NC64A.
Since $CO_2$ concentrations in industrial flue gases are usually 10%-20%, one of the prerequisites for efficient $CO_2$ removal by algae is the level of tolerance of microalgal species to exposure to high concentrations of $CO_2$. A newly isolated microalgal strain, Chlorella sp. MRA-1, could retain growth with high concentrations of $CO_2$ up to 15%. The highest lipid productivity for Chlorella sp. MRA-1 was 0.118 g/l/day with a 5% $CO_2$ concentration. Octadecenoic acid and hexadecanoic acid, the main components of biodiesel, accounted for 70% of the total fatty acids. A lipid content of 52% of dry cell weight was achieved with limited amounts of nitrogen. Chlorella sp. MRA-1 seems to be an ideal candidate for biodiesel production when cultured with high concentrations of $CO_2$.

Three new species of the genus Micractinium were collected from five localities on the South Shetland Islands in maritime Antarctica, and their morphological and molecular characteristics were investigated. The vegetative cells are spherical to ellipsoidal and a single chloroplast is parietal with a pyrenoid. Because of their simple morphology, no conspicuous morphological characters of new species were recognized under light microscopy. However, molecular phylogenetic relationships were inferred from the concatenated small subunit rDNA, and internal transcribed spacer (ITS) sequence data indicated that the Antarctic microalgal strains are strongly allied to the well-supported genus Micractinium, including M. pusillum, the type species of the genus, and three other species in the genus. The secondary structure of ITS2 and compensatory base changes were used to identify and describe six Antarctic Micractinium strains. Based on their morphological and molecular characteristics, we characterized three new species of Micractinium: M. simplicissimum sp. nov., M. singularis sp. nov., and M. variabile sp. nov.
The variations of species compositions, standing crops and seasonal succession of phytoplanktons including Cyanophyceae, Chlorophyceae and Bacillariophyceae were investigated with physico-chemical elements of water to clarify the causes of water bloom according to the water depth in the Lake Unmun. The increased amounts of turbidity, T-N, T-P and SS originated from heavy rain and typhoon in the middle of June provoked to produce phytoplanktons. In July and August, the dominant species was Peridinium sp., while the subdominant species was Microcystis aeruginosa. In October, Aulacoseira distans was dominant and Asterionella formosa was subdominant. During the period of water shortage, the concentrated pollutants caused the decrease of precipitation, the long stagnation time of water body and the mixed pollutants by upwelling as decreasing water temperature could accelerate the water bloom. The preventives to decrease microalgal generation should be controled by the stagnation time of water body by increasing outflow to intercept water bloom such as ulacoseira sp. in October as well as summer.
A taxonomic survey of the dinoflagellate family Dinophysiaceae Stein was conducted on 17 locations off the coast of Korea. A total of twelve species have been identified and described, of which eight species, Dinophysis dens Pavillart D. infundibulus Schiller, D. irregulare Lebour, D. lapidistrigiliformis Abe, D. mitra(Schutt) Abe vel Balech, D. parvula(Schutt) Jorgensen, D. rapa(Stein) Balech, Phalacroma sphaeroideum Schiller, are new records for Korea and six are potentially toxic.

The combination of Simian Virus40 (SV40)'s large T antigen with its replication origin is commonly used in molecular studies to enhance the expression of heterogeneous genes through multiplying the plasmid copy number. There are no reports related to the impact of the SV40 T antigen on plant, multiple fissional, cell-type. This study explores the response of two multiple-fission microalgal cells, Scenedesmus quadricauda and Chlorella vulgaris, to the expression of the T-antigen, with aim of applying SV40 T-antigen to increase the expression efficiency of foreign genes in the two species. Different levels of low-expression have been constructed to control the expression of SV40 T antigen using three heterogenous promoters (NOS, CaMV35S, and CMV). Chlorella cultures showed slowdown in the growth rate for samples harboring the T antigen under the control of CaMV35S and CMV promoters, unlike Scenedesmus cultures which showed no significant difference between samples and could have silenced the expression.

A eukaryotic marine microalga was isolated from Jungmun Saekdal Beach, Jeju Island, Korea and an integrated approach, including molecular phylogeny and morphology, was used to determine its taxonomical status. Molecular phylogenetic evidence inferred from the small subunit (SSU) 18S rRNA sequence and internal transcribed spacer (ITS) secondary structure analysis clearly showed that the isolate belonged to the recently described species, Jaagichlorella roystonensis. Distinctive morphological keys of the species were also observed by light microscopy and scanning/transmission electron microscopy(S/TEM). In this study, a Korean marine J. roystonensis species was described for the first time and was subsequently added to the national culture collections in Korea.
Marine microalgae have different collection methods depending on their habitat. In the case of adhesive microalgae, it is difficult to separate organisms from the substrate surface, and contamination is likely to occur during the sampling process. In this study, we analyzed the collection efficiency of adhesive microalgae using three artificial fiber materials (nylon, blend fabric, and viscose rayon). Each fiber showed different fiber diameter and pore characteristics (nylon 26.09 ㎛, blend fabric 56.6 ㎛, viscose rayon 101.3 ㎛). In addition, attached organisms were collected on the surface of artificial substrates at Bukseong Port in Incheon using each tested fiber material. After that, we investigated the population and species composition. The highest number of cells was found in nylon, which was 8 times higher than in the least collected viscose rayon material. In addition, we identified 24 microalgal species from the substrate, demonstrating that the species composition differed from that of surface water. The number of collected microalgae species varied depending on the fiber materials, with nylon containing all the adhesive microalgae. In contrast, only a few microalgae were observed in other fibers. These results suggest that, of the tested fibers, nylon material may be suitable for collecting adhesive microalgae. As a result, this study may be useful for future research on adhesive microalgae.
