This study investigated the effects of the light conditions on the productivity of scenedesmus dimorphus in the continuous mass cultivation system. To compare the algal productivity according to the light conditions, S. dimorphus was cultivated continuously under the wide range of light intensity(200-600 PPFD) and various light wavelength(white light and red-blue mixed light). After 100 days of cultivation under the different light intensity, the productivity of S. dimorphus increased as light intensity decreased. So, the productivity was maximized as 100 mg/L/d when light intensity was 200 PPFD. In case of light wavelength, the productivity of S. dimorphus was enhanced about 20% with the white light compared to that of the red-blue mixed light. Consequently, the optimal light conditions for the continuous mass cultivation of S. dimorphus were 200 PPFD as light intensity and white light as light wavelength.
Heavy reliance on fossil fuels has been associated with increased climate disasters. As an alternative, microalgae have been proposed as an effective agent for biomass production. Several advantages of microalgae include faster growth, usage of non-arable land, recovery of nutrients from wastewater, efficient CO2 capture, and high amount of biomolecules that are valuable for humans. Microalgae Chlorella spp. are a large group of eukaryotic, photosynthetic, unicellular microorganisms with high adaptability to environmental variations. Over the past decades, Chlorella has been used for the large-scale production of biomass. In addition, Chlorella has been actively used in various food industries for improving human health because of its antioxidant, antidiabetic, and immunomodulatory functions. However, the major restrictions in microalgal biofuel technology are the cost-consuming cultivation, processing, and lipid extraction processes. Therefore, various trials have been performed to enhance the biomass productivity and the lipid contents of Chlorella cells. This study provides a comprehensive review of lipid enhancement strategies mainly published in the last five years and aimed at regulating carbon sources, nutrients, stresses, and expression of exogenous genes to improve biomass production and lipid synthesis.

Microalgae are currently a very promising source of biomass and triacylglycerol (TAG) for biofuels. In a previous study, we identified Chlorella saccharophila as a suitable source of oil for biodiesel production because it showed high biomass and lipid content with an appropriate fatty acid methyl esters profile. To improve the TAG accumulation in C. saccharophila, in this study we evaluated the effect of abscisic acid (ABA) addition on cell concentration, lipid content and TAG production in this microalga. First, we evaluated the effects of four ABA concentrations (1, 4, 10, and 20 μM) added at the beginning of a single-stage cultivation strategy, and found that all concentrations tested significantly increased cell concentration and TAG content in C. saccharophila. We then evaluated the addition of 1 μM ABA during the second stage of a two-stage cultivation strategy and compared it with a nitrogen deficiency treatment (ND) and a combination of ND and ABA (ND + ABA). Although ABA alone significantly increased lipid and TAG contents compared with the control, ND showed significantly higher TAG content, and ND + ABA showed the highest TAG content. When comparing the results of both strategies, we found a superior response in terms of TAG accumulation with the addition of 1 μM ABA at the beginning of a single-stage cultivation system. This strategy is a simple and effective way to improve the TAG content in C. saccharophila and probably other microalgae as a feedstock for biodiesel production.
Open raceway ponds are cost-efficient for mass cultivation of microalgae compared with photobioreactors. Although low-cost options like wastewater as nutrient source is studied to overcome the commercialization threshold for biodiesel production from microalgae, a cost analysis on the use of wastewater and other incremental increases in productivity has not been elucidated. We determined the effect of using wastewater and wavelength filters on microalgal productivity. Experimental results were then fitted into a model, and cost analysis was performed in comparison with control raceways. Three different microalgal strains, Chlorella vulgaris AG10032, Chlorella sp. JK2, and Scenedesmus sp. JK10, were tested for nutrient removal under different light wavelengths (blue, green, red, and white) using filters in batch cultivation. Blue wavelength showed an average of 27% higher nutrient removal and at least 42% higher chemical oxygen demand removal compared with white light. Naturally, the specific growth rate of microalgae cultivated under blue wavelength was on average 10.8% higher than white wavelength. Similarly, lipid productivity was highest in blue wavelength, at least 46.8% higher than white wavelength, whereas FAME composition revealed a mild increase in oleic and palmitic acid levels. Cost analysis reveals that raceways treating wastewater and using monochromatic wavelength would decrease costs from 2.71 to 0.73 $/kg biomass. We prove that increasing both biomass and lipid productivity is possible through cost-effective approaches, thereby accelerating the commercialization of low-value products from microalgae, like biodiesel.

High production and efficient harvesting of microalgae containing high omega-3 levels are critical concerns for industrial use. Aeration can elevate production of some microalgae by providing CO2 and O2. However, it may lower the production of others by generating shear stress, causing severe cell damage. The mixotrophic dinoflagellate Gymnodinium smaydae is a new, promising microalga for omega-3 fatty acid production owing to its high docosahexaenoic acid content, and determining optimal conditions and methods for high omega-3 fatty acid production and efficient harvest using G. smaydae is crucial for its commercial utilization. Therefore, to determine whether continuous aeration is required, we measured densities of G. smaydae and the dinoflagellate prey Heterocapsa rotundata in a 100-L semi-continuous cultivation system under no aeration and continuous aeration conditions daily for 9 days. Furthermore, to determine the optimal conditions for harvesting through centrifugation, different rotational speeds of the continuous centrifuge and different flow rates of the pump injecting G. smaydae + H. rotundata cells into the centrifuge were tested. Under continuous aeration, G. smaydae production gradually decreased; however, without aeration, the production remained stable. Harvesting efficiency and the dry weights of omega-3 fatty acids of G. smaydae + H. rotundata cells at a rotational speed of 16,000 rpm were significantly higher than those at 2,000-8,000 rpm. However, these parameters did not significantly differ at injection pump flow rates of 1.0-4.0 L min-1. The results of the present study provide a basis for optimized production and harvest conditions for G. smaydae and other microalgae.
The aim of this study was to evaluate a temperature-induced two-stage cultivation (TTC) strategy for the regulation of lipid and carbohydrate production by two microalgae, Chlorella vulgaris and Dunaliella salina, for biofuel production. The microalgae were grown under several temperature conditions (15, 25, 35, and $45^{\circ}C$) and optimal growth was observed at $25^{\circ}C$ for both microalgae. To test the TTC, aseptically cultured microalgae were incubated under optimal conditions ($25^{\circ}C$) for 20 days, and then divided into four aliquots that were incubated at 15, 25, 35, and $45^{\circ}C$ for 5 days. Similar but somewhat decreased growth rates were observed at the non-optimal temperatures (15, 35, and $45^{\circ}C$). In addition, while total lipid accumulation increased in a temperature-dependent manner in both microalgae, total carbohydrate increased with temperature in C. vulgaris but decreased in D. salina. However, for lipid and carbohydrate production, while the highest lipid productions of C. vulgaris and D. salina were observed at $25^{\circ}C$ and $35^{\circ}C$, respectively, the highest total carbohydrate productions of C. vulgaris and D. salina were obtained at $15^{\circ}C$ and $25^{\circ}C$, respectively. These results suggest that the TTC strategy may be easily and efficiently applied to bioprocessing for biofuel production.
본 연구 목적은 미세조류 대량생산을 위한 ICT와 BT기술이 융합된 계단식 연속 배양 시스템(CCCS)을 개발함에 있다. 본 시스템은 미세조류 성장에 필요한 주요 운전변수인 pH, 온도, 이산화탄소와 조도 제어를 통해 실시간 배양조건 유지를 통해 세포성장 증진과 계절과 장소에 관계없이 미세조류를 생산할 수 있어 경제성이 확보 된 시스템이다. 또한, 안정적이고 높은 생산성을 제공하는 장점을 갖는다. 본 연구에서는 이 시스템을 활용하여 71일간 미세조류를 배양하고 실험 데이터를 분석하였다. 그 결과, 배양초기 O.D.는 수조1에서 0.006로 측정되었으며 이후 배양 71일, O.D.는 수조1: 0.399, 수조2: 0.961, 수조3: 0.795 그리고 수조4: 0.438로 측정되었다. 따라서 배양기간 동안 연속적인 배양이 가능함이 확인되었다. 대량 배양 방식인 ISMC (In-situ monitoring and control)기반 스마트팜을 제시하고, 본 개발기술을 통해 미세조류 이외의 수경재배 기반의 약용식물 등에 적용하여 식품, 화장품 그리고 의료 소재용 고부가가치 식물 재배에도 적용이 가능한 상업화 적합 기술이라 사료된다.
최근, 미세조류에 대한 생물공학적 관심이 급격히 증가하고 있으며 이의 응용범위는 식품이나 제약, 화장품 등 다양한 구도로 확장되어지고 있다. 고농도의 미세조류 배양을 위해서는 빛이 핵심적 제한요소로 작용되어지며 빛의 투과 깊이나 강도에 따라 균체의 성장속도가 결정되어지게 된다. 본 연구에서는 다양한 빛의 투과경로와 빛을 받는 면적/배양액의 부피 비율, 조도 그리고 단계적 조사에 따른 Chlorella sp.의 성장률을 조사하여 빛이 미세조류에 미치는 영향을 알아보았으며, 본 연구에 적용된 값들 중 4 cm의 직경, 57.6%의 면적/부피 비율, 62 $\mu$mo1/$m^2$/s의 조도에서 Chlorella sp. 성장에 필요한 빛 에너지를 가장 효율적으로 이용함을 확인할 수 있었다.
