Molecular biology and microalgal biotechnology have the potential to play a major role in improving the production efficiency of a vast variety of products including functional foods, industrial chemicals, compounds with therapeutic applications and bioremediation solutions from a virtually untapped source. Microalgae are a source of natural products and have been recently studied for biotechnological applications. Efficient genetic transformation systems in microalgae are necessary to enhance their potential to be used for human health. A microalga such as Chlarella is a eukaryotic organism sharing its metabolic pathways with higher plants. This microalga is capable of expressing, glycosylating, and correctly processing proteins which normally undergo post-translational modification. Moreover, it can be cultured inexpensively because it requires only limited amount of sunlight and carbon dioxide as energy sources. Because of these advantages, Chlarella may be of great potential interest in biotechnology as a good candidate for bioreactor in the production of pharmaceutical and industrial compounds for human functional foods. Here, we briefly discuss recent progress in microalgal transgenesis that has utilized molecular biology to produce functional proteins and bioactive compounds.

Cell growth of the marine microalga Tetraselmis suecica was regulated by the addition of seaweed extracts in its culture medium. Of 26 species of seaweed tested, water-soluble extracts from Monostroma nitidum and Pachymeniopsis elliptica and methanol-soluble extract from Sargassum confilsum enhanced the growth of T suecica cells. The water extract of M. nitidum was the most effective, producing up to a 2-fold increase in cell density with the addition of 1 mg/mL of extract to the culture medium. Cell size, gross biochemical composition, fatty acids, and digestion efficiency all differed marginally between cultures of T suecica grown with and without the M. nitidum extract.
A highly $CO_2$ tolerant microalga, Chlorella KR-1, has been isolated and used to fix $CO_2$ from actual flue gas. Growth of Chlorella KR-1 with the supply of flue gas from a liquified natural gas boiler was comparable to that obtained with 10% $CO_2$. Chlorella KR-1 produced from $CO_2$ fixation using the flue has about 50% crude protein with balanced amino acid profiles. Toxicity was not detected when the microalga was used as a feedstuff for chicks. These results indicate that the KR-1 cells could be a favorable protein source for poultry.

Increasing demands on fossil fuel have led to the unprecedented attraction to microalgal biofuel as an alternative energy. In this study, we investigated growth and lipid productions of microalga Nannochloris oculata under various carbon dioxide or nitrogen source concentrations and irradiance conditions. Biomass production of N. oculata was highest under 2% $CO_2$ with 0.3 flow rate (vvm). In addition, biomass productivities were proportional to the concentration of nitrogen source, whereas lipid biosynthesis was suppressed under higher nitrogen concentration (up to 50 mg/L). High irradiation ($160{\sim}180\;{\mu}mol/m^2{\cdot}s$) enhanced growth rate and lipid production of N. oculata.

A series of experiments were carried out with three native strains of microalgae to measure growth rates, biomass, and lipid productivities. Scenedesmus sp. IMMTCC-6 had better biomass growth rate and higher lipid production. The growth, lipid accumulation, and carbon dioxide ($CO_2$) consumption rate of Scenedesmus sp. IMMTCC-6 were tested under different NaOH concentrations in modified BBM. The algal strain showed the maximum specific growth rate (0.474/day), biomass productivity (110.9 mg $l^{-1}d^{-1}$), and $CO_2$ consumption rate (208.4 mg $l^{-1}d^{-1}$) with an NaOH concentration of 0.005 M on the $8^{th}$ day of cultivation. These values were 2.03-, 6.89-, and 6.88-fold more than the algal cultures grown in control conditions (having no NaOH and $CO_2$). The $CO_2$ fixing efficiency of the microalga with other alternative carbon sources like $Na_2CO_3$ and $NaHCO_3$ was also investigated and compared. The optimized experimental parameters at shake-flask scale were implemented for scaling up the process in a self-engineered photobioreactor. A significant increase in lipid accumulation (14.23% to 31.74%) by the algal strain from the logarithmic to stationary phases was obtained. The algal lipids were mainly composed of $C_{16}/C_{18}$ fatty acids, and are desirable for biodiesel production. The study suggests that microalga Scenedesmus sp. IMMTCC-6 is an efficient strain for biodiesel production and $CO_2$ biofixation using stripping solution of NaOH in a cyclic process.

Ultrasonified extracts from seawater-based cultures of the microalga Spiyulina platensis were obtained using water and ethanol at 60 and 100$^{\circ}C$. The yield of the aqueous fraction of S. platensis extracted using ultrasonification was about 33.46%. The cytotoxicity against HEK293 and inhibition ratios of the cancer cell lines A549, AGS, MCF7, and Hep3B were measured using the sulforhodamine-B (SRB) assay. The cytotoxicity of all extracts at 1.0 mg/mL was below 26%. The cytotoxicity of the ultrasonified extracts from the seawater-based culture of the microalga Spirulina platensis was about 4% less than that of Spirulina platensis without ultrasonification. The inhibition ratio of cancer cell growth was approximately 80% for 1.0 mg/mL extracts. The inhibitory effect on cancer cell growth was greater for seawater containing ultrasonified Spirulina platensis extracts than for extracts without ultrasonification. The differentiation ratio of HL-60 cells was 160.9%. Densitometric analysis of Bcl-2 revealed that the ultrasonified extracts had greater anticancer activity than the extracts without ultrasonification.
The gene encoding squalene synthase (SQS) of the lipid-producing heterotrophic microalga Aurantiochytrium sp. KRS101 was cloned and characterized. The krsSQS gene is 1,551 bp in length and has two exons and one intron. The open reading frame of the gene is 1,164 bp in length, yielding a polypeptide of 387 predicted amino acid residues with a molecular mass of 42.7 kDa. The deduced krsSQS sequence shares at least four conserved regions known to be required for SQS enzymatic activity in other species. The protein, tagged with $His_6$, was expressed into soluble form in Escherichia coli. The purified protein catalyzed the conversion of farnesyl diphosphate to squalene in the presence of NADPH and $Mg^{2+}$. This is the first report on the characterization of an SQS from a Thraustochytrid microalga.
