  • An in vitro gas production technique was used in this study to elucidate the effect of two strains of active live yeast on methane ($CH_4$) production in the large intestinal content of pigs to provide an insight to whether active live yeast could suppress $CH_4$ production in the hindgut of pigs. Treatments used in this study include blank (no substrate and no live yeast cells), control (no live yeast cells) and yeast (YST) supplementation groups (supplemented with live yeast cells, YST1 or YST2). The yeast cultures contained $1.8{\times}10^{10}$ cells per g, which were added at the rates of 0.2 mg and 0.4 mg per ml of the fermented inoculum. Large intestinal contents were collected from 2 Duroc${\times}$Landrace${\times}$Yorkshire pigs, mixed with a phosphate buffer (1:2), and incubated anaerobically at $39^{\circ}C$ for 24 h using 500 mg substrate (dry matter (DM) basis). Total gas and $CH_4$ production decreased (p<0.05) with supplementation of yeast. The methane production reduction potential (MRP) was calculated by assuming net methane concentration for the control as 100%. The MRP of yeast 2 was more than 25%. Compared with the control group, in vitro DM digestibility (IVDMD) and total volatile fatty acids (VFA) concentration increased (p<0.05) in 0.4 mg/ml YST1 and 0.2 mg/ml YST2 supplementation groups. Proportion of propionate, butyrate and valerate increased (p<0.05), but that of acetate decreased (p<0.05), which led to a decreased (p<0.05) acetate: propionate (A: P) ratio in the both YST2 treatments and the 0.4 mg/ml YST 1 supplementation groups. Hydrogen recovery decreased (p<0.05) with yeast supplementation. Quantity of methanogenic archaea per milliliter of inoculum decreased (p<0.05) with yeast supplementation after 24 h of incubation. Our results suggest that live yeast cells suppressed in vitro $CH_4$ production when inoculated into the large intestinal contents of pigs and shifted the fermentation pattern to favor propionate production together with an increased population of acetogenic bacteria, both of which serve as a competitive pathway for the available H2 resulting in the reduction of methanogenic archaea.

  • This study was carried out to elucidate residual characteristics of environment friendly agricultural material rotenone in chilli pepper and calculate processing factors by drying. The test material was sprayed twice onto chilli peppers at an interval of seven days and then the chilli peppers were harvested at 0, 1, 3, 5 and 7 days after final spray. Limits of quantitation (LOQs) of rotenone in fresh and dried chilli peppers were 0.03 and 0.07 mg/kg, respectively. Recoveries of the test material in fresh and dried chilli peppers ranged from 89.52 to 97.86% and from 85.76 to 91.61%, respectively. As a results of residual material analysis, amounts of rotenone in fresh and dried chilli peppers ranged from 0.03 to 0.39 mg/kg and from 0.07 to 0.75 mg/kg, respectively, representing that the residual amounts of rotenone decreased time-coursely. Processing factors of rotenone in fresh chilli pepper by drying were found to be from 2.03 to 3.13, indicating that the residual concentration of rotenone in dried chilli pepper increased from two to three times by drying. However, the reduction factor of rotenone in fresh chilli pepper by drying ranged from 0.38 to 0.59, representing that some of rotenone in fresh chilli pepper disappeared during the drying process.

  • This study was performed to examine the combined effects of gamma linolenic acid and isoflavone supplementation on menopausal symptoms and serum lipids in 73 postmenopausal women. A total subjects were randomly assigned to isoflavone (30 mg) + gamma-linolenic acid (110 mg) group or placebo group. We measured menopausal symptoms by modified Kupperman Index (KI) and oxidized LDL, lipid peroxides, blood components and anthropometric parameters before and after the 12 week intervention period. After the 12 weeks of supplementation, supplement group and placebo group showed a significant reduction of modified kupperman index (p < 0.001). Isoflavone (30 mg) + gamma-linolenic acid (110 mg) supplement group showed a significant reduction of oxidized LDL cholesterol concentration (p = 0.006) whereas placebo group did not show significant change. Isoflavone and gamma-linolenic acid consumption did not significantly affect plasma concentrations of total, LDL, HDL cholesterol, triglyceride, apo A1, B and blood components. The result of present study demonstrated the supplementation of 30 mg isoflavone and 110 mg gamma-linolenic acid per day for 12 weeks may protect LDL cholesterol from oxidative stress.

  • Objective: This experimental study was performed to examine if Hang-Am-Dan non-boiled water extracts (HAD-N) induce apoptosis in human lung carcinoma NCI-H460 cells in vitro and inhibits the growth of NCI-H460 cell-transplanted solid tumor in vivo. Materials and Methods: We cultured NCI-H460 cell lines and xenografted them to nude mice. The mice were divided into 3 groups, NCI-H460 cell alone, NCI-H460 + 90 mg/kg HAD-N treated group, and NCI-H460 + 180 mg/kg HAD-N treated group, with seven mice per group. HAD-N was orally administrated every day for four weeks. We checked their body weight and tumor weight and volumes two times a week and their absolute organ weight and biochemical blood analysis at the final day by sacrificing them. We also calculated their tumor inhibition rate (IR), mean survival time and percent increase in life span (% ILS). Results: In this study, we observed that all of the HAD-N treated mice got smaller tumors. The more doses of HAD-N used, the less IR showed at the 8th day after starting this experiment. Tumor weight and volume of HAD-N treatment groups also decreased. Mean survival time and percent increase in life span (% ILS) in the high-dose HAD-N treatment groups were higher than those of other groups. The test substances in the blood level UN results showed reduction in the significance in both HAD-N 90 mg/kg and HAD-N 180 mg/kg (p<0.01). The blood level phosphatase results in HAD-N 90 mg/kg group compared to NCI-H460 cell alone group showed a reduction in significance (p<0.05). AST levels HAD-N 180 mg/kg group compared to NCI-H460 cell alone group significance as well (p<0.05). Conclusion: We suggest that the results of the in vivo study showed that HAD-N may have potential as a growth inhibitor of tumor-induced NCI-H460 of nude mice in spite of the shortcomings of this study. More studies to overcome those shortcomings and to find out significant antitumor mechanism will be needed.

  • Broadly speaking, in order to analyze the water quality improvement effects of the implementation of the Total Water Pollutant Management System in the Sapgy-Lake waterways, a reference was made to the [Plans for implementation of the Total Maximum Daily Load(TMDL)] in 3 cities (Cheonan, Asan, Dangjin). The results of the investigation into the plans to reduce the pollutant load show in that region show that there are plans to reduce pollution for a total of 16 reduction facilities. As for the result of the computation of the reduction in the load, these measurements were computed at the Gokgyo-stream basin and Namwon-stream basin, with BOD and T-P at the Gokgyo-stream basin reduced by 13.9 % and 13.3 %, respectively, while BOD and T-P at the Namwon-stream were reduced by 3.7 % and 3.3 %, respectively. In this way, thus using the results of the water quality forecast of Sapgyo-Lake in measures for the improvement of water quality (in accordance with the implementation of the TMDL), and using the QUAL-MEV model and EFDC model, it is noted that BOD will be improved by 26.4 % from 6.1 mg/L to 4.5 mg/L 0.0 %, T-P by 36.7 % from 0.168 mg/L to 0.107 mg/L and TOC by 26.4 % from 7.7 mg/L to 5.6 mg/L. However, it is forecasted that the targeted standards for the medium influence area will not be achieved. Evidently, Gokgyo-stream and Namwon-stream have been implementing the Total Water Pollutant Management System for the BOD items since January 1, 2019, but the Sapgyo-stream and Muhan-stream were excluded from being designated as subject regions. As such, it is noted now that it is necessary to implement the TMDL for the entire Sapgyo-Lake water systems including Sapgyo-stream and Muhan-stream in order to improve the water quality of Sapgyo-Lake, and likewise the T-P should be designated as the substance subjected to management in addition to BOD.

  • This study was carried out to examine effect of post-chlorine dosage reduction by ozonation and GAC process in the field plant for 3years in the "G" water purification plant in Seoul. And it is to compare GAC with BAC process in removal effects of TOC, THMs, THMFP, $UV_{254}$. As a result, chlorine dosage of ozonation and GAC(=BAC) is less demand than GAC. Seasonal reduction of chlorine demand is from about 37% to 59% with BAC, and from 24 to 46% with GAC. Higher reduction in BAC could be achieved. The efficiency of chlorine demand reduction with ozonation was depending on the organic carbon removal. $UV_{254}$ concentration is less about 0.13~0.74L/mg.m in BAC than GAC. Therefore, the combination of ozonation and GAC was more effective in reducing post-chlorine than the single GAC. TOC was also monitored, and results show that a linear relationship between TOC and chlorine demand is appropriate under each treatment process. It means that removal of organic matter(TOC) from finished water is necessary to reduce post-chlorine dosage in clear well and to minimize order of chlorine in distribution systems.

  • Hexavalent chromium ($Cr^{6+}$) is a highly harmful pollutant, which can be detoxified and precipitated through reduction to $Cr^{3+}$. Bacillus megaterium TKW3 previously isolated from chromium-contaminated marine sediments was capable of reducing $Cr^{6+}$ in concomitance with metalloids ($Se^{4+}$, $Se^{6+}$, and $As^{5+}$). Notwithstanding approximately 50% inhibition, it was the first report of simultaneous bacterial reduction of $Cr^{6+}$ and $Se^{4+}$ (to elemental Se). No significant difference was observed among electron donors (glucose, maltose, and mannitol) on $Cr^{6+}$ reduction by B. megaterium TKW3. The reduction was constitutive and determined to be non-plasmid mediated. Peptide mass fingerprints (PMF) revealed a novel aerobic membrane-associated reductase with $Cr^{6+}$-induced expression and specific reductive activity (in nmol $Cr^{6+}$/mg protein/min) of 0.220 as compared with 0.087 of the soluble protein fraction. Respiratory inhibitor $NaN_3$ did not interfere with the reductase activity. Transmission electron microscopy with energy dispersive X-ray (TEM-EDX) analysis confirmed the aggregation of reduced chromium along the intracellular membrane region. Future identification of the N-terminal amino acid sequence of this reductase will facilitate purification and understanding of its enzymatic action.

  • We study on the dynamic interaction with a simulated physical-biological coupled model response to nutrient reduction scenario in Jinhae Bay. According to the low relative errors, high regression coefficients of COD and DIN, and realistic distribution in comparison to the observation, our coupled model could be applicable for assessing the marine ecosystem response to nutrient input reduction in Jinhae Bay. Due to the new construction and expansion of sewage treatment plant from our government, we reduce 50% nutrient inputs near Masan Bay and sewage treatment plant. COD achieves Level II in Korea standard of the water quality from the middle of the Masan Bay to all around Jinhae Bay except the inner Masan Bay remaining at Level III. When our experiment reduces 50% nutrient inputs near Masan Bay and Dukdong sewage treatment plant simultaneously, COD decreases to about 0.1-1.2 mg/L $(128^{\circ}30'{\sim}128^{\circ}40'\;E,\;35^{\circ}05'{\sim}35^{\circ}11'\;N)$. The COD from the middle of the Masan Bay to Jinhae Bay achieves Level II.

  • From this research, the performances of a sludge reduction in the sewage sludge aerobic digestion was experimented by using a sludge pretreatment and membrane bioreactor. The submerged plate membrane was used as the solid-liquid separation membrane. After drawing small amounts of sludge in a bioreactor and then doing the alkaline treatment and ozone treatment, the sludge was sent to back to the reactor. The HRT in the reactor was set as 5 days and the operation in the reactor was carried out at the DO of 1mg/L on average. After 100 days of operation in the reactor, it was shown that the reduction efficiency of total solids was more than 83%. Most of volatile solids were removed through mineralization, and the considerable portion of the non-volatile solids was dissolved and then flowed out with the effluent. Only about 16.3% of total solids in the sludge was accmulated in the reactor even without the loss of volatile fraction. Also, by deriving nitrification and denitrification in one reactor simultaneously, more than 90% of nitrogen removal effect was realized and the experiment was run smoothly without fouling of membrane, even in the high concentration of MLSS. Based on this experiment, sludge can be reduced considerably at a low HRT by these two newly suggested approach.
