Effect of p38 inhibitor on the proliferation of chicken muscle stem cells and differentiation into muscle and fat

  • Objective: Inhibiting the p38 mitogen-activated protein kinase (MAPK) signaling pathway delays differentiation and increases proliferation of muscle stem cells in most species. Here, we aimed to investigate the effect of p38 inhibitor (p38i) treatment on the proliferation and differentiation of chicken muscle stem cells. Methods: Chicken muscle stem cells were collected from the muscle tissues of Hy-line Brown chicken embryos at embryonic day 18, then isolated by the preplating method. Cells were cultured for 4 days in growth medium supplemented with dimethyl sulfoxide or 1, 10, 20 μM of p38i, then subcultured for up to 4 passages. Differentiation was induced for 3 days with differentiation medium. Each treatment was replicated 3 times. Results: The proliferation and mRNA expression of paired box 7 gene and myogenic factor 5 gene, as well as the mRNA expression of myogenic differentiation marker gene myogenin were significantly higher in p38i-treated cultures than in control (p<0.05), but immunofluorescence staining and mRNA expression of myosin heavy chain (MHC) were not significantly different between the two groups. Oil red O staining of accumulated lipid droplets in differentiated cell cultures revealed a higher lipid density in p38i-treated cultures than in control; however, the expression of the adipogenic marker gene peroxisome proliferator activated receptor gamma was not significantly different between the two groups. Conclusion: p38 inhibition in chicken muscle stem cells improves cell proliferation, but the effects on myogenic differentiation and lipid accumulation require additional analysis. Further studies are needed on the chicken p38-MAPK pathway to understand the muscle and fat development mechanism.

The Adenylyl Cyclase Activator Forskolin Increases Influenza Virus Propagation in MDCK Cells by Regulating ERK1/2 Activity

  • Vaccination is the most effective method for preventing the spread of the influenza virus. Cell-based influenza vaccines have been developed to overcome the disadvantages of egg-based vaccines and their production efficiency has been previously discussed. In this study, we investigated whether treatment with forskolin (FSK), an adenylyl cyclase activator, affected the output of a cell-based influenza vaccine. We found that FSK increased the propagation of three influenza virus subtypes (A/H1N1/California/4/09, A/H3N2/Mississippi/1/85, and B/Shandong/7/97) in Madin-Darby canine kidney (MDCK) cells. Interestingly, FSK suppressed the growth of MDCK cells. This effect could be a result of protein kinase A (PKA)-Src axis activation, which downregulates extracellular signal-regulated kinase (ERK)1/2 activity and delays cell cycle progression from G1 to S. This delay in cell growth might benefit the binding and entry of the influenza virus in the early stages of viral replication. In contrast, FSK dramatically upregulated ERK1/2 activity via the cAMP-PKA-Raf-1 axis at a late stage of viral replication. Thus, increased ERK1/2 activity might contribute to increased viral ribonucleoprotein export and influenza virus propagation. The increase in viral titer induced by FSK could be explained by the action of cAMP in assisting the entry and binding of the influenza virus. Therefore, FSK addition to cell culture systems could help increase the production efficiency of cell-based vaccines against the influenza virus.

만성 당뇨발에서 표재조직 및 심부조직 세균배양검사의 비교 (Difference of Microbiology according to Tissue Sampling in Diabetic Ulcers)

  • Purpose: Diabetic foot infection is one of the most common and severe complications of diabetes mellitus that delays healing of the wound. Deep tissue biopsy is considered to be the gold standard method for antibiotic selection in treating infected chronic diabetic ulcers. However, swab culture or superficial tissue biopsy is often performed for a microbiologic test since deep tissue biopsy has limitations in application. The purpose of this study is to find out whether microbiologic results of swab culture or superficial tissue biopsy could be used for selection of antibiotics in treating diabetic ulcers. Methods: This study involved 42 patients with diabetic foot ulcers with negative results in bone probing test. Tissue samples for microbiologic tests were collected from all the patients by using superficial cotton swab, superficial tissue, and deep tissue. The microbiologic results of deep tissue biopsy were compared with swab culture and superficial tissue biopsy statistically. Results: Microbiology of the deep tissue showed the same results with those of the swab culture and superficial tissue in 67% and 71%, respectively. Statistical analysis demonstrated that the microbiology of the swab culture and superficial tissue does not coincide with that of the deep tissue. Conclusion: These results suggest that the microbiology of the swab culture and superficial tissue is not concordant with that of the deep tissue in infected chronic diabetic ulcers. To select appropriate antibiotic regimen, the speci specimen for the microbiologic test should be obtained from deep tissue.

Evaluation of Haemagglutinin Content by RP-HPLC to Generate Pandemic Influenza Vaccine

  • The potency of influenza vaccine is determined based on its hemagglutinin (HA) content. In general, single radial immunodiffusion (SRID) assay has been utilized as the standard method to measure HA content. However, preparation of reagents for SRID such as antigen and antibody takes approximately 2~3 months, which causes delays in the development of influenza vaccine. Therefore, quantification of HA content by other alternative methods is required. In this study, we measured HA contents of H1N1 antigen and H1N1 influenza vaccine by reverse phase-high performance liquid chromatography (RP-HPLC) methods. The presence of HA1 and HA2 was investigated by silver staining and Western blot assay. In addition, accuracy and repeatability of HA measurement by RP-HPLC were evaluated. Comparison of HA concentration by SRID and RP-HPLC revealed a precise correlation between the two methods. Our results suggest that RP-HPLC assay can replace SRID in the event of a pandemic flu outbreak for rapid vaccine development.

풍쇄 슬래그의 샌드매트 대체 가능성에 대한 실험적 연구 (An Experimental Study for Substitutability of Sand Mat with Precious Slag Ball)

  • 연약지반 개량을 위한 수평배수공법으로 샌드매트 공법이 활발하게 사용이 되고 있다. 그러나 현재 압밀침하를 촉진시키는 재료로써 샌드매트에 관련하여 문제점들이 발생하고 있다. 무엇보다도, 샌드매트공법에 요구되는 모래의 양이 증가함에 따라 공급이 원활하게 이루어지고 있지 않은 상태이다. 또한, 수많은 건설현장에서 모래 및 골재의 무분별한 채취로 인하여 생태환경의 파괴 및 천연적인 자연경관의 손상을 초래하고 있다. 따라서 본 연구에서는 이러한 문제점을 해결하고자 샌드매트 대체제로서 풍쇄 슬래그의 사용이 적합한지를 판단하기 위하여 기본 물성시험, 실내 통수능 시험, 침하특성 분석과 현장모형을 실험을 통한 통수능과 침하특성, 프로그램을 통한 침하특성을 통해서 확인하고자 한다.

  • Cognitive model, that is cognitive architecture, is the model expressed with computer program to show the process how human solve a certain problem and it is continuously under investigation through various fields of study such as cognitive engineering, computer engineering, and cognitive psychology. In addition, the much extensive applicability of cognitive model usually helps it to be used for quantitative prediction of human Behavior or Natural programming of human performance in many HCI areas including User Interface Usability, artificial intelligence, natural programming language and also Robot engineering. Meanwhile, when a system designed, an usability test about conceptual design of interface is needed and in this case, analysis evaluation using cognitive model like GOMS or ACT-R is much more effective than empirical evaluation which naturally needs products and subjects. In particular, if we consider the recent trend of very short-end term between a previous technology development and the next new one, it would take time and much efforts to choose subjects and train them in order to conduct usability test which is repeatedly followed in the process of a system development and this finally would bring delays of development of a new system. In this study, we predicted quantitatively the human behavior processes which contains cognitive processes for menu selection in touch screen interface through ACT-R, one of the common method of usability test. Throughout the study, it was shown that the result using cognitive model was equal with the result using existing empirical evaluation. And it is expected that cognitive model has a possibility not only to be used as an effective methodology for evaluation of HCI products or system but also to contribute the activation of HCI cognitive modeling in Korea.

Preliminary Report of Validity for the Infant Comprehensive Evaluation for Neurodevelopmental Delay, a Newly Developed Inventory for Children Aged 12 to 71 Months

  • Objectives: Early detection of developmental issues in infants and necessary intervention are important. To identify the comorbid conditions, a comprehensive evaluation is required. The study's objectives were to 1) generate scale items by identifying and eliciting concepts relevant to young children (12-71 months) with developmental delays, 2) develop a comprehensive screening tool for developmental delay and comorbid conditions, and 3) assess the tool's validity and cut-off. Methods: Multidisciplinary experts devised the "Infant Comprehensive Evaluation for Neurodevelopmental Delay (ICEND)," an assessment method that comes in two versions depending on the age of the child: 12-36 months and 37-71 months, through monthly seminars and focused group interviews. The ICEND is composed of three parts: risk factors, resilience factors, and clinical scales. In parts 1 and 2, there were 41 caretakers responded to the questionnaires. Part 3 involved clinicians evaluating ten subscales using 98 and 114 questionnaires for younger and older versions, respectively. The Child Behavior Checklist, Strengths and Difficulties Questionnaire, Infant-Toddler Social Emotional Assessment, and Korean Developmental Screening Test for Infants and Children were employed to analyze concurrent validity with the ICEND. The analyses were performed on both typical and high-risk infants to identify concurrent validity, reliability, and cut-off scores. Results: A total of 296 people participated in the study, with 57 of them being high-risk (19.2%). The Cronbach's alpha was positive (0.533-0.928). In the majority of domains, the ICEND demonstrated a fair discriminatory ability, with a sensitivity of 0.5-0.7 and specificity 0.7-0.9. Conclusion: The ICEND is reliable and valid, indicating its potential as an auxiliary tool for assessing neurodevelopmental delay and comorbid conditions in children aged 12-36 months and 37-71 months.

A Study of 0.5-bit Resolution for True-Time Delay of Phased-Array Antenna System

  • This paper presents the analysis of increasing the resolution of True-Time-Delay (TTD) by 0.5-bit for phased-array antenna system which is one of the Multiple-Input and Multiple Output (MIMO) technologies. For the analysis, a 5.5-bit True-Time Delay (TTD) integrated circuit is designed and analyzed in terms of beam steering performance. In order to increase the number of effective bits, the designed 5.5-bit TTD uses Single Pole Triple Throw (SP3T) and Double Pole Triple Throw (DP3T) switches, and this method can minimize the circuit area by inserting the minimum time delay of 0.5-bit. Furthermore, the circuit mostly maintains the performance of the circuit with the fully added bits. The idea of adding 0.5-bit is verified by analyzing the relation between the number of bits and array elements. The 5.5-bit TTD is designed using 0.18 ㎛ RF CMOS process and the estimated size of the designed circuit excluding the pad is 0.57×1.53 mm2. In contrast to the conventional phase shifter which has distortion of scanning angle known as beam squint phenomenon, the proposed TTD circuit has constant time delays for all states across a wide frequency range of 4 - 20 GHz with minimized power consumption. The minimum time delay is designed to have 1.1 ps and 2.2 ps for the 0.5-bit option and the normal 1-bit option, respectively. A simulation for beam patterns where the 10 phased-array antenna is assumed at 10 GHz confirms that the 0.5-bit concept suppresses the pointing error and the relative power error by up to 1.5 degrees and 80 mW, respectively, compared to the conventional 5-bit TTD circuit.

Electroacupuncture Delays Development of Hypertension through Increase of NO Level in Spontaneously Hypertensive Rats

  • Objective : Using a spontaneously hypertensive rat (SHR) model of essential hypertension, this study investigated whether electroacupuncture (EA) could reduce early stage hypertension by examining whether EA increased nitric oxide (NO) levels in plasma, which compensates for elevated blood pressure (BP). Methods : EA was applied to the acupoint, Baekhoe (GV20), and to a non acupoint in the tail at 10 Hz and an intensity of 1 mA for 10 minutes on the first and fourth day of the week for three weeks under isoflurane anesthesia. In conscious SHR and normotensive Wistar Kyoto (WKY) rats, blood pressure was determined the day after EA treatment by the tail cuff method using an automatic BP monitoring system. We also measured NO concentration of blood serum in SHR and WKY. Results : Systolic blood pressure (SBP) and diastolic blood pressure (DBP) were lower after 3 weeks of GV20 treatment than in non EA treated or non acupoint treated SHR rats. The NO level of plasma was significantly lower in hypertensive SHR than in normotensive WKY. EA prevented the augmentation of blood pressure, and also increased NO concentrations from $7.91{\pm}0.42$ ${\mu}M$ to $11.50{\pm}0.93$ ${\mu}M$ in SHR serum. Conclusions : We suggest that acupuncture may be an early intervention to delay the development of hypertension and enhance NO/NOS activity.

화재 현장에서 효과적인 구조활동을 위한 휴대전화의 무선 신호 활용 방안 연구 (A Study on the Use of Wireless Signals of Mobile Phones for Effective Rescue Activities at Fire Sites)

  • 2017년 말에 발생한 제천 복합건물 화재 사고를 분석한 결과, 고립된 사람들의 위치정보 확인이 지연되어 사상자가 많이 발생하였다. 이와 같은 사고를 방지하기 위해, 재난 현장에 존재하는 휴대 전화기의 무선 통신 정보를 검출하여, 요구조자들의 위치를 추정하는 연구가 진행중이다. 주요 개념은 화재 현장에서 이동 통신 단말의 RF 신호를 탐지하고, 이를 기반으로 LOS/NLOS 상에서 휴대전화기의 위치를 정밀하게 측위하는 것이다. 그러나 화재 현장은 연기로 인해 사야 확보가 어렵고, 건물 내 통로가 화점에 의해 유실되는 등의 어려움이 존재한다. 따라서 요구조자들의 위치를 추정하는 것과 별개로, 화재 현장 내에서 진입하는 방향을 결정하는 연구 역시 필요하다. 본 논문에서는 휴대 전화기의 무선 통신 정보를 이용하여 효과적인 구조 활동을 제공하기 위한 진입 방향을 결정하는 방안을 제시한다.
