Comparision of Food Components in the Raw, Cooked Meat and Cooked Meat Extracts of Cookie Shell -2. Nitrogenous compounds and minerals- (새조개 생육과 자숙육 및 자숙액즙의 식품성분 비교 -2. 함질소엑스성분 및 무기성분의 비교-)

  • The nitrogenous compounds and minerals in the raw and cooked meat of cockle shell were analyzed, and compared with those of cooked meat extracts. In abundant free amino acids, the content of glutamic acid was $129mg\%$ in raw meet, $105mg\%$ in cooked meat, $28mg\%$ in cooked meet extracts, aspartic acid, glycine, arginine, lysine, leucine, and alanine in order. The major components were lysine, arginine and leucine, and the minor components of essential amino acids were proline, tyrosine, serine and cystine. Some of ATP, ADP, AMP, inosine and hypoxanthine were identified in raw and cooked meat, but IMP and inosine were not detected in cooked meat extracts. A slight drop in content of ATP was showed in cooked meat and those had a higher content in inosine and hypoxanthine compared with raw meat. TMA, TMAO and betaine were also checked in all meat products and TMA slightly increased during cooking. Minerals in cooked cockle shell products were phosphorous, potassium, calcium and zinc. The content of phosphorous showed the highest value($16mg\%$ in raw, $185mg\%$ in cooked meat, and $25mg\%$ in extracts).

  • Ascidian, Halocynthia roretzi, mainly inhabiting the subtidal rocky bottom as a sessile form, has been favored a tasty sea food in abundance along the east and south coast of Korea. The study was attempted to establish the basic data for evaluating the processing suitability of ascidian. The height, width, total weight, integument weight and edible portion of the ascidian were $9.2{\sim}12.2cm,\;4.7{\sim}6.1cm,\;100.2{\sim}163.0g,\;18.2{\sim}41.4g$ and $20.2{\sim}62.0g$, respectively. The moisture content, which showed a minimum value as low as $80.0\%$ in October. On the contrary, glycogen content of edible portion revealed the maximum value, $3.1{\sim}4.1\%$ from July to August. The protein and fat showed a similar change to glycogen. The level of ash was rather constant throughout the period, being $2.2{\sim}3.8\%$. Among the 7 kinds of mineral analyzed in all the samples of edible portion and integument, Na, K, Mg and Ca contents were predominent in order being more than $96.14{\sim}99.10\%$, of the total ash. In the amino acid composition of ascidian, the predominent ones were asparagine, glutamic acid, taurine, aspartic acid, proline, and lysine in order and the sum of these amino acids occupied $85.0\%$ of the total free amino acid while methionine and arginine were poor.

Fatty Acid and Amino Acid Compostions of Gugiseun (Lycuim chinense Miller) Depending on Variety and Harvest Time (채취시기 및 품종에 따른 구기순의 지방산과 아미노산조성)

  • As a part of studies on the development of a Gugisuen(Lycium chinense M.), which is one of the unutilization of waste resources, we investigated the composition of fatty acid and amino acid according as picking period and varieties of Gugisuen. The chemical components of Gugisuen were as follows : 1. Proximate composition of Gugisuen were Cheongyang native had the highest contents of crude lipid, crude protein, total sugar, and the contents of crude protein, total sugar, and reducing sugar was the highest picked on June 20. 2. The contents of $P_2O_5$, $K_2O$, CaO, MgO, Cu, and Mn were higher in Cheongyang native than that of other varieties. And mineral contents of Gugisuen picked on May 10 were the highest. 3. The major fatty acids were linoleic, linolenic acids, and these unsaturated fatty acids composed about 50% of total fatty acids. Fatty acid compositions between each varieties were not significant, and those components were the highest in Gugisuen picked on May 10. 4. Eighteen amino acids were identified from Gugisuen, glutamic acid, the highest content was $10.05\sim10.94%$ and these contents increased in the order to aspartic acid > glycine > alanine > leucine > lysine. The contents of serine, arginine, methionine, cystine-2 and isoleucine were higher in late harvest time and glutamic acid and tryptophane were higher in early harvest time.

Studies on the Changes of Taste Compounds during Soy Paste Fermentation (된장 숙성중 정미성분의 변화에 관한 연구 (I) - 유리아미노산과 핵산 관련물질 -)

  • For the purpose of supplying the imformation to improve the acceptability of soy paste as the condiment, the changes of enzyme activity, general component and flavor compounds (Free amino acid, Nucleic acid related compounds, and peptide) during improved soy paste fermentation were determined. The results were as follows; 1. The protease activity during fermentation were increased continuously, but amylase activity were decreased in 45 day fermentation. Cellulase activity were slowly increased until 45 day, and then slowly decreased. 2. Total nitrogen contents were almost constant during fermentation, but amino nitrogem were increased rapidly. Reducing sugar were not constant, but increased in the end of fermentation. PH were decreased to pH 4.97. 3. Total contents of free amino acid as flavor compound were rapidly increased in 10 day fermentation, but were constant in $30{\sim}60$ day. Aspartic acid contents were increased continuously, but glutamic acid were increased slowly until 30 day fermentation and were almost constant. IMP and GMP contents showed increasing pattern during fermentation.

Studies on Nutritional Components of Liver Sausage (간소시지의 영양성분에 관한 연구)

  • The purposes of this study were to investigate the applicability of pork liver in manufacturing sausages and to examine the nutritional components of liver sausages prepared. Sausages containing different proportions of pork liver were manufactured with various processes and were analysed for their nutritional value. Based on the basic recipe for manufacturing sausages, fifteen recipes were proposed with different combinations of ingredients. Sausages manufactured with different methods (sliceable, spreadable, smoked spreadable) and different proportions of liver also were evaluated. The results were obtained as follows : 1. The average contents of moisture, protein, fat, carbohydrate, fiber, ash, and energy were 62.31 %, 15.71 %, 17.12%, 3.88%, 0.24%, 1.48%, and 234.04 kcal/100 g respectively. As the percentage of liver increased, the fat content and total energy were decreased significantly. 2. As the percentage of liver in sausage increased, the content of amino acids including valine, leucine, isoleucine, threonine, lysine, phenylalanine, arginine, aspartic acid, glycine, proline, tyrosine, and cysteine was increased. However, the contents of histidine, methionine, glutamic acid, and alanine were decreased. 3. The vitamin A content of liver sausage was increased by 11 times compared with the control, however, the content of vitamin B was slightly decreased. 4. As the percentage of the liver was increased in sausage, the content of minerals such as calcium, magnesium, phosphorus, sodium, potassium, and iron were increased.

Changes in contents of general components and amino acids of comfrey during growth (한국산(韓國産) Comfrey의 성숙중(成熟中)의 일반성분(一般成分)과 아미노산(酸)의 함량변화(含量變化))

  • Changes in contents of general components of comfrey cultured in Korea were experimented and amino acids were analyzed by thin layer chromatography (T.L.C.) and gas chromatography (G.L.C.). The results obtained were as follows; 1. General components of comfrey such as moisture, fat, protein, carbohydrate, crude fiber and total ash were 13.20, 2.22, 22.30, 37.62, 9.38 and 15.06%. respectively, after 60 days growth. 2. The root of comfrey after 60 days contained 6.03% of alanine, 2.24% valine, 10.77% arginine, 2.96% glycine, 4.08% histidine. 1.54% isoleucine, 0.58% cystein, 1.72% methionine, 7.55% aspartic acid, 7.81% glutamic acid and 4.65% lysine in the gas chromatographic analysis of amino acid composition 3. The crude protein was decreased after 60 days of growth whereas the contents of carbohydrate, crude fiber and total ash were increased. 4. The total amount of amino acids in root was greater than that in leaf of comfrey.

Development of Growing Pig Diets for Environment-Friendly by Using Phytase (Phytase를 이용한 환경친화성 육성돈사료개발)

  • An experiment was carried out to evaluate the effects of adding phytase on nutrient digestibility in growing pigs. Three barrow pigs averaging 28.5$\pm$3.1kg of body weight were allotted to three treatments by 3$\times$3 latin square. Treatments included 1) com-soybean meal based-control diet and 2) and 3) control diet with phytase 500 and 1,000 unit. There were no effects of treatments on dry matter and nitrogen digestibility (P〉0.05). Ash, Ca and P digestibility in pigs fed diet with phytase were greater than those in pigs fed control diet(P〈0.05). An increased phosphorus intake was observed from pigs fed diet with treatments compared to that from pigs fed control diet. Pigs fed diet with adding phytase had improved P absorption compared to pigs fed control diet(P〈0.05). There was no significantly effect on fecal and urine excretion(P〈0.05). Ca intake was increased for pigs fed adding phytase compared to pigs fed control diet. No evident effect was observed on Ca fecal excretion among dietary treatments, however, urine excretion of pigs fed diet with treatments was decreased compared to pigs fed control diet. Cystine, isoleucine, tyrosine and phenylalanine digestibility of amino acid were greater for pigs fed diet with treatments than those for pigs fed control diet(P〈0.05). Asparagine, threonine, serine, methionine and leucine digestibility were improved for pigs fed with phytase 500 unit compared to pigs fed control diet. Lysine, histidine, arginine and other animo acids digestibility were higher for pigs fed diet with phytase than control diet, but there were no significant effects by the treatments(P〉0.05). In conclusion, the results of the experiments suggest that phytase supplementation in the diets could be a very beneficial strategy in environmental aspects.

Effects of Cow에s Milk Addition on the Quality of Soybean Curd (우유 첨가가 두부 품질에 미치는 영향)

  • Soybean curds were made from the mixture of soybean milk and cow's milk (raw milk) at the ratio of 10, 20, 30, 40, 50% (v/v), and then the general composition and mineral content, pH, yield, firmness by sensory evaluation and amino acid composition were investigated. As cow's milk addition increased, total sugar crude fat, crude protein, ash content, firmness, weight and volume of the soybean curd were increased. Whereas moisture content decreased, $Ca^{++}$, $K^{+}$, and Na$^{+}$ were increased, but $Mg^{++}$ and Fe$^{++}$ decreased. In sensory evaluation, color (white), firmness, flavor, and taste were higher than those of control as cow's milk increased. By adding cow's milk at the ratio of 10, 20, 30, 40%, and 50%, sulfur containing amino acid content such as methionine and cystine were enriched 1.31, 1.58, 1.67, 1.85, and 1.95 times, respectively.

Role of PI3-kinase and MAP Kinases in the ARE-mediated Glutathione S-Transferase Induction by Phytochemicals: Comparison with the Oxidative Stress Caused by Decreased Glutathione

  • The expression of phase II detoxifying enzymes is affected by a variety of compounds and the induction of the enzymes plays an essential role in chemoprevention. A variety of phytochemicals such as sulfur-containing chemoprotective agents (SCC) may trigger cellular signals and activate phase II gene expression through ARE activation. see induces glutathione S-transferases. Studies were conducted to investigate the role of mitogen-activated protein (MAP) kinase and phosphatidylinositol 3-kinase (PI3-kinase) in the induction of GST (e.g. rGSTA2) by sec. We also studied the MAP kinase pathway responsible for the GST expression by see and compared that with the pathway activated by oxidative stress as a result of sulfur amino acids deprivation (SAAD). see inhibited phosphorylation of ERK1/2 although the effect of see on JNK and p38 MAP kinase was minimal. Wortmannin and LY294002. PI3-kinase inhibitors. abolished the increases in rGSTA2 mRNA and protein levels by SCC. Deprivation of cystine and methionine caused oxidative stress in H4IIE cells. as evidenced by a decrease in the reduced glutathione and an increase in prooxidant production. Electrophoretic mobility shift assay revealed that the ARE complex consisting of Nrf-1/2 and Maf proteins was activated 12~48 h. The rGSTA2 mRNA and protein levels were increased by SAAD. Activation of ARE and induction of rGSTA2 were both completely inhibited by PI3-kinase inhibitors. Inhibition of p38 MAP kinase by SB203580 prevented the ARE-mediated rGSTA2 induction. The results of this study showed that PI3-kinase might play an essential role in the ARE-mediated rGSTA2 induction by see or SAAD and that the dual MAP kinase pathways were responsible for the enzyme induction.

Antioxidant and ACE Inhibiting Activities of the Rockfish Sebastes hubbsi Skin Gelatin Hydrolysates Produced by Sequential Two-step Enzymatic Hydrolysis

  • This study was conducted to obtain hydrolysates with potent antioxidative activity from rockfish skin gelatin. Gelatin was extracted under high temperature/high pressure using a two-step enzymatic hydrolysis with commercial enzymes such as Alcalase, Flavourzyme, Neutrase, and Protamex. The second rockfish-skin gelatin hydrolysate (SRSGH) was prepared by further incubating the first gelatin hydrolysate (FRSGH), which had been hydrolyzed with Alcalase for 1-h (FRSGH-A1), with Flavourzyme for 2-h (SRSGH-F2). The second gelatin hydrolysate showed higher antioxidative activity of 3.72 as measured by a Metrohm Rancimat and superior angiotensin I-converting enzyme (ACE) inhibiting activity of 0.82 mg/mL. Compared with the gelatin, the relative proportion in SRSGH-F2 was markedly decreased in the 100-kDa peak, whereas it was increased in that less than 100-kDa. The amino acid composition of SRSGH-F2 was rich in glycine (25.9%), proline (10.8%), alanine (9.1%), and glutamic acid (9.1%). In contrast, it was poor in cystine (not detected), methionine (1.6%), tyrosine (0.4%), hydroxylysine (0.9%), and histidine (0.9%). In recent years, demand for natural functional foods has been increasing, and SRSGH-F2 can be used as a functional food ingredient in the food industries. However, further detailed studies on SRSGH-F2 with regard to its antioxidant activity in vivo and the various antioxidant mechanisms are needed.
