Mychonastes sp. 246 Suppresses Human Pancreatic Cancer Cell Growth via IGFBP3-PI3K-mTOR Signaling

  • Previously, we confirmed that Mychonastes sp. 246 methanolic extract (ME) markedly reduced the viability of BxPC-3 human pancreatic cancer cells. However, the underlying mechanism ME remained unclear. Hence, we attempted to elucidate the anticancer effect of ME on BxPC-3 human pancreatic cancer cells. First, we investigated the components of ME and their cytotoxicity in normal cells. Then, we confirmed the G1 phase arrest mediated growth inhibitory effect of ME using a cell counting assay and cell cycle analysis. Moreover, we found that the migration-inhibitory effect of ME using a Transwell migration assay. Through RNA sequencing, Gene Ontology-based network analysis, and western blotting, we explored the intracellular mechanisms of ME in BxPC-3 cells. ME modulated the intracellular energy metabolism-related pathway by altering the mRNA levels of IGFBP3 and PPARGC1A in BxPC-3 cells and reduced PI3K and mTOR phosphorylation by upregulating IGFBP3 and 4E-BP1 expression. Finally, we verified that ME reduced the growth of three-dimensional (3D) pancreatic cancer spheroids. Our study demonstrates that ME suppresses pancreatic cancer proliferation through the IGFBP3-PI3K-mTOR signaling pathway. This is the first study on the anticancer effect of the ME against pancreatic cancer, suggesting therapeutic possibilities and the underlying mechanism of ME action.

Anti-nociceptive Effects of Clerodendrum Trichotomum (누리장나무의 진통 효과)

  • Clerodendrum trichotomum has been widely used as a traditional medicine for the treatment of numerous diseases, including pain management. However, there have been extremely limited pharmacological and phytochemical studies on Clerodendrum trichotomum up to now. In this study, we investigated the effects of the methanolic extract of Clerodendrum trichotomum (MCT) on nociceptive pain in mice. Our finding demonstrate that MCT treatment significantly extended the latency time in both the tail-immersion test and hot-plate test. Additionally, MCT treatment reduced acetic acid-induced writhing motions. These results suggest that MCT possesses strong anti-nociceptive activities against thermal and chemical nociception. In the formalin test, mice fed with MCT exhibited reduced licking time during both the early and late phases, thereby confirming the therapeutic potential of MCT in central and peripheral nociception. Furthermore, in combination tests using naloxone, the MCT-mediated anti-nociception was slightly reduced, indicating that MCT might act as a partial opioid receptor agonist. Based on these results, MCT may be a valuable candidate for the development of anti-nociceptive agents.

Inhibition of Food-derived Lactic Acid Bacterial Biofilm Formation Using Eisenia bicyclis-derived Nanoparticles (식품 유래 Biofilm 형성 유산균에 대한 대황(Eisenia bicyclis) 유래 Nanoparticle의 Biofilm 형성 저해)

  • Lactic acid bacteria (LAB) growth in processed meat products produces slime. In this study, 10 different biofilm-forming LAB, including Leuconostoc mesenteroides, Lacticaseibacillus paracasei, Levilactobacillus brevis, Lactiplantibacillus plantarum, Leuconostoc citreum, Weissella viridescens, and Latilactobacillus sakei, were isolated from various meat products and identified based on 16S rRNA gene analysis. To inhibit biofilm formation by LABs, Eisenia bicycles methanolic extract (EB) and ethyl acetate soluble fraction (EA) were used as antibacterial and antibiofilm agents, respectively. Furthermore, EA and EB were employed to synthesize gold nanoparticles (AuNPs) such as EB-AuNPs and EA-AuNPs, which could serve as antibiofilm agents against the isolated LAB. These findings demonstrate that EA, EB-AuNPs, and EA-AuNPs exhibit significant antibacterial activity against the isolated LAB. Furthermore, EB-AuNPs reduced L. citreum biofilm production, whereas EA-AuNPs inhibited L. mesenteroides and L. brevis biofilm formation. The current results suggest that EB-AuNPs and EA-AuNPs can be used as nanomaterials to inhibit LAB that form biofilms on meat products.

Antioxidant Properties and Ubiquinone Contents in Different Parts of Several Commercial Mushrooms (시판버섯의 부위별 항산화능과 유비퀴논 함량)

  • Antioxidant properties and antioxidant compound contents in different parts of 14 commercial mushrooms were evaluated. Methanolic extracts from the entire mushroom, the pileus and the stipe, separately, were screened for their DPPH and ABTS free radical scavenging activities. Total ubiquinones (Coenzyme Qs; CoQs), total phenolic, and flavonoid contents were determined, in order to assess the extract's antioxidant activity. The portion of the mushroom selected had an effect on the results, with pileus methanolic extract exhibiting the greatest antioxidant effect (p<0.05). The analyzed mushrooms contained powerful antioxidants such as phenols (144.5~536.6 mg of gallic acid equivalents, mg GAE/100 g of dried weight, dw), flavonoids (3.7~31.2 mg of quercetin equivalents, mg QE/100 g dw) and ubiquinones (65.6~485.1 ${\mu}g$/100 g dw). Content of CoQ9 and CoQ10 in the 14 commercial mushrooms varied from 23.1 to 256.2 ${\mu}g$/100 g and from 16.1 to 238.3 ${\mu}g$/100 g, respectively. Phellinus linteus showed the highest antioxidant activity among all species due to the contribution of antioxidants such as phenols (530.5 mg GAE/100 g dw) and ubiquinones (308.8 ug/100 g dw). A positive linear correlation was demonstrated between free radical scavenging activity and total phenolic ($R^2=0.79$) and ubiquinone ($R^2=0.59$) contents in the pileus of mushrooms (p<0.05). Our data indicate that commercial mushrooms have potential as dietary sources of CoQs and phenolic antioxidants.

Inhibitory Effect of the Methanolic Extract of Symphyocladia latiuscula on the Growth of HT-29 Human Colon Cancer Cells (보라우무 메탄올추출물의 HT-29 대장암세포 증식 억제 효과)

  • In the present study, twenty eight marine algae species were evaluated for their antiproliferative effect on HT-29 human colon cancer cells. Among these, the methanolic extract of Symphyocladia latiuscula (SL Ex) showed the highest inhibitory activity on HT-29 cell growth. In this study, we examined the mechanism by which SL Ex inhibited the HT-29 cell growth. Cells were cultured with various concentrations of $(0{\sim}20{\mu}g/mL)$ SL Ex. The SL Ex substantially decreased the viable cell numbers and induced apoptosis of HT-29 cells in a dose-dependent manner Western blot analyses of total cell lysates revealed that SL Ex increased the levels of cleaved caspase-8, -9, -7, and -3, and poly (ADP-ribose) polymerase in HT-29 cells. In addition, SL Ex increased truncated Bid levels but moderately decreased Bax levels at only $20{\mu}g/mL$. Furthermore, SL Ex did not affect Bcl-2 protein levels but increased the levels of Fas in HT-29 cells. The present results indicate that SL Ex inhibits cell growth via inducing apoptosis in human colon cancer cells. The mechanism of apoptosis induction by SL Ex involves caspase-8 activation leading to changes in mitochondrial events and subsequent activation of the caspase-7/caspase-3 cascade. Our finding may lead to the development of new therapeutic strategies for the treatment of colon cancer.

Identification of Antioxidant Compound Derived from Methanolic Extract of Houttuynia Cordata (어성초 메탄올 추출물로부터 항산화 효능을 가진 활성물질의 확인)

  • This study was carried out to evaluate the antioxidant effect of methanolic extract of Houttuynia cordata (HCME) and to identify a compound having antioxidant effect. The ethyl acetate fraction of HCME showed the highest antioxidant effect in organic solvent fractions. The fraction was then separated into 12 fractions by open column chromatography. Among these fractions, the fraction 10 (Fr. 10) with the highest antioxidant activity was isolated, and its antioxidant effect was evaluated by DPPH radical scavenging activity, reducing power, TBARS, cell viability, DNA oxidation and DCF fluorescence. The Fr. 10 at a $64{\mu}g/ml$ showed 60% of inhibitory effect similar to that of vitamin C at $10{\mu}g/ml$, compared with blank group. The Fr. 10 at $64{\mu}g/ml$ showed 264% of reducing power, compared with blank group. TBARS assay showed that the Fr. 10 at $64{\mu}g/ml$ had 35.5% of inhibitory effect similar to that of vitamin E at $1,000{\mu}g/ml$, compared with blank group. The Fr. 10 above $32{\mu}g/ml$ displayed cytotoxicity. However, it was observed that the Fr. 10, above $1{\mu}g/ml$ reduced DNA damage. DCF fluorescence assay showed that the Fr. 10 inhibited oxidative stress by $H_2O_2$ in a dose dependent manner. The compound of Fr. 10 was identified to be rutin whose molecular weight is 610 by the IR and LC-MS analyses. Therefore, these results suggest that the rutin of Fr. 10 could use as a natural antioxidant for development of cosmetics and functional foods.

Nutritional Composition and Cytoprotective Effect of Moringa oleifera Lam. (Drumstick-tree (Moringa oleifera Lam.)의 주요 영양성분 및 추출물의 신경세포 보호 효과)

  • The cytoprotective effect of Moringa oleifera Lam. (drumstick tree) on neuronal cells was investigated to confirm the physiological benefits associated with this natural food resource. First, the drumstick tree extract was chemically analyzed to determine inherent nutritional constituents. Calcium and potassium were identified as the major mineral constituents, and palmitic acid (C16:0, 16.33%) and gadoleic acid (C20:01, 66.34%) were detected as the major fatty acids. Moreover, drumstick tree extract contained 94.78 mg/100 g vitamin E and 112.61 mg/100 g niacin. PC12 cells were used to study the cytoprotective effects of drumstick tree extract. Intracellular accumulation of reactive oxygen species was significantly reduced when $H_2O_2$ treated-neuronal cells were cultured in a medium containing the methanolic extract of drumstick tree, compared to cells treated with only $H_2O_2$. Cell viability assay using MTT showed that the extract protected cells against $H_2O_2$-induced neurotoxicity and inhibited LDH leakage from the cell membrane. Caspase assay showed that the extract exerted cytoprotective effect against apoptosis. Consequently, these data suggest that drumstick tree is a useful natural resource with positive effects on human health.

Antioxidant and Anticancer Activities of Traditional Kochujang Added with Garlic Porridge (마늘죽 첨가 고추장의 항산화 및 항암효과)

  • In order to improve the functional properties of kochujang, garic porridge was added to traditional kochujang during manufacturing. Changes in physiochemical properties of kochujang by garic porridge addition were then investigated. No big differences in general chemical compositions was observed between three kinds of kochujangs tested in this study, general kochujang purchased from a market (GK), kochujang added with raw garlic (RGK) and kochujang added with garlic porridge (GPK). However, GPK showed higher level of antioxidant and anticancer activities than those of others. The methanolic extract of GPK showed 66.38% of DPPH radical scavenging activity, while the extracts of GK and RGK exhibited 38.44% and 50.97%, respectively. Also, the effects of three different extracts of kochujangs on cell proliferation of stomach cancer cell (MKN 45), colon cancer cell (HCT116), and lung cancer cell (NCI-H460) were investigated using MTT assay. All of three extracts exhibited the highest anti-proliferative activity against stomach cancer cell, even though the proliferation of colon cancer cell and lung cancer cell were also inhibited. Among them, the extract of GPK showed the highest anti-proliferative activity (62.35%) against stomach cancer cell. From the results obtained in the present study, we concluded that the antioxidant and anticancer activity of GPK mainly originated from garlic because GPK was consisted of 23% garlic (w/w) compared to 10% (w/w) of RGK.

Antimicrobial Effect of Extract of Glycyrrhiza uralensis on Methicillin-Resistant Staphylococcus aureus (감초 추출물이 항생제 내성균주의 항균활성에 미치는 영향)

  • Antimicrobial drug-resistance is natural response to antimicrobial stress based on selection, which weakens chemotherapy effect. Introduction of large numbers of chemotherapeutic agents to clinical practice has generated strains of microorganisms that survive and multiply in vivo with high-drug concentrations. Methicillin-resistant Staphylococcus aureus (MRSA), bacteria found in normal daily life, can be easily ingested through milk vegetables, and meats, etc. MRSA emerged in many port of the world, increasing complex clinical problems. Therefore, new agents are needed to treat MRSA. Glycyrrhiza uralensis was extracted using 80% MeOH to investigate its antimicrobial activity against MRSA stains KCCM 11812, 40510, and 40512 through bacterial measurement, disc diffusion, and O.D. methods, MIC values, MRSA gene expression investigation, and scanning electron microscope observation. Results revealed MecA, Mecl, MecRI, and FemA were the most highly manifested MRSA genes. Methanolic extract of G. uralensis significantly inhibited MRSA and thus could be used in development of antibacteria.

Antioxidative Compounds in Extracts of Acer ginnala Max. (신나무 추출물의 항산화 활성물질)

  • To search for antioxidative compounds from plant resources, methanol extracts of 45 plant species were investigated using DPPH method. The highest activity was shown in the methanol extract of Acer ginnala($RC_{50}\;:\;15{\mu}g$), followed by Stewartia koreana($RC_{50}\;:\;28{\mu}g$) and Carpinus laxiflora($RC_{50}\;:\;33{\mu}g$). Two antioxidative compounds were isolated from the methanolic extract of Acer ginnala Max and identified as acertannin(2, 6-di-O-galloyl-1, 5-anhydro-D-glucitol) and gallicin (methyl-3, 4, 5-trihydroxybenzoic acid) on the basis of mass spectroscopy, $^1H-\;and\;^{13}C-NMR$ data. The DPPH free radical scavenging activities of acertannin($RC_{50}\;:\;3.5{\mu}g$) and gallicin($RC_{50}\;:\;2.8{\mu}g$) were more effective than those of BHA($RC_{50}\;:\;14{\mu}g$) and ${\alpha}-tocopherol$ ($RC_{50}\;:\;12{\mu}g$).
