Antimicrobial Effect of Lithospermum erythrorhizon Extracts on the Food-borne Pathogens (지치추출물의 식중독성 미생물에 대한 항균효과)

  • Antimicrobial effect of Lithospermum erythrorhizon extracts against food-borne pathogens was investigated. L. erythrorhizon was extracted with methanol at room temperature, and the extraction was sequentially fractionated using petroleum ether, chloroform, ethyl acetate, and methanol. Antimicrobial activity of L. erythrorhizon extracts was determined using paper disc method against food-borne pathogens and food spoilage bacteria. Ethyl acetate extracts of L. erythrorhizon showed the highest activity against Staphylococcus aureus and Shigella dysenteriae. Synergistic effect was found in combined extracts of L. erythrorhizon and Sophora subprostrata as compared with each extract alone. Growth inhibition curve was determined using ethyl acetate extracts of L. erythrorhizon, against S. aureus and S. dysenteriae. Ethyl acetate extract of L. erythrorhizon, showed strong antimicrobial activity against S. aureus at 4,000 ppm, retarding growth of S. aureus more than 48 hr and S. dysenteriae up to 12 hr.

Effects of Gamma-Irradiation on Biological Activities and Color Changes of Extracts of Schizandrae fructus (감마선 조사가 오미자의 생리 활성과 색상 변화에 미치는 영향)

  • This study was carried out to examine the effects of gamma irradiation on the color removal, antioxidation, DPPH radical scavenger, and antimicrobial activity of Schizandra fructus extracts by different solvents. Schizandra fructus was extracted by hot water, ethanol, acetone and methanol, and the extracts were irradiated 10, 20 and 30 kGy with gamma rays. Hunter color L-value was increased by irradiation in a dose-dependent manner, resulting in brighter color for all solvents used. The a and b values were decreased by irradiation in a dose-dependent manner. Antioxidant activity of the ethanol extract of Schizandra fructus against soybean oil was the highest. The free radical scavenging activity was the strongest in hot water extract. All solvent extracts from Schizandra fructus had the strongest antimicrobial activities to B. subtilis, B. natto, B megaterium, S. aureus, Sal. typhymurium and E. coli. These results suggested that Schizandra fructus extracts have a strong potential as natural food preservatives and cosmetic raw materials. Furthermore, irradiation may not influence adversely on biological activites of the extracts when irradiated up to 30 kGy.

Effect of Extracts from Root Bark of Ulmus parvifolia on Inhibition of Growth and DNA Synthesis of Human Cancer Cells. (느릅나무 근피 추출물에 의한 인체 암세포 증식 및 DNA 합성 억제효과)

  • Growth and DNA synthesis inhibitory effects of extracts from root bark of Ulmus parvifolia on MG-63 human osteosarcoma cells, HT-29 human colon cancer cells and K-562 leukemia cancer cells were studied. The root bark extract of Ulmus parvifolia was extracted with methanol, hot water and juice. The methanol extract showed the highest inhibitory effect on growth of MG-63, HT-29 and K-562 cancer cells by >85%. The treatment of hot water and juice extracts from root bark of Ulmus parvifolia also inhibited growth of the above cancer cells with increasing concentration. DNA synthesis of MG-63 and HT-29 cancer cells was significantly inhibited by adding methanol, hot water and juice extracts from root bark of Ulmus parvifolia with increasing concentration, showing that the inhibitory effect of growth was more effective on HT-29 cancer cells. These results suggest that the methanol extract from root bark of Ulmus parvifolia may have specific active com-pounds on anticancer effect. The hot water extract also showed a strong inhibitory effect on growth of cancer cells, indicating that the active compounds may be stable to heat.

Antioxidant Activity and Contents of Bioactive Components in Polar Microalgae (극지미세조류의 유용성분 함량 및 항산화 활성)

  • In this study, bioactive components such as polyohenols, flavonoids and tocopherols were determined in cultured polar microalgae (Fragilariopsis pseudonana, Chaetoceros neogracile, Stellarima microtrias, Porosiara pseudodenticular). Antioxidant activity of methanol extracts of polar microalgae was also investigated. ${\alpha}-Tocopherol$ contents in Fragilariopsis pseudonana were almost two times higher than those of Chaetoceros neogracile in. The antioxidant activity of methanol extracts of Fragilariopsis pseudonana methanol extracts determined by ABTS assay was higher than other algae. Total polyphenol contents of methanol extracts also showed a similar trend as antioxidant activity. The protective activity against oxidative damages induced by glutamate in PC 12 cells was shown in only Chaetoceros neogracile.

Morinda citrifolia Inhibits Both Cytosolic $Ca^{2+}$-dependent Phospholipase $A_2$ and Secretory $Ca^{2+}$-dependent Phospholipase $A_2$

  • This study investigated the effects of the methanol extracts of Morinda citrifolia containing numerous anthraquinone and iridoid on phospholipase $A_2$ ($PLA_2$) isozyme. $PLA_2$ activity was measured using various $PLA_2$ substrates, including 10-pyrene phosphatidylcholine, 1-palmitoyl-2-[$^{14}C$]arachidonyl phosphatidylcholine ([$^{14}C$]AA-PC), and [$^3H$]arachidonic acid (AA). The methanol extracts suppressed melittin-induced [$_3H$]AA release in a concentration-dependent manner in RAW 264.7 cells, and inhibited $cPLA_2/sPLA_2$-induced hydrolysis of [$^{14}C$]AA-PC in a concentration- and time-dependent manner. A Dixon plot showed that the inhibition by methanol extracts on $cPLA_2$ and $sPLA_2$ appeared to be competitive with inhibition constants ($K_i$) of $3.7{\mu}g/ml$ and $12.6{\mu}g/ml$, respectively. These data suggest that methanol extracts of Morinda citrifolia inhibits both $Ca^{2+}$-dependent $PLA_2$ such as, $cPLA_2$ and $sPLA_2$. Therefore, Morinda citrifolia may possess anti-inflammatory activity secondary to $Ca^{2+}$-dependent $PLA_2$ inhibition.

Multidrug resistance reversal in mouse lymphoma cells by indian tea leaves, indian coffee seeds and chicory

  • Systematic analysis of caffeine from the commercial samples of Indian tea leaves was performed by a routine method and the content of caffeine was found to be 19.0-37.4 mg/100 g leaves. The caffeine contents from coffee seeds and chicory from Indian origin were analyzed and found to be 0.6540-1.4920 g/100 g seeds. Caffeine contents of roasted Indian chicory roots were lower than either those of Indian tea leaves or Indian coffee seeds. The multidrug resistance (MDR) reversing effects were tested on a mouse leukemia cell line of L-5178 cells by methanol extracts [M1-M15] of Indian tea leaves and coffee seeds, comparing to a control of $({\pm})-verapamil$. The effects were measured by fluorescence ratio between treated and untreated group cells. Among fifteen methanol extracts, a Gemini tea [M6] (fluorescence activity ratio 5.26) had the most potent effect for L-5178 cells. The extract M6 was 0.63-fold of $({\pm})-verapamil$. We suggest that one of mechanisms of reversal by M6 might have strong affinity to dopamine $D_1$ and D_2$ receptors. Further studies with many more tumor and normal cell lines are necessary to confirm the MDR reversal specificity of coffee methanol extracts.

Antioxidant Activity of Several Cabbage (Brassica oleracea L.) Cultivars

  • Total phenol, flavonoid and antioxidant components of cabbage leaf samples derived from different cultivar were determined. Total phenol compound content showed the highest amount in methanol extracts from ‘YR Howol’ cultivar (11.72 ㎎/g), followed by ‘Harutame’ (10.66㎎/g), ‘Winstar’ (10.34 ㎎/g) and YR Hero (10.20 ㎎/g). The highest amount of total flavonoid content was observed from the methanol extracts of Harutame (5.39 ㎎/g), followed by Winstar (4.28 ㎎/g), Wialhowol (4.10 ㎎/g). The SOD enzyme activity showed a high activity of ‘YR Hogeo’ cultivar, and the cultivar of ‘YR Howol’ cultivar showed the lowest activity of SOD. The activity of CAT and APX showed higher values ‘Ogane’ and ‘YR Hogeol’ cultivars than the other cultivars. The POD activities showed relatively high values ‘Ogane’ and ‘YR Howol’ cultivars compared with other cultivars. The free radical (DPPH) scavenging activity showed lower IC50 values of ‘Harutame’ (15.71) and ‘YR Howol’ cultivar (16.88), however methanol extract of ‘YR Hero’ cultivar (22.49) being the highest. The extracts of all cabbage cultivars in the reaction solution of pH 1.2 could be decomposed nitrite more than 50%. Especially, the cultivar ‘YR Hogeol’ and ‘Ogane’ showed a relatively high nitrite scavenging activity for each 60.13% and 57.20% respectively. The IC50 values of antioxidant activity determined by ABTS were lower in ‘Harutame’ (17.04) and ‘YR Howol’ cultivar (17.97), and its results observed similar with values obtained from the same extracts by DPPH method. The result of this study suggests that the methanol extract of Brassica oleracea L. contains the high amount of phenolic and higher radical scavenging activities.

Comparison of Antioxidant Activities of Black Onion Extracts (추출용매에 따른 흑양파의 항산화 활성 비교)

  • The antioxidant activities of the ethanol, methanol, and water extracts of black onion were investigated. The highest extraction yield (28.56%) and total polyphenol content (13.5 mg/g) were found in the water extract. The water extract also showed the highest protective effect on the RAW 264.7 cell anti-inflammatory activity, and the water and ethanol extracts showed the highest reducing power. The water extract showed higher DPPH and ABTS radical scavenging activities (65.98 and 89.69%, respectively) than the other solvent extracts (ethanol, 47.77%; methanol, 66.12%). Hence, black onion can be used as a potent natural antioxidative source.

In vitro Antioxidant Activity and Anticancer Effects of The Extracts from Eleutherococcus senticosus Max

  • Ethanol and butanol fractionation of E. senticosus showed strong anti-oxidant activity, and methanol and water extracts also had high anti-oxidant activity. The anti-oxidant activities in ethanol and butanol fractionation were higher than or similar to those of ${\alpha}-tocopherol$. The cytotoxic effect of root extract of E. senticosus was evaluated on seven different human cancer cell lines, The extracts of leaf and stem of E. senticosus also had strong antioxidant activity, but the antioxidant activity in root extract was higher than those in leaf and stem extracts. Methanol, hexane, and aqueous fraction layer had much higher inhibitory activities on lipid peroxidation in rat liver microsomes compared with ${\alpha}-tocopherol$. The effect of root extract of E. senticosus was evaluated on six human cancer cell lines. The values of 50% growth inhibition $(GI_{50})$ for the extracts were mostly below $30{\mu}g/ml$, and the extracts are considered as active inhibitory compounds on cancer cells.

Studies for Component Analysis and Antioxidant Effect, Antimicrobial Activity in Acanthopanax senticosus HARMS (가시오갈피 (Acanthopanax senticosus) 열매의 성분 분석과 항산화 효과 및 항균활성)

  • In the present studγ, we investigated the component analγsis, measurements of antioxidant activities and anti-microbial activities from fruit of Acanthopanax senticosus HARMS in order to detect the biological activities and develop novel functional resources. Different solvents extraction ratios were different according to different solvent extraction of Acanthopanax senticosus HARMS. Composition analysis we highly composed as 72.33%. The minerals of different organs were highly composed of potasium as 5951.3 mg/100g. The monosaccharldes are composed of the arabinose, xylose, mannose, galactose, glucose. Antioxidamt activity was measured in water extracts: $57.3\;{mu}/ml$, ethanol extracts $630.1\;{\mu}g/ml$, methanol extracts: $248.5\;{\mu}g/ml$, 71% ethanol extracts: $198.97\;{\mu}g/ml$, 75% methanol exracts: $96.77\;{\mu}g/ml$, chloroform extracts. $1194.83\;{\mu}g/ml$ at $IC_{50}$ value. The antimicrobial activities were observed in extracts from Acanthopanax senticosus HARMS against am negative bacteria and gram positive bacteria.
