Antioxidant and Anti-Inflammatory Activities of Methanol and Hot Water Extracts of Pholiota nameko Fruiting Bodies (맛버섯 자실체의 메탄올 및 열수추출물의 항산화 및 항염증 활성)

  • Pholiota nameko is an edible mushroom belonged to Family Strophariaceae of Agaricales, Basidiomycota. The purpose of this study was to investigate the antioxidant and anti-inflammatory activities for the methanol and hot water extracts prepared from fruiting bodies of Pholiota nameko. Besides measuring for 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity, a reducing power and a chelating activity on ferrous ions were also measured to evaluate the antioxidant activity for those extracts. To measure the anti-inflammatory activities for the extracts, nitric oxide(NO) production from lipopolysaccharide treated RAW 264.7 macrophage cells and carrageenan-induced acute hind paw edema of rats were investigated. The results showed that those extracts has a excellent chelating activity on the ferrous ions compared with positive controls. And it also turned out that extracts had a good DPPH activity and a reducing power. The NO production in LPS-treated RAW 264.7 macrophage cells were decreasing as we increased concentration of those mushroom extracts. Significant reduction of paw edema were also observed at 2~6 h after we treated methanol and hot-water extracts at the 50 mg/kg concentration to the rats which are induced acute hind paw edema by carrageenan treatment. The experimental results suggested that methanol and hot-water extracts of Pholiota nameko fruiting bodies might be used for potential source of antioxidant and anti-inflammatory agents.

Antioxidant Activities of Various Solvent Extracts from Canola Meal (추출용매별 canola meal의 항산화 활성)

  • This study was performed to investigate antioxidant activity of various solvent extracts (methanol, ethanol, acetone and water) from canola meal. As the concentration of all extracts increased, antioxidant activities increased gradually. Among solvent extracts from canola meal, methanol extract showed the highest antioxidant activities with $EC_{50}$ values of 0.9, 3.5, 1.3, and 8.5 mg/mL for DPPH radical scavenging ability, ABTS radical scavenging ability, reducing power, and chelating ability, respectively, and it had the highest total phenolics and total flavonoids contents. Furthermore, methanol extract was fractionated into three fractions (MF 1, MF 2, and MF 3) using Sephadex LH-20 column. The highest antioxidant activity was found on MF 2 for DPPH radical scavenging ability, on MF 3 for ABTS radical scavenging ability and reducing power, and on MF 1 for chelating ability at the same concentration of 1.5 mg/mL. The results indicated that each fraction containing various antioxidant materials seemed to have different influence at each antioxidant activity.

Studies on Immunomodulatory and Antioxidant Activities of Astragali membranacei Radix according to the Cultivated Years (황기(黃芪)의 재배 년수에 따른 면역 및 항산화 활성 연구)

  • Purpose: Contents of astragaloside I, II and IV, cytotoxicity, anticancer activity, immunomodulatory activity and antioxidant capacity were to be compared as a function of the cultivated years as one, three, five and seven years. Method: Major components of Astragali membranacei Radix were separated as astragaloside I, astragaloside II, astragaloside IV by HPLC analysis. Cytotoxicity and anticancer activities were measured by MTT and SRB assay. For immunomodulatory activity, the secretion of IL -6 and $TNF-{\alpha}$, NK cell activation and macrophage activation were observed as well as kinetics of responding to human T cells by a microphysiometer. In vitro antioxidant activities were measured by several radical scavenging activities of superoxide anion radican, DPPH, LDL and linoleic acid. For in vivo activity, the activation of SOD, GSH-px, catalase, ALDH and ADH was measured as well the relative weight of liver. Result : 1. For HPLC analysis, the contents of all of astragaloside I, astragaloside II, astragaloside IV were in order of three, five, one and seven years. 2. The cytotoxicity of normal human lung cell line, HEL299 showed lower than 18% in adding 0.25 mg/ml, and 28.9% in adding 1.0 mg/ml of water extract of seven year root. For methanol extracts, three year root showed highest cytotoxicity as 35.2 % and there was no difference between the cultivated years. 3. For anticancer activities, methanol extracts of one and three year roots showed relatively high inhibition of human stomach cancer cells, AGS, breast cancer cells, MCF-7, lung cancer cells, A549 and liver cancer cell, Hep3B as well as high selectivities. 4. The water extract of seven year root could yield high secretion of IL-6 from both human Band T cells while the methanol extracts of three and five year roots secreted high amounts of IL-6 and $TNF-{\alpha}$ from both Band T cells. 5. As a result of in vitro antioxidant activities, both water and methanol extracts from five and seven year roots showed high activities for superoxide anion radical scavenging activity, inhibiting linoleic acid peroxide and contents of total phenols. 6. For in vivo tests, Mn-SOD and GSH-px activities and weight of liver were better in adding seven year root. For ALDH activity one year root was better and for ADH activity five year root. Overall speaking, seven year root showed relatively better antioxidant activities. Conclusion:There was difference of the contents of astragaloside I, astragaloside II, astragaloside IV according to cultivation year. Methanol extract showed better activities of anticancer and immune activation rather than water extract Interestingly enough, for methanol extracts, overall activities were improved as the cultivation year increased. There might be further investigation required for the clinical uses of the results as several biological activities varied according to the cultivated year of Astragali membranacei Radix.

Inhibitory Effect of Methanol Extract of Doenjang on Growth and DNA Synthesis of Human Cancer Cells (된장 메탄올 추출물의 인체 암세포 성장 억제 효과 및 DNA 합성 저해 효과)

  • Doenjang (Korean soy paste) is one of the popular soybean based fermented foods in Korea. This study investigated the growth and DNA synthesis inhibitory effect of doenjang methanol extracts on AGS human gastric adenocarcinoma cells, Hep 3B human hepatocellular carcinoma cells and HT-29 human colon cancer cells. In order to determine an anticancer effect of doenjang methanol extracts, other soybean fermented foods and original materials were compared. The treatment of doenjang methanol extracts (200 $\mu\textrm{g}$/mL) to the AGS, Hep 3B and HT-29 cancer cells inhibited the growth of cancer cells by 80%, 77% and 86%, respectively. Compared to other soybean fermented foods and original materials, doenjang methanol extracts showed the highest growth inhibitory effect on different cancer cells. In addition, doenjang methanol extracts (200 $\mu\textrm{g}$/mL) significantly inhibited DNA synthesis of AGS and Hep 3B cancer cells by 76% and 59%, respectively. These results suggested that this anticancer effect of doenjang may be due to specific active compounds, which will be newly produced during soybean fermented process and not contained in soybean.

Study on The Anti-HSV(Herpes Simplex Virus) Activity of Natural complex Products (한약 탕제를 이용한 항 Herpes virus 제제의 개발 연구)

  • In order to search for anti-HSV agents from natural complex products, we extended the number of specimens. Both methanol extract and boiling water extract of the natural complex products were screened to detect anti-HSV activity by MTT assay. Anti-HSV activities of thirteen natural complex products extracted by methanol and boiling water were screened. Three of 13 natural complex products extracted by methanol showed efficacy against HSV. Natural complex products showing anti-HSV activities as methanol extracts were No.3, 6, 11 and their Sl were 323.809, 2811.041 and 708.20. As water boiling extracts, No.8 and No.11 have displayed Sl of 16.45 and 60.39 respectively. Especially anti-HSV activities of natural complex products extracted by methanol No.6 was stronger than other ones.

In vitro Antimutagenic Activity of Brown Rice and its Physico-Chemical Characteristics (현미의 in vitro 항돌연변이 활성 및 물리화학적 특성)

  • In vitro antimutagenic activity of methanol extract from brrwn rice and its physico-chemical characteristics were investigated using Salmonella typhimurium reversion assay and SOS chromotest. Methanol extracts of brown rice were not mutagenic compared with direct and indirect, mutagenicities of 4NQO (4-nitroquinoline oxide), 2NF(2-nitrofluorene), Trp-p-1(3-Amino-1,4-dimethyl-5H-pyrido-[4,3-b]indole), and Trp-p-2(3-Amino-1-methy-5H-pyrido-[4,3-b]indole). Antimutagenic activity against the indirect mutagenicties induced by Trp-p-1, Trp-p-2 and AFB1 (aflatoxin B1) was found in methanol extract. Even though antimutagenic activity showed dose-dependent, it remained constant at inhibition rate ranging 60~90% when the concentration was abov 3mg/plate in the S. typhimurium reversion assay and 0.2~0.6 mg/assay in the SOS chromotest. The antimutagenic activity of the methanol extracts was stable at various pH (2, 7 and 10), temperatures (60, 80 and 10$0^{\circ}C$)and heation times (2, 4, 6, 8, 10 min at 10$0^{\circ}C$).

Anticomplementary Activity and Immune-Stimulating Effect of the Extracts from Barley (Hordeum Vulgare) (보리(Hordeum vulgare)추출물의 항보체 및 면역증강 효과)

  • By using several solvents, barley extracts containing the anticomplementary activities in classical pathway were prepared (250 $\mu\textrm{g}$/ml): methanol (83.1%), ethanol (71.9%), water extract (25.4%), M-1 (250 $\mu\textrm{g}$/ml), and the soluble part of methanol extract which showed the highest activity (83.4%) and the yield. Anticomplementary activity of methanol extract as well as protease digestion in classical pathway showed 82.4% and 78.4% in the concentration of 250 $\mu\textrm{g}$/ml, respectively. It was found that protein was not involved in anticomplementary activity in the classical pathway and the methanol extract made an impact on classical pathway, but not on alternative pathway. For the immune-stimulating effect, the T cell proliferation effect of the protease digestion displayed little effect irrespective of the dose. In addition, the T cell proliferation effect of methanol extract showed 13-fold higher proliferation effect compared with positive control. It was revealed that the substance containing protein serves as an important factor for the immune proliferation. Therefore, the anticomplementary activity ${\beta}$-glucan in classical pathway and alternative pathway displayed the lowest activity, showing 2.2%, 22.3% respectively. However, the immune-stimulating effect of ${\beta}$-glucan showed the T cell stimulating effect 13 times higher than positive control.

A Study on Skin Care Effects of Rapeseed Meal Extract (유채박 추출물의 피부미용 효과에 관한 연구)

  • This research was to investigate the inhibitory effects of tyrosinase, elastase and collagenase using rapeseed meal extract for the functional cosmetics. Gamma-tocopherol and alpha-tocopherol contents were 304.9 and 212.2 mg/kg, respectively. In the case of delta-tocopherol and plastochromanol-8, they were 12.1 and 35.7 mg/kg, respectively. The total phenol content of methanol extract was the highest (49.6 mg/g) which was about 4.96 fold higher than that of water extract. The maximum nitrite scavenging activities of methanol and acetone extract at pH 1.2 were 85.2 and 80.1%, respectively, at 8.0 mg/mL. When the extract concentration of rapeseed meal increased upto 2.0 mg/mL, cell viabilities did not appear to have any significant cytotoxic effect, irrespective of extracts. Tyrosinase and elastase inhibition activities increased from 25.2 to 42.5% and 25.3 to 48.0%, respectively, as methanol extract concentration increased from 0.5 to 1.0 mg/mL. The collagenase inhibition activities of methanol and acetone extracts at 1.0 mg/mL were 67.2 and 68.0%, respectively. These results showed that the methanol and acetone extract of rapeseed meal can be used as a new source of functional cosmetic agent.

Cytotoxic Effects of Methanol Extracts from Saururus Chinensis Bail and Herba Houttuyniae on Cancer Cell Lines (삼백초(三白草)와 어성초(魚腥草)의 암세포(癌細胞)에 대한 독성억제(毒性抑制) 효과(效果))

  • This study was performed to determine the cytotoxic effect of methanol extract from medicinal plants. 1. The cell viability was determined by MTT method. Their cytotoxic activities against three cancer cell lines such as A549, MDA-MB-231 and SNU-C4 cell line were tested. 2. Among them, The methanol extract of Saururus Ohinensis Bail showed the strongest cytotoxic effect against SNU-C4 cells. These results suggest that the methanol extract of Saururus Ohinensis Bail possessed a potential antitumorous agent. 3. The free radical scavenging activity using DPPH method was the strongest of Saururus Chinensis Bail extract.

Antioxidant Activities of Liriope platyphylla L. Extracts Obtained from Different Solvents (유기용매별 맥문동 추출물의 항산화 활성 연구)

  • This study was conducted to investigated the antioxidant activities of extract for Megmoondong fruit. Liriope platyphylla L. was extracted with 70% methanol at room temperature for 48 hr and concentrated under reduced pressure to measure its total polyphenol contents and electron donating ability. The total polyphenol contents were $7,253.50{\pm}335.43{\mu}g\;GAE/mL$ in 70% methanol extract, $1,239.77{\pm}9.30{\mu}g\;GAE/mL$ in methylene chloride fraction, $919.30{\pm}50.83{\mu}g\;GAE/mL$ in methanol fraction, $105.44{\pm}2.04{\mu}g\;GAE/mL$ in ethyl acetate fraction. The total polyphenol contents showed significant differences (p<0.05) between the solvents. The electron donating ability was $69.17{\pm}12.61%$ for 70% methanol extract, $33.11{\pm}1.77%$ for methylene chloride fraction, $5.19{\pm}2.59%$ for ethyl acetate fraction, and $20.16{\pm}1.04%$ for methanol fraction. The electron donating abilities were significantly different (p<0.05) between the solvents. The removal ability of superoxide radical was $0.0174{\pm}0.0007$ for 70% methanol extract, $0.0164{\pm}0.0007$ for methylene chloride fraction, $0.0172{\pm}0.0007$ for ethyl acetate fraction, and nothing for methanol fraction. The measurement of hydrogen peroxide decomposition was $0.0985{\pm}0.1021$ for 70% methanol extract, $0.0896{\pm}0.0893$ for methylene chloride fraction, $0.0115{\pm}0.0085$ for ethyl acetate fraction, and $0.0170{\pm}0.0180$ for methanol fraction. The Liriope platyphylla L. extracts obtained from methylene chloride showed significantly relevant results in the total polyphenol contents and electron donating ability, which was higher than the original extract.
