Antimicrobial, Antihypertensive and Anticancer Activities of Garlic Extracts (마늘 추출물의 항균, 항고혈압 및 항암활성)

  • Antibacterial, antifungal, anticancer, and ACE inhibitory activities of methanol, ethyl acetate, chloroform, and hexane extracts of garlic were investigated. Methanol extract showed highest yield of 7.9-8.8 brix. Ethyl acetate and chloroform extracts showed strong antibacterial activities as determined by inhibition zone (8-16mm) against B. subtilis and P. aeruginosa, and good antifungal activities as determined by inhibition zone (8-30mm) against A. niger, M. miehei, and T. reesei. Wando, Banyans, Chinese, and Seosan garlic methanol extracts showed 65.6, 60.4, 70.1, and 55.3% ACE inhibitory activities, 42.3, 53.8, 50.4, and 54.5% anticancer activities against SNU-1, and 11.3, 20.2, 36.6, and 30.5% anticancer activities against HeLa, respectively.

Antioxidant and Anticancer Activities of Extracts from Styela plicata (주름 미더덕(Styela plicata) 추출물의 항산화력 및 항암활성)

  • The antioxidant and anticancer activities of Styela plicata extracts were evaluated. When extracts were prepared with fresh Styela plicata (FR), extraction yield was in the order of methanol > ethanol = acetone > water among treated solvents. However, the extraction order was methanol > water > ethanol > acetate in freeze dried Styela plicata (FD). Radical scavenging activity was the highest in acetone extracts $(37.39\%)$ from FR, while in ethanol extracts $(78.40\%)$ from FD. Reducing power of FR was the greatest in methanol extracts (1.076), and that of FD in ethanol extracts (1.360). The acetone extracts from FD showed significant anticancer activity when revealed with human colon cancer cell line HT-29. These results indicated that extraction yields and properties of extracts from Styela plicata were variable depending on solvent and/or physicochemical state, and appropriate extraction process could provide some valuable bioactive materials from Styela plicata.

Screening of Seaweed Extracts for Algicidal Substances Using a Photosensitization Effect (해조류 추출물로부터의 Photosensitizing 효과를 지니는 항 적조물질 탐색)

  • Photosensitized extracts of 28 seaweed species were tested in vitro to examine the variation in their algicidal activities against the toxic microalga Cochlodinium polykrikoides. When both seaweed extracts and microalgae were exposed to UV and visible light, methanol extracts of Porphyra yezoensis showed 5.7- and 4.4-fold increasesin light-dependent algicidal activity, respectively. When only seaweed extracts were exposed to both UV (365 nm) and visible light (white lamp) at the same time, methanol extracts of Enteromorpha linza and Carpopeltis affinis showed 3.3- and 3.4-fold increases in algicidal activity, respectively. When UV-photosensitized extracts were left in the dark, the algicidal activity of Ecklonia cava increased 13-fold after 5 h. When visible light-photosensitized extracts were left in the dark, the algicidal activity of Monostroma nitidum increased by 3.3-fold in 1 h.

Effect of Defatted Sesame and Perilla Methanol Extracts on Cognitive Function and Antioxidant Activity in SAMP8 Mice (노화촉진마우스에 있어서 참깨박 및 들깨박의 메탄올 추출물이 인지기능 및 항산화능에 미치는 영향)

  • Effects of defatted sesame and perilla methanol extracts on cognitive function and antioxidant activity of learning- and memory-impaired animal model SAMP8 mice. Animals were divided into 4 groups and fed with diets containing 0.3%(w/w) defatted sesame (S) or defatted perilla methanol extracts (P) for 12 weeks. Step through latency of SAMP8 control group was significantly higher than that of SAM R1 normal group, whereas significantly increased in S and P groups compared with SAMP8 control on passive avoidance test (p<0.001). Acetylcholinesterase activity of brain in SAMP8 increased compared with SAMR1 but no difference between SAMP8 control group and sample-treated group. Brain TBARS contents of SAMP8 control significantly increased compared with SAMR1 and were lowered significantly by supplementation of defatted sesame and perilla methanol extracts. Defatted sesame and perilla methanol extracts attenuated increased brain superoxide dismutase and glutathione peroxidase activities in SAMP8. These results suggest defatted sesame and perilla methanol extracts could attenuate cognitive deficits induced by aging possibly through activation of antioxidant activity of defatted sesame and perilla methanol extracts.

Antimicrobial Effect of Forsythiae Fructus Extracts on Several Food-Borne Pathogens (연교 추출물의 항균활성 검색 및 일부 식중독균의 증식에 미치는 영향)

  • This study was performed to investigate the antimicrobial effect of the Forsythiae Fructus extracts against food-borne pathogens. First, Forsythiae Fructus was extracted with methanol at room temperature and the methanol extracts were fractionated by using petroleum ether, chloroform, ethyl acetate, and methanol. The antimicrobial activity of the Forsythiae Fructus extracts was determined by using a paper disc method against food-borne pathogens and food spoilage bacteria. The methanol extracts of Forsythiae Fructus showed the highest antimicrobial activity against Salmonella paratryphimurium and Salmonella typhimurium. A synergistic effect in inhibition was observed when Forsythiae Fructus extract was mixed with Ulmus davidiana Japonica extract as compared to each extract alone. Finally, the growth inhibition curves were determined by using ethyl acetate extracts of Forsythiae Fructus against Shigella flexneri and Salmonella paratyphimurium. The aqueous extract of Forsythiae Fructus had strong antimicrobial activity against Staphylococcus epidermidis at the concentration of 10,000 ppm. At this concentration, the growth of Shigella fexneri was retarded for more than 24 hours and for up to 12 hours for Staphylococcus epidermidis. In conclusion, the methanol extracts of Forsythiae Fructus efficiently inhibited Staphylococcus epidermidis and Shigella flexneri.

Inhibitory effect of Erythronium japonicum on the human breast cancer cell metastasis

  • BACKGROUND/OBJECTIVES: In this study, the inhibitory effect of Erythronium japonicum extracts on the metastasis of MDA-MB-231 human breast cancer cell line was determined. MATERIALS/METHODS: Cells were cultured with DMSO or with 50, 75, 100 or $250{\mu}g/ml$ of Erythronium japonicum methanol or ethanol extract. RESULTS: Both methanol and ethanol extracts significantly inhibited the growth and induced apoptosis of MDA-MB-231 cells in a dose-dependent manner. Erythronium japonicum extracts inhibited the adhesion of MDA-MB-231 cells. The invasion of breast cancer cells was suppressed by Erythronium japonicum extracts in a dose-dependent manner. The motility and MMP-2 and MMP-9 activities were also inhibited by both methanol and ethanol extracts. CONCLUSIONS: Our results collectively indicate that Erythronium japonicum extracts inhibit the growth, adhesion, migration and invasion as well as induce the apoptosis of human breast cancer cells. Clinical application of Erythronium japonicum as a potent chemopreventive agent may be helpful in limiting breast cancer invasion and metastasis.

Antifungal and Insecticidal Activity of Methanol Extracts from 17 Medicinal Plants against Biological Deterioration of Organic Cultural Heritage (약용식물 17종 메탄올 추출물의 유기질문화재 생물학적 열화원에 대한 항진균 및 살충활성 조사)

  • To investigate bioactive materials for development of natural conservative agent on organic cultural heritage, methanol extracts from 17 medicinal plants were screened for antifungal and insecticidal activity against 6 wood rot fungi and adult of Lasioderma serricorne. Antifungal activity of extracts was tested by using paper disc soaking method against wood-rot fungi. Among these extracts, the most significant antimicrobial activity was observed from the extract of Coptis japonica at 5 mg/disc against all wood rot fungi tested. The insecticidal activity of extracts was examined by topical application against L. serricorne adults. The extracts from Asarum sieboldii gave 83.3 % mortality at $25{\mu}g/adult$ for 24hr. From these result, the methanol extract from C. japonica and A. sieboldii described could be useful for conservation of organic cultural heritage against biological deterioration by wood rot fungi and L. serricorne.

In Vitro Screening of Anti-lice Activity of Pongamia pinnata Leaves

  • Growing patterns of pediculocidal drug resistance towards head louse laid the foundation for research in exploring novel anti-lice agents from medicinal plants. In the present study, various extracts of Pongamia pinnata leaves were tested against the head louse Pediculus humanus capitis. A filter paper diffusion method was conducted for determining the potential pediculocidal and ovicidal activity of chloroform, petroleum ether, methanol, and water extracts of P. pinnata leaves. The findings revealed that petroleum ether extracts possess excellent anti-lice activity with values ranging between 50.3% and 100% where as chloroform and methanol extracts showed moderate pediculocidal effects. The chloroform and methanol extracts were also successful in inhibiting nymph emergence and the petroleum ether extract was the most effective with a complete inhibition of emergence. Water extract was devoid of both pediculocidal and ovicidal activities. All the results were well comparable with benzoyl benzoate (25% w/v). These results showed the prospect of using P. pinnata leave extracts against P. humanus capitis in difficult situations of emergence of resistance to synthetic anti-lice agents.

  • The antioxidant activity of various solvent extracts from Jeju camellia mistletoe (Korthalsella japonica (Thunb.) Engl.) was investigated using various in vitro assays as the 1,1-diphenyl-2-picrylhydrazyl radical scavenging, ferrous ion chelating and reducing power assays. Methanol and ethanol extracts showed the most potent antioxidant activity in all assays tested followed by water extract. The inhibitory effect of the Jeju camellia mistletoe extracts on pancreatic lipase and $\acute{a}$-glucosidase was also evaluated and the results showed that methanol and ethanol extracts markedly reduced both enzyme activities. Therefore, the methanol and ethanol extracts of Jeju camellia mistletoe is definitely worthy of further investigation for these beneficial effects on nutraceutical medicine.

Anti-aging and Anti-dementia Activities of Different Solvent Extracts from Silvetia siliquosa (뜸부기(Silvetia siliquosa) 추출물의 항노화 및 항치매 활성)

  • This study investigated the ingredients, anti-aging and anti-dementia activities of the Korean marine algae Silvetia siliquosa. The S. siliquosa solvent extracts were prepared with 70% ethanol, 80% methanol, and distilled water. The extraction yield range of various solvent extracts was 15.82-49.98%. The ethanol and methanol extracts had higher tyrosinase and collagenase inhibitory activities than those of the water extract. Meanwhile, all extracts exhibited high elastase inhibitory activity. Conversely, the methanol and water extracts exhibited the highest acetylcholinesterase inhibitory activity (IC50, 0.40 mg/mL) and β-secretase inhibitory activity (IC50, 0.81 ㎍/mL), respectively. These results indicate that S. siliquosa may be useful in food and pharmaceutical materials as a cosmetic and functional.
