Structural Investigation of the Hydrolysis-Condensation Process of Modified Titanium Isopropoxide

  • The structures of modified Ti$(OPr^i)_4$ with chelating ligands (L) such as ethylacetoacetate (Etac), acetylacetone (Acac) and methylacetoacetate (Mtac) were identified by using IR, ¹H NMR and $^13C$ NMR spectroscopies, and the octahedral structure was confirmed after modification. The pre-edge peaks of XANES spectra of modified metal alkoxides also denoted the mixture of five-fold and six-fold structures. The EXAFS fitting results showed the local structure around Ti atom after alkoxide modification. The hydrolysis-condensation rates of modified Ti alkoxide with organic additives were investigated by ¹H NMR spectroscopy. The Ti$(OPr^i)_4$ modified by Acac was less reactive toward hydrolysis-condensation reaction than those modified by the other alkoxides, which can be attributed to the stable ligand structure between Ti$(OPr^i)_4$ and Acac. The small particle size of modified Ti$(OPr^i)_4$ sol was obtained when Acac was employed.

Effect of Zinc on Vascular Smooth Muscle Cell Death Mediated by PDTC

  • Pyrrolidinedithiocarbamate (PDTC) and N-Acetylcysteine (NAC) are metal and nonmetal-chelating antioxidant which can induce rat and human smooth muscle cell death. When the smooth muscle cells from mouse aorta (MASMC) that we successfully cultured recently was exposed to PDTC and NAC in a normal serum state, the cells were induced to death by these compounds. However, PDTC did not induce the cell death in a serum depleted medium. This data suggests that certain factors in the serum may mediate the cytotoxic effect of PDTC. The metal chelator, Ca-EDTA blocked PDTC-induced cell death, but Cu-, Fe-, and Zn-EDTA did not block the PDTC-induced cell death. This data indicated that copper, iron, and zinc in the serum may lead to the cytotoxic effect of PDTC. Investigation of the intracellular zinc level in PDTC-induced smooth muscle cell death using the zinc probe dye N-(6-methoxy-8-quinolyl)-p-toluenesulfonamide shows that only the muscle-containing layers of the arteries have higher level of zinc. As expected, PDTC increased the intracellular fluorescence level of the zinc. In agreement with these results, the addition of an exogenous metal, zinc, induced the vascular aortic smooth muscle cell death which led to an increased intracellular zinc level. We concluded that PDTC induced mouse aortic smooth muscle cell death required not only zinc level but also intracellular copper and iron level. The mechanism of this antioxidant to induce vascular smooth muscle cell death may provide a new strategy to prevent their proliferation in arteriosclerotic lesions.

pT7MT, a Metallothionein 2A-Tagged Novel Prokaryotic Fusion Expression Vector

  • In the present article, a novel fusion expression vector for Escherichia coli was developed based on the pTORG plasmid, a derivative of pET32a. This vector, named pT7MT(GenBank Accession No DQ504436), carries a T7 promoter and it drives the downstream gene encoding Metallothionein 2A(MT2A). There are in-framed multiple cloning sites(MCS) downstream of the MT2A gene. A target gene can be cloned into the MCS and fused to the C-terminal of the MT2A gene in a compatible open reading frame(ORF) to achieve fusion expression. The metal-binding capability of MT2A allows the purification of fusion proteins by metal chelating affinity chromatography, known as $Ni^{2+}$-affinity chromatography. Using this expression vector, we successfully got the stable and high-yield expression of MT2A-GST and MT2A-Troponin I fusion proteins. These two proteins were easily purified from the supernatant of cell lysates by one-step $Ni^{2+}$-affinity chromatography. The final yields of MT2A-GST and MT2A-Troponin I were 30mg/l and 28mg/l in LB culture, respectively. Taken together, our data suggest that pT7MT can be applied as a useful expression vector for stable and high-yield production of fusion proteins.

Organic amendment-driven removal and speciation of metals using wormwood in two contrasting soils near an abandoned copper mine

  • To test the hypothesis that humic acid (HA), anaerobically digested pig slurry filtrate (APS), and their combination would differently affect the chemical speciation and extractability of metals (cadmium, copper, and zinc) and their uptake by plants, we conducted a pot experiment using wormwood in two texturally contrasting soils (sandy loam and clay loam) collected from a field near an abandoned Cu mine. Four treatments were laid out: HA at $ 23.5g\;kg^{-1}$ (HA), APS at $330mL\;kg^{-1}$ (APS), HA at $ 23.5g\;kg^{-1}$ and APS at $330mL\;kg^{-1}$ (HA + APS), and a control. Each treatment affected the chemical speciation and mobility of the metals, and thereby resulting in variable patterns of plant biomass yield and metal uptake. The APS supported plant growth by increasing nutrient availability. HA supported or hindered plant growth by impacting the soil's water and nutrient retention capacity and aeration, in a soil texture-dependent manner, while consistently enhancing the immobilization of heavy metals. Temporal increases in whole-plant dry matter yield and metal accumulation suggested that the plants were capable of metal hyperaccumulation. The results were discussed in terms of the mobility of metals and plant growth and corroborated by the $^{15}N$ recovery of soil- and plant-N pools under H and HS treatments. Therefore, for effective phytoremediation of polluted soils, an appropriate combination of plant growth promoters (APS) and chelating agents (HA) should be predetermined at the site where chemical stabilization of pollutants is desired.

Anti-Melanogenesis and Anti-Wrinkle Effects of Sargassum micracanthum Extracts (잔가시 모자반 추출물의 주름 개선 및 미백 효과)

  • Sargassum micracanthum (SM) is a member of the family Sargassaceae and commonly occurs along the coast of Korea. Extracts from SM were evaluated for their antioxidant and collagenase and tyrosinase inhibitory activities based on their total phenolic concentration (TPC), 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging effect, and metal-chelating effect. The TPC of SM ethanol and water extracts used in the study was 11.20 and 11.70 mg/g of dry sample, respectively. Both SM ethanol and water extracts had high DPPH radical-scavenging effect. The metal-chelating effect of SM ethanol extract (40.47% at 0.5 mg/ml) was higher than that of water extract (23.28% at 0.5 mg/ml). With regard to the anti-wrinkling effect, SM ethanol extract showed collagenase inhibitory activity with an $IC_{50}$ value of $488.20{\mu}g/ml$. Lastly, regarding the anti-melanogenesis effect, SM ethanol extract showed higher tyrosinase inhibitory activity (45.08% at 5 mg/ml) than that shown by the water extract (21.29% at 5 mg/ml). These results suggest that SM has the potential to be a resource with natural anti-melanogenesis, anti-wrinkle, and anti-oxidant effects.

Extraction Characteristics and Antioxidant Activity of Ethanol Extract of Rhus javanica Bark (붉나무 껍질 에탄올 추출물의 추출특성과 항산화 활성에 관한 연구)

  • This study was conducted to investigate the several physicochemical extraction characteristics and the antioxidant activities of ethanol extract from Rhus javanica bark. The contents of soluble solid and sugar in extract was measured to 73.5 mg/100g dry basis and $17.8^{\circ}Brix$, respectively. The contents of total aromatics, total flavonoids and total phenolic compounds was measured to 0.508 in absorbance, 49.88 mg/100g and 296.6 mg/100g, respectively. The reducing power of extract was about 27.5 % of ascorbic acid with the same soluble solid contents of the extract. But the ferric reducing antioxidant power and the DPPH radical scavenging ability of extract were measured to equivalent to those of ascorbic acid. The metal ion chelating ability of the extract was 81.58 % whereas that of ascorbic acid was 74.73 %. The nitrite scavenging ability of the extract was measured to 51.76 % at pH 2. And the antioxidative effect of the extract on soybean oil was observed with Rancimat test.

Comparison of Total Polyphenols, Total Flavonoids, and Biological Activities of Black Chokeberry and Blueberry Cultivated in Korea (국내산 Black Chokeberry와 Blueberry의 총 폴리페놀, 총 플라보노이드 함량 및 생리활성 비교)

  • This study investigated the biological activities of 70% methanol extracts from black chokeberry and blueberry by measuring DPPH, superoxide anion and ABTS radical scavenging activities, metal chelating effect, reducing power, and nitrite scavenging activity. Extraction yields of black chokeberry and blueberry were 62.90% and 67.09%, respectively. Total polyphenol and total flavonoid contents were 117.20 mg/g and 32.50 mg/g for black chokeberry and 42.26 mg/g and 26.39 mg/g for blueberry, respectively. Black chokeberry had higher antioxidant activity than blueberry. DPPH and ABTS radical scavenging activities, nitrite scavenging activity, and metal chelating effect of black chokeberry were 98.29%, 96.68%, 97.64%, and 91.33%, respectively, at a concentration of 5 mg/mL, which were equal to those of ascorbic acid (positive control). These results suggest that black chokeberry has potent biological activities, and in the future, the availability of black chokeberry will increase in the field of value added food products.

  • Amberlite XAD-7 and XAD-4 resins impregnated with DXHQ (5,7-dihalo-8-hydroxyquinoline) were prepared for the adsorption, separation and recovery of heavy metal ions from aqueous solutions. The characteristics of the impregnated resins, DXHQ (X : Cl, Br, I)-XAD were studied to find out the proper pairs of resin and DXHQ for the adsorption of metal ions. The increasing order of the impregnated amount of DXHQ onto XAD-7 resin was as follows: DCHQ < DBHQ < DIHQ. It was observed from the plot of log $K_d$ vs. pH that the optimum pH range for the adsorption of DIHQ onto XAD-4 resin was from 3.0 to 7.0. The stabilities of the DXHQ-XAD resins were investigated by measuring the amount of DXHQ remained on the XAD resin after shaking the DXHQ-XAD resins in various solutions of pH ranging from 2 to 12 and hydrochloric acid solutions. The impregnated resins were considerably stable in both acidic and neutral solutions. The amount of DIHQ leached from DIHQ-XAD-4 resin by eluting with various HCl solutions (1 ∼ 5M) was negligible, but in the case of XAD-7 resin it increases as the concentration of HCl solution increases. The optimum pH ranges, absorption mole ratio (M : DXHQ) and adsorption capacities (mmol metal per gram of resin) for the adsorption of metal ions onto the DXHQ-XAD resins were determined respectively. The stability of metal ion absorbed by the DXHQ-XAD resins was observed as the following order: M-DCHQ-XAD-7 < M-DBHQ-XAD-7 < M-DIHQ-XAD-7. The adsorbed metal ions were quantitatively recovered by eluting with HCl (0.5 ∼ 5M) and DXHQ-XAD resins could be reused over 5 times without re-impregnation of DXHQ.

A Study on the Adsorption and Recovery of Metal Ions by Amberlite XAD Resins Impregnated with Oxime Compounds (Oxime 화합물을 침윤시킨 Amberlite XAD 수지에 의한 금속이온의 흡착 및 회수에 관한 연구)

  • The adsorption behaviors of some oxime compounds well known as metal chelating agents on the Amberlite XAD resins were compared by measuring their distribution coefficients (log Kd) in various media, respectively. Among the oxime compounds, salicylaldoxime (SAO) and $\alpha-benzoinoxime(${\alpha}$-BzO)$ which showed large log Kd values were chosen. The characteristics of XAD-4 resins impregnated with SAO and ${\alpha}$-BzO have been studied to apply them for the adsorption and recovery of minute quantities of metal ions in aqueous solution. The optimum conditions for adsorption of SAO and ${\alpha}$-BzO on the resin were 30% methanol media having pH range of 1~8(for SAO) and 1~9 (for ${\alpha}$-BzO), respectively. The distribution coefficients of two oxime compounds were decreased as temperature increased. From the adsorption enthalpy data of SAO and ${\alpha}$-BzO, ranging from 4.96 to 6.66 Kcal/mol, it is suggested that their adsorption mechanism on XAD-4 resin is likely due to molecular adsorption equivalent to dipole-dipole interaction. The impregnated resins were considerably stable in the aqueous solutions of pH 5.0~10.0 and in 0.1~5M hydrochloric acid solutions. The former is the medium for adsorption of metal ions, while the latter is for recovery of the adsorbed metal ions. The adsorption mole ratio of Mn(II), Co(II), Ni(II), Zn(II) ions on SAO-XAD-4 and ${\alpha}$-BzO-XAD-4 resins were about 1 : 2 at the optimum conditions, respectively. The adsorbed metal ions were recovered completely by eluting with 3M HCl-50% methanol solution

  • The adsorption behavior of 8-hydroxyquinoline (8HQ) on Amberlite XAD-4 and-7 resins was investigated by measuring its distribution coefficients under various experimental conditions, such as shaking time, pH and concentration of methanol in the medium. The application of 8HQ-impregnated-XAD resins for the absorption and separation of metal ions was studied. The maximum adsorption of 8HQ on XAD resins was observed in the 30% methanol solution having pH range from 6.0 to 9.0. The impregnation capacities of XAD resins for 8HQ were 3.81${\times}$10-2mmol, 8HQ/g, XAD-4 resins and 2.60${\times}$10-2mmol, 8HQ/g, XAD-7 resin, respectively. The 8HQ-impregnated-XAD resins were stable in pH range from 6.0 to 10.0 and the amount of 8HQ leached from XAD-4 resin by eluting with hydrochloric acid(above 5M) was negligible. The optimum pH range for the adsorption of metal ions on 8HQ-impregnated XAD resin was also 6.0 to 10.0, and the adsorption mole ratio of metal ion to 8HQ were 1 : 2 for Cu(II), Cd(II) and Ni(II), and 1 : 3 for Fe(III) at the above pH range. It was found that the absorbed metal ions on 8HQ-impregnated-XAD resins were recovered quantitatively with 5M HCl and 8HQ-impregnated-XAD-4 resin could be reusable over 5 times without decrease in its impregnation capacity.
