This work describes a new strategy for optimal design of Multiplex-PCR primer sequences. The process is based on the Particle Swarm Optimization-Simplex algorithm (Mult-PSOS). Diverging from previous solutions centered on heuristic tools, the Mult-PSOS is selfconfigured because it does not require the definition of the algorithm's initial search parameters. The successful performance of this method was validated in vitro using Multiplex-PCR assays. For this validation, seven gene sequences of the most prevalent bacteria implicated in urinary tract infections were taken as DNA targets. The in vitro tests confirmed the good performance of the Mult-PSOS, with respect to infectious disease diagnosis, in the rapid and efficient selection of the optimal oligonucleotide sequences for Multiplex-PCRs. The predicted sequences allowed the adequate amplification of all amplicons in a single step (with the correct amount of DNA template and primers), reducing significantly the need for trial and error experiments. In addition, owing to its independence from the initial selection of the heuristic constants, the Mult-PSOS can be employed by non-expert users in computational techniques or in primer design problems.
This study presents a particle swarm optimization algorithm integrated with weighted particle concept and improved fly-back technique. The rationale behind this integration is to utilize the affirmative properties of these new terms to improve the search capability of the standard particle swarm optimizer. Improved fly-back technique introduced in this study can be a proper alternative for widely used penalty functions to handle existing constraints. This technique emphasizes the role of the weighted particle on escaping from trapping into local optimum(s) by utilizing a recursive procedure. On the other hand, it guaranties the feasibility of the final solution by rejecting infeasible solutions throughout the optimization process. Additionally, in contrast with penalty method, the improved fly-back technique does not contain any adjustable terms, thus it does not inflict any extra ad hoc parameters to the main optimizer algorithm. The improved fly-back approach, as independent unit, can easily be integrated with other optimizers to handle the constraints. Consequently, to evaluate the performance of the proposed method on solving the truss weight minimization problems with discrete variables, several benchmark examples taken from the technical literature are examined using the presented method. The results obtained are comparatively reported through proper graphs and tables. Based on the results acquired in this study, it can be stated that the proposed method (integrated particle swarm optimizer, iPSO) is competitive with other metaheuristic algorithms in solving this class of truss optimization problems.
Optimizing composite structures to exploit their maximum potential is a realistic application with promising returns. In this research, simultaneous maximization of the fundamental frequency and frequency separation between the first two modes by optimizing the fiber angles is considered. A high-fidelity design optimization methodology is developed by combining the high-accuracy of finite element method with iterative improvement capability of metaheuristic algorithms. Three powerful nature-inspired optimization algorithms viz. a genetic algorithm (GA), a particle swarm optimization (PSO) variant and a cuckoo search (CS) variant are used. Advanced memetic features are incorporated in the PSO and CS to form their respective variants-RPSOLC (repulsive particle swarm optimization with local search and chaotic perturbation) and CHP (co-evolutionary host-parasite). A comprehensive set of benchmark solutions on several new problems are reported. Statistical tests and comprehensive assessment of the predicted results show CHP comprehensively outperforms RPSOLC and GA, while RPSOLC has a little superiority over GA. Extensive simulations show that the on repeated trials of the same experiment, CHP has very low variability. About 50% fewer variations are seen in RPSOLC as compared to GA on repeated trials.

The paper presents problems of optimization of the synthesis of embedded systems, in particular Pareto optimization. The model of such a system for its design for high-level of abstract is based on the classic approach known from the theory of task scheduling, but it is significantly extended, among others, by the characteristics of tasks and resources as well as additional criteria of optimal system in scope structure and operation. The metaheuristic algorithm operating according to this model introduces a new approach to system synthesis, in which parallelism of task scheduling and resources partition is applied. An algorithm based on a genetic approach with simulated annealing and Boltzmann tournaments, avoids local minima and generates optimized solutions. Such a synthesis is based on the implementation of task scheduling, resources identification and partition, allocation of tasks and resources and ultimately on the optimization of the designed system in accordance with the optimization criteria regarding cost of implementation, execution speed of processes and energy consumption by the system during operation. This paper presents examples and results for multi-criteria optimization, based on calculations for specifying non-dominated solutions and indicating a subset of Pareto solutions in the space of all solutions.
Reliability-Based Design Optimization (RBDO) is an appropriate framework for obtaining optimal designs by taking uncertainties into account. Large-scale problems with implicit limit state functions and problems with discrete design variables are two significant challenges to traditional RBDO methods. To overcome these challenges, this paper proposes a hybrid method to perform RBDO of structures that links Firefly Algorithm (FA) as an optimization tool to advanced (finite element) reliability methods. Furthermore, the Genetic Algorithm (GA) and the FA are compared based on the design cost (objective function) they achieve. In the proposed method, Weighted Simulation Method (WSM) is utilized to assess reliability constraints in the RBDO problems with explicit limit state functions. WSM is selected to reduce computational costs. To performing RBDO of structures with finite element modeling and implicit limit state functions, a First-Order Reliability Method (FORM) based on the Direct Differentiation Method (DDM) is utilized. Four numerical examples are considered to assess the effectiveness of the proposed method. The findings illustrate that the proposed RBDO method is applicable and efficient for RBDO problems with discrete and continuous design variables and finite element modeling.
Due to the importance of accurate analysis of bearing capacity in civil engineering projects, this paper studies the efficiency of two novel metaheuristic-based models for this objective. To this end, black hole algorithm (BHA) and multi-verse optimizer (MVO) are synthesized with an artificial neural network (ANN) to build the proposed hybrid models. Based on the settlement of a two-layered soil (and a shallow footing) system, the stability values (SV) of 0 and 1 (indicating the stability and failure, respectively) are set as the targets. Each model predicted the SV for 901 stages. The results indicated that the BHA and MVO can increase the accuracy (i.e., the area under the receiving operating characteristic curve) of the ANN from 94.0% to 96.3 and 97.2% in analyzing the SV pattern. Moreover, the prediction accuracy rose from 93.1% to 94.4 and 95.0%. Also, a comparison between the ANN's error decreased by the BHA and MVO (7.92% vs. 18.08% in the training phase and 6.28% vs. 13.62% in the testing phase) showed that the MVO is a more efficient optimizer. Hence, the suggested MVO-ANN can be used as a reliable approach for the practical estimation of bearing capacity.

In this paper, a numerical procedure is proposed for achieving the minimum cost design of reinforced concrete polygonal column cross-sections under compression and biaxial bending. A methodology is developed to integrate the metaheuristic algorithm Quantum Particle Swarm Optimization (QPSO) with an algorithm for the evaluation of the strength of reinforced concrete cross-sections under combined axial load and biaxial bending, according to the design criteria of Brazilian Standard ABNT NBR 6118:2014. The objective function formulation takes into account the costs of concrete, reinforcement, and formwork. The cross-section dimensions, the number and diameter of rebar and the concrete strength are taken as discrete design variables. This methodology is applied to polygonal cross-sections, such as rectangular sections, rectangular hollow sections, and L-shaped cross-sections. To evaluate the efficiency of the methodology, the optimal solutions obtained were compared to results reported by other authors using conventional methods or alternative optimization techniques. An additional study investigates the effect on final costs for an alternative parametrization of rebar positioning on the cross-section. The proposed optimization method proved to be efficient in the search for optimal solutions, presenting consistent results that confirm the importance of using optimization techniques in the design of reinforced concrete structures.

An efficient optimization algorithm and damage-sensitive objective function are two main components in optimization-based Finite Element Model Updating (FEMU). A suitable combination of these components can considerably affect damage detection accuracy. In this study, a new hybrid damage-sensitive objective function is proposed based on combining two different objection functions to detect the location and extent of damage in structures. The first one is based on Generalized Pseudo Modal Strain Energy (GPMSE), and the second is based on the element's Generalized Flexibility Matrix (GFM). Four well-known population-based metaheuristic algorithms are used to solve the problem and report the optimal solution as damage detection results. These algorithms consist of Cuckoo Search (CS), Teaching-Learning-Based Optimization (TLBO), Moth Flame Optimization (MFO), and Jaya. Three numerical examples and one experimental study are studied to illustrate the capability of the proposed method. The performance of the considered metaheuristics is also compared with each other to choose the most suitable optimizer in structural damage detection. The numerical examinations on truss and frame structures with considering the effects of measurement noise and availability of only the first few vibrating modes reveal the good performance of the proposed technique in identifying damage locations and their severities. Experimental examinations on a six-story shear building structure tested on a shake table also indicate that this method can be considered as a suitable technique for damage assessment of shear building structures.
Structural design, in general, is developed through trial and error technique which is guided by standards criteria and based on the intuition and experience of the engineer, a context that leads to structural over-dimensioning, with uneconomic solutions. Aiming to find the optimal design, structural optimization methods have been developed to find a balance between cost, structural safety, and material performance. These methods have become a great opportunity in the steel structural engineering domain since they have as their main purpose is weight minimization, a factor directly correlated to the real cost of the structure. Assuming an objective function of minimum weight with stress and displacement constraints provided by Brazilian standards, the present research proposes the sizing optimization and combined approach of sizing and shape optimization, through a software developed to implement the Simulated Annealing metaheuristic algorithm. Therefore, two steel plane frame layouts, each admitting four typical truss geometries, were proposed in order to expose the difference between the optimal solutions. The assessment of the optimal solutions indicates a notable weight reduction, especially in sizing and shape optimization combination, in which the quantity of design variables is increased along with the search space, improving the efficiency of the optimal solutions achieved.
