Prevalence of Clonorchis sinensis Metacercariae in Fish from Water Systems of Seomjin-gang (River)

  • The prevalence of Clonorchis sinensis metacercariae (CsMc) was examined in freshwater fish from the water systems of Seomjin-gang (River), the Republic of Korea. Total 1,604 fish from 7 local sites of Seomjin-gang were examined by artificial digestion methods. The metacercariae of C. sinensis were detected in 102 (39.8%) out of 256 fish (14 species) from the upper reaches of Seomjin-gang, i.e., Osucheon (22.3% in 6 fish species) in Imsil-gun, and Seomjin-gang (63.9% in 9 fish species) in Sunchang-gun, Jeollabuk-do. Their average density was 9.0 per infected fish. They were also found in 132 (48.0%) out of 275 fish (12 spp.) from the middle reaches of Seomjin-gang, i.e., Songdaecheon (58.9% in 4 fish species) in Namwon-si, Jeollabuk-do, and Seomjin-gang (45.2% in 10 fish species) in Gokseong-gun, Jeollanam-do. Their average density was 21.0 per infected fish. CsMc were detected in 77 (56.6%) out of 136 fish (11 species) from the lower reaches of Seomjin-gang, i.e., Seomjin-gang (73.3% in 11 fish species) in Gurye-gun, Jeollanam-do, and Namsancheon (8.6% in 1 fish species) in Hadong-gun, Gyeongsangnam-do. Their average density was 64.9 per infected fish. The metacercariae of Metorchis orientalis were also detected in 6 fish species from 4 sites of Seomjin-gang. Conclusively, it has been confirmed that CsMc are more or less prevalent in fish from some water systems of Seomjin-gang in Korea.

Infections with Centrocestus armatus Metacercariae in Fishes from Water Systems of Major Rivers in Republic of Korea

  • The infection status of Centrocestus armatus metacercariae (CaMc) was broadly surveyed in freshwater fishes from major river systems in the Republic of Korea (Korea) during 2008-2017. A total of 14,977 fishes was caught and examined by the artificial digestion method. CaMc were detected in 3,818 (97.1%) (2,114 Z. platypus: 96.1% and 1,704 Z. temminckii: 98.4%) out of 3,932 Zacco spp. examined and their density was 1,867 (2,109 in Z. platypus and 1,567 in Z. temminckii) per fish infected. The prevalences with CaMc were high, 93.7-100%, in Zacco spp. from all surveyed areas. However, their densities were more or less different by the surveyed areas and fish species. They were most high in Nakdong-gang in Gyeongsangnam-do (4,201 in average), and followed by Geum-gang (2,343), Nakdong-gang in Gyeongsangbuk-do (1,623), Han-gang (1,564), Tamjin-gang and Yeongsan-gang (1,540), streams in the east coast (1,028), Seomjin-gang (488) and Mangyeong-gang (170). In another species of rasborinid fish, Opsariichthys uncirostris amurensis, CaMc were detected in 222 (74.8%) out of 297 ones examined and their density was 278 (1-4,480) per fish infected. CaMc were also detected in total 41 fish species except for the rasborinid fish, Z. platypus, Z. temminckii and O. uncirostris amurensis. Conclusively, it was confirmed that among the 3 species of rasborinid fish, Z. platypus and Z. temminckii are highly prevalent and O. uncirostris amurensis is moderately prevalent with CaMc. Additionally, we could know that variety of fish species act as the second intermediate hosts of C. armatus in Korea.

Infection Status with Metagonimus spp. Metacercariae in Fishes from Seomjin-gang and Tamjin-gang in Republic of Korea

  • To grasp the infection status of Metagonimus spp. metacercariae (MsMc), the freshwater fishes were surveyed from Seomjin-gang (river) and Tamjin-gang in the Republic of Korea. Total 1,604 fishes from 7 local sites of Seomjin-gang and 1,649 fishes from 2 sites of Tamjin-gang were examined for 6 years (2012-2017) by the artificial digestion method. MsMc were detected in fishes from 7 sites, i.e., Osucheon in Imsil-gun (36.3% fish in 6 spp.), Seomjin-gang in Sunchanggun (49.8% in 18 spp.), Songdaecheon in Namwon-si (64.5% in 8 spp.), Seomjin-gang in Gokseong-gun (72.4% in 14 spp.) and in Gurye-gun (78.8% in 17 spp.), Hoengcheon (75.9% in 11 spp.) and Namsancheon (58.9% in 7 spp.) in Hadong-gun. Their average densities were 4.2, 86.8, 39.9, 43.1, 246.5, 173.6 and 67.5 per fish infected respectively. Prevalence with MsMc in rasborinid fish from Seomjin-gang was 89.2% and their intensity was 73.6 per fish infected. Endemicities of MsMc in rasborinid fish from Seomjin-gang were significantly higher in fishes from the lower reaches (prevalence: 98.7%; density: 137) rather than in fishes from the middle (93.5%; 38) and upper (72.4%; 13) reaches. MsMc were also detected in 56.1% and 66.4% fishes from 2 surveyed areas, i.e., the middle reaches in Jangheung-gun and the lower reaches in Gangjin-gun, of Tamjin-gang, and their densities were 147 and 121 per fish infected. In susceptible fishes from Tamjin-gang, the prevalence was 84.1% and density was 227 per fish infected. By the present study, it was confirmed that MsMc is highly prevalent in the fishes from Seomjin-gang and Tamjin-gang in Korea.

강화군(江華郡)의 폐흡충중간숙주(肺吸虫中間宿主)에 관(関)한 역학적(疫學的) 추이(推移) (Epidemiological change on Paragonimiasis 2nd intermediate host in Kang Hwa Gun)

  • Paragonimiasis is a common trematodiasis of medical importance and one of public health problems in many areas of sob thern part of Korea. During the last sixty years, several on the prevalence among the Korean population have been carried out by various investigators, although the general distribution and prevalences of paragonimiasis are now yet clearly determind. This study was undertaken to evaluate the status of human paragonimiasis in Kang Hwa Gun, Kyung gi Province, Korea by the use of intradermal screening test whih Veronal buffered saline antigen. A total number of 208 residents (male 119, female 89) of all age group in Kilgeong-Ri, Yangdo Myun, Kang Hwa Gun were examined by the intradermal test with paragonimus antigen. The infestation rate of crayfish with Paragonimus metacerearia was also examine in Konneung and Jeoungha village, Kilgeong-Ri from March 1973 to October 1978 by the interval of two to five years. 1. In the resnlt of 208 residents, Kilgeong-Ri, the positive rate showed in 29.3% in as a whole(36.1% in male, 20.2% in female). The highest positive rate showed in 10 to 19 age group(41.5%). 2. Among the residents in Kilgeong-Ri, Paragonimus eggs were detected in 11 residents by the sputum and the stool examinations without the intradermal test result. 3. The infestation rate of crayfish with Paragonimus inetacercaria was 90.6%(116 out of 128 crayfishes) in Konneung village, and 61.5%(24 out of 39) crayfishes) in Jeongha village, Kilgeng-Ri. 4. The fluctation of the infestation tate and density of crayfish with Paragonimus metacercaria was decreased by the interval of two to five years in Konneung and Jeongha village in Kilgeong-Ri.

한국에서의 오리극구흡충 (Echinoparyphium recurvatum)의 생활사 (Life history of Echinopowphium recurvatum (Trematoda: Echinostomatidae) in Korea)

  • 이 연구는 자연계 및 실험실 내 조건 하에서 오리극구흡충의 생활사를 관찰하기 위하여 수행하였다. 1992년 8월과 9월에 부산시 강서구에 소재하는 선암천에서 다랑의 물달팽이 (Rndix owe,cdwiacoreona)를 채집하였다. 채집한 물달팽이 중 106개를 파쇄법으로 조사하였던 바, 52개 (89.0%)에서 오리극구흡충의 유충 (레디아, 유미유충, 피낭유충)이 한 종류 이상씩 검출되었다. 물달팽이로부터 자연 유출되는 유미유충을 접촉감염시킨 물달팽이, 논우렁 및 미꾸리 중 물달팽이와 미꾸리에서 피낭유충이 검출되었다. 물달팽이로부터 검출한 피낭유충을 경구감염시킨 병아리, 오리, 흰쥐, 마우스 중 병아리와 오리에서 성충이 검출되었다. 물달팽이로부터 검출한 피낭유 충을 경구감염시킨 병아리에서의 충체 회수율은 평균 13. 1%이었다. 레디아는 주머니 모양으로 평균 $2.437{\;}{\times}{\;}0.317{\;}mm$ 크기이었으며 근육성의 인두와 체장의 약 112 부위까지 내려와 있는 맹장을 하나씩 갖고 있었다. 유미유충의 체부위는 방추형으로 평균 $0.262{\;}{\times}{\;}0.129{\;}mm$ 크기이었고, 꼬리부위는 긴 막대모양으로 평균 $0.528{\;}{\times}{\;}0.056{\;}mm$ $\times$ 크기이었다. 유미유충의 체부위에서는 45개의 두극이 배열되어 있는 두관과 미세한 과립을 다수 보유하고 있는 배설관이 특징적으로 관찰되었다. 피낭유충뜬 구형으로 평균 $0.144{\;}{\times}{\;}0.142{\;}mm$ 크기이었으며 2중의 낭벽을 가지고 있었다. 성충은 비교적 길쭉하였고 평균 $0.2760{\;}{\times}{\;}0.550{\;}mm$ 크기이었으며 두관에는 특징적인 45개의 두극이 배열되어 있었다. 이상의 결과로 우리 나라에서 물달팽이가 오리극구흡충의 제1 및 제2중간숙주로서 생활사 유지에 중요한 역할을 수행하고 있음을 확인하였다.

Infection Status of Zoonotic Trematode Metacercariae in Fishes from Vientiane Municipality and Champasak Province in Lao PDR

  • The infection status of fishborne zoonotic trematode (FZT) metacercariae was investigated in fishes from 2 localities of Lao PDR. Total 157 freshwater fishes (17 species) were collected in local markets of Vientiane Municipality and Champasak Province in December 2010 and July 2011, and each fish was examined by the artificial digestion method. Total 6 species of FZT metacercariae, i.e., Opisthorchis viverrini, Haplorchis taichui, Haplorchis yokogawai, Haplorchis pumilio, Centrocestus formosanus, and Procerovum varium, were detected in fishes from Vientiane Municipality. The metacercariae of O. viverrini were detected in 50 (49.5%) out of 101 fishes (6 species), and their average number was 154 per fish infected. The remaining 5 species of heterophyid metacercariae were detected in 36.8%, 65.8%, 9.4%, 23.9%, and 5.1% fishes examined, and their average densities were 12, 1,038, 4, 15, and 13 per infected fish, respectively. In fishes from Champasak Province, 3 species of FZT metacercariae, i.e., O. viverrini, H. taichui, and H. yokogawai, were detected. Only 2 O. viverrini metacercariae were found in only 1 Barbonymus schwanefeldi. The metacercariae of H. taichui and H. yokogawai were detected in 60.0% and 50.0% of fishes examined, and their average densities were 47 and 28 per fish infected. By the present study, it has been confirmed that several species of FZT metacercariae are prevalent in fishes from Vientiane Municipality, with P. varium being a new member of FZT in Lao PDR. In comparison, FZT metacercariae are less prevalent in fishes from Champasak Province.

  • 실험실에서 사육한 수종 패류를 이용하여 Echinostoma cinetorchis의 생활환을 형성시키 며 발육과정을 관찰하였다. 성충체(흰쥐에서 얻음)에서 분리한 충란을 배양하여 miracidium을 얻고 제 1중간숙주인 Hippeutis sp. 우렁이에 20개씩 감염시킨 바 58.3%(12개 중 7)는 cercaria 유출 전에 감염사하였고 나머지는 25일 째부터 7∼9일간 cercaria를 유출하였다. 이 cercaria를 수종의 패류에 접촉감염(패당 40∼50마리)시켰더니 피낭유충 형성은 Hippeutis sp.에서 100%(접촉 cercaria에 대한 피낭 비율은 56.7%), Radix auricuzaria coreana 100% (66.4%), Phyla acute 66.7% (37.3%) 및 자연산 Cipangopaludina sp. 50% (8%) 감염으로 전 실험 패류가 피낭유충을 형성하였다. Cercaria는 침입시 노출부인 두부, 족부에 부착한 후 포복하여 외투강 내로 이행하며, 패 체내의 피낭부위는 외투막, 원체강내, 내부 기관 및 조직 사이였다. 그리고 이를 흰쥐에 감염시켜 성충을 검출할 수 있었다. 이상의 결과로 한국에 있어서 E. cinetorchis의 주요 인체 감염원 역할을 할 것으로 믿어지는 논우렁이(Cipnngopaludina sp.)에 있어서 피낭유충 형성을 확인할 수 있었고 이 극구흡충은 타종(특히 E. hortense)과 달리 여러 담수 패류가 제2중간숙주 역할을 하고 있음을 확인하였다.

Centrocestus armatus에 관한 연구 1. 피라미와 갈겨니의 피낭유충 감염상 (Study on Centyocestus armatus in Korea I. Infection status of Zucco plutupus and Z. temminckii with the metacercariae of C. armatus)

  • 우리 나라 5대 강의 19개 하천에서 피라미와 갈겨니 총 370마리를 1988년 6월부터 9월까지 포획하고 개체별로 인공소화시켜 C. armatus 피낭유충 감염상을 조사하였다. 피라미의 피낭유충 감염률은 평균 86.8%, 어체당 피낭유충 수는 평균 224개(1∼5,431)이었으며, 갈겨니에서는 78.5% 및 131개(1∼2,711)이었다. 한강 수계에서 포획된 어류의 피낭유충 감염률과 어체당 피낭유충 수는 평균 73.4% 및 32개(1∼289)이었으며, 영산강산 어류는 각각 88.0% 및 44개(2∼176)이었다. 금강, 탐진강 및 섬진강산 어류는 모두 피낭유충에 감염되어 있었으며 어체당 평균 피낭유충 수는 지역별로 각각 45개(2∼243), 59개(3∼311) 및 119개(1∼1,682)이었다. 낙동강 수계에서 포획된 어류의 피낭유충 감염률은 81.0%이었으며 어체당 피낭유충 수는 평균 348개(1∼5,431)이었다. 이 연구로 C. armatus 는 우리 나라 5대 강 유역에 널리 분포하고 있음을 확인하였다.

Gymnophaiioides seoi (Digenea: Gymnophallidae) 피낭유충의 시험관내 배양 (In vitro cultivation of Gymnophalloides seoi metacercariae (Digenea: Gymnophallidae))

  • 참굴큰입흡충(GMmophQHoidesseoi)은 새로운 인체 기생 장흡충으로서 최근 우리 나라에서 인체 감염의 유행이 보고된 바 있다. 이 연구는 참굴큰입흡충의 피낭유충을 시험관내에서 배양하여 성충으로 발육시킬 수 있는지 알아보고자 시행하였다. 피낭유충은 자연산 감염 굴에서 회수하였고. NCTC 109 배지에 우태아 혈청(FBS) 20%와 항생제-항진균제(Gibco) 1%를 첨가하여 사용하되 $37^{\circ}C$/5%{\;}CO_2,{\;}41^{\circ}C/8%{\;}CO_2{\;}및{\;}41^{\circ}C/5%{\;}CO_2$의 3가지 배양조건 하에서 배양하였다 배양 2일에서 5일까지 각 조건 하에서 얻은 충체를 실험 감염 C3H 마우스에서 회수한 충체와 발육 및 성숙도를 비교하였다. 배양군의 충체 크기는 길이 $200-300{\;}\mu\textrm{m}$로서 피낭유충과 비교할 때 유의한 차이를 보이지 않았으나. 마우스로부터 회수한 충체는 $300-400{\;}\mu\textrm{m}$에서 유의한 차이를 보였다. 마우스 체내, $41^{\circ}C/5%{\;}CO_2{\;}및{\;}41^{\circ}C/8%{\;}CO_2$ 조건 하에서는 모두 2일째에 충란이 형성되기 시작하였으나, $37^{\circ}C/5%{\;}CO_2$ 하에서는 배앙 5일까지도 충란이 형성되지 않았다 배양군 중에서는 $41^{\circ}C/5%{\;}CO_2$ 조건이 충체 성숙 및 충란 생산에 가장 적합하였으나 마우스 체내에서 자란 충체에 비해 자궁내 충란 수가 약 1/2에 불과하였다. 결론적으로 참굴큰입흡충의 시험관내 배양은 $41^{\circ}C/5%{\;}CO_2$ 또는 $41^{\circ}C/8%{\;}CO_2$ 조건에서 부분적으로 성공적임을 알 수 있었다.

Clonorchis sinensis metacercarial infection in the pond smelt Hypomesus olidus and the minnow Zacco platypus collected from the Soyang and Daechung Lakes

  • The pond smelt Hypomesus olidus and minnow Zacco platypus were collected from the Soyang and Daechung Lakes in January 2003, and their metacercarial infections was examined by the muscle compression and artificial digestion techniques. In the Soyang Lake, 161 metacercariae of Clonorchis sinensis (0.35 per fish) were harvested from 459 pond smelts examined. Also, 13 metacercariae of C. sinensis (0.43 per fish), 1 of Metagonimus sp., 4 of Echinostoma sp., 148 of Centrocestus armatus and 44 unidentified species were collected from 30 minnows. In the Daechung Lake, 369 metacercariae of C. sinensis (3.69 per fish) and 51 unidentified species were recovered from 100 pond smelts. The metacercariae of C. sinensis were fed to experimental rats, in which the adult flukes were identified. The pond smelts and minnows collected from the Soyang and Daechung Lakes were verified to be the second intermediate hosts and the sources of human C. sinensis infection.
