Polyacetylenes from the Tissue Cultured Adventitious Roots of Panax ginseng C.A. Meyer

  • Five polyacetylenes, ginsenoyne K (1), (Z)-1-methoxyheptadeca-9-en-4,6-diyne-3-one (2), panaxydol (3), panaxydiol (4), and (E)-heptadeca-8-en-4,6-diyne-3,10-diol (5) were isolated from the adventitious roots of Panax ginseng and their chemical structurFive polyacetylenes, ginsenoyne K (1), (Z)-1-methoxyheptadeca-9-en-4,6-diyne-3-one (2), panaxydol (3), panaxydiol (4), and (E)-heptadeca-8-en-4,6-diyne-3,10-diol (5) were isolated from the adventitious roots of Panax ginseng and their chemical structures were elucidated by interpretation of spectroscopic data. Among the isolated compounds, compounds 2 and 5 were reported for the first time as a natural product. Ginsenoyne K (1) showed dose-dependent inhibitory effect on dopa oxidase activity of tyrosinase.es were elucidated by interpretation of spectroscopic data. Among the isolated compounds, compounds 2 and 5 were reported for the first time as a natural product. Ginsenoyne K (1) showed dose-dependent inhibitory effect on dopa oxidase activity of tyrosinase.

  • 4-Hydroxy-3-(3'-methyl-2'-butenyl)-benzoic acid (HMBA) was previously isolated from Curvularia sp. KF119 as a cell-cycle inhibitor. However, the present study used a novel and practical synthetic method to prepare a large quantity of HMBA. The synthetic HMBA was found to inhibit the cell-cycle progression of HeLa cells with a comparable potency to the natural fungal metabolite. The inhibition of the cell-cycle progression by the synthetic HMBA involved both the activation of $p21^{WAF1}$ and the inhibition of cyclin D1 expression in the cells. Consequently, this new synthetic procedure provides an easy and convenient way to produce or manipulate the original fungal metabolite.

  • A high-performance liquid chromatographic assay using ion-pair reverse-phase system was developed for the separation of acebutolol and acebutolol acetyl metabolite in plasma. A ion-pair reversephase system consisting of an ODS-bonded silica column and a mixture of 20% $CH_3CN$, 0.1% $H_3PO_4$, 0.035 M heptanesulfonic acid and 0.005 M tetrabutylammonium hydrogen sulfate as the mobile phase were used. Triamterene was employed as an internal standard. Based on 0.2 ml of plasma, the detection limits were 10.4 ng/ml for acebutolol and 10.3 ng/ml of acebutolol acetyl metabolite at the signal-to-noise ratio of 3:1.

Fragrance and Metabolite Components of Flowers from Korean Native Apocynum lancifolium Russanov

  • This study characterizes the volatile aromatic and metabolite components of domestic native Apocynum lancifolium blossom. The accurate characterization of fragrances collected from the blossom was carried out using gas chromatography-mass. A total of 70 chemical components were identified, including ketones of acetophenone (29.22%), phenylethyl alcohol (10.54%), methyl-benzenemethanol (8.43%), benzyl alcohol (7.97%), natural bicyclic sesquiterpene types of caryophyllene (6.08%), gurjunene (6.20%), humulene (1.90%), and ocimene (1.04%). Overall, the content of ketones, alcohols, and terpenes was higher than that of others. The major metabolite components were pentanoic acid, malic acid, fructofuranoside, quinic acid, tagatose, sorbose, galactose, inositol, galactaric acid, glucopyranoside, and octadecenoic acid.

Identification of an ISR-Related Metabolite Produced by Pseudomonas chlororaphis O6 against the Wildfire Pathogen Pseudomonas syringae pv. tabaci in Tobacco

  • Pseudomonas chlororaphis O6 exhibits induced systemic resistance (ISR) against P. syringae pv. tabaci in tobacco. To identify one of the ISR metabolites, O6 cultures were extracted with organic solvents, and the organic extracts were subjected to column chromatography followed by spectroscopy analyses. The ISR bioassay-guided fractionation was carried out for isolation of the metabolite. High-resolution mass spectrometric analysis of the metabolite found $C_{9}H_{9}O_{3}N$ with an exact mass of 179.0582. LC/MS analysis in positive mode showed an $(M+H)^{+}$ peak at m/z 180. Nuclear magnetic resonance ($^{1}H,\;^{13}C$) analyses identified all protons and carbons of the metabolite. Based on the spectroscopy data, the metabolite was identified as 4-(aminocarbonyl) phenylacetate (4-ACPA). 4-ACPA applied at 68.0 mM exhibited ISR activity at a level similar to 1.0 mM salicylic acid. This is the first report to identify an ISR metabolite produced by P. chlororaphis O6 against the wildfire pathogen P. syringae pv. tabaci in tobacco.

  • Rosiglitazone metabolites in rat plasma were analyzed after intraperitoneal and oral administration to rats. Seven metabolites (M1-M7) were detected in rat plasma (IP and PO), and the structures were confirmed using liquid chromatography-triple time of flight (TOF) mass spectrometry; as a result, the most abundant metabolite was M5, a de-methylated rosiglitazone. Other minor in vivo metabolites were driven from monooxygenation and demethylation (M2), thiazolidinedione ring-opening (M1, M3), mono-oxygenation (M4, M7), and mono-oxygenation followed by sulfation (M6). Among them, M1 was found to be a 3-{p-[2-(N-methyl-N-2-pyridylamino)ethoxy]phenyl}-2-(methylsulfinyl)propionamide, which is a novel metabolite of rosiglitazone. There was no significant difference in the metabolic profiles resulting from the two administrations. The findings of this study provide the first comparison of circulating metabolite profiles of rosiglitazone in rat after IP and PO administration and a novel metabolite of rosiglitazone in rat plasma.

First of all, to analyze the metabolite(s) of CKD-712, a simple and sensitive detection method for CKD-712 was developed by using gas chromatography-mass spectrometry GC/MS). Urine was collected from adult male Sprague-Dawley rats 250${\pm}$10g) in metabolic cage for 24hr after oral administration of 100 mg/kg of CKD-712. The recovery of CKD-712 after extraction and concentration with AD-2 resin column was above 90 % from rat urine. The detection limits of CKD-712 in urine was approximately 0.1 ng/mL. It has well been suggested that isoquinoline possessing catechol moiety such as CKD-712 should be subjected to the catechol-O-methyl kransferase activity in vivo. We detected three major peaks of presumed CKD-712 metabolites in the total ion chromatogram obtained from the rat urine sample after oral administration of CKD-712. From these results, it is assumed that the urinary metabolites are mono-methylation in the naphthyl moiety (metabolite I ), methylation at the C-6 or 7 hydroxy group in the isoquinoline moiety and hydroxylation at in the naphthyl moiety (metaboliteII), and methylation at the C-6 or 7 hydroxy group in the isoquinoline moiety (metaboliteIII).

Metabolite Profiling and Microbial Community of Traditional Meju Show Primary and Secondary Metabolite Differences Correlated with Antioxidant Activities

  • Meju, a type of fermented soybean paste, is used as a starter in the preparation of various Korean traditional soybean-based foods. In this study, we performed Illumina-MiSeq paired-end sequencing for microbial communities and mass spectrometry analysis for metabolite profiling to investigate the differences between 11 traditional meju products from different regions across Korea. Even though the bacterial and fungal communities showed remarkable variety, major genera including Bacillus, Enterococcus, Variovorax, Pediococcus, Weissella, and Aspergillus were detected in every sample of meju. The metabolite profile patterns of the 11 samples were clustered into two main groups: group I (M1-5) and group II (M6-11). The metabolite analysis indicated a relatively higher amino acid content in group I, while group II exhibited higher isoflavone, soyasaponin, and lysophospholipid contents. The bioactivity analysis proved that the ABTS (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)) radical-scavenging activity was higher in group II and the FRAP (ferric reducing antioxidant power) activity was higher in group I. The correlation analysis revealed that the ABTS activity was isoflavonoid, lipid, and soyasaponin related, whereas the FRAP activity was amino acid and flavonoid related. These results suggest that the antioxidant activities of meju are critically influenced by the microbiome and metabolite dynamics.

GC/MS를 이용한 요 중 resveratrol과 그 대사체에 관한 연구 (Studies on resveratrol and its metabolite in human urine by GC/MS)

  • Resveratrol을 GC/TOF-MS를 이용하여 대사체를 확인한 결과, 두 개의 phenyl기를 연결하는 이중결합이 단일결합으로 환원된 구조로 추정되었다. 또한, GC/MSD를 이용하여 resveratrol 및 내인성스테로이드의 분석법에 대한 유효성을 점검한 결과, 회수율은 96.47 - 114.74%의 범위로 나타났으며, intra-day와 inter-day의 정밀도는 1.40 - 10.87%과 1.10 - 10.93% 그리고 정확도는 80.03 - 119.92%과 80.02 - 119.56%로 조사되었고, 모두 0.996이상의 직선성을 나타내어 유효한 분석 방법으로 검증되었다. 한편, 지원자들에게 resveratrol을 경구투여 한 요시료로부터 resveratrol과 그 대사체에 대한 상관성을 조사해본 결과, 요중 최대농도 도달시간이 일반적인 약물(1 - 2시간) 보다 긴 10 - 15시간에 나타났으며, 대사체로의 전환율은 남성보다 여성이 높게 나타났다. 한편, 내인성 스테로이드는 약물 복용 후 20시간 까지는 resveratrol 및 그 대사체와 다소 유사한 분비형태를 나타내었으며, estrone과 estradiol의 경우 여성이 남성에 비해 이 약물에 대한 민감성이 높게 나타났다. 그 외의 내인성 스테로이드는 유의할만한 차별된 분비형태의 변화가 나타나지 않았다. 따라서 resveratrol의 경우 약물의 활성이 남성보다는 여성에게 유의한 영향을 미치는 것으로 추측되었다.
