인돌을 이용한 잔류성 농약 endosulfan의 소장 흡수 억제 연구 (A study to reduce the intestinal transport of endosulfan by indole)

  • Endosulfan은 높은 잔류성과 소수성을 가진 유기염소계 잔류성 농약이다. 이 연구는 십자화과 식물에 다량 함유되어 있고 심혈관계질환의 예방에 효과가 있는 것으로 알려진 인돌을 이용하여 소장에서 endosufan의 흡수에 미치는 영향을 조사하였다. 흰쥐를 이용해서 37 kBq의 $^{14}C$-endosulfan이 함유된 지질유화액을 시간당 3 mL씩 8시간 동안 지속적으로 공급받은 동물군을 대조군으로 하였고, 인돌이 함유된 지질유화액을 같은 조건에서 공급받은 동물군을 실험군으로 하여 8시간 동안 흡수율을 측정하였다. 림프순환계로 흡수된 대조군의 $^{14}C$-endosulfan 양에 비해 인돌을 공급받은 실험군의 $^{14}C$-endosulfan의 양이 유의적으로 감소되었다. 이와 유사하게 콜레스테롤의 흡수도 인돌에 의해서 유의적으로 감소되는 경향을 보였다. 또한, 8시간 동안 흡수되지 않고 장벽에 잔류하는 $^{14}C$-endosulfan 양이 인돌을 공급받은 실험군에서 유의적으로 증가되는 것으로 나타났다. 이러한 림프순환계로의 endosulfan 흡수 감소현상은 인돌공급에 의해 소장의 점막에 잔류하는 endosulfan의 양이 증가되었기 때문인 것으로 판단된다. 이상의 실험결과들을 종합해 볼 때, 십자화과 식물에 존재하는 인돌 성분의 섭취가 대표적인 잔류성 농약인 endosulfan의 소장에서의 흡수를 감소시켜 체내 잔류 저감화에 효과적으로 영향을 미치는 것을 이 실험을 통해서 확인할 수 있었다.

  • Phellinus linteus (PL), one of the immune-regulatory substances, is recognized to play the role in the metabolic process on inflammation and immunity. It has been traditionally used in the oriental medicine to treat inflammatory related disease. The purpose of this study was to evaluate the effects of water extracts of PL on the mesenteric lymph node lymphocytes immune function in the ICR male mice. Control mice received vehicle only. The PL treated mice were administered the respective extract by oral gavages for 4 weeks. IgE concentrations in serum and MLN lymphocytes were significantly lower in PL treated mice than in control mice. PL increased the proportion of $CD4^+\;and\;CD8^+$ T cells in MLN lymphocytes. PL significantly decreased Th2 cytokine concentrations and mRNA expression levels in cytokine secretions. Therefore, water extracts of PL modulate inflammatory parameters through regulation of immunoglobulin production resulting from decreased Th2 cytokine secretion and mRNA expression levels and reduce pro-inflammatory cytokine secretion and mRNA expression in MLN lymphocytes.

돼지에서 살모넬라균(菌)을 분리(分離)하는 방법(方法)에 관한 연구(硏究) (Studies on the Methods for the Isolation of salmonella from Pigs)

  • 건강도쇄돈(健康屠殺豚)의 장간막임파선(腸間膜淋巴膳)과 분(糞)(대장내용물(大腸內容物))으로부타 살모넬라균(菌)의 분리방법(分離方法)을 비교연구하여 다음과 같은 결론을 얻었다. 1. 비교적 다른 세균에 의한 오염도(汚染度)가 낮은 장간막임파선(腸間膜淋巴膳)을 검사하였을 경우, 증균배지(增菌培地)로서는 tetrathionate broth(TB)가 selenite broth(SB) 보다 살모넬라균(菌) 분리율(分離率)이 월등히 높았으며, 선택배지(選擇培地)로서는 brilliant green agar (BG)가 SS agar(SS) 보다 역시 월등히 높았다. 2. 여러가지 장내세균(腸內細菌)에 의한 오염도(汚染度)가 높은 분(糞)을 검사하였을 경우에는, 살모넬라균(菌)의 분리율(分離率)에 있어서 증균배지(增菌培地)로 사용한 TB와 SB의 우열(優劣)을 인정할만한 차이를 보지 못하였으며, 선택배지(選擇培地)로 사용한 BG와 SS의 성적도 비슷하였다. 증균배지(增菌培地)나 선택배지(選擇培地)를 어느 한가지 배지(培地)만 사용한 경우보다는 두가지 배지(培地)를 동시에 같이 사용함으로써 살모넬라균(菌)의 분리율(分離率)이 크게 높았다. 3. Brilliant green McConkey broth 를 증균배지(增菌培地)로 사용하였을 경우에는 장간막임파선(腸間膜淋巴膳)과 분(糞)의 두 가지 재료중 어느것이나 TB 보다 살모넬라균(菌)의 분리율(分離率)이 훨씬 낮았다. 분(糞)을 brilliant green McConeky agar 에 직접 배양(培養)하는 방법은, TB나 SB에 증균배양(增菌培養)을 한 뒤에 BG나 SS의 증균배지(增菌培地)를 사용하는 방법보다 또한 훨씬 분리율(分離率)이 낮았다. 4. TB 및 SB의 증균배양시간(增菌培養時間)은 $37^{\circ}C$에서 24, 48 및 72 시간(時間)등 세가지를 비교해본 결과 장간막임파선(腸間膜淋巴膳)은 48 시간(時間) 배양(培養)했을 때에 살모넬라균(菌)의 분리율(分離率)이 가장 높았으나 분(糞)은 24 시간(時間)배양했을 때에 가장 분리율(分離率)이 높았다.

Cholera Toxin Disrupts Oral Tolerance via NF-κB-mediated Downregulation of Indoleamine 2,3-dioxygenase Expression

  • Cholera toxin (CT) is an ADP-ribosylating bacterial exotoxin that has been used as an adjuvant in animal studies of oral immunization. The mechanisms of mucosal immunogenicity and adjuvanticity of CT remain to be established. In this study, we investigated the role of indoleamine 2,3-dioxygenase (IDO), which participates in the induction of immune tolerance, in CT-mediated breakdown of oral tolerance. When IDO-deficient ($IDO^{-/-}$) mice and their littermates were given oral ovalbumin, significant changes in antibody responses, footpad swelling and $CD4^+$ T cell proliferation were not observed in $IDO^{-/-}$ mice. Feeding of CT decreased IDO expression in mesenteric lymph nodes (MLN) and Peyer's patch (PP). CT-induced downregulation of IDO expression was reversed by inhibitors of nuclear factor-kappa B (NF-${\kappa}B$), pyrrolidine dithiocarbamate and p50 small interfering RNA. IDO expression was downregulated by the NF-${\kappa}B$ inducers lipopolysaccharide and tumor necrosis factor-${\alpha}$. CT dampened IDO activity and mRNA expression in dendritic cells from MLN and PP. These data indicate that CT disrupts oral tolerance by activating NF-${\kappa}B$, which in turn downregulates IDO expression. This study betters the understanding of the molecular mechanism underlying CT-mediated abrogation of oral tolerance.

페렛에서 발생한 coronavirus 감염 증례 보고 (Ferret coronavirus infection in a domestic ferret (Mustela putorius furo))

  • A female domestic ferret (Mustela putorius furo) presented to a veterinary clinic with a clinical history of anorexia and poor body condition. Due to gradual deterioration of the body condition, explorative laparotomy was performed. Diffusely, the mesentery was severely thickened and adhered with prominent mesenteric lymph nodes. A portion of the mesentery and mesenteric lymph nodes were biopsied and fixed. Microscopic analysis revealed severe pyogranulomatous peritonitis and lymphadenitis, but staining revealed no bacterial organisms. However, immunohistochemistry for feline coronavirus exhibited strong immunoreactivity, primarily in the macrophages. Based on these results, the case was diagnosed as ferret coronavirus infection.

무플론 요네병 발생 사례 (Paratuberculosis in mouflon (ovis musimon) : A case report)

  • A 2-years-old female domesticated mouflon with a clinical history of chronic diarrhea and emaciation was submitted to NVRQS. Grossly, there were severe thickening of small intestine wall and enlargement of mesenteric lymph nodes. Microscopically, severe granulomatous inflammation was found in small and large intestine, mesenteric lymph nodes, spleen and liver. By Ziehl-Neelsen stain, innumerable acid-fast rod bacteria were found in the cytoplasm of epitheloid and Langhans type giant cells present in these organs. By PCR assay with primer pair specific for Mycobacterium avium subspecies paratuberculosis(IS 900) with small intestine sample, strong positive reaction was detected, although the organism was not isolated from this organ. Based on the results of histopathology and PCR, we concluded that the case was a typical paratuberculosis in mouflon. As far as we know, this is the first case report of paratuberculosis in mouflon Korea.

궁치화담전과 청신화담전의 면역조절효과 (Immunoregulatory Effects of Gungchihwadam-jeon and Cheongsinhwadam-jeon)

  • This study was performed to evaluate the immunoregulatory effects of Gungchihwadam-Jeon(궁치화담전, GCHDJ) and Cheongsinhwadam-Jeon(청신화담전, CSHDJ) was administered(p.o.) once a day for 14 days to mice. By the treatment of GCHDJ or CSHDJ was increased the cell viability of cultured mice splenocytes, thymocytes and mesenteric lymph node cells. Administration of GCHDJ or CSHDJ was increased the splenic T lymphocyte, especially the Tc cell subpopulation was increased by the GCHDJ, on the while the TH cell was increased by the CSHDJ. The administration of GCHDJ or CSHDJ was significantly increased the apoptosis of transplanted-L1210 leukemia cells to mice peritoneal cavity. These results suggest that GCHDJ and CSHDJ have an immunoregulatory action

Gastrointestinal Stromal Tumor with Extensive Lymphatic Metastasis: A Case Report

  • Gastrointestinal stromal tumor is a rare tumor which arises from the whole gastrointestinal tracts and most of it is detected in the stomach. It is uncommon with small intestine originated gastrointestinal stromal tumor and more uncommon with lymphatic metastasis. We experienced an unusual case of the small bowel gastrointestinal stromal tumor during experimental autopsy. Two primary tumors with central necrosis were detected in the ileum. The sizes of each tumor were $6.1{\times}3.4{\times}4.0$ cm and $3.7{\times}4.2{\times}3.2$ cm. There was extensive lymphatic metastasis on the greater omentum and mesenteric, iliac lymph nodes were also involved. With histologic findings, the eosinophilic spindle cells were densely distributed. Immunohistochemical findings were CD117 (-), CD34 (+), desmin (-), and S-100 protein (-). Therefore, we diagnosed the tumors as small bowel gastrointestinal stromal tumors with broad lymph node mestasis.

국내에서 분리된 돼지 유행성 설사증 바이러스 접종자돈의 병리학적 소견 (Pathological findings of experimentally infected piglets with a porcine epidemic diarrhea virus isolated in Korea)

  • Twelve colostrum-deprived piglets were inoculated oral1y with porcine epidemic diarrhea virus isolated from Korea. The piglets were euthanatized from at 12 hour postinoculation(PI) at 6-hour intervals. At 24 to 36 hours PI, all infected piglets showed severe yellowish to watery diarrhea, dehydration, depression, and anorexia. At necropsy, the intestinal wall looked thin, mesenteric vessels congested, and mesenteric lymph nodes edematous. The histological findings showed marked villous atrophy and fusion, severe degeneration of enterocytes and monomuclear cell infiltration in the lamina propria. On electron microscopy, villous epithelial cells of infected jejunum contained viral particles at 18 hour PI. The viral particles were pleomorphic spheres with a mean diameter of 95∼180nm including 18nm projections.
