Detection and Genetic Differentiation of Megalocytiviruses in Shellfish, via High-Resolution Melting (HRM) Analysis (HRM 분석법을 이용한 패류 내 Megalocytiviruses의 검출과 유전적 분석)

  • Viruses in the genus Megalocytivirus have been subdivided into four subgroups. Among these subgroups 2 and 4, represented by the red sea bream iridovirus (RBIV) and the olive flounder iridovirus (FLIV), respectively, are non-exotic. subgroups 1 and 3, represented by the red sea bream iridovirus (RSIV) and the infectious spleen and kidney necrosis virus (ISKNV), respectively, have not been detected in Korea and are known as exotic. Shellfish are filter-feeders, and can thus filter and accumulate Megalocytivirus in their digestive glands, allowing us to track viral contamination in surrounding aquatic environment. In this study, we developed a high-resolution melting (HRM) analysis to differentiate among subgroups of Megalocytivirus accumulated in shellfish, and confirmed the convenience and efficiency of this method. More than two subgroups of Megalocytivirus were found in the digestive gland of a single shellfish. We classified all Megalocytivirus viruses from shellfish in Korea into subgroups 2 and 4, although proportions of subgroups were different among regions. Compared to nucleotide sequencing analysis, HRM analysis is a simple and rapid method for differentiating of Megalocytivirus subgroups.

  • Nowadays, It is required human body-friendly, good mechanical properties, and economical efficiency material, simultaneously. The material to meet above requirement condition rear up high nitrogen stainless steel(HNS). However, HNS have a lot of problem such as poor workability, hot crack sensitivity. So, It is needed the condition of plastic working to overcome above many problem. In this study, VIM ingot with 100kg was made by pressurized vacuum induction melting. And then, The slab perform for hot rolling was prepared by open-die forging. Hot rolling process was performed by computer simulation according to change of height reduction, rolling temperature, heating numbers, rolling pass and so forth. The results of analysis were investigated between analysis and lab-scale rolling product.

  • For the fabrication of poly-crystalline silicon ingot, CCCC (Cold Crucible Continuous Casting) method under a high frequency alternating magnetic field, was utilized in order to prevent crucible consumption and ingot contamination and to increase production rate. In order to effectively and continuously melt and cast silicon, which has a high radiation heat loss due to the high melting temperature and a low induction heating efficiency due to a low electric conductivity, Joule and pinch effects were optimized. Throughout the present investigation, poly-crystalline Si ingot was successfully produced at the casting speed of above 1.5 mm/min under a non-contact condition.

  • A three-dimensional Gaussian heat source model is modified to include the effects of the gap and thickness-difference for the laser keyhole welding. The gap of the butt joint influences the welding efficiency such that the melting area decreases linearly with the gap. When the different plate thickness is used such as the tailored blank welding, melting areas of the thick and thin plates are predicted by introducing the thickness-difference factor. The calculated results using the modified heat source show reasonably good agreements with the experimental results.

  • Yttria stabilized zirconia coating is an attractive material for several engineering applications. In order to produce coatings with consistent and reliable performance it is important to understand the influence of spraying parameters on the coating properties and optimize the spraying parameters. In this paper the low pressure plasma spray(LPPS) deposition of as-received zirconia powder has been investigated using simple one-factor-at-a-time approach. The deposition efficiency was chosen to evaluate the melting characteristics of the as-received zirconia powder. The results obtained indicated that the deposition efficiency of zirconia powder is very sensitive to the spraying parameters such as plasma gas flow rate and ranges from 24% to 57% The microstructure and the phase composition of zirconia coating deposited with the different plasma spraying parameters were also examined by SEM and XRD respectively. The relationship between deposition efficiency and the microstructure of zirconia coating was discussed.

  • Selective laser melting (SLM) is an additive manufacturing process by melting metallic powders and stacking into layers, and can product complex shapes or near-net-shape (NNS) that are difficult to product by conventional processes. Also, SLM process is able to raise the efficiency of production by creating a streamlined manufacturing process. For manufacturing in SLM process using Ti-6Al-4V powder, analysis of microstructural evolution and evaluation of mechanical properties are essential because of rapid melting and solidification process of powders according to high laser power and rapid scan speed. In addition, it requires a post-processing because the soundness and mechanical properties are degraded by defects such as pore, un-melted powder, lack-of-fusion, etc. In this study, hot isostatic press (HIP) was conducted as a post-processing on SLM-printed Ti-6Al-4V alloy. Microstructure of post-processed Ti-6Al-4V alloy was compared to as-built Ti-6Al-4V, and the evolution of quasi-static (Vickers hardness, room temperature tensile characteristic) and dynamic (high-cycle fatigue characteristic) mechanical properties were analyzed.

  • In this study, basic design data which were required for development of highly efficient ice storage system with low temperature latent heat were experimentally obtained. The ice storage system considered in this study was the one that has been widely used in the developed country and called the ice-on-coil type. Using the system, the ice storage performance for various design parameters which were the flow direction and the inlet temperature of the secondary fluid was tested. In addition, the clockwise variation of the heat transfer characteristics of the PCM in the ice storage tank were investigated. During the melting processes in the ice storage tank with several vertical tubes, decrease of the solid-liquid interface area, which was the heat transfer area, between the floating ice and the water made the decreasing rate of IPF less. Also, the total melting energy for the upward flow of the secondary fluid was higher than that for the downward flow during the melting process, but this trend did not appear if the initial temperature of the PCM was $4^{\circ}C$. The average ice recovery efficiency for the upward flow of the secondary fluid was higher than that for the downward flow.

  • The aim of this study is to make the technical standards for certificating a high efficiency metal-halide lamps and reflectors using the high intensity discharge lamps, to accomplish the practical energy saving effect. Metal-halide lamps are certificated by stabilzed and restarting time, lamp voltage, current, power and light output efficiency. The high illumination reflectors using HID lamps are certificated by testing reflective index, fading, the melting point, glare and install spacing factor.

  • Copper die casting motor (CDM) has a better design flexibility than aluminium die casting motor (ADM) because of the higher conductivity of copper. However, CDM has an economic and manufacturing weak point due to high copper price and high melting temperature compared to ADM. This paper describes a comparison of the design parameters, specifications, performances and material cost between CDM and ADM of 5.5kW_6P.
