In total, 170 carrot lines developed in Korea were screened for resistance to Meloidogyne incognita race 1 to select parental genetic resources useful for the development of nematode-resistant carrot cultivars. Using the gall index (GI), gall formation was examined on carrot roots inoculated with approximately 1,000 second-stage juveniles of the nematode 7 weeks after inoculation. Sixty-one carrot lines were resistant (GI ${\leq}1.0$), while the other 109 were susceptible (GI > 1.0) with coefficient of variance (CV) of GI for total carrot lines 0.68, indicating low-variation of GI within the lines examined. The histopathological responses of two carrot plants from resistant and susceptible lines were examined after nematode infection. In susceptible carrots, giant cells formed with no discernible necrosis around the infecting nematodes. In the resistant carrot line, however, no giant cells formed, although modified cells were observed with extensive formation of necrotic layers through their middle lamella and around the infecting nematodes. This suggested that these structural modifications were related to hypersensitive responses governed by the expression of true resistance genes. Therefore, the Korean carrot lines resistant to the nematode infection are potential genetic resources for the development of quality carrot cultivars resistant to M. incognita race 1.

In the course of screening for microbes with nematicidal activity, we found that Enterobacter asburiae HK169 displayed promising nematicidal activity against the root-knot nematode Meloidogyne incognita, along with plant growth-promoting properties. Soil drenching of a culture of HK169 reduced gall formation by 66% while also increasing root and shoot weights by 251% and 160%, respectively, compared with an untreated control. The cell-free culture filtrate of the HK169 culture killed all juveniles of M. incognita within 48 h. In addition, the nematicidal activity of the culture filtrate was dramatically reduced by a protease inhibitor, suggesting that proteolytic enzymes contribute to the nematicidal activity of HK169. In order to obtain genomic information about the HK169 isolate related to its nematicidal and plant growth-promoting activities, we sequenced and analyzed the whole genome of the HK169 isolate, and the resulting information provided evidence that the HK169 isolate has nematicidal and plant growth-promoting activities. Taken together, these observations enable the future application of E. asburiae HK169 as a biocontrol agent for nematode control and promote our understanding of the beneficial interactions between E. asburiae HK169 and plants.
Pasteuria penetrans KW1 (PP), parasitic bacterium of nematode, was isolated from oriental melon greenhouse soil in Korea and evaluated for the suppression effect on the reproduction of southern root-knot nematode, Meloidogyne incognita (MI), in lettuce (Lactuca sativa L. var. Chungchima) and oriental melon (Cucumis melo L. var. Eunchun). Pot experiments were conducted by planting the lettuce seedlings in medium inoculated with 5,000 MI juveniles/pot (J), J +100,000 PP endospores/l g medium, and J +200,000 PP endospores/1 g medium. After 11 weeks of plantation, number of root galls in J +200,000 PP endospores/1 g medium was decreased to 92/root (38.9%, control effect), compared to the J of 150/root. In the second plantation of lettuce in the same pots, the numbers of root gall were significantly decreased in PP treated pots with 75 (77.2%, control effect) and 150/root (54.4%, control effect) in J +200,000 and J +100,000 PP endospores/1 g medium, respectively, compared to the J of 330/root when harvested at 10 weeks after planting. In oriental melon, root gall percentages were 32.1 (60.2%, control effect) and 52.9% (34.5%, control effect) in J +200,000 and J + 1(10,000 endospores/l g medium which were significantly lower than that of 80.7% in J.

Control of nematode has become difficult owing to the restricted use of effective soil fumigant, methyl bromide, and other non-fumigant nematicides. Therefore, it is urgently necessary to develop microbial nematicide to replace chemical nematicides. In this study, the 50% aqueous methanol extraction solution of fermentation broths of 2,700 actinomycete strains were tested for their nematicidal activity against second stage of juveniles (J2s) of Meloidogyne incognita. As the results, only the 50% aqueous methanol extraction solution of AN110065, at 20% equivalent to 10% fermentation broth, showed strong nematicidal activity with 78.9% of mortality 24 h after treatment and 94.1% of mortality at 72 h. The 16S rRNA gene sequencing showed that the strain sequence was 99.78% identical to Streptomyces netropsis. The extract of S. netropsis AN110065 fermentation broth was successively partitioned with ethyl acetate and butanol and then the ethyl acetate, butanol and water layers were investigated for their nematicidal activity against the M. incognita. At $1000{\mu}g/ml$, ethyl acetate layer showed the strongest activity of 83.5% of juvenile mortality 72 h after treatment. The pot experiment using the fermentation broth of AN110065 on tomato plant against M. incognita displayed that it evidently suppressed gall formation at a 10-fold diluent treatment. The tomato plants treated with the fermentation broth of S. netropsis AN110065 did not show any phytotoxicity. The results suggest that S. netropsis AN110065 has a potential to serve as microbial nematicide in organic agriculture.
Survey on soybean parasitic nematodes has been undertaken in order to get an ideas about the distribution of nemic fauna. Total 71 soil samples were collected from soybean fields in Gyeongbug and Chungbug provinces. Eighteen species belonging to fourteen different genera were found associated with soybean, Glycine max. The results were as follows: Aphelenchus avenae, Circonemoides informis, Ditylenchus dipsaci, Helicotylenchus pseudorobustus, Heterodera glycines, Hirschmanniella imamuri, Meloidogyne hapla, Meloidogyne incognita, Paratylenchus pandus, Pratylenchus thornei, Pratylenchus vulnus, Psilenchus hilarulus, Rotylenchus sp. Tylenchorhynchus clay toni, Tylenchorhynchus dubius, Tylenchorhynchus nudus, Tylenchus sp. Xiphinema americanum were found in soybean fields. In frequency of occurring nematodes by localities, soybean cyst nematode was higher in Chungbug by $70\%$, while $50\%$ in Gyeongbug. But rook-knot nematode was higher in Gyeongbug by $54.1\%$, while $19.1\%$ in Chungbug. In density of nematode, Heterodera was the most dominant as 200.4 populations per 100ml soil, Meloidogyne as 107.1 populations, Rotylenchus, Xiphinema, Aphelenchus in decreasing order. In survey of loss, density of soybean cyst nematode in heavily infested fields was 155 cysts per 100ml soil while 35 cysts in slightly infested fields. Seventy percent of soybean yields was decreased when comparing with average 64.2kg/10a in heavily infested fields and average 209.5kg/10a in slightly infested. Correlation coefficients among number of soybean cyst and yields, pod weight, number of pod was negative as r- 0.57, 0.47, 0.38 over all localities.

A preliminary survey on the incidence and intensity of root disease complex (association of Meloidogyne incognita and root rot pathogens) was carried out in the sericultural areas of Karnataka. A total of 280 mulberry gardens were surveyed in 14 districts of Karnataka belonging to different types of soil (red sandy, red loamy and black cotton), farming systems (irrigated and rainfed), varieties (V-1, K-2, Local and S-13) and age of the plants (0-5, 5-10 and 10-15 years). It was observed that the association of M. incognita with Botryodiplodia theobromae and Fusarium solani causes the root disease complex in mulberry. Of the 280 gardens visited, 94 were infested with the disease complex and incidence was recorded as 33.6%. The higher intensity of root disease complex was observed when the root system had more than 100 galls/plant with infection of mixed population of B. theobromae and F. solani in sandy soil under irrigated farming. The 5-10 years old mulberry plantation with V-I variety was found to be most susceptible to root disease complex. Districts like Mysore, Kolar, Mandya, Tumkur, Chitradurga and Bangalore were observed as sensitive areas. Further, the wounds caused by M. incognita in mulberry roots favour the easy entry of root rot pathogens, which increased the severity of the disease very fast.

Soil and root samples were collected form the rhizoshpere of 11 different medicinal plants to determine the incidence, density and identification of root-knot nematode species associated with medicinal herbs. About 55% of medicinal herbs examined was found to be infested with root-knot nematodes. As a result of infection casued by three root-knot nematodes, M. hapla recorded 43.3% in medicinal herba whereas M. incognita and M. arenaria showed 7.9% and 3.7%, repectively. Forsythia koreana, Hemerocalis fulva, Hibuscus mutabilis and Petasites japonicus were the most severely infested herbs whereas Acanthopanax sessilflorus was least infested. Population of the second stage younger plants. Meloidogyne hapla, M. incognita and M. arenaria were the species associated with the medicinal herbs. The most abundant nematode observed in medicinal herbs was M. hapla and followed by M. incognita and M. arenaria. M. arenaria was observed firstly on Ficus carica, one of medicinal plant.
Meloidogyne arenaria race 2 (59%) is widely distributed, followed by M. incognita race 1 (23%), and an unknown race of M. incognita (18 %) in greenhouses in southern Korea. The key character to distinguish between M. arenaria and M. incognita is excretory pore in female head. When oriental melon, Cucumis melo L., grafted on Shintozoa (Cucurbit maxima x Cu. moschata) is transplanted in February in a plastic tunnel inside a greenhouse infested with M. arenaria, nematodes produced egg masses on roots at 40 days after transplanting and the soil juveniles (J2) population reach maximum in July to 3,817/100 ㎤. Juveniles are distributed relatively uniform over the 180-cm-wide row horizontally and the highest density occurs at 0-25 cm soil depth. For the control of root-knot nematodes, rice rotation, solarization, and soil addition treatments are the most effective (P=0.05); treatments reduce number of J2 over 90% and increase yield two times. Corn retation, fosthiazate, and soil drying treatment are moderately effective, while sesame and green onion rotations are not effective. The relationship between M. arenaria and yield of oriental melon is adequately described by a linear regression model. In the test with wild Cucumis genetic sources introduced from U.S.Dept. of Agriculture (USDA), one of C.heptadactylus, two of C.anguria, two of C. anguria var. longaculeatus, nine of C. metuliferus are resistant to both species of root-knot nematodes.
Plant disease caused by root-knot nematode is a major problem in crop production. Using of chemical pesticides, one of the most efficient methods to control nematodes, have raised issues in toxicity to humans and animals and environmental pollution. In this study, to select actinomycete strains that have potential to serve as a microbial agent for control of nematodes, we investigated nematicidal activity of culture broth from 670 Streptomyces isolates. A culture filtrate of KRA15-528 isolate that was identified as S. flavogriseus on the basis of 16S rRNA sequence analysis, showed strong nematicidal activity against second stage of juveniles of Meloidogyne incognita and inhibited egg hatching; exposure to 10% of culture filtrate resulted in 71% juvenile mortality at 48 hours afters treatment and suppressed egg hatching by 54% at 9 days after treatment. When the KRA15-528 culture filtrate was partitioned with ethyl acetate and butanol, ethyl acetate layer exclusively showed strong activity; 91%, 53%, 30% of mortality at 1,000, 500, $250{\mu}g/ml$, respectively. Additionally, the culture filtrate suppressed gall formation on cucumber plant by M. incognita with no phytotoxicity. These results suggest that S. flavogriseus KRA15-528 has potential to serve as a microbial nematicide for the control of root-knot nematode disease.
This study examined the control effects of amino acid biochemical agent extracted from chicken feathers on Meloidogyne incognita for the purpose of developing an environmentally friendly nematicidal agent that can replace chemical control of root-knot nematodes (RKN). We investigated the lethal effects of J2 juveniles for 19 types of commercial amino acids. As a result, five kinds of amino acids (L-asparagine, L-aspartic acid, L-methionine, L-tyrosine, L-cysteine) showed mortality rate of more than 50% at a concentration of 50 mM. L-asparagine showed the highest mortality rate at 94%. We also investigated the lethal effect of J2 juveniles and suppressive effects of egg hatching by feather amino acids (FAA) biochemical agent. It showed that the mortality rate of J2 juveniles was more than 80% and suppression rate of egg hatching was 74% at 1/50 concentrations of FAA. As a result of conducting a tomato pot culture experiment for 60 days after treating 1/50 concentrations of FAA biochemical agent in rhizosphere soil, it showed that the control effects were 63% of juveniles density in the soil, 59% of egg mass and 61% of root gall index, respectively. Based on the above results, it is considered that the FAA biochemical agent extracted from chicken feathers can be used as an environmentally friendly nematicidal agent of RKN.
