Objectives : This study was carried out for the development of medicine for vitiligo treatment and focused on the effect of Psoraleae fructus extract on melanin synthesis of B16 melanoma cells. Methods : Activity of tyrosinase playing a vital role in the synthesis and quantity of melanin, which is the final product in cultured B16 melanoma cells, the effects of Psoraleae fructus extract were measured. Results : The results indicated that Psoraleae fructus extract increased beth the amount of melanin and the activity of tyrosinase according to concentration, also supported by western blot analysis. Conclusions : The results suggest that Psoraleae fructus extract has an advantageous effect on the promotion of melanin synthesis and will contribute to the development of vitiligo treatment through further related studies.
Objective : This research was carried out to compare the effect of Gorynju and a Soju extract Psoraleae fructus on melanin synthesis of B16 melanoma cells. Methods : To investigate melanin synthesis of B16 melanoma cells, this research was measured cell survival, tyrosinase activity, melanin synthesis, western blot. Results : Both Gorynju and Soju extract Psoraleae fructus, cell toxicity depended on the density. Tyrosinase activity depended on the density of Gorynju extract Psoraleae fructus and statistic was showed significant(0.5, 1, 2, 3 ${\mu}g/ml$), in a Soju extract Psoraleae fructus, 1 ${\mu}g/ml$ were showed significant. Melanin synthesis was showed significant in a Soju extract Psoraleae fructus(3, 4 ${\mu}g/ml$). Western blot was showed to depend on the density of Gorynju and a Soju extract Psoraleae fructus. Conclusions : In a tyrosinase activity and a melanin synthesis, the intermediate alcohol of Gorynju and a Soju may be suitable to use.
To investigate the relationship between structure and biological activity of phenylpropanoids, we measured effects of phenylpropanoids on anti-oxidant and whitening activity, In DPPH radical scavenging activity, caffeic acid analogues showed the significant anti-oxidant activity. Although phenylpropanoids did not inhibit purified-tyrosinase activity, they significantly inhibited tyrosinase activity and melanin production in MSH-stimulated B16 melanoma cells. However, phenylpropanoids did not affect tyrosinase expression in MSH-stimulated B16 melanoma cells, which suggest that inhibition of MSH-induced melanin production was due to tyrosinase inhibition mediated via other signal pathways but not expression of tyrosinase. Phenylpropanoids also significantly inhibited both hyaluronidase and elastase activity, suggesting that phenylpropanoids may be used as whitening, hydration and anti-wrinkling agents. Hydroxyl residue of aromatic ring in phenylpropanoids plays an important role in anti-oxidant and whitening activity.
Salicylamide, a non-steroidal anti-inflammatory drug (NSAID), is used as an analgesic and antipyretic agent. We have previously shown that several NSAIDs have anti-melanogenic properties in B16F1 melanoma cells. In contrast, we have found that salicylamide enhances melanin contents in B16F1 melanoma cells; however, the underlying mechanism is not known. Therefore, we investigated the mechanism through which salicylamide stimulates melanogenesis. Interestingly, salicylamide enhanced diphenolase activity in a cell-free assay. Western blotting and real-time RT-PCR revealed that salicylamide increased tyrosinase expression via transcriptional activation of the Mitf gene. Together, our results indicate that salicylamide could be used as an anti-hypopigmentation agent for skin and/or hair.
To examine the potential of Terminalia chebula as a whitening agent, we measured antioxidant activity using DPPH$\cdot$, ABTS${\cdot}^+$ assays and ferric-reducing antioxidant power (FRAP) assays, and depigmenting activity using B16F10 melanoma cells. The intracellular reactive oxygen species (ROS) level was monitored by $H_2DCFDA$ fluorescence labeling, and melanin contents in B16F10 melanoma cells by 960 $J/m^2$ dose of UVA-induced oxidative stress. The radical-scavenging activities of T. chebula extract (TCE) were measured in terms of $EC_{50}$ values using DPPH$\cdot$, ABTS${\cdot}^+$ assays and FRAP value were 280.0 ${\mu}g/mL$, 42.2 ${\mu}g/mL$ and 113.1 ${\mu}mol$$FeSO_4{\cdot}7H_2O/g$, respectively. We found that ROS and melanin concentrations were reduced by TCE treatments of 25 ${\mu}g/mL$ under UVA-induced oxidative stress. Tyrosinase activity and melanin contents in $\alpha$-melanocyte stimulating hormone (MSH)-induced melanoma cells both decreased dose-dependently in the treatment groups. TCE similarly reduced melanogenesis in B16F10 melanoma cells stimulated by $\alpha$-MSH as compared to arbutin as a positive control. T. chebula may prove to be a useful therapeutic agent for hyperpigmentation and an effective component in skin whitening and.or lightening cosmetics.
Purpose: This study was performed to determine the inhibitory effect of Sihosogansangagambang (SS) on melanin synthesis in B16F10 melanoma cells (B16F10). Methods: The inhibitory effects of Sihosogansangagambang on melanin synthesis were used by in vitro assay. To elucidate inhibitory effects of SS on melanin synthesis, we determined the melanin release in B16F10. And to investigate the mechanism of inhibitory effect of SS, we assessed the gene expression of tyrosinase, TRP-1, TRP-2 and ERK-1 in B16F10. Results: 1. SS decreased the release of melanin in B16F10 melanoma cells. 2. SS inhibited mushroom tyrosinase activity in vitro. 3. SS decreased the expression of tyrosinase, TRP-2 in B16F10 melanoma cells, but did not decreased the expression of TRP-1 in B16F10 melanoma cells. 4. SS decreased the expression of ERK-1 in B16F10 melanoma cells. Conclusion: From these results, it may be suggested that SS is possesed of the antimelanogenetic effects.
A five-year-old female Yorkshire Terrier dog presented with a perianal mass. Fine needle aspiration revealed that the mass comprised two different types of cells: hepatoid epithelial cells and melanin-containing melanocytes. Histopathological examination confirmed perianal gland adenoma with malignant melanoma. Evidence of metastasis was found on thoracic radiography with soft-tissue densities observed within the pulmonary parenchyma. The dog survived for three months after diagnosis of malignant melanoma. This report describes the clinical findings, diagnostics used, cytological and histopathological findings, and the potential prognosis for a dog diagnosed with malignant anal sac melanoma.
To assess inhibition mechanisms of a Phellinus igniarius (PI) extract on cancer, C57BL/6 mice were orally treated with PI extractive after or before implanting H22 (hepatocellular carcinoma ) or B16 (melanoma) cells. Mice were orally gavaged with different doses of PI for 36 days 24h after introduction of H22 or B16 cells. Mice in another group were orally treated as above daily for 42 days and implanted with H22 cells on day 7. Then the T lymphocyte, antibody, cytokine, LAK, NK cell activity in spleen, tumor cell apoptosis status and tumor inhibition in related organs, as well as the expression of iNOS and PCNA in tumor tissue were examined. The PI extract could improve animal immunity as well as inhibit cancer cell growth and metastasis with a dose-response relationship. Notably, PI's regulation with the two kinds of tumor appeared to occur in different ways, since the antibody profile and tumor metastasis demonstrated variation between animals implanted with hepatocellular carcinoma and melanoma cells.
The purpose of this study was to analyze inhibitory effects of anti-4-1BB monoclonal antibody on melanoma metastasis The 4-1BB (CD137) T cell molecule is a member of the TNF receptor family and its activation by either 4-1BB ligand or antibody induces T cell activation and growth. In the present study, administration of anti-4-1BB mAb induced inhibition of melanoma metastasis. Agonistic anti-4-1BB mAb induced not only CD$8^+$4-1BBT cells but also CD$8^+$IFN-${\gamma}$$^{+}$ T cell population. In the presence of anti-CD3 antibody, lymphocytes produced high levels of IFN-${\gamma}$ and low levels of IL-4 in anti-4-1BB mAb treated group. Exposure of melanoma cells to IFN-${\gamma}$ induced expression of MHC-I molecules. Thus, the increase in number of CD$8^+$T cells and enhanced MHC-I expression on B16F10 cells by augmented IFN-${\gamma}$ production in response to anti-4-1BB mAb may result in suppression of tumor growth and metastasis.s.
The effect of methamphetamine on the pulmonary metastasis was investigated in C57BL/6 mice injected with Bl6 melanoma cells. Bl6 melanoma cells (2$\times$10$^{5}$ cells) were injected intravenously into 5~7 weeks old C57BL/6 mice. Mice were then treated intraperitoneally with methamphetamine either acutely (two times with one week interval) or subchronically (daily for 14 days). Degree of pulmonary metastasis was investigated and specific immunologic parameters such as natural killer cell cytotoxicity(NKCC), antibody-dependent cellular cytotoxicity(ADCC) and blastogenic responses of splenocytes were examined. Mice which had been subchronically treated with methamphetamine showed significant decreases in the number of pulmonary metastasis of Bl6 melanoma cells, NKCC and ADCC without a significant change in blastogenic responses. In the acutely-treated group, slight trends of decrease in the numbers of pulmonary metastasis, NKCC and ADCC were observed without statistical significances whereas there was a significant increase in blastogenic responses. The mechanism underlying the decrease in the degree of metastasis despite diminished NKCC and ADCC after methamphetamine treatment and the relationship between the degree of pulmonary metastasis and duration of methamphetamine treatment remain to be investigated.
