알러젠 제거 옻나무 추출물 투여로 호전된 악성흑색종(惡性黑色腫) 환자 1례 (A Case of Malignant Melanoma Patient Treated with Allergen-removed Rhus Verniciflua Stokes)

  • Malignant melanoma is rare in Asians and one of the poorer prognostic diseases. Malignant melanoma accounts for only a small ratio of all skin cancers, but it causes the greatest number of skin cancer - related deaths worldwide. Melanoma incidence has continued to increase worldwide and in Korea, like other countries, the number of patients has increased compared with previous cancer incidence data. In this case report, we describe a case of a malignant melanoma patient who showed neck node metastasis (right supraclavicular area) after wide excision of facial melanoma 2 years before the diagnosis of relapse. The patient refused to get conventional western medical treatment including chemotherapy and the size of cancer increased over the 5 months of clinical follow-up. After 4 months of traditional Korean medical treatment using allergen-removed Rhus verniciflua Stokes(A-RVS), however, the size of cancer mass decreased and the patient showed no side effects. Further study will be needed in order to determine the long-term effect of allergen-removed Rhus verniciflua Stokes on patients with malignant melanoma.

Antitumor Effect of Metformin in Combination with Binimetinib on Melanoma Cells

  • Cutaneous melanoma is a fatal disease for patients with distant metastasis. Metformin is the most widely used anti-diabetic drug, and proved to suppress cell proliferation and metastasis in diverse cancers including melanoma. We previously reported that MEK inhibitor trametinib increases the expression of epithelial-mesenchymal transition (EMT) regulators and melanoma cell motility, which are suppressed by addition of metformin in A375 melanoma cells. To confirm our findings further, we first evaluated the effect of metformin in combination with another MEK inhibitor binimetinib on cell viability in G361 melanoma cells. We then investigated whether binimetinib affects the expression of EMT regulators and cell motility. We finally monitored the effect of metformin on binimetinib-induced cell migration. Cell viability assay showed that combination index (CI) value at ED50 is 0.80, suggesting synergy for the combination of metformin with binimetinib. Our results also revealed that binimetinib increased the expression of EMT regulators such as integrin αV, fibronectin and slug, which correlate well with the enhanced cell migration in wound healing assay. Metformin, on the contrary, suppressed the expression of sparc, integrin αV, fibronectin and N-cadherin with the reduced cell motility. The combination treatment showed that metformin counteracts the binimetinib-induced increase of cell motility. Overall, these results suggest that metformin with binimetinib might be useful as a potential therapeutic adjuvant against cell survival and metastatic activity in melanoma patients.

Primary Spinal Cord Melanoma

  • Primary central nervous system (CNS) melanoma is a rare condition that accounts for only 1% of all melanomas. A 34-year-old Korean female presented with a two-month history of progressive weakness in both legs. Spinal magnetic resonance image (MRI) revealed a spinal cord tumor at the level of T4, which was hyperintense on T1-weighted imaging and hypointense on T2-weighted imaging. The intradural and extramedullary tumor was completely resected and diagnosed as melanoma. There were no metastatic lesions. At three years after surgery, the patient is still alive, with no evidence of tumor recurrence. We present the details of this case along with a comprehensive review of spinal cord melanoma.

Quinazolinylmethoxybenzene 유도체 합성 및 흑색종 세포증식 저해효능 (Synthesis and Antiproliferative Activity of Quinazolinylmethoxybenzene Derivatives against Melanoma Cell Line)

  • Melanoma is the most aggressive form of skin cancer and is the fastest growing type of cancer in the United States. We report here the synthesis of a novel series of quinazolinylmethoxybenzene derivatives 1a-c and their antiproliferative activities against A375 human melanoma cell line. Among them, urea compound 1a ($IC_{50}\;=\;4.8\;{\mu}M$) having 4-chloro-3-trifluoromethylphenyl moiety showed superior antiproliferative activity to Sorafenib ($IC_{50}\;=\;5.5\;{\mu}M$) as a reference compound. These results will helpful for designing structure of a therapeutic agent for the treatment of melanoma.

흑색종 세포증식 저해효능의 새로운 Benzaminoquinoline 유도체의 합성 (Synthesis of New Benzaminoquinoline Derivatives with Antiproliferative Activity against Melanoma Cell Line)

  • Melanoma is the most serious type of skin cancer as a malignant tumor of melanocytes. In this work, the syntheses of a novel series of benzaminoquinoline derivatives 1a-c and their antiproliferative activities against A375 human melanoma cell line were described. All the compounds ($IC_{50}=0.78-1.02{\mu}M$) showed superior antiproliferative activities to Sorafenib ($IC_{50}=5.58{\mu}M$) as a reference compound. These results suggested that benzaminoquinoline derivatives have potentials as a therapeutic agent for the treatment for melanoma.

  • Micro plasma has been recently studied to investigate the effects on various cells. We study a micro-plasma produced by a plasma needle that is operated with RF power and its effects on G361 melanoma cells. The micro plasma size ranges from sub-mm to several mm at a few watts of RF power. For the bio-medical treatment, low-temperature plasma is obtained and gas temperature is controlled within several tens of degrees $(^{\circ}C)$ in order not to disturb cell activities. Elementary spectroscopic studies to obtain plasma characteristics are presented for Ar and He plasma with different frequencies of RF power. Also the preliminary results of the micro plasma effects on G361 melanoma cells are presented. It was observed that the irradiation of micro plasma induces cell death through the deprivation of tyrosine phosphorylation in the G361 cells.

Malignant Anal Sac Melanoma in a Dog

  • A five-year-old female Yorkshire Terrier dog presented with a perianal mass. Fine needle aspiration revealed that the mass comprised two different types of cells: hepatoid epithelial cells and melanin-containing melanocytes. Histopathological examination confirmed perianal gland adenoma with malignant melanoma. Evidence of metastasis was found on thoracic radiography with soft-tissue densities observed within the pulmonary parenchyma. The dog survived for three months after diagnosis of malignant melanoma. This report describes the clinical findings, diagnostics used, cytological and histopathological findings, and the potential prognosis for a dog diagnosed with malignant anal sac melanoma.

  • An 11-year-old castrated male miniature Schnauzer, weighing 8 kg, presented with multiple masses on the humeri. Grossly, the masses were separated from the surrounding soft tissues. Desmoplastic melanoma is an uncommon tumor in dogs. Fine-needle aspiration cytology showed large spindle-shaped cells with a high nuclear-cytoplasmic ratio. Histopathology showed interlacing bundles of fibroblast-like cells, highly pleomorphic multinuclear cells, occasional atypical spindle cells separated by fibrocollagenous stroma, and lymphocyte aggregates admixed with abundant collagen. The tumors were diffusely positive for S100a, a strong marker for desmoplastic melanoma, and negative for desmin. Herein, we report the pathobiology of a rare, uncommon variant of melanoma in a dog.

  • Pedunculagin is an ellagitannin purified from Alnus hirsuta var. microphylla. Betulaceae. The effects of pedunculagin on immune system have been characterized to induce enhancement of NK (natural killer) cell cytotoxicities against tumor cells. Here, we report the evaluation of the effects of pedunculagin on the growth of murine Bl6-F10 melanoma in vivo. After the intradermal inoculation of Bl6-F10 melanoma, Bl6-F10 tumors grew progressively in immunocompetent syngenic C57BL/6 mice. The mice treated with pedunculagin(10 mg/kg, every 48 hrs) resulted in a significant improvement in survival. Inhibitory effects of pedunculagin on lung metastasis in C57BL/6 mice were also detected. Summarizing treatment with pedunculagin has a significant antitumor effect upon Bl6-F10 murine melanoma.

B16 Melanoma 세포에서 Citrus Essential Oil이 Melanin 생성에 미치는 영향 (Effects of Citrus Essential Oils on Melanin Production in B16 Melanoma Cells)

  • This study is performed to investigate the effects of citrus essential oils on melanin production in B16 melanoma cells. Five kinds of citrus essential oils (Bergamot, Grapefruit, Lemon, Mandarin, Petigrain) did not have any influence on DPPH radical scavenger activity, cell growth and cytotoxicity in B16 melanoma cells. Both mandarin and petigrain essential oils dose-dependently inhibited purified tyrosinase activity, but bergamot did not. In 1$\mu$M MSH-stimulated B16 melanoma cells, all of 5 citrus essential oils inhibited melanin production in a dose dependent manner. From the above results, it is possible that citrus essential oils may be developed to be an anti-melanogenesis agent on the basis of their inhibitory effect on MSH-induced melanin production.
