Effect of Chitosan Oligosaccharide on Melanin Production in B16 Melanoma Cells (B16 Melanoma 세포에서 Chitosan Oligosaccharide가 Melanin 생성에 미치는 영향)

  • To investigate the effect of chitosan oligosaccharide on melanin synthesis, we measured tyrosinase activity and melanin production in B16 melanoma cells. Chitosan oligosacchaide itself did not have any anti-oxidant activity in DPPH radical scavenging, and did not affect the proliferation of B16 melanoma cells. Chitosan oligosaccharide dose-dependently increased melanin production in the absence or presence of MSH. However, chitosan oligosaccharide did not have any influence on the tyrosinase activity and tyrosinase expression in B16 melanoma cells. These results suggest that chitosan oligosaccharide-induced melanin production may be independent on tyrosinase activity in B16 melanoma cells. From the above results. chitosan oligosaccharide dose-dependently appears to increase melanin production in B16 melanoma cells, suggesting that chitosan oligosaccharide may be used as a tanning agent.

Effects of Protein Kinase Inhibitors on Melanin Production in B16 Melanoma Cells Stimulated via Cyclic AMP-dependent Pathway (B16 Melanoma 세포에서 Protein Kinase 억제제들이 Cyclic AMP 경로를 통한 멜라닌 생성에 미치는 영향)

  • To investigate the effect of protein kinase on melanin production via cAMP-dependent pathway, we measured the melanin amount and tyrosinase activity in B16 melanoma cells stimulated by alpha-melanocyte stimulating hormone (MSH), forskolin and 8-Br-cAMP. MSH, forskolin and 8-Br-cAMP significantly increased both melanin production and tyrosinase activity in B16 cells. Melanin production and tyrosinase activity by MSH are significantly inhibited by cyclic AMP-dependent protein kinase inhibitor (KT5720) and protein kinase C down-regulation treated with PMA. Bisindolmaleimide (1$\mu$M), protein kinase C inhibitor, significantly inhibited melanin production and tyrosinase activity stimulated by MSH, forskolin and 8-Br-cAMP with the following order of potency: MSH>forskolin>8-Br-cAMP. Tyrosine kinase inhibitor, genistein and DHC, significantly inhibited both, but the inhibitory effect was more potent in 8-Br-cAMP-stimulated B16 cells than MSH-stimulated cells. NFkB inhibitor (parthenolide) significantly inhibited melanin production and tyrosinase activity. Neither melanin production nor tyrosinase activity induced by MSH, forskolin and 8-Br-cAMP were affected by KN-62 (calmodulin-dependent protein kinase II inhibitor), PD098059 (mitogen-activated protein kinase inhibitor, MAPKK) and worthmannin (phosphatidylinositol 3-kinase inhibitor). These results suggest that both protein kinase C and tyrosine kinase are involved in melanin production by cyclic AMP-dependent pathway and NFkB pathway may play an important role in cyclic AMP-dependent melanin production in B16 melanoma cells.

  • Hair graying is processed by loss of melanin production caused by the decrease of activity and number of melanocyte and the accumulation of hydrogen peroxide ($H_2O_2$) in the hair follicle with increase of age. The purpose of this study was to investigate the effect the Black oryzasativa ethanolic extract (BLEE) on the melanin production. In this study BLEE at $8{\mu}g/ml$ or more showed a significant 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging activity and reduction power. BLEE at $16{\mu}g/ml$ or more showed promoted tyrosinase activity and melanin production. In addition BLEE scavenged intracellular $H_2O_2$ in 2',7'-dichlorodihydrofluorescein (DCF) fluorescence assay in B16F1 cells. However, Western blot analyses displayed that BLEE decreased the expression level of catalase, but no effect on the expression level of tyrosinase, tyrosinase associated protein-1 (TRP-1), tyrosinase associated protein-2 (TRP-2) and microphthalmia-associated transcription factor (MITF) transcription factor involved in melanogenesis. Thus, the promotive effect of BLEE on melanin production is attributed to the increase of tyrosinase activity and the reduction of intracellular $H_2O_2$ level. In conclusion, BLEE played a key role in in promoting melanin production, which suggests that the BLEE could be applied as a potential functional material in the development of hair care cosmetics related to the promotion of melanin production for the growth of black hair.

  • Objective : The aim of this study is to assess the effect of Schizandrae Fructus on melanin synthesis inhibition and whitening effect. Methods : We assessed inhibitory effects of Schizandrae Fructus on melanin-release from B16F10, on melanin production in B16F10, on mushroom tyrosinase activity in vitro, on tyrosinase activity in B16F10 and effect of Schizandrae Fructus on the expression tyrosinase, TRP-1, TRP-2, PKA, ERK-1 ERK-2, AKT-1, MITF in B16F10. Results and Conclusion : 1. Schizandrae Fructus inhibited melanin-release, melanin production in B16F10. 2. Schizandrae Fructus inhibited tyrosinase activity in vitro and in B16F10. 3. Schizandrae Fructus suppressed the expression of tyrosinase, TRP-1, TRP-2, PKA, ERK-2 in B16F10.

  • Objective : This study was performed to assess the whitening effect of Bombysis Corpus on melanin synthesis. Methods : The whitening effects of Bombysis Corpus were examined by in vitro melanin production assay. We assessed inhibitory effects of Bombysis Corpus on melanin-release from B16F10, on melanin production in B16F10, on mushroom tyrosinase activity in vitro, on tyrosinase activity in B16F10, effect of Bombysis Corpus on the expression tyrosinase, TRP-1, PKA, ERK-1 ERK-2, AKT-1, MITF in B16F10. Results : 1. Bombysis Corpus inhibited melanin-release, melanin production in B16F10. 2. Bombysis Corpus inhibited tyrosinase activity in vitro and in B16F10. 3. Bombysis Corpus suppressed the expression of tyrosinase, TRP-1 in B16F10. 4. Bombysis Corpus suppressed the expression of PKA in B16F10. 5. Bombysis Corpus suppressed the expression of ERK-1, ERK-2, AKT-1 in B16F10. 6. Bombysis Corpus suppressed the expression of MITF in B16F10. Conclusion : The study shows that Bombysis Corpus inhibited melanin production on the melanogenesis.

  • This study is performed to investigate the effects of citrus essential oils on melanin production in B16 melanoma cells. Five kinds of citrus essential oils (Bergamot, Grapefruit, Lemon, Mandarin, Petigrain) did not have any influence on DPPH radical scavenger activity, cell growth and cytotoxicity in B16 melanoma cells. Both mandarin and petigrain essential oils dose-dependently inhibited purified tyrosinase activity, but bergamot did not. In 1$\mu$M MSH-stimulated B16 melanoma cells, all of 5 citrus essential oils inhibited melanin production in a dose dependent manner. From the above results, it is possible that citrus essential oils may be developed to be an anti-melanogenesis agent on the basis of their inhibitory effect on MSH-induced melanin production.

Effect of Arctigenin on Tyrosinase Activity and Melanin Production in B16 Melanoma Cells (B16 Melanoma 세포에서 Arctigenin이 Tyrosinase 활성과 Melanin 생성에 미치는 영향)

  • To investigate the possibility of development as a whitening agent using arctigenin, we measured DPPH assay, NBT/XO assay, intracellular ROS scavenging assay, tyrosinase assay and MSH-induced melanin production in B16 melanoma cells. Arctigenin dose-dependently had anti-oxidant activity in DPPH, NBT/XO and intracellular ROS assay. Although arctigenin did not inhibit purified tyrosinase activity, it dose-dependently inhibited tyrosinase activity and melanin production in B16 melanoma cells stimulated by $1{\mu}M$ ${\alpha}$-MSH. In particular, arctigenin at a concentration $100{\mu}M$ inhibited ${\alpha}$-MSH-stimulated tyrosinase activity and melanin production by $50.9{\pm}2.9%$ and $69.0{\pm}6.5%$ respectively. And typical tyrosinase inhibitor, arbutin, inhibited $57.7{\pm}2.9%$ and $65.1{\pm}5.0%$ respectively. Such an similar inhibitory effect of arctigenin and arbutin in B16 melanoma cells may be due to the inhibition of MSH signal pathway rather than the direct inhibition of tyrosinase. Therefore, these results suggest that arctigenin may be useful for the development as whitening agents.

  • Melanin is a black pigment, responsible for hair and skin color. In order to find the melanin stimulatory technique which prove useful for a gray and a white hair-preventive agent or tanning agent, we developed atmospheric pressure plasma jet (APPJ) and tested for tyrosinase activity and melanin production in melanoma (B16F10) cells in vitro. We found plasma dose dependent increase in melanin production. To explore the contributing mechanism in melanin synthesis, intracellular reactive oxygen species (ROS) and MAP kinase signaling pathways were studied. Furthermore, the development of plasma technology for melanin synthesis and planning for in-vivo future studies will be discussed.

Ethanolic Extract of Oryza sativa Displays Antioxidative Activity and Promotes Melanin Synthesis (현미 주정 추출물의 항산화 활성 및 melanin 합성 촉진 효과)

  • Hair loses melanin with aging, which leads to hair graying. The change in hair color is caused by a reduction in tyrosinase activity and an accumulation of hydrogen peroxide ($H_2O_2$) in hair follicles. The purpose of this study was to investigate the effect of ethanolic extract of Oryza sativa (OREE) on melanin production and antioxidative activity in B16F1 cells. In this study, OREE showed low DPPH radical scavenging activity and reducing power. However, it displayed a strong antioxidative effect against intracellular $H_2O_2$ in live cells. OREE did not inhibit DOPA oxidation activity in vitro, but it increased tyrosinase activity at a concentration of $64{\mu}g/ml$. OREE at a concentration higher than $32{\mu}g/ml$ showed cell toxicity in B16F1 cells. However, OREE at a concentration higher than $8{\mu}g/ml$ not only increased melanin synthesis in a dose-dependent manner in B16F1 cells but also increased melanin synthesis in cells treated with $H_2O_2$ inhibiting melanin synthesis. To confirm the effect of OREE on melanin production, Western blot analysis was performed. The results revealed that OREE increased the expression levels of tyrosine hydroxylase and tyrosinase-related protein-2 (TRP-2) involved in melanin production in the $H_2O_2$-treated cells in which melanin production was inhibited. The findings suggest that OREE could improve melanin synthesis and be available for development of hair cosmetics aimed at improving melanin production.

The Study of Antioxidant Effects in Melanins Extracted from Various Tissues of Animals

  • This study was carried out to investigate the antioxidant effects of melanin extracts, including silkie fowl skin melanin (SS-melanin), silkie fowl comb melanin (SC-melanin), sepia ink sac melanin (SE-melanin), octopus ink sac melanin (OC-melanin) and synthetic melanin (SY-melanin). The results showed that with the addition of melanin extracts, linoleic acid peroxide significantly, decreased (p<0.05) with the increase in the irradiative time of UV and that OC-melanin had the highest efficiency on antioxidant activity (p<0.05). Melanin extract had reducing power and chelating power to $Fe^{2+}$, which increased with the increase in the different melanin concentration. Therefore, it could be concluded that the antioxidant action of melanin extracts did not come from one single function, but is a result of many characteristic functions.
