Anticancer Activity of the Branch Extracts from Vaccinium oldhamii through Cyclin D1 Proteasomal Degradation in Human Cancer Cells

  • In this study, we investigated the effect of the extracts from Vaccinium oldhamii on cell proliferation and the regulatory mechanisms of cyclin D1 protein level in human cancer cells. The branch extracts from Vaccinium oldhamii (VOB) showed higher inhibitor effect against the cell growth than leave extracts (VOL) and fruit extracts (VOF) in human colorectal cancer, breast cancer, prostate cancer, non-small lung cancer, pancreatic cancer and liver cancer cells. In addition, VOB decreased cyclin D1 level at both protein and mRNA level. MG132 treatment attenuated VOB-mediated cyclin D1 downregulation. A point mutation of threonine-286 to alanine attenuated cyclin D1 degradation by VOB. In addition, the inhibition of nuclear export by leptomycin B (LMB) attenuated cyclin D1 degradation by VOB. But, the treatment of PD98059 (ERK1/2 inhibitor), SB203580 (p38 inhibitor), SP600125 (JNK inhibitor), LiCl ($GSK3{\beta}$ inhibitor), LY294002 (PI3K inhibitor) or BAY 11-7082 ($I{\kappa}K$ inhibitor) did not affect VOB-induced cyclin D1 degradation. In conclusion, VOB induced cyclin D1 degradation through redistribution of cyclin D1 from the nucleus to cytoplasm via T286 phosphorylation of cyclin D1, which resulted in the inhibition of cancer cell proliferation.

  • The roots of Codonopsis lanceolata afforded tangshenoside I(1) and $\beta$-adenosine (2) as $\alpha$-glucosidase inhibitors. Their structures were unambiguously determined by 1D and 2D NMR data including HMQC and HMBC experiments. Compounds 1 and 2 exhibited weak $\alpha$-glucosidase inhibitory activities in vitro with $IC_{50}$ of 1.4 and 9.3 mM, respectively.

  • NAD(P)H:quinone oxidoreductase (quinone reductase: QR: EC1.6.99.2), a cytosolic FAD-containing flavoprotein, form one of the important component of the phase II drug-metabolizing enzyme systems. It is found in all mammalian species tested and is expressed in many organs including the liver. QR catalyses two-electron reduction of qui nones to hydroquinones thereby suppresses the formation of superoxide anion radical. (omitted)

In Vitro Propagation of Commonly Used Medicinal Trees in Korea

  • Forest medicinal resources, which constitute one of the non-timber forest products, have been regarded as healthy and highly valued products. To meet the increasing demand of the medicinal resources, it is necessary to improve the propagation methods of medicinal plants. In vitro propagation not only allows an opportunity for propagating plants in large numbers but also allows for enhancing the quality and quantity of the desired functional component of a plant by altering the growth factors, such as medium, carbon source, and plant growth regulators influence plant. There have been several studies of in vitro propagation methods, such as axillary bud culture, shooting, and embryogenesis, on Kalopanax septemlobus, Eleutherococcus sessiliflorus, Hovenia dulcis, and Schisandra chinensis in Korea between from 2000 through 2010. Furthermore, there have been attempts to proliferate callus and plantlets for producing useful natural compounds by using bioreactors. Here, we provide an account of the in vitro propagation methods of medicinal trees in Korea based on a review of several micropropagation studies.

일천궁과 토천궁에서 발생하는 총채벌레류의 분자동정 (Molecular Identification of Thrips in Two Medicinal Crops, Cnidium officinale Makino and Ligusticum chuanxiong Hort)

  • Background: Cnidium officinale Makino and Ligusticum chuanxiong Hort. are important medicinal crops in Korea. However, there is insufficient information on the identification of thrips, which attack these plants. Until now, one species of thrips has been recorded as a main pest. Methods and Results: To identify the thrips emerging in C. officinale Makino and L. chuanxiong Hort., these plants were independently cultivated in two local areas. Thirty individuals of each plant species were selected randomly and surveyed for the presence of thrips. After confirming the existence of thrips, 100 thrips individuals were collected from each crop using the beating method. To identify thrips species, we performed PCR-restriction fragment length polymorphism (RFLP)-based analysis using ITS2 primer sets. Six thrips species were identified: western flower thrips (Frankliniella occidentalis), flower thrips (F. intonsa), onion thrips (Thrips tabaci), chrysanthemum thrips (T. nigropilosus), chilli thrips (Scirtothrips dorsalis), and grass thrips (Anaphothrips obscurus). The proportion of these species differed between the host plant species. Conclusions: Six thrips species were major pests of two medicinal crops. Integrated pest management is required to control these thrips species, and will enhance the yield and quality of C. officinale and L. chuanxiong.

Immunostimulatory Activity of Solanum nigrum Through TLR4-Mediated JNK Activation in RAW264.7 Cells

  • In this study, we investigated the effect of Solanum nigrum aerial parts (SNAP) on macrophage activation and macrophage autophagy in RAW264.7 cells. SNAP increased the production of immunostimulatory factors and phagocytosis in RAW264.7 cells. TLR4 inhibition blocked SNAP-mediated production of immunostimulatory factors. In addition, the JNK inhibition reduced the SNAP-mediated production of immunostimulatory factors, and the SNAP-mediated JNK activation was blocked by the TLR4 inhibition. SNAP activated macrophage autophagy. TLR4 inhibition blocked SNAP-mediated macrophage autophagy and inhibition of p38 and JNK attenuated SNAP-mediated macrophage autophagy. These findings indicate that SNAP may induce TLR4/JNK-mediated macrophage activation and TLR4/p38 and JNK-mediated macrophage autophagy.

Antioxidant and Anticancer Activities of Leaves Extracts from Acer tegmentosum

  • In this study, we were performed to elucidate the antioxidant and anticancer activity by leaves extracts from Acer tegmentosum (AT-L). In DPPH, ABTS radical scavenging activity, the AT-L revealed the high scavenging activity. Especially, the AT-L measured the highest ABTS radical scavenging activity, which is higher than ascorbic acid. The types of human cancer cells for evaluating the anticancer activity were colorectal cancer (SW480), prostate cancer (PC-3), breast cancer (MCF-7), pancreatic cancer (AsPC-1), lung cancer (A549) and liver cancer (HepG2). Human cancer cell viability was measured using MTT assay. Treatment of the AT-L decreased the cell viability and induced apoptosis in SW480 cells. These results suggest that extracts of the AT-L can be used as supplementary material for developing the natural antioxidant and anticancer drug for human cancer cells.
