In this paper, the effect of Ganopoly(extracts of Ganoderma lucidum) and Ganopoly/C+(70% Ganopoly + 30% chitosan) on cisplastin-induced nephrotoxicity was investigated in Sprague-Dawley rats. A single dose of cisplastin(5 ㎎/㎏) kg) was administered intraperitoneally after pretreatment of saline, Ganopoly and Ganopoly/C+ for 7 days. The nephrotoxicity and renal function were manifestated by the changes of body weight, blood pressure, biochemical changes and solute in urine and plasma. After the treatment of CDDP(cis-dichlorodiamineplatinum), a significant elevation of kidney weight, serum urea, cretinine, urine volume for 24 hours, urine magnesium, and a severe or significant decrease in body weight, blood pressure, creatinine clearance, urine osmolarity, serum albumin, etc. The nephrotoxicity was further confirmed by a significant decrease in glutathione S-transferase(GSH) in urine and kidney homogenate, GSH, glutathione peroxidase(GSH-Px) and catalase in kidney tissue. And also the lipid peroxidation was significantly increased in kidney homogenate. These signs of nephrotoxicity was ameliorated by the pretreatment and consecutive administration of Ganopoly and Ganopoly/C+ for 14 days after the Lp. injection of CDDP on 7th day after pretreatment of Ganopoly and Ganopoly/C+. The amelioration of nephrotoxicity was evidenced by significant reduction in serum urea and creatinine concentration, and improvement of other index of renal function. And The activity of antioxidant enzymes were partially recovered in kidney tissue of rats treated by CDDP and the administration of Ganopoly and Ganopoly/C+. These results indicate the cispastin induced nephrotoxicity is due to an impairment of tubular reabsorption systems enhanced by necrosis of proximal tubule, and the Ganopoly and Ganopoly/C+ has a partial protective effect on nephrotoxicity induced by CDDP. The polysacchride of Ganoderma lucidum may improve the therapeutic index of nephrotoxicity induced by CDDP. However, it is needed to elucidate the mechanism for confirming the therapeutic effect.

In this report, we present a case study of how pharmacogenomics and pharmacometabolomics can be useful to characterize safety and pharmacokinetic profiles in early phase new drug development clinical trials. During conducting a first-in-human trial for a new molecular entity, we were able to determine the mechanism of dichotomized variability in plasma drug concentrations, which appeared closely related to adverse drug reactions (ADRs) through integrated omics analysis. The pharmacogenomics screening was performed from whole blood samples using the Affymetrix DMET (Drug-Metabolizing Enzymes and Transporters) Plus microarray, and confirmation of genetic variants was performed using real-time polymerase chain reaction. Metabolomics profiling was performed from plasma samples using liquid chromatography coupled with quadrupole time-of-flight mass spectrometry. A GSTM1 null polymorphism was identified in pharmacogenomics test and the drug concentrations was higher in GSTM1 null subjects than GSTM1 functional subjects. The apparent drug clearance was 13-fold lower in GSTM1 null subjects than GSTM1 functional subjects (p < 0.001). By metabolomics analysis, we identified that the study drug was metabolized by cysteinylglycine conjugation in GSTM functional subjects but those not in GSTM1 null subjects. The incidence rate and the severity of ADRs were higher in the GSTM1 null subjects than the GSTM1 functional subjects. Through the integrated omics analysis, we could understand the mechanism of inter-individual variability in drug exposure and in adverse response. In conclusion, integrated multi-omics analysis can be useful for elucidating the various characteristics of new drug candidates in early phase clinical trials.
Purpose: The purpose of this study is to identify the real condition of urinary incontinence(UI), interstitial cystitis(IC), atrophic viginitis(AV) for elderly women and analyze the cognition of traditional korean medicine(TKM) for them. Methods: We utilized questionnaire from May to June, 2010. Questionnaires were taken from 125 women using senior welfare center and nursing home, aged over 65 up to 92. The data were analyzed by $X^2$-test using SPSS/PC ver 18.0 program. Results: The prevalences of UI, IC and AV symptoms were 50.4%, 40.8%, 56%, respectively. The average I-QoL score for UI was $82.62{\pm}21.16$, and the average ICSI score for IC was $8.16{\pm}2.50$. After adjustment for each of the variables considered in this study, alcohol was associated with UI and age, BMI(body mass index) were associated with IC. Most of respondents have no experience(94.4%) or don't know (79.2%) about TKM for UI, IC and AV. 44 women(37.3%) indicated that they weren't willing to use TKM for UI, IC and AV. Reasons for not taking TKM were because of 'no knowledge of TKM(34.1%)' and 'more accustomed to western treatment(34.1%)'. Conclusion: Although the prevalence of UI, IC, AV for elderly women was high, the actual percentage of treatment for these diseases was low, furthermore, patients were not aware of TKM and had very few experiences of TKM for these diseases. The development and increased promotion about TKM program for elderly women's urogenital diseases is needed.
This study aims to determine the effects of mental health on obesity among Korean adolescents. The data in this study were obtained from the 2013 Korean Youth Health Risk Behavior Web-based Survey conducted in 70,354 adolescents aged twelve to eighteen (35,575 males and 34,779 females) from June 1 to 30, 2013. The variables included age, the economic status of household, academic achievement, health status, perceived happiness, perceived stress, satisfaction of sleeping, perceived depression, and suicidal ideation and the data were processed using t-test, chi square test, and logistic regression analysis. The general characteristics affecting obesity for both Korean male and female adolescents included age (p<0.001), the economic status of households (p<0.001), academic achievement (p<0.001), and health status (p<0.001); the variable of mental health affecting obesity was suicidal ideation among females (p<0.05). It is expected that female adolescents' experience of suicidal ideation can be used as information to predict the likelihood of obesity in managing an obesity program for adolescents and it is necessary to provide obesity education suitable for age and to include obesity guidance in consideration of gender and the economic status.

Hypertension is the most prevalent disease worldwide and is itself a risk factor for cerebral, cardiac, and renal diseases. The inconsistency of candidate genes suggested by previous genomewide association studies (GWASs) may be due to not only differences in study design and genetic or environmental background but also the difference in the power of analysis between continuous traits and discrete traits. We analyzed 352,228 single nucleotide polymorphisms (SNPs) in 8842 unrelated Koreans obtained from Ansan and Ansung cohorts. We performed a series of GWA analyses using three different phenotype models; young hypertensive cases (278 subjects) versus elderly normotensive controls (680 subjects); the upper 25% (2211 hypertensive cases) versus the lower 25% of the SBP distribution (2211 hypotensive controls); and finally SBP and DBP as continuous traits (8842 subjects). The numbers of young hypertensive cases and elderly normotensive controls were not large enough to achieve genomewide significance. The model comparing the upper 25% subjects to the lower 25% of subjects showed a power that was approximate to that of QTL analysis. Two neighboring SNPs of the ATP2B1 gene, rs17249754 (SBP, p=$2.53^{-10}$; DBP, p=$1.28{\times}10^{-8}$) and rs7136259 (SBP, p=$1.30{\times}10^{-9}$; DBP, p=$6.41{\times}10^{-8}$), were associated with both SBP and DBP. Interestingly, a SNP of the RPL6 gene, rs11066280, revealed a significant genomewide association with SBP in men only (p=$3.85{\times}10^{-8}$), and four SNPs located near the MAN2A1 gene showed a strong association with DBP only in elderly men aged 60-70 years (e.g., rs6421827, p=$4.86{\times}10^{-8}$). However, we did not observe any gene variant attaining genomewide significance consistently in the three phenotype models except for the ATP2B1 gene variants. In general, the association signal with blood pressure was stronger in women than in men. Genes identified in GWASs are expected to open the way for prevention, early diagnosis, and personalized treatment of hypertension.
Purpose: A quantitative descriptive study aimed to identify the relationships between spiritual well-being and perception attitudes of death in nursing students. Methods: A survey was conducted and 175 data were collected. Collected data were computed and analyzed using SPSS10.0 for Win. 1) Descriptive statistics were used to identify demographic data, and 2) both t-test and ANOVA statistics were used to figure out the relationships between spiritual well-being /or perception attitudes of death and demographic data of the participants. 3) Pearson's correlation coefficient was used to identify the relationships between the two main variables. Results: 1) The mean of spiritual well-being score of the participants was $50.9943{\pm}10.7235$. Significant relationships between religion /or doing economic activity and spiritual well-being were found. 2) The mean of perception attitudes of death was $20.4914{\pm}2.6280$. There were significant relationships between gender /or age and perception attitudes of death among participants. 3) A strong positive relationship between spiritual well-being and perception attitudes of death of participants was also found(r=.261, P=.000). Conclusion: The study results shows that spiritual well-being of nursing students as caregivers of terminal patients is important when the perception of death of terminal patients is considered. Previous studies indicate that caregivers' perception attitudes impact on those of terminal patients. Therefore, it is suggested that not only perception attitudes of death and spiritual well-being in terminal patients, but also those of caregivers are importantly required to consider in educational programs in relation to spiritual care of terminal patients.

Objectives: Synchronous electroencephalogram (EEG) and functional magnetic resonance imaging (fMRI) has been used to explore sleep stage dependent functional brain networks. Despite a growing number of sleep studies using EEG-fMRI, few studies have conducted network analysis on whole night sleep due to difficulty in data acquisition, artifacts, and sleep management within the MRI scanner. Methods: In order to perform network analysis for whole night sleep, we proposed experimental procedures and data processing techniques for EEG-fMRI. We acquired 6-7 hours of EEG-fMRI data per participant and conducted signal processing to reduce artifacts in both EEG and fMRI. We then generated a functional brain atlas with 68 brain regions using independent component analysis of sleep fMRI data. Using this functional atlas, we constructed sleep level dependent functional brain networks. Results: When we evaluated functional connectivity distribution, sleep showed significantly reduced functional connectivity for the whole brain compared to that during wakefulness. REM sleep showed statistically different connectivity patterns compared to non-REM sleep in sleep-related subcortical brain circuits. Conclusion: This study suggests the feasibility of exploring functional brain networks using sleep EEG-fMRI for whole night sleep via appropriate experimental procedures and signal processing techniques for fMRI and EEG.
This study was performed to explore the difference in communication anxiety and class satisfaction after taking the traditional lecture and flipped learning lecture. Fifty four nursing students participated in this study. The study design was one group pretest-posttest design. 4 weeks traditional lecture and 4 weeks flipped learning lecture was applied. Flipped learning was ineffective in improving communication anxiety (t=1.85, p=.069) of nursing students. But emotional state variables and activity variables in the emotional domain were significantly higher after taking the flipped learning lecture(t=-3.80, p=.000; t=-3.35, p=.001). In addition, all of the variables were higher in the flipped learning based team, in the control of the class activities (t=-3.07, p=.003), personal ability (t=-2.48, p=.016), and class participation(t=-3.25, p=.002). Flipped learning is therefore considered to be effective in training nursing students. This study suggested to investigate the effectiveness of flipped learning and learners' satisfaction.
Purpose: Biliary atresia (BA) is a disease that manifests as jaundice after birth and leads to progressive destruction of the ductal system in the liver. The aim of this study was to investigate histopathological changes and immunohistochemically examine the expression of glial cell line-derived neurotrophic factor (GDNF), synaptophysin, and S-100 protein in the gallbladder of BA patients. Methods: The study included a BA group of 29 patients and a control group of 41 children with cholecystectomy. Gallbladder tissue removed during surgery was obtained and examined immunohistochemically and histopathologically. Tissue samples of both groups were immunohistochemically assessed in terms of GDNF, S-100 protein, and synaptophysin expression. Expression was classified as present or absent. Inflammatory activity assessment with hematoxylin and eosin staining and fibrosis assessment with Masson's trichrome staining were performed for tissue sample sections of both groups. Results: Ganglion cells were not present in gallbladder tissue samples of the BA group. Immunohistochemically, GDNF, synaptophysin, and S-100 expression was not detected in the BA group. Histopathological examination revealed more frequent fibrosis and slightly higher inflammatory activity in the BA than in the control group. Conclusion: We speculate that GDNF expression will no longer continue in this region, when the damage caused by inflammation of the extrahepatic bile ducts reaches a critical threshold. The study's findings may represent a missing link in the chain of events forming the etiology of BA and may be helpful in its diagnosis.
This study was conducted to identify the subjective happiness and self-esteem of nursing students and how these variables affect clinical practice stress. Participants consisted of 199 nursing students who were in the third and fourth years who had experienced clinical practice at two nursing universities located in Gyeongnam, and agreed to the survey. Data were collected using questionnaires from June 15 to August 29, 2020. Collected data were analyzed using the SPSS 22.0 statistics program with descriptive statistics, an independent t-test, ANOVA, Scheffe's test, and multiple regression. The mean score for subjective happiness was 4.87 (out of 7), the mean for self-esteem was 3.84 (out of 5), and clinical practice stress was 2.77 (out of 5). The significant predictors of the clinical practice stress were gender, major satisfaction and self-esteem, and the explanatory power of variance was 14.1%. Therefore, we need differentiated clinical practice stress reduction programs according to gender and various strategies that can enhance major satisfaction in order to reduce the clinical practice stress of nursing students.
