콩 주요 노린재류의 약제감수성 (Susceptibility of Stink Bugs Collected in Soybean Fields in Milyang to Some Insecticides)

  • 밀양의 콩 포장에서 채집한 5종 노린재류의 7종 약제에 대한 감수성을 평가하였다. 7종 약제에 대한 노린재의 반수치사농도($LC_{50}$)는 노린재의 종류에 관계없이 deltamethrin에서 0.4${\sim}$2.3 ppm으로 가장 낮았으며, 톱다리개미허리노린재의 fenthion을 제외하면 가로줄노린재, 풀색노린재, 알락수염노린재 및 썩덩나무노린재는 phenthoate에서 가장 높았다. 7종 약제에 대한 노린재류의 평균 반수치사농도는 톱다리 개미허리노린재에서 7.5 ppm으로 가장 낮았으며, 다음은 썩덩나무노린재(16.6 ppm)>알락수염노린재 (17.6 ppm)>풀색노린재(19.1 ppm)>가로줄노린재(28.4 ppm)의 순으로 낮았다. 또한 약제별 추천농도에 대한 노린재의 90% 치사농도의 비로 나타낸 상대적 약제내성비 (상대적 약제내성비=$LC_{90}$/약제별 추천농도)는 톱다리개미허리노린재, 가로줄노린재 및 알락수염노린재는 fipronil에서, 풀색노린재는 fenitrothion에서 썩덩나무노린재는 fenthion에서 가장 낮았다. 노린재류의 7종 약제에 대한 평균 약제내성비는 톱다리개미허리노린재에서 0.09로 가장 낮았으며, 다음은 알락수염노린재(0.26)>풀색노린재(0.30)>가로줄노린재(0.37)>썩덩나무노린재(0.39)의 순으로 낮았다. 그리하여 노린재류의 약제감수성은 톱다리개미허리노린재에서 가장 높았으며, 가로줄노린재에서 가장 낮았고, 상대적 약제내성비는 톱다리개미허리노린재에서 가장 낮았으며, 썩덩나무노린재에서 가장 높았다. 따라서 공시약제에 대한 노린재류의 상대적 내성비가 1.0 미만으로 약제감수성이 매우 높아 콩 포장에서 약제살포에 의한 노린재류의 방제효과는 매우 높을 것으로 여겨진다.

중금속, 암모니아, 유기주석화합물이 피조개 (Scapharca broughtonii) 의 생존에 미치는 영향 (Influence of Heavy Metals, Ammonia, and Organotin Compounds on the Survival of Arkshell Clams, Scapharca broughtonii)

  • Arkshell clams, Scapharca broughtonii, are economically important edible bivalves and widely cultivated in the Southern coast of Korea. Recently, the production of S. broughtonii has been dramatically decreased and various reasons including chemical pollution were suspected to be related to the production declines. However, it remains unknown whether the chemical pollution levels of the surrounding environments were high enough for the biological and ecological disturbance for the population of S. broughtonii, because no systematic toxicological study using S. broughtonii has been conducted previously. In the present study, we exposed arkshell clams, S. broughtonii to various waterborne pollutants including heavy metals (Cd, Cu and Hg), ammonia and organotins (tributyltin and triphenyltin) to determine the effect concentrations of these pollutants for the survival of S. broughtonii for 20 days. The median lethal concentrations ($LC_50$) of S. broughtonii were 2.1 mg/l for Cd, 0.065 mg/l for Cu, 0.40 mg/l for Hg, 79.4 mg/l for total ammonia (1.9 mg/l for unionized ammonia), 0.5 ${\mu}$g/l for TBT, and 14${\mu}$g/l for TPhT. Lethal toxicity of the most pollutants increased with both exposure duration and concentration. The toxicity of TBT was greatest for S. broughtonii, followed by TPhT > Cu > Hg > Cd > ammonia. The sensitivities of S. broughtonii to heavy metals and TBT were comparable to those of other aquatic organisms, but they were relatively tolerable to ammonia. The environmental concentrations of the tested pollutants were compared with the effect concentrations of those for the survival of S. broughtonii to assess the potential risks of the pollutants in the field conditions.

화학물질 관리 연구-1. 산업안전보건법상 관리 화학물질의 특성과 노출기준 비교 (Study on the Chemical Management - 1. Chemical Characteristics and Occupational Exposure Limits under Occupational Safety and Health Act of Korea)

  • Objectives: This study aims to compare the physicochemical characteristics, toxicological data with Occupational Exposure Limits (OELs) of chemicals under the Occupational Safety and Health Act(OSHA) regulated by the Ministry of Employment and Labor of Korea. Methods: Information on chemicals which have OELs on physicochemical characteristics and toxicological data was collected using Material Safety Data Sheet(MSDS) from Korea Occupational Safety and Health Agency(KOSHA) and the Korea Information System for Chemical Safety Management(KISChem) in 2014. Statistical analyses including correlation and simple regression were performed to compare the OELs with chemical characteristics including molecular weight, boiling point, odor threshold, vapor pressure, vapor density, solubility and octanol-water partition coefficient(OWPC) and toxicological data such as median lethal dose($LD_{50}$) and median lethal concentration($LC_{50}$). Results: A total of 656 chemicals have OELs under OSHA in Korea. The numbers of chemicals which have eight-hour time weighted average(TWA) and short term exposure limits(STEL) are 618 and 190, respectively. TWA was significantly correlated with boiling point and STEL was only correlated with vapor pressure among physicochemical characteristics. Solubility and OWPC between "skin" and "no skin" substances which indicate skin penetration were not significantly different. Both $LD_{50}$ and $LC_{50}$ were correlated with TWA, while the $LC_{50}$ was not with STEL. As health indicators, health rating and Emergency Response Planning Guidelines(ERPG) rating as recommended by the National Fire Protection Association(NFPA) and American Industrial Hygiene Association(AIHA) were associated with OELs and reflect the chemical hazards. Conclusions: We found relationships between OEL and chemical information including physicochemical characteristics and toxicological data. The study has an important meaning for understanding present regulatory OELs.

Characterization of Mamestra brassicae Nucleopolyhedrovirus (MabrNPV)-K1 Isolated in Korea

  • The purpose of this study was to investigate the characteristics of Mamestra brassicae nucleopolyhedrovirus (MabrNPV)-K1 isolated in Korea. Polyhedra of MabrNPV-K1 showed irregular appearance in shape with the average diameter $1.8{\mu}m$. MabrNPV-K1 contained a number of nucleocapsids within a viral envelope embedded in polyhedron. The polyhedrin of MabrNPV-K1 was composed of single polypeptide with a M.W. of approximate 31 kDa which is identical to the commercialized MabrNPV, Mamestrin, as a biological control agent. The nucleotide and amino acid sequences within the coding region of MabrNPV-K1 polyhedrin shared 99.0% similarity with the polyhedrin gene from previous reported MabrNPVs. The median lethal concentrations ($LC_{50}$) of MabrNPV-K1 and Mamestrin to M. brassicae larvae were $3.9{\times}10^3$ PIBs/larva and $6.0{\times}10^4$ PIBs/larva, respectively. Mortality of the MabrNPV-K1 against to the third instars larvae was 15 times higher than that of the Mamestrin. The median lethal times ($LT_{50})$ of MabrNPV-K1 by the concentration of polyhedra were lower ($4.4{\sim}6.1$ days) than those of Mamestrin ($4.1{\sim}8.6$ days). These results suggest that a local strain MabrNPV-K1 has high pathogenicity to M. brassicae and may be useful for the development of biological control agent to control this.

Mass Production of Aphicidal Beauveria bassiana SFB-205 Supernatant with the Parameter of Chitinase

  • Beauveria bassiana SFB-205 supernatant can effectively control cotton aphid populations, which is closely associated with its chitinase activity. The present work extends to optimizing a culture medium to produce more efficacious supernatant in flask conditions, followed by scale-up in 7 L, 300 L and 1.2 KL fermentors with the parameter of chitinase. In flask conditions, a combination of soluble starch and yeast extract produced the greatest amount of chitinase (5.1 units/ml) and its supernatant had the highest aphicidal activity. An optimal quantitative combination of the two substrates, estimated by a response surface method, enabled the supernatant to have 15.7 units/ml of chitinase activity and 3.7 ml/l of median lethal concentration ($LC_{50}$) of toxicity against cotton aphid adults in laboratory conditions. In the scale-up conditions, overall supernatant had 25-28 units/ml of chitinase activity. Decrease in pH and limitation of dissolved oxygen (DO) during cultures were significantly related to the yield of chitinase. These results suggest that the substrate-dependent chitinase production can be background information for optimizing a culture medium, and pH and DO are critical factors in maximizing the production in scale-up conditions.

SD Rats를 이용 1-hexene의 흡입독성 연구 (Study on Inhalation Toxicity of 1-Hexene in SD Rats)

  • The purpose of this study was to investigate the acute (4 hours) and repeated-dose (6 hours a day, 5 days a week, 4 weeks) toxic effects of 1-hexene on Sprague-Dawley (SD) rats which were treated by inhalation. The results were as follows; 1. The median lethal concentration(LC$_{50}$) was estimated 52,694 ppm (confidence limit 95%; 49,494~55,447 ppm) in acute inhalation. Abnormal clinical signs related to the 1-Hexene were not observed with the acute inhalation dose. Cross findings of necropsy revealed on evidence of specific toxicity related to the 1-hexene. II. By repeated inhalation exposure the body weight of male were more or less reduced by the dose of 2,500 ppm and 5,000 ppm compared with control group. However there were no significant variation hematology and blood biochemistry for the exposed rats compared with the control rats. Abnormal clinical signs and gross findings of necropsy related to the 1-hexene were not shown. In conclusion when we exposed 1-hexene to SD rats for 4 weeks, 5 days per week, 6 hours per day, the Lowest observed effect level (LOEL) was over 2,500 ppm and Non observed effect level (NOEL) was below 500 ppm.

Acute Toxicity to Peptone Concentrations in the Polychaete Perinereis aibuhitensis under Laboratory Culture

  • Organic pollution causes eutrophication and dystrophication, which occur when excessive amounts of organic matter enters seawater. Eutrophication can contaminate sediment and harm aquaculture. Polychaeta species have been shown to restore eutrophic sediment. In this study, we used peptone to simulate a eutrophic environment and detect the levels at which eutrophication became toxic to the polychaete Perinereis aibuhitensis. Peptone concentrations were 0, 100, 200, and 500 mg/L. The median lethal concentrations were 950.35 mg/L at 48 h, 340.34 mg/L at 72 h, and 120.22 mg/L at 96 h, which are much higher than those of other aquatic species. Polychaeta species are highly tolerant of eutrophication. During the 15-day long-term experiment, sediment loss on ignition, as well as seawater total organic carbon and total nitrogen all decreased significantly (P<0.05). However, $NH_4^+$ concentration increased with time. Perinereis aibuhitensis slowed the increment of $NH_4^+$ but could not prevent its increase. Our results indicate that this polychaete is helpful in the recovery of seawater and sediment from eutrophication.

Quantification of the Sub-lethal Toxicity of Metals and Endocrine-disrupting Chemicals to the Marine Green Microalga Tetraselmis suecica

  • Microalgae are sensitive indicators of environmental changes, and hence they are widely used in environmental risk assessments and for the development of discharge guidelines. Here we evaluated the toxicity of metals and endocrine-disrupting chemicals (EDCs) to the marine green microalga, Tetraselmis suecica. The toxicants investigated included the metals, Cu, Ni, and Pb; and the EDCs, bisphenol A (BPA), endosulfan (ES), and polychlorinated biphenyl (PCB). The endpoints were variations in cell counts and chlorophyll a levels. T. suecica displayed a varied pattern of sensitivity to the toxicants. Based on the 72-h median effective concentration ($EC_{50}$), ES (0.045 mg/L) was most toxic to T. suecica, followed by PCB (3.96 mg/L) and Pb (9.62 mg/L). Interestingly, T. suecica was relatively tolerant to Cu (43.03 mg/L). The 72-h $EC_{50}$ values of Ni and BPA were approximately 16 mg/L. Our data suggest that this species may be relatively tolerant to most of the chemicals within their permissible limits in the environment.

Chlorothalonil이 미역(Undaria pinnatifida) 배우체의 생존 및 상대성장률에 미치는 영향 (The impact of chlorothalonil on female gametophyte survival rate and relative growth rate of Undaria pinnatifida)

  • TBT 사용이 금지된 이후, 방오도료의 방오능력을 보완하기 위하여 booster biocides를 추가하여 사용하고 있다. Booster biocides 중에서 chlorothalonil은 해양환경 내 지속적으로 유입되며 다양한 해양생물에게 심각한 독성영향을 미치고 있지만 해조류에 대한 연구는 제한적이 다. 따라서 우리나라의 주요 양식생물인 미역(Undaria pinnatifida)의 초기생활사에 chlorothalonil이 미치는 영향을 분석하였다. U. pinnatifida의 암배우체를 chlorothalonil(0, 0.03, 0.05, 0.10, 0.20, 0.40 mg L-1)에 노출하여 생존율과 상대성장률을 분석하였다. 암배우체 생존율의 무영향 농도(NOEC), 최소영향농도(LOEC), 반수치사농도(LC50)는 0.05, 0.10, 0.141 (0.121~0.166) mg L-1이었고 상대성장률의 NOEC, LOEC, 반수영향농도(EC50)은 0.10, 0.20, 0.124 (0.119~0.131) mg L-1로 분석되었다. 따라서, 미역의 암배우체는 0.05~0.10 mg L-1 이상의 농도에서 독성영향을 받기 시작할 것으로 판단된다. 본 연구의 결과는 chlorothalonil에 대한 U. pinnatifida 초기생활사의 건강도를 평가하기 위한 기준자료로 활용될 것으로 기대된다.

CuCl2 노출에 의해 유도되는 제브라피시의 행동과 내분비계의 변화 (Exposure to Copper (II) Chloride Induces Behavioral and Endocrine Changes in Zebrafish)

  • 본 연구의 목표는 CuCl2가 지브라피시에 미치는 영향을 조사하는 것이다. CuCl2에 대한 지브라피시의 반수치사농도를 조사하기 위하여 각기 다른 시간에 따른 농도를 설정하여 노출시켰다. 본 연구에서는 스트레스 반응의 증가를 보기 위하여 whole-body cortisol level 측정과 행동약리학적 변화를 측정하기 위해서 open field test와 novel tank test를 실시하였다. Whole-body cortisol의 sample을 수집하거나 행동실험을 실시하기 전에 CuCl2 또는 vehicle을 1시간 동안 처리하였다. CuCl2의 반수치사농도는 24시간, 48시간, 96시간에서 각각 30.3, 25.3, 14.8 ㎍/l으로 측정되었다. CuCl2에 노출된 지브라피시의 novel tank test의 결과는 총 이동거리, 움직이는 시간 그리고 속도는 유의성이 있게 감소한 반면, turn angle은 최고농도에서 유의성이 있게 증가하였다(p<0.05). 다른 행동실험인 open field test의 결과에서도 총 이동거리, 움직이는 시간 속도가 CuCl2의 노출에 의해서 감소하였으며, turn angle과 meandering이 유의하게 증가하였다(p<0.05). CuCl2에 노출된 지브라피시의 whole-body cortisol은 1.5 or 15 ㎍/l의 농도에서는 유의하게 증가하였으나, 150 ㎍/l의 농도에서는 유의성 있는 변화를 발견하지 못하였다(p<0.05). 본 연구의 결과는 CuCl2의 노출이 지브라피시에서 치사율 증가, 행동 및 내분비의 변화 등을 포함하는 독성 반응을 야기하는 것을 나타낸다.
