In this study, we analyzed the densities and taxonomic characteristics of various microorganisms that play important roles in Agaricus bisporus culture medium composting, and examined changes in the levels of decomposition-related enzymes secreted by these microorganisms. Various microorganisms such as thermophilic bacteria, actinomycetes, fluorescent Pseudomonas spp., and filamentous bacteria are closely associated with culture medium composts of Agaricus bisporus. The population densities of microorganisms change, and harmful bacteria disappear during thermophilic composting. Psychrobacter sp., Pseudomonas sp., Bacillus sp., and Pseudoxanthomonas sp. accounted for the highest proportion of bacteria in the culture media during outdoor composting, whereas Bacillus sp. and Psychrobacillus sp. were dominant after pasteurization. Cellulose and hemicellulose enzymes of the microorganisms were important at an early stage of rice straw composting and after decomposition of carbon sources, respectively. Microorganisms that secreted these enzymes were present in the second and third turning stage of composting.
The objective of this study was to investigate the treatment efficiency, kinetics, and morphological characteristics of biofilm in upflow $Biobead^{(R)}$ process, a kind of biological aerated filter(BAF). The $Biobead^{(R)}$ system showed high removal rates of $COD_{Mn}$(76~83%), $BOD_5$(67~88%) and SS(71~91%) for food wastewater with high salt concentration ($>4,000mg/{\ell}$) under short reaction times(2~3hrs). Even at aerobic condition, the system had high treatment efficiency for both T-N (51~63%) and T-P(62~81%). The removal kinetics of $COD_{Mn}$, $BOD_5$, T-N, T-P, and $Cl^-$ in the $Biobead^{(R)}$ system showed a plug-flow pattern with reaction rate constants($hr^{-1}$) of 0.58, 0.63, 0,30, 0.48, and 0.38 respectively. A backwashing process to remove excess biomass and filtered solids was needed at least once during 22-hour operation at $0.5kg\;BOD\;m^{-3}{\cdot}d^{-1}$ loading. At the higher loading($1.0kg\;BOD\;m^{-3}{\cdot}d^{-1}$) the backwashing interval was shorten by 8 hours. The COD, BOD, T-N, and T-P were removed from 43 to 66% only by aerobic biodegradation. The SS was removed over 70% by the filtering of $Biobead^{(R)}$ media in the treatment system. The first one of three serial Biobead reactors showed the highest removal values for $COD_{\alpha}$(52.3%), $COD_{Mn}$(38.8%), BOD(62.5%), and T-N(40.0%). The SS and T-P had the highest removal values(47.5% and 29.2%) at the second one of the serial reactors. The biofilm had non-homogeneous spatial distribution and the colonies were embedded in the sunk area of the Biobead. The thickness of the biofilm was very thin ($5.0{\sim}29.4{\mu}m$) compared to the biofilm thickness($200{\sim}300{\mu}m$) used in other BAF systems.

Nuclease was secreted to the environmental media from the Escherichia coli JM107 tranformant harboring the extracellular nuclease gene of Serratia marcescens in the plasmid of pNUC4. Under the growth conditions, the amount of secreted enzyme was increased in parallel with bacterial growth conditions, the amount of secreted enzyme was increased in parallel with bacterial growth. The enzyme was purified using chromatofraphic procedures of Matrex green gel and heparin agarose affinity gel, resulted in 50-fold purification with 15% recovery of the enzyme. The apparent molecular weight of the enzyme was estimated to be 29Kda by sodium dodecylsulfate denaturing gel electrophoresis. Using the purified enzyme, polyclonal antibody was obtained from the rabbit. The specificity of the antibody was confirmed by immunoblotting and immunoprecipitaion. For the investigation of cellular distribution of the enzyme, cells were fractionated into three fractions; cytoplasm, periplasm and extracellular fluid. While more than 80% of the enzymatic activity was detected in the extracellular fluid and periplasm, a little was found in the cytoplasm, indicating that the enzyme was likely to be immediately exported to the membrane for excretion after biosynthesis. These results were confirmed again by immunocytochemistry technique using the antibody.
A clinical study was made on 71cases of Mycoplasma pneumoniae pneumonia from March, 86 to February, 89. The results were as follows; 1. Among the 315 cases of pneumonia, the incidence of mycoplasmal infection was 22.5%. 2. The peak incidence of age was between 5 to 9years of age(53.5%). 3. The sex ratio of male to female was 1.3:1. 4. Monthly distribution showed relatively high frequency from October to January(59.2%). 5. Most common clinical symptoms were cough(98.6%)and then followed by fever(49.3%), coryza(19.7%). Rales were the most common finding(95.7%) and followed by pharyngeal injection (49.3%) and wheezing(18.3 %). 6. The leukocyte counts in peripheral blood were most common in the range of 5000-10000/$mm^3$(47.9%) and the ESR was increased in 57.7%, and positive CRP cases were 87.3%. 7. The most common radiologic finding of pulmonary infiltration was interstitial infiltration(45.1) and then followed by disseminated lobular(39.4%) and lobar pneumonia(15.5%). 8. There are a few cases associated disease or complication: otitis media (5.6%), hepatitis(4.2%) acute glomerulonephritis. bronchial asthma and sinusitis(2.8%), thrombocytopenia(1.4%).
In this study the migration experiment using packed column with crushed tuff was conducted as a basic research to develop migration model of radionuclides through geologic media. The main emphasis was put on evaluating the validity of migration models. For this, two models were introduced: one is the model which is based on the assumption of instantaneous equilibrium reaction and the other the model based on kinetic process such as intraparticle diffusion. The coefficient of hydrodynamic dispersion in packed column was determined using iodine as nonsorbing tracer. The hydrodynamic dispersion coefficient, D$_{L}$ was shown to be 0.11$\times$10$^{-2}$$\textrm{cm}^2$/min under the condition of the column porosity of 0.483 and the average water velocity of 0.915$\times$10$^{-2}$ cm/min. The distribution coefficient, Kd of Cs-137 on crushed tuff was 11.3 cc/g at the concentration of 2$\times$10$^{-6}$ M and the temperature of 2$0^{\circ}C$. The breakthrough curve of Cs-137 through packed column was shown to have an asymmetric curve in which long trailing tail appears at the end part of the curve. The results obtained from the comparison of introduced models with experimental data indicated that the mass transfer model with intraparticle diffusion as rate-controlling step simulated the behaviors of Cs-137 migration more adequately, when compared with the bulk reaction model in which the assumption of instantaneous equilibrium reaction was maded. Consequently, the intraparticle diffusion was found to be an important factor in the migration of Cs-137 through packed column.n.

This study surveyed how retirees' evaluation of starting food service business affects the effectiveness their new business and quality of life, based on personal factors such as entrepreneurship and business-starting capability and environmental factors by using questionnaires. Bootstrapping was carried out in order to find out factors affecting rapidly changing new business environments and retirees' initial intent to start a business so as to verify basic hypothesis about relation between retirees' evaluation of starting food service business (social, economic, and psychological effects) and the effectiveness of their new business and quality of life and confirm whether the effectiveness of the new business acts as a medium between the evaluation of starting food service business and quality of life. In addition, PLS-MGA was performed in order to verify whether the correlations among test factors can be varied according to the kind of job the target retirees had. Having examined the basic thesis, it was found that social and economic factors had significant positive effect on the effectiveness of the new business, and social and psychological factors had significant positive effect on the quality of life. Having analyzed whether the effectiveness of new business acted as a media between evaluation of starting food service business and quality of life, there was no significant effect as a medium. Having studied whether the kind of job of the retirees controlled or affected the relations among evaluation of starting food service business, effectiveness of new business and quality of life, the results were as follows: in the office job retiree group, the greater economic factor led to increase of effectiveness of new business, while social and psychological factors influenced the quality of life; In the physical labor group, the higher social factor resulted in higher effectiveness of new business, which showed significant positive effect on the quality of life. Having researched about which element is considered to be most important in starting food service business, the most important element was found to be dish/menu, followed by staff management, accounting management, business management, and service education. Having analyzed relation between accomplishment and important consideration for starting food service business, "managers with entrepreneurship," "appropriate distribution of time to prepare for starting business," and "operation of practical field education programs" showed higher importance compared to the degree of satisfaction, so it is needed to more concentrate on the above matters. This study intends to raise retirees' awareness of starting business and help them live better life based on the analysis results, and further suggest detailed mechanism and specific operations of factors affecting retirees' decision making on starting business, such that they can use the information as basic materials to make better choices that can lead to successful business.
To prevent the secondary hospital-acquired infection (cross-infection) from occurring in the general radiographic room in the department of radiology, the microbial measurement was conducted at the points making direct close contact with radiologists and patients. For the case of radiologists, the microbial measurement and incubation were focused on the x-ray tube handle of the radiation generating device, and, for the case of patients, the microbial measurement and incubation were focused on the chin supporting device, chest-contact point, and handle. Once disinfected with Aniosurf, the sterilized media were gathered and identified, and the microorganisms were confirmed. Based on the identification results, it was confirmed that the points making direct close contact with radiologists showed a value of 103 CFU for Proteus mirabilis, Staphylococcus epidermidis, Bacillus spp. and Candida spp., and that the points making direct close contact with patients showed a value of 103~5 CFU for Proteus mirabilis, Enterococcu faecium, Pseudomonas aeruginosa, NTM(Non-Tuberculosis Mycobacteria) and Candida spp.. It was also confirmed that the types and number of microorganisms gathered from the points making direct close contact with patients were greater. Fortunately, most of the involved microorganisms were observed to be on the skin surface and are known to become extinct when disinfected in accordance with the hospital-acquired infection control rules. However, since even minor exposure to such microorganisms may be lethal for patients with reduced immunity, caution must be taken. In particular, since the points making contact with patients showed a high level of microbial measurement, it was thought that it would be necessary for radiologists and personnel having frequent access to strictly disinfect the parts, such as instruments and handles, making frequent contact with patients. The purpose of this study was to announce the importance of safe microbial control in the radiographic inspection room in hospital, and this study is expected to be used as the baseline data for preventing hospital-acquired secondary infection and Nth infectious diseases.
El Niño is a typical ocean and atmospheric interaction phenomenon that causes climate variability on a global scale, so it has been used as a very important teaching and learning material in the field of earth science. This study summarized the distribution and dynamics of the equatorial current system. The variability of the equatorial current system during the El Niño period and the associated misconceptions were also investigated. The North Equatorial Current, South Equatorial Current, and Equatorial Under Current significantly weaken during El Niño years. However, the variability of the North Equatorial Counter Current (NECC) during the El Niño period cannot be generalized because the NECC shows southward movement with weakening in the northern area and strengthening in the southern area, along its central axis. In the western Pacific, the NECC is further south during El Niño years, and thus, it has an eastward flow in the equatorial western Pacific. Our analysis of a mass media science article, a secondary school exam, and a survey for incumbent teachers confirmed disparate ideas about the equatorial current system's variability during El Niño periods. This is likely due to inaccurate interpretations of the existing El Niño schematic diagram and insufficient understanding of the equatorial current and wave dynamics.

From the 25 fresh fruits and vegetable products, 1,250 isolates grown on MRS agar media were screened for the inhibitory activity on the growth of Escherichia coli 0157:H7, Listeria monocytogenes, and Bacillus cereus as well as Lactobacillus plantarum, L. casei, and Lactococcus lactis subsp. lactis. Among them, 607 isolates (49% of total isolates) from 23 different foods produced growth inhibitory activity on the E. coli 0157:H7, L. monocytogenes, or B. cereus. When these isolates were screened for the inhibitory activity on the growth of L. plantarum, L. casei, and Lactococcus lactis subsp., 24 isolates (3% of total isolates) from 7 food samples showed bacteriocin-like activity. These isolates had typical physiological characteristics of lactic acid bacteria, which indicated these isolates were strains of lactic acid bacteria. The inhibitor from 3 out of 24 revealed as nicin. From the RAPD-PCR profiles, 24 strains was classified and it was also indicated that most of the strains isolated from same produce showed similar phylogenetic profile.
Flammulina velutipes, which is a white rot fungus, is a commercially important edible mushroom and is produced in large quantities with the help of an automated and mechanized cultivation system in Korea. F. velutipes has the lowest distribution rate among domestic cultivars, estimated at about 20 percent. As most white cultivars of F. velutipes produced and exported to Korea were introduced from Japan, farmers pay large amounts of royalties. Therefore, we have developed a new, purely domestic cultivar, "Baeke," to substitute for Japanese cultivars, which has improved storage characteristics for export. Baeke was bred by mating two monokaryotic strains isolated from ASI 4216-18 (Hansol) and ASI 4217-26 (Baekjung). Baeke showed faster mycelial growth and higher mycelial density upon incubation for seven days at $25^{\circ}C$ on PDA media than the control variety. The mycelial growth of Baeke was even maintained at $30^{\circ}C$ unlike the control. The lengths of pilei and stipes in Baeke harvested in the optimal stage were $11.2{\pm}0.5mm$ and $125{\pm}5.4mm$, respectively, and they were $11{\pm}0.5mm$ and $141.9{\pm}5.7mm$, respectively, in the control harvested in the optimal stage. The yields of Baeke ($257.4{\pm}13.5g$) and control ($270.7{\pm}17.8g$) per 1,100ml in bottle cultivation showed no significant difference. Overall, our results showed that Baeke was at par with foreign varieties of Flammulina velutipes in terms of quality and yield and had a uniformly shaped fruitbody, which added to its commercial value.
