Effect of Milk Protein Isolate/κ-Carrageenan Conjugates on Rheological and Physical Properties of Whipping Cream: A Comparative Study of Maillard Conjugates and Electrostatic Complexes

  • With increasing consumer demand for "clean label" products, the use of natural ingredients is required in the food industry. Protein/polysaccharide complexes are considered good alternatives to synthetic emulsifiers and stabilizers for formulating stable emulsion-based foods. Milk protein and carrageenan are widely used to improve the physical properties and stability of dairy food products. In a previous study, milk protein isolate (MPI) was conjugated with 𝛋-carrageenan (𝛋-Car) in a wet-heating system through the Maillard reaction, and the Maillard conjugates (MC) derived from MPI and 𝛋-Car effectively improved the stability of oil-in-water emulsions. Therefore, MPI/𝛋-Car conjugates were used in whipping cream as natural emulsifiers in this study, and the physical and rheological properties of whipping creams stabilized using MPI/𝛋-Car MC and MPI/𝛋-Car electrostatic complexes (EC) were investigated. The whipping creams stabilized with MPI/𝛋-Car MC have lower rheological parameters (ηa,50, K, G', and G'') than those of whipping creams stabilized with MPI/𝛋-Car EC. Although the overrun value was slightly reduced owing to the addition of MPI/𝛋-Car MC, the stability of the whipped creams with MC was effectively improved due to enhanced water-holding ability by conjugation.

듀록 품종의 Melanocortin-4 Receptor(MC4R) 유전자와 성장형질과의 연관성 분석 (Associations of the Porcine Melanocortin-4 Receptor (MC4R) Gene with Growth Traits in Duroc Pigs)

  • 본 연구는 축산과학원에서 보유중인 듀록 품종내에서 MC4R 유전자의 SNP 발현 특성과 육종가에 의한 선발 후 MC4R 유전자 빈도의 변화 및 경제형질에 대한 유전자형간 육종가의 차이를 구명하고자 수행하였다. 1999년부터 2005년까지 검정된 검정성적을 바탕으로 일당증체량, 등지방두께, 90kg 도달일령 및 사료요구율에 대하여 유전력과 유전상관 및 육종가를 추정하였으며, 2003년과 2004년에 출생한 660두에 대한 혈액을 채취하여 MC4R 유전자에 대한 유전자형 및 대립유전자 분석을 실시하였다. 또한 육종가에 의한 선발 후 유전자의 빈도변화를 세대별 그리고 선발군 및 도태군에 대하여 분석하였으며, 육종가의 분석결과를 이용하여 MC4R의 유전자형 효과를 보았다. 분석결과 육종가를 근거하여 선발한 MC4R 유전자형은 세대당 그리고 선발군과 도태군에서 차이를 보였으며, 각 형질별 육종가의 분산분석결과 사료요구율을 제외한 기타 경제형질에서 유의적으로 MC4R 유전자의 효과가 있는 것으로 나타났다. MC4R 유전자의 효과에 대한 보고는 상이한 부분도 있지만 자체 축군에 대한 다형성 분석 및 경제형질과의 연관성 분석에 의하여 축군 및 개량하고자 하는 경제형질에 따라 표지 유전인자로 활용하여 선발반응 및 정확도를 높일 수 있을 것으로 사료된다.

  • This study was carried out to develop an immunoassay for the diagnosis of the pregnancy and ovarian function of domestic animals. Using 2E92C10 monoclonal antibody(McAb) generated against estrone-3-glucuronide(E1-3-G) and appeared a high cross-reactivity with estrone-3-sulfate(E1-3-S), chemiluminesence immunoassay (CIA) to detect E1-3-S was developed. 2E92C10 McAb cross-reacted with E1-3-S (30%) was purified from ascites fluid using protein G sepharose gel column. The purity of purified antibody fraction was monitored by SDS-PAGE and was better compared to that of crude ascite fluid. The soild and liquid phase CIA for E1-3-S were established utilizing 2E92C10 antibody and E1-3-G-ABEI conjugate used as a tracer. As the results, the titer of 2E92C10 antibody was 5g/ml in soild phase and 1:2000 in liquid phase. The sensitivity on soild and solid phase CIA were about 200 pg/ml. These results indicate that CIA for measurement of E1-3-S was successfully developed by using ant-E1-3-G McAb cross-reacted with E1-3-S and could be usefully used to research this area.

방향가공시 유연제 전처리에 의한 면편성물의 태변화 (Effect of Pretreatment by Softener on the Cotton Knit Handle in Fragrant Finishing)

  • The fragrant fabrics are prepared by the attaching microcapsules using acrylic binder. To improve the handle of fragrant fabrics, pretreatment by softener is suggested. The durability of fragrant fabrics and the change of their handle are investigated. It is carried out One-way ANOVA and Duncan test to determine bending properties, shear properties, surface properties of untreated, SO-MC and BI-MC by Kawabata system. BI-MC shows significant differences in B and 2HB. SO-MC shows significant differences in G and 2HG. It turned out the pretreatment by softener mitigated the effect of binder and makes fabrics softer than untreated fabrics.

Association between expression levels and growth trait-related SNPs located in promoters of the MC4R and MSTN genes in Spinibarbus hollandi

  • Melanocortin 4 receptor: (MC4R) and Myostatin (MSTN) are two important growth trait-related genes in animals. In this study, we showed that two SNPs, MC4R-719A>G and MSTN-519C>T, found in the promoters of the MC4R and MSTN genes, respectively, are both associated with growth traits in Spinibarbus hollandi. Furthermore, we observed that there were significant associations between the expression levels of the MC4R and MSTN genes and these two growth trait-related SNPs. The expression level of MC4R gene in brain was lower in GG genotype fish with extremely high growth performance than that in AA genotype fish with extremely low growth performance. Expression level of the MSTN gene in muscle was lower in TT genotype fish with extremely high growth performance than that in CC and CT genotype fish with lower growth performance. The results indicated that these SNPs located in the promoters of MC4R and MSTN are associated with growth-related traits through modification of gene expression levels. The MSTN and MC4R SNPs may have useful application in effective marker-assisted selection aimed to increase output in S. hollandi.

Relationships between solar/interplanetary (IP) parameters and Dst index, according to IP sources

  • We have investigated interplanetary (IP) structures of 82 intense geomagnetic storms (Dst $\leq$ -100 nT) that occurred from 1998 to 2006. According to their interplanetary origins, we classified them as four groups: 20 sMC events (IP shock and MC), 19 SH events (sheath field), 12 SH+MC events (Sheath field and MC), and 8 nonMC events (non-MC type ICME). For each group, we examined the relationships between Dst index and solar/IP parameters, namely, direction parameter (DP), CME speed ($V_{CME}$), solar wind speed ($V_{SW}$), minimum of IMF $B_z$ component($Bz_{min}$), and maximum of $E_y$ component ($Ey_{max}$).We found that the relationships strongly depend on their IP source. Our main results can be summarized as follows: 1) The correlation between Dst and DP is the best for the SH+MC events (r = -0.61). 2) The relationship between Dst and $V_{CME}$ gives the best correlation for the sMC events (r = -0.56). 3) There is the best correlation between Dst and $V_{SW}$ for the sMC events (r = -0.61), while there is a very weak correlation (r=-0.17) for the SH events. 4) The relationship between Dst and $Bz_{min}$ gives the best correlation (r = -0.87) for the SH+MC events. 5) The correlation between Dst and $Ey_{max}$ is the best for the SH+MC events (r = -0.87). Summing up, the sMC and SH+MC events give us good correlations, but the SH events, weak correlations. From this study, we suggest that this tendency should be caused by the characteristics of IMF southward components, e.g., smooth field rotations for the MC events and highly IMF fluctuations for the SH events.

비만세포에서 은행잎 플라보놀에 의한 Th2 Cytokine 발현 및 신호전달 억제 기전 효과 (Suppressive effects of Th2 cytokines expression and the signal transduction mechanism in MC/9 mast cells by flavonol derived from Ginkgo biloba leaves)

  • 은행잎 유래 플라보놀 성분이 갖는 항아토피 활성에 대해 입증된 바는 드물다. 이 논문에서는 MC/9 비만세포에서의 은행잎 플라보놀의 항아토피 효과를 조사하기 위하여 ELISA와 Real-time PCR, western blot으로 은행잎 플라보놀을 분석하였다. 분석 결과 IL-13, MIP-1a의 생성량은 농도 의존적으로 현저하게 감소되었으며 IL-4, IL-5, IL-13, TNF-a 유전자 발현량은 25, 50, $100{\mu}g/m{\ell}$의 농도에서 효과적으로 억제되었다. western blot 분석 결과 c-jun과 NFAT-1단백질 발현이 감소되었음을 확인하였다. 이러한 결과들은 은행잎 유래 플라보놀 성분이 NFAT-1, c-jun 전사인자의 전사를 억제함으로써 MC/9 비만세포에서의 Th2 싸이토카인의 생성을 감소시키는 효과를 갖는 것을 나타낸다. 따라서 은행잎 유래 플라보놀 성분이 아토피 피부염 치료제로서 활용가능성이 있음을 보고하고자 한다.

Allelic variation of melanocortin-1 receptor locus in Saudi indigenous sheep exhibiting different color coats

  • Objective: This study was designed to characterize the DNA polymorphisms of the melanocortin-1 receptor (MC1R) gene in indigenous Saudi Arabian sheep breeds exhibiting different color coats, along with individuals of the Sawaknee breed, an exotic sheep imported from Sudan. Methods: The complete coding region of MC1R gene including parts of 3' and 5' untranslated regions was amplified and sequenced from three the indigenous Saudi sheep; Najdi (generally black, n = 41), Naeimi (generally white with brown faces, n = 36) and Herri (generally white, n = 18), in addition to 13 Sawaknee sheep. Results: Five single nucleotide polymorphisms (SNPs) were detected in the MC1R gene: two led to nonsynonymous mutations (c.218 T>A, p.73 Met>Lys and c.361 G>A, p.121 Asp>Asn) and three led to synonymous mutations (c.429 C>T, p.143 Tyr>Tyr; c.600 T>G, p.200 Leu>Leu, and c.735 C>T, p.245 Ile>Ile). Based on these five SNPs, eight haplotypes representing MC1R $E^d$ and $E^+$ alleles were identified among the studied sheep breeds. The most common haplotype (H3) of the dominant $E^d$ allele was associated with either black or brown coat color in Najdi and Sawaknee sheep, respectively. Two other haplotypes (H6 and H7) of $E^d$ allele, with only the nonsynonymous mutation A218T, were detected for the first time in Saudi indigenous sheep. Conclusion: In addition to investigating the MC1R allelic variation in Saudi indigenous sheep populations, the present study supports the assumption that the two independent nonsynonymous Met73Lys and Asp121Asn mutations in MC1R gene are associated with black or red coat colors in sheep breeds.

동물세포내에서의 유체성 세균의 증식 (Amplification of Chlamydia trachomatis in Animal Cell Host)

  • Chlamydia trachomatis는 인간에게 각종 질병을 일으키는 병원균이며 최근 그 중요성 이 크게 증가하였다. Chlamydia trachomatis감염에 대한 재래식 진단방법은 낮은 정밀도와 낮은 특이성을 갖고 있으며 방법의 수행 자체도 어렵다는 문제점을 지니고 있다. 이들 재래식 진단방법들이 안고 있는 문제점들을 해결하기 위하여 항-병원균 모노클로날 항체를 생산하는 하이브리도마 세포주를 만들 최종목표를 갖고 우선 생쥐에 면역시킬 항원으로 사용할 Chlamydia trachomatis를 동물 숙주 세포내에서 증식시키는데 끼치는 각종 인자들에 대한 영향을 조사연구하였다. 동물 세포 유래의 McCoy 세포내에서 충분한 양의 Chlamydia tracthomatis를 증식시킨 후 불연속 Urografin 구배 원심분리에 의해 정제하였다. Mc-Coy 세포 내에서의 Chlamydia trachomatis의 생성 개체수를 늘리기 위하여 화학제제를 사용했던 바 IUdR 처리가 cycloheximide 처리보다 더 효과적이었으며 Chlamydia trachomatis 증식을 위해 쓸 수 있는 방법이었다. 원심력의 증가가 Chlamydia tracho chomatis의 McCoy 세포 표면에의 흡착을 증진시켰으며 약 3000g 근처에서 최대 감염율을 보였다. 분리 정제한 Chlamydia trachomatis는 SPG 용액 내에서 4$^{\circ}C$에서 48시간 동안 저장될 수 있었으며 더욱 오래 동안 저장시키기 위해서는 -7$0^{\circ}C$까지 냉동시키는 것이 필요하였다.

The quality of subgroup analyses in chronic pain randomized controlled trials: a methodological review

  • The quality of subgroup analyses (SGAs) in chronic non-cancer pain trials is uncertain. The purpose of this study was to address this issue. We conducted a comprehensive search in MEDLINE and EMBASE from January 2012 to September 2018 to identify eligible trials. Two pairs of reviewers assessed the quality of the SGAs and the credibility of subgroup claims using the 10 criteria developed by Sun et al. in 2012. The associations between the quality of the SGAs and the studies' characteristics including risk of bias, funding sources, sample size, and the latest impact factor, were assessed using multivariable logistic regression. Our search retrieved 3,401 articles of which 66 were eligible. The total number of SGAs was 177 of which 52 (29.4%) made a subgroup claim. Of these, only 15 (8.5%) were evaluated as being of high quality. Among the 30 SGAs that claimed subgroup effects using an appropriate method of performing interaction tests, the credibility of only 5 were assessed as high. None of the subgroup claims met all the credibility criteria. No significant association was found between the quality of SGAs and the studies' characteristics. The quality of the SGAs performed in chronic pain trials was poor. To enhance the quality of SGAs, scholars should consider the developed criteria when designing and conducting trials, particularly those which need to be specified a priori.
