Analysis for Concentration Range of Fluorescein Sodium (플루오레신나트륨의 농도 범위 분석)

  • Brain tumors or gliomas are fatal cancer species with high recurrence rates due to their strong invasiveness. Therefore, the goal of surgery is complete tumor resection. However, the surgery is difficult to distinguish the border because tumors and blood vessels have the same color tone and shape. The fluorescein sodium is used as a fluorescence contrast agent for boundary separation. When the external light source is irradiated, yellow fluorescence is expressed in the tumor, which helps distinguish between blood vessels and tumor boundaries. But, the fluorescence expression of fluorescence sodium depends on the concentration of fluorescein sodium and such analytical data is insufficient. The unclear fluorescence can obscure the boundaries between blood vessels and tumors. In addition, reduce the efficiency of fluorescence sodium use. This paper proposes a protocol of concentration range for fluorescence expression conditions. Fluorescent expression was observed using a near-infrared (NIR) color camera with corresponding dilution using normal saline in 1 ml microtube. The flunoresence emission density range is 1.00 mM to 0.15 mM. The fluorescence emission begin to 1.00 mM and the 0.15 mM discolor. The discolor is difficult to fluorescence emission condition obserbation. Thus, the maximum density range of the bright fluoresecein is 0.15 mM to 0.30 mM. When the concentration range of fluorescein sodium is analyzed based on the gradient of fluorescence expression and the power measurement, the brightest fluorescence is expected to facilitate the complete resection of the tumor. For the concentration range protocol, setting concentration ranges and analyzing fluorescence expression image according to saturation and brightness to find optimal fluorescence concentration are important. Concentration range protocols for fluorescence expression conditions can be used to find optimal concentrations of substances whose expression pattern varies with concentration ranges. This study is expected to be helpful in the boundary classification and resection of brain tumors and glioma.

  • Effects of several different petroleum fractions (LGO, HGO, VGO, Diesel oil, SP(E), HGO-wax, L/M-wax), stepwise addition of calculated amounts of HGO at defined intervals, recycling of spent media on cell growth of Candida tropicalis KIST 351 were studied using $2.5{\ell}$ fermenter by batch process. In addition, continuous cultivation of the yeast was also performed in the light of biomass production using $28{\ell}$ fermenter with LGO. 1) Cell concentration, yield on the basis of gas oil and n-paraffin with the petroleum fractions were in the range of $11{\sim}15g/{\ell}$, $10{\sim}12%$ and $77{\sim}82%$, respectively. 2) By stepwise addition of the gas oil, cell concentration and yield on the oil were increased up to 18.9 g/land 13%, respectively. 3) Spent medium slowed emulsifying ability of hydrocarbon and stimulating effect on the cell growth. Without additional supplementation of $Mg^{++}$ up to 20% of spent medium could be reused, while by adding of the $Mg^{++}$, 50% of medium could be recycled. 4) Optimum condition of continuous cultivation for biomass production was attained at the dilution rate of $D=0.1{\sim}0.125\;hr^{-1}$. Maximum yield coefficient on consumed n-paraffin was 0.94 at $D=0.1\;hr^{-1}$, however, 24% of supplied n-paraffin in the media was not utilized at this dilution rate.

Effect of Soluble-silicate or Chitosan Foliar Spray on Ginseng Cultivated in Blue-white Plastic Film House

  • The experiments were performed in the Jinan (elevation: 300 meters above sea level), Jeollabuk-do. Seedlings (n = 63 per $3.3m^2$) of ginseng cultivar (Cheonpung, Yeonpung) were planted on April 10, 2015. Shading material of plastic film house was blue-white film. Before the Planting seedling, silicate (3 kg/10 a) or chitosan (40 kg/10 a) was fertilized and foliar sprayed on the leaves 1000 times dilution solution once a month from May to September every year. The growth results of 5-year old ginseng surveyed in 2018 are as follows. The average air temperature in the plastic film house was the highest at $26.6^{\circ}C$ and $26.5^{\circ}C$ in July and August, respectively, and the highest temperature was $40.5^{\circ}C$ in July. The maximum daily temperature of $35^{\circ}C$ or more was 30 days, with the average soil temperature being $24.9^{\circ}C$ in August. The chemical properties of the test soil are as follows. pH was 6.4~6.9 level and EC was 0.35~0.46 dS/m. The organic matter content was 33.5~41.4 g/kg, and available-P content was 251.9~306.8 mg/kg. Exchangeable cations contents, such as K, Ca and Mg were all the appropriate ranges. The soil microbial density surveyed by the dilution plate method was 10~50 times higher than that of control (Non-treatment) and actinomycete density was 3~6 times higher. Pathogens of the genus Fusarium by Metagenome analysis decreased 91.3% and 68.2% respectively in the foliar sprayed of chitosan and soluble-silicate. The light intensity (PAR) in the blue-white film plastic film house gradually increased until July and then decereased, with the average of light intensity in March-October was $120.3umol/m^2/s$. The growth of aerial parts such as plant height and stem length was better than non-sprayed group in silicate or chitosan treatments and Yeonpung cultivar was superior to the Cheonpung cultivar. The SPAD value was higher in Yeonpung cultivar foliar sprayed with soluble-silicate. The growth of underground parts such as root length and taproot length were better in chitosan and soluble-silicate treatment than control, especially in Yeonpung cultivar foliar sprayed with chitosan was good in taproot length and taproot diameter, and fresh weight of root was 60.1 g. Ginsenoside contents were 24.9 mg/g and 22.4 mg/g, in the Cheonpung cultivar foliar sprayed with soluble-silicate or chitosan respectively, 28% and 15% higher than control (19.5 mg/g). The incidence of disease by Alteraria panax and Botrytis cinerea was 3~9% and 4~9%, respectively. High temperature damage rate was 3~5%.

Influence of the Asian Monsoon on Seasonal Fluctuations of Water Quality in a Mountainous Stream (산간 계류성 하천의 계절적 수질변동에 대한 몬순강우의 영향)

  • The present study was to determine how seasonal rainfall intensity influences nutrient dynamics, ionic contents, oxygen demands, and suspended solids in a lotic ecosystem. Largest seasonal variabilities in most parameters occurred during the two months of July to August and these were closely associated with large spate of rainfall. Dissolved oxygen (DO) had an inverse function of water temperature (r = = = - 0.986, p<0.001). Minimum pH values of<6.5 were observed in the late August when rainfall peaked in the study site, indicating an ionic dilution of stream water by precipitation. Electrical conductivity (EC) was greater during summer than any other seasons, so the overall conductivity values had direct correlation (r = 0.527, p<0.01) with precipitation. Ionic dilution, however, was evident 4 ${\sim}$ 5 days later in short or 1 ${\sim}$ 2 weeks in long after the intense rain, indicating a time-lag phenomenon of conductivity. Daily COD values varied from 0.8 mg $L^{-1}$ to 7.9 mg $L^{-1}$ and their seasonal pattern was similar (r = 0.548, p<0.001) to that of BOD. Total nitrogen (TN) varied little compared to total phosphorus (TP) and was minimum in the base flow of March. In contrast, major input of TP occurred during the period of summer monsoon and this pattern was similar to suspended solids, implying that TP is closely associated (r = 0.890, p<0.01) with suspended inorganic solids. Mass ratios of TN : TP were determined by TP (r= -0.509, p<0.01) rather than TN (r= -0.209, p<0.01). The N : P ratios indicated that phosphorus was a potential primary limiting nutrient for the stream productivity. Overall data suggest that rainfall intensity was considered as a primary key component regulating water chemistry in the stream and maximum variation in water quality was attributed to the largest runoff spate during the summer monsoon.

Inhibitory effect on Streptococcus mutans and mechanical properties of the chitosan containing composite resin

  • Objectives: This study evaluated the antibacterial effect and mechanical properties of composite resins ($L_{CR}$, $M_{CR}$, $H_{CR}$) incorporating chitosan with three different molecular weights (L, Low; M, Medium; H, High). Materials and Methods: Streptococcus (S). mutans 100 mL and each chitosan powder were inoculated in sterilized 10 mL Brain-Heart Infusion (BHI) solution, and was centrifuged for 12 hr. Absorbance of the supernatent was measured at $OD_{660}$ to estimate the antibacterial activities of chitosan. After S. mutans was inoculated in the disc shaped chitosan-containing composite resins, the disc was cleansed with BHI and diluted with serial dilution method. S. mutans was spread on Mitis-salivarius bacitracin agar. After then, colony forming unit (CFU) was measured to verify the inhibitory effect on S. mutans biofilm. To ascertain the effect on the mechanical properties of composite resin, 3-point bending and Vickers hardness tests were done after 1 and 3 wk water storage, respectively. Using 2-way analysis of variance (ANOVA) and Scheffe test, statistical analysis was done with 95% significance level. Results: All chitosan powder showed inhibition effect against S. mutans. CFU number in chitosan-containing composite resins was smaller than that of control resin without chitosan. The chitosan containing composite resins did not show any significant difference in flexural strength and Vickers hardness in comparison with the control resin. However, the composite resin, $M_{CR}$ showed a slightly decreased flexural strength and the maximum load than those of control and the other composite resins $H_{CR}$ and $L_{CR}$. Conclusions: $L_{CR}$ and $H_{CR}$ would be recommended as a feasible antibacterial restorative due to its antibacterial nature and mechanical properties.

  • Some of the cultural conditions were improved in order to obtain the higher biomass of Lactobacillus bulgaricus NLS-4 which has the higher lactic acid producing activity as well. Among eight media including 11% non-fat milk medium as a control, the TIP medium was selected. By a batch experiment, the maximum cell concentration could be increased to 1.0$\times$10$^{9}$ cells per $m\ell$ when the organism was grown at 38$^{\circ}C$ for 18 hours with agitation speea of 200 rpm and under the constant level of pH 6.5 con-trolled with 1 N KOH solution in the selected medium. The cell concentration was further increased to 2.3$\times$10$^{9}$ cells per me in the steady state of continuous culture at the dilution rate of 0.17 hr$^{-1}$ for 18 hours.

Degradation of Salicylic Acid by Free and Immobilized Cells of Pseudomonas sp. Strain NGK1

  • A Pseudomonas sp. strain NGK1 (NCIM 5120) capable of utilizing salicylate was immobilized in alginate and polyurethane foam (PUF). The degradation rate of salicylate by freely suspended cells was compared with the degradation rate by immobilized cells. In an initial 20 and 40 mM salicylate, free cells ($2{\times}10^{11}\;cfu\;ml^{-1}$) degraded to 16 and 14 mM, alginate-entrapped cells degraded to 18 and 26 mM, and PUF-entrapped cells degraded to 20 and 32 mM salicylate, respectively, in batch cultures. The alginate-and PUF-entrapped cells were used in repeated batch and continuous culture systems. The efficiency of both the immobilized systems f3r the degradation of salicylate was compared. It has been observed that the PUF-entrapped cells could be reused for more than 20 cycles whereas alginate-entrapped cells could be reused for a maximum of only 12 cycles, after which a decrease in degradation rat was observed with the initial 20 and 40 mM salicylate. The continuous degradation of sallcylate by freely suspended cells showed a negligible degradation rate of salicylate when compared with immobilized cells. With the immobilized cells in both alginate and polyurethane foam, the degradation rate increased with an increase in the dilution rate up to $2\;h^{-1}$ for 20 mM, and $1.5\;h^{-1}$ for 40 mM salicylate. The results revealed that PUF-entrapped cells were more efficient for the degradation of salicylate than alginate-entrapped cells and freely suspended cells.

Numerical study of a conical MILD combustor with varing the fuel flow rate (연료유량 변화에 따른 원추형 MILD 연소로의 수치적 해석)

  • MILD combustion is a highly favored technology for solving the trade-off relation between high thermal efficiency and low pollutant emissions. The system has low NOx concentration in high temperature combustion by recirculating the combustion gas, as well as improving the thermal efficiency by making the internal temperature in a combustion furnace uniform. This study describes the combustion characteristics of a conical MILD combustor in a laboratory-scale furnace by adjusting the equivalence ratio with the fuel gas flow rate while maintaining a constant air flow rate of the furnace. The MILD regime in the furnace is well characterized and the in-furnace temperature and emissions were predicted, respectively, for the range of equivalence of 0.69 - 0.83. For the range of equivalence ratios, this study confirmed the existence of a stable flame region that has an approximately $300^{\circ}C$ temperature difference between the maximum flame temperature region and main reaction region.

Comparison of Biomass Productivity of Two Green Microalgae through Continuous Cultivation (두 종 미세 녹조류의 연속배양을 통한 바이오매스 생산성 비교)

  • In the present study, the biomass productivity of two green microalgae (Chlorella sp. and Dunaliella salina DCCBC2) were assessed in a 12 L tubular photobioreactor under optimum culture conditions. In the batch culture optimization process, the Chlorella sp. biomass was obtained as 1.2 g/L under atmospheric air as a sole $CO_2$ source and other culture conditions as follows: light intensity, temperature, pH, $NH_4Cl$ and $K_2HPO_4$ were 100 ${\mu}E/m^2/s$, $27^{\circ}C$, 7.0, 20.0 mM and 2.0 mM, respectively. On the other hand, 2.9 g/L of D. salina DCCBC2 biomass production was observed under the following conditions: light intensity, temperature, pH, $KNO_3$ and $K_2HPO_4$were 80 ${\mu}E/m^2/s$, $27^{\circ}C$, 8.0, 3.0 mM and 0.025 mM, respectively. At 1% $CO_2$ supply to the reactor, the Chlorella sp. production was reached 1.53 g/L with 25% increment under the same operating conditions. In addition, the maximum D. salina DCCBC2 biomass was observed as 3.40 g/L at 3% $CO_2$ concentration. Based on the aforementioned optimized conditions, the dilution rate and maximal biomass productivity of Chlorella sp. and D. salina DCCBC2 in the continuous cultivation were 0.4/d and 0.6 g/L/d and 0.6/d and 1.5 g/L/d, respectively.

Reduction of Nitrous Oxide Emission by EGR Method on Diesel Engine (디젤엔진에서 배기가스 재순환 방법을 이용한 아산화질소의 배출률 저감)

  • Nitrous oxide($N_2O$) concentration in the atmosphere has been constantly increased by the human activities with industrial growth after the industrial revolution. One of factors to increase $N_2O$ concentration in the atmosphere is the $N_2O$ emission caused by the combustion of marine fuel. Especially, a sulfur component included in marine fuel oils is known as increasing the $N_2O$ formation in diesel combustion. Form this point of view, $N_2O$ emission from a ship is not negligible. On the other hand, Exhaust gas recirculation(EGR) that have thermal, chemical and dilution effect is effective method for reducing the NOx emission. In this study, an author investigated $N_2O$ reduction by using EGR on a direct injection diesel engine. The test engine was a 4-stroke diesel engine with maximum output of 12 kW at 2600rpm, and operating condition of the engine was a fixed load of 75%. The experimental oil was a blend-fuel that were adjusted with sulfur ratio of 3.5%, and EGR ratio of 0%, 10%, 20% and 30%. In conclusion, diesel fuel that contained 3.5% sulfur component increased $SO_2$ emission in exhaust gas, and increment of EGR ratio reduced NO emission. Moreover, $N_2O$ emission was decreased as over 50% at EGR ratio of 10% and reduced 100% at EGR ratio of 30% compared with $N_2O$ emission of 0% EGR ratio.
