Structural characterization and anti-inflammatory activity of fucoidan isolated from Ecklonia maxima stipe

  • Enzyme-assisted hydrolysis is frequently used as a cost-effective and efficient method to obtain functional ingredients from bioresources. This study involved the enzyme-assisted hydrolyzation and purification of fucoidan from Ecklonia maxima stipe and the investigation of its anti-inflammatory activity in lipopolysaccharide (LPS)-induced RAW 264.7 cells. Fucoidans of Viscozyme-assisted hydrolysate from E. maxima (EMSFs) harvested in Jeju, Korea. Structural and chemical characterizations were performed using fourier transform infrared spectroscopy, scanning electron microscope, and monosaccharide analysis. Among fucoidans, EMSF6 was rich in fucose and sulfate and had a similar structural character to commercial fucoidan. EMSF6 showed a strong inhibitory effect on nitric oxide generation in LPS-induced RAW 264.7 cells and significantly decreased the production of LPS-induced pro-inflammatory cytokines, including interleukin-6, interleukin-1β, and tumor necrosis factor α. The anti-inflammatory potential of EMSF6 was mediated through the downregulation of inducible nitric oxide synthase and cyclooxygenase-2 expression. Thus, fucoidans from E. maxima stipe are promising candidates for functional food products.

Processing and Shelf-life Stabilities of Flavoring Substances of the Smoke-Dried Oysters (훈건 굴을 이용한 분말조미소재의 가공 및 품질안전성)

  • This study was conducted to evaluate the optimal processing conditions of smoke-dried powdered oysters and to determine their shelf-life during storage for development of a natural oyster flavoring substance. The optimal conditions for processing of smoke-dried oyster powder with freshy oyster were as follows. Raw shelled oysters were rinsed with 3% saline solution, drained, boiled for 10 minutes at $98^{\circ}C$, and then smoked for 1 hour at $50^{\circ}C$, followed by drying for 4 hours at $80^{\circ}C$ Smoke-dried oyster powder with oyster scraps were prepared as flavoring material. The smoked oyster scraps were submerged in oyster sauce far 10 minutes at room temperature and then dried with hot air for 5 hours at $50^{\circ}C$. The smoke-dried oysters and smoke-dried oyster scraps were then pulverized to 50 mesh and packed in tea bags or vacuum-packed in laminated plastic film bags (PE/PVDC/CPP, $12{\mu}m/15{\mu}m/50{\mu}m$). Compared to non smoke-dried powdered oysters, the smoking and dipping in oyster sauce enhanced the flavor and prevented lipid oxidation of the smoke-dried powdered oyster product. Shelf-life tests indicated that the vacuum-packaging method preserved the quality of smoke-dried powdered oysters stored for 150 days at room temperature.

  • While modern illustration designs require originality, the shape and color of marine life provide rich materials for illustration creation. Illustration has a very high artistic creation value. Based on the knowledge of color science and visual form design, this study takes illustration design and marine biology as the main research object. The purpose of this study is: ① The typical features of Marine life are sorted by color and form. ② Lead out graphic symbols representing Marine life. ③ Combine Marine life with illustration design to make design cases. Through research, it is highlighted that the curve feature is a typical feature for distinguishing marine organisms from terrestrial organisms. The classification and comparative analysis of the color phenomena of tropical fish help to extract and process the color of marine tropical fish on the basis of understanding the color characteristics of tropical fish, and apply it to the illustration design.

  • The marine ecosystem represents a vast and dynamic array of bio-resources attributed with its huge diversity and considered as potential untapped reservoirs for the development of functional foods for future health markets. Basically, marine microorganisms, sponges, algae, invertebrates such as crustaceans and mollusks along with marine fish species can be considered as marine bio-resources, which can be utilized to obtain different health benefits for humans, directly or after processing. Most of the bio-molecular components, such as lipids and proteins from these marine bio-resources, which can be extracted in large scale using the modern and advanced biotechnological approaches, are suitable drug candidates for the pharmaceutical industry as well as functional food ingredients for the food industry. Moreover, the furtherance of high throughput molecular biological techniques has already been incorporated with identification, mining and extraction of molecular components from marine bio-resources. In this review, potential marine bio-resources with respect to their extractable bio-molecules were described in details, while explaining the present and prospective methods of identification and extraction, which are integrated with advanced techniques in modern biotechnology. In addition, this provides an overview of future trends in marine biotechnology.

Cell proliferation effect of brown marine algae extracts on Mouse Fibroblast (해조류 추출물이 섬유아세포의 증식에 미치는 영향)

  • We examined cell regeneration efficiency of brown marine algae living in Jeju coast for search of a novel therapeutic device with cutaneous wound healing materials. The five algae were collected and compared with epidermal growth factor (EGF) as a positive control in the assays of cell proliferation and cell migration of NIH3T3 fibroblast cells. Among the 80% methanol extracts of these brown algae, the two algal extracts from Ishige foliacea and Colpomenia bullosa showed the proliferative effects of the cells similar to the effect of EGF. Besides it was found that Colpomenia bullosa extract significantly enhanced cell migration of NIH3T3 cell. In the study, therefore, we confirmed that the Colpomenia bullosa extract improved proliferation of NIH3T3 cell and a potential candidate for cultaneous wound healing.

  • Despite its economic importance as an aquaculture species, the molecular and genetic information regarding physiologically important elements in rock bream (Oplegnathus fasciatus) is not completely understood. Rab proteins play a vital role in cellular mechanisms and immunity as one of the key regulators of membrane trafficking. In this investigation, a Rab gene, named as RbRab-5C-like, was identified from Oplegnathus fasciatus. RbRab-5C-like protein exhibited high homology with Rab proteins of other species and possessed signature characteristics of Rab proteins with four conserved cysteine residues. Phylogenetic analysis showed that RbRab-5C-like clustered with other fish counterparts. The RbRab-5C-like genomic sequence possesses six exons and five introns. Transcriptional analysis revealed that RbRab-5C-like was ubiquitously expressed in all examined tissues with the highest expression occurring in the liver. While the structural and homologic characteristics of RbRab-5C-like suggest a strong conservation of this element in different species, its mRNA distribution implies a wide range of biological significance in rock bream.

  • An analog of antibacterial sulfated alkene against Bacillus subtilis was isolated from a species of tunicate (Mideoduck), Styela clava, cultured at Jindong Bay, Korea. The structure was determined as 4,8-dimethyl-3-nonenyl sulfate by MS and spectral analysis such as UV IR and NMR.

Physiological Characteristics of Olive Flounder (Paralichthys olivaceus) and Korean Rockfish (Sebastes schlegelii) According to the Concentration of Suspended Load (부유사 농도에 따른 넙치와 조피볼락의 생리학적 특성)

  • After exposing Olive flounder and Korean rockfish to the concentration ranges of control (0 mg/L), 100 mg/L, 250 mg/L, 500 mg/L, and 1,000 mg/L, to investigate the physiological properties of suspended load on fish AST/GOT, ALT/GPT, ALP and cortisol were analyzed. As a result of the analysis, the AST/GOT, ALT/GPT, ALP and cortisol values were higher in the suspended load concentration section than in the control group. After artificial infection with S. parauberis for Olive flounder and V. harveyi for Korean rockfish exposed to suspended load, the survival rate was higher in the control group than in the suspended load concentration section. These results are related to the previously study results of suspended load that adversely affect fish, suggesting that suspended load affects the health of fish.

  • In order to aid the researchers who analyze the biotoxins in marine molluscs, this paper introduces the official names and ecological characteristics of the domestically produced or distributed bivalve and gastropods that can be contaminated with marine biotoxins. Also, the paper intends to inform the location and scientific name of each organ of the representative group of species through anatomical maps. In the future, it is necessary to standardize and normalize the names of the species and the research institutions in food codes, scholarly papers and reports on the marine biotoxin analysis.

Ecklonia cava (Laminariales) and Sargassum horneri (Fucales) synergistically inhibit the lipopolysaccharide-induced inflammation via blocking NF-κB and MAPK pathways

  • Ecklonia cava (EC) has been widely utilized as an ingredient in commercial products such as functional foods and cosmeceuticals. Recently it has been found that Sargassum horneri (SH) has been invading on Jeju Island coast area by its huge blooming. Moreover, both seaweeds are considering as important ingredients in traditional medicine specifically in East-Asian countries (China, Japan, and Korea). In the present study, we attempted to compare anti-inflammatory properties of 70% ethanolic extracts of EC (ECE), SH (SHE), and their different combinations on lipopolysaccharide (LPS)-activated RAW 264.7 cells. Results indicated that 8 : 2 combinations of ECE : SHE significantly inhibited LPS-activated inflammatory responses (cytokines, protein, and gene expression) in RAW 264.7 macrophage cells compared to the respective extracts and other combinations. The synergistic effect of ECE and SHE was found to be prominent than the effects of ECE or SHE alone. These observations provide useful information for the industrial formulation of functional materials (functional foods and cosmeceuticals) using these two particular seaweeds in Jeju Island of South Korea.
