The genus Aspergillus is commonly isolated from various marine and terrestrial environments; however, only a few species have been studied in rhizosphere soil. As part of the Korean indigenous fungal excavation project, we investigated fungal diversity from rhizosphere soil, focusing on Aspergillus species. A total of 13 strains were isolated from the rhizosphere soil of three different plants. Based on phylogenetic analysis of β-tubulin and calmodulin and morphological characteristics, we identified five Aspergillus species. A. calidoustus and A. pseudodeflectus were commonly isolated from the rhizosphere soil. Four species were confirmed as unrecorded species in Korea: A. calidoustus, A. dimorphicus, A. germanicus, and A. pseudodeflecuts. The detailed morphological descriptions of these unrecorded species are provided.
BACKGROUND: Marine algae is a productive organism that is consumed as a nutritious food. However, large amounts of unused portions of the algae are incinerated as trash or dumped in the sea, causing pollution. Recycling algae is important for saving resources and conserving the environment. In this study, the fucoidan which is a major carbohydrate of marine algae was tested as a source of fertilizer for farming. METHODS AND RESULTS: The growth rate of the melon was examined after treating fucoidan and the melon growth factors, weight and length of stem were measured. To discover the mechanism of melon growth promotion of fucoidan, bacteria that decomposed fucoidan were isolated from soil and abalone. Bacillus wiedmannii and Stenotrophomonas pavanii were isolated from terrestrial soil and Pseudomonas sp. was isolated from abalone. Among these three bacteria, Pseudomonas sp. had the highest and most specific fucoidan-decomposing activity. When Pseudomonas sp. was treated with fucoidan on melon-growing soil, the growth of melon was relatively improved compared to the treatment with fucoidan alone. CONCLUSION: We found that fucoidan, the main carbohydrate of marine algae, promoted melon growth. Fucoidan-decomposing microorganisms were isolated from terrestrial soil and marine organism, and we found that these bacteria stimulated the effect of melon growth promotion of marine algae. This is the first report that confirms the fertilizer effect of marine algae and shows the use of bacteria with marine algae.
This study examined the growth, fatty acid (FA) content, and carotenoids of a newly isolated freshwater microalga, Mychonastes sp. 246, in various culture media. The appropriate temperature and light intensity for culturing Mychonastes sp. 246 were determined as 18℃-22℃ and 200-250 µmol/m2/s using a high throughput photobioreactor. The microalgal cells were cultivated in 0.5 L bubble column photobioreactors using BG11, Bold's Basal media, and f/2 media. According to the growth results of the microalgae, BG11, among the tested media, showed the highest biomass concentrations (3.5 ± 0.1 g/L in 10 d). To enhance the biomass growth of the microalgae, the N:P ratio in BG11 was manipulated from 45:1 to 7:1 based on the stoichiometric cell composition. The biomass concentrations of Mychonastes sp. 246 grown on the manipulated BG11 (MBG) increased to 38% (4.6 ± 0.3 g/L in d) compared with the original BG11 (3.3 g/L). The FA content of the microalgae grown on the MBG was lower (8.4%) than that of the original BG11 (10.1%) while the FA compositions did not exhibit any significant differences. Furthermore, three kinds of carotenoids were identified in Mychonastes sp. 246, zeaxanthin, lutein, and β-carotene. These results suggest an effective strategy for increasing biomass concentrations, FA content, and carotenoids of microalgae by performing a simple N:P adjustment in the culture media.

In 2016 and 2017, as part of a comprehensive investigation to identify the prokaryotic species in Korea, a total of 12 bacterial strains were isolated from the gastrointestinal tract and/or fecal samples of four endangered species, including reptile, bird, and marine and terrestrial mammals. Phylogenetic analysis with the 16S rRNA gene sequence was used to assign these strains to the phyla, Firmicutes, Actinobacteria or Proteobacteria. Furthermore, most of the strains Firmicutes belonged to the order Lactobacillales. Interestingly, 12 of the isolated strains have not been previously reported from the Korean Peninsula. Also, based on their high 16S rRNA gene sequence similarities(>98.7%) and formation of strong monophyletic clades with the closest type species, each isolated strain of isolates was assigned to an independent, predefined bacterial species. Gram-stain reaction, colony and cell morphology, biochemical characteristics, isolation source, and NIBR IDs are described in the species description section.

Plant-derived antioxidants are used as a healthy diet and are known to inhibit various human diseases. In this study, we investigated free radical scavenging and antioxidant activity of extracts from three plants (Ammannia multiflora, Ammannia coccinea and Salix gracilistyla) with the most DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging activity from 196 plant extracts inhabiting along Nakdong River in Republic of Korea. The three extracts also have strong total antioxidant activity. Moreover, the extracts inhibited hydrogen peroxide (H2O2)-induced reactive oxygen species production and depolarized mitochondrial membrane potential in RAW264.7 macrophages. In zebrafish larvae, 2',7'-dichlorodihydrofluorescein diacetate (DCFDA) fluorescent intensity, induced by H2O2, was markedly reduced by the extracts of A. multiflora, A. coccinea and S. gracilistyla. Meanwhile, the extracts were upregulated Nrf2 and HO-1 expression, and an HO-1 inhibitor reversed the extract-induced oxidative responses both in vivo and in vitro. The data suggest that the extracts of A. multiflora, A. coccinea, and S. gracilistyla exert potential free radical scavenging and antioxidant capacities both in vivo and in vitro by activating the Nrf2/HO-1 signaling pathway.
A cellulolytic bacterial strain, S2-3, was isolated from sea water collected in Jeju island, Republic of Korea. The strain was aerobic and gram negative, and formed yellow colored colonies on marine agar medium. S2-3 cells were long rod-shaped, 0.5 × 0.25 ㎛ (width x length) in size, and did not have flagella. The optimal growth conditions for S2-3 were 30-35℃ and pH 6.5-7.0. Analysis of the 16S rRNA gene sequence of S2-3 revealed that it had the highest identity with those of Seonamhaeicola algicola Gy8 (97.08%), Hyunsoonleella udonensis JG48 (95.01%), and Aestuariibaculum scopimerae I-15 (94.86%). In phylogenetic analysis, S2-3 formed the same clade as S. algicola Gy8, implying that S2-3 belongs to the genus Seonamhaeicola. The major fatty acids (>10%) comprised C15:1 iso G (22.29%), C15:0 iso (17.71%), C17:0 iso 3OH (16.06%), and C15:0 iso 3OH (10.7%), resulting in quite different ratio of the component from those of S. algicola Gy8. Moreover, its biochemical characteristics, including acid production and enzyme activities, were different from those of S. algicola Gy8. Therefore, putting all these results together, we concluded S2-3 is distinct species from S. algicola Gy8, and thus named it Seonamhaeicola sp. S2-3. In liquid culture, S2-3 produced extracellular cellulases that can hydrolyze cellulose or cellooligosaccharides into cellobiose, which is a good enzyme resource that deserves further research.

Obesity, which is characterized by excessive fat accumulation in adipose tissues, occurs by fat absorption by lipase and sequential fat accumulation in adipocyte through adipocyte differentiation. Thus, inhibition of pancreatic lipase activity and adipocyte differentiation would be crucial for the prevention and progression of obesity. In the present study, we attempted to evaluate anti-adipogenic activity of several algae extracts employing preadipocytes cell line, 3T3-L1 as an in vitro assay system. The effects on pancreatic lipase activity in vitro were also evaluated. Total methanolic extracts of Cladophora wrightiana and Costaria costata showed significant inhibitory activity on adipocyte differentiation as assessed by measuring fat accumulation using Oil Red O staining. Related to pancreatic lipase, C. wrightiana and Padina arborescens showed significant inhibition. Further fractionation of C. wrightiana, which showed the most potent activity, suggested that $CHCl_3$ and n-BuOH fraction are responsible for adipocyte differentiation inhibition, whereas n-BuOH and $H_2O$ fraction for pancreatic lipase inhibition. Our study also demonstrated that n-BuOH fraction was effective both in early and middle stage of differentiation whereas $CHCl_3$ fraction was effective only in early stage of differentiation. Taken together, algae might be new candidates in the development of obesity treatment.
세계적으로 생물자원 연구데이터는 그 자체로 중요할 뿐만 아니라, 공유되고 활용되어야 한다. 본 논문에서는 명확한 기준 없이 각각의 연구목적과 특성에 따라 개별적으로 구축, 관리되고 있는 생물자원 연구데이터를 공동 활용 할 수 있도록 정보시스템의 구축 단계부터 적용 가능한 데이터 참조모델을 제시한다. 이를 위해 기존 관련 정보시스템의 데이터 모델을 국내외 표준 및 데이터 관리 정책을 기반으로 확장하여 개별 정보시스템에서 공동 활용 할 수 있는 데이터 참조모델을 개발하고 그 적용 절차를 제안한다. 또한, 제안하는 데이터 참조모델의 우수성을 입증하기 위하여 Krogstie의 데이터모델 평가모형을 적용하여 품질수준을 검증하고 국내외 표준들과의 데이터 공유수준을 비교한다. 실험 결과 기존 데이터 모델보다 데이터를 자원, 대상, 활동, 성과의 4단계로 분류하고 엔티티 도출 및 관계를 정의한 데이터 참조모델에서 데이터의 품질과 공유수준이 높게 나타나는 것을 확인 할 수 있었다.
바다는 지구 표면의 70% 이상을 차지하고 있으며, 그 자체가 대부분 탐사되지 않은 미지와 기회의 공간으로 제시되고 있다. 특히, 우리나라는 삼면이 바다로 둘러싸인 반도로 해양 연구의 중요성이 강조되고 있다. 매우 복잡하고 다양한 환경을 가지고 있는 해양은 막대한 생물학적 다양성을 보이고 있으며 미생물학적 측면에서도 해양 환경은 다양하고 극단적인 온도, 압력, 일사량, 염분, pH 등을 가지고 있어 생태학적으로 특이한 서식처를 제공한다. 이로 인해 육상과는 달리 계통분류학적으로 매우 다르며, 다양한 미생물들이 서식하나 그 다양성, 분리, 배양 그리고 이들이 생산해 내는 2차 대사산물 등에 대한 연구는 아직도 미진한 상황이다. 1990년대까지도 거의 연구되지 않던 해양 환경 자생 미생물의 생리활성물질에 대한 연구는 2000년대 들어 해양 방선균이 생산해 내는 천연물에 대한 연구가 가속화 되기 시작했다. 이후, 박테리아, 고세균, 조류 등을 활용한 항균제, 항암제, 항산화제, 항염증제 등과 같은 의약품 개발 분야 뿐만 아니라 및 바이오 플라스틱 생산, 바이오 연료 생산, 이산화탄소 포집, 생균제 발굴 및 개발 등의 다양한 산업분야에서 해양 미생물을 활용한 연구가 가속화 되고 있는 실정이다. 본 총설에서는 해양미생물을 활용한 생명과학 및 생명공학 기술 분야의 연구 성과 및 최신 동향을 소개하고자 한다. 이를 통해 독자들이 의약소재 개발 외 제반 천연물 관련 분야의 기반 및 응용 연구의 중요성을 인식하고 미래 해양 유래 소재를 이용한 바이오 연구 개발의 최적화 및 실용화 연구에 적극 도움이 되길 기대한다.
Two new marine sponges, Haliclona (Haliclona) tonggumiensis n. sp. and H. (Reniera) sinyeoensis n. sp., in the family Chalinidae were collected from Ulleungdo Island and Gageodo Island, Korea from 2007 to 2009. Haliclona (Haliclona) tonggumiensis n. sp. is similar to H. (H.) simulans (Johnston, 1842) in shape, but the former differs in its ectosomal skeleton structure and spicules' shape and size. The ectosomal skeleton of H. (H.) tonggumiensis n. sp. is absent, but that of H. (H.) simulans is very regularly arranged, and has tangential reticulation with oxea. The spicule shape of H. (H.) tonggumiensis n. sp. is slender, but that of H. (H.) simulans is short and cigar-shape. The new species have two sizes of oxea, but H. (H.) simulans has one size of oxea. Haliclona (Reniera) sinyeoensis n. sp. resembles H. (R.) tubifera (George and Wilson, 1919) in the growth form and choanosomal skeleton structure. However, the new species has two kinds of oxea in size, but H. (R.) tubifera has only one size.
