The vegetative and male reproductive anatomy of a marine alga, Laurencia intercalaris sp. nov. (Rhodomelaceae, Rhodophyta), is described from subtidal habitats of eastern and southern Korea. This species has terete thalli with entangled fibrous holdfasts and regularly alternate branching of ultimate branchlets, and is inseparable from L. okamurae Yamada on the basis of habit. Vegetative axial cells produce a trichoblast and four pericentral cells in an alternating sequence. Spermatangia are produced intercalary or subterminally from one of two laterals on suprabasal cells of trichoblasts arising from axial cells in apical pits of branchlets. The other lateral remains sterile. In this sterile lateral, budding-like regeneration occurs on older segments that are oabscised. Comparison is made with other related Laurencia species, particularly those with terete thalli. The vegetative anatomy and the regeneration in sterile laterals of male trichoblasts, with the mode of spermatangial formation, distinguish the new species from previously described species of Laurencia including L. okamurae.

In continuing search for BChE-inhibitory compounds from Korean marine algae. we found a highly potent inhibitory activity in the methanolic extract of Sargassum species. After partition of the MeOH extract between $CHCl_3$ and 30% MeOH. the former layer was subjected to a series of ODS flash chromatography. silica column chromatography. and preparative TLC to afford three compounds (1-3). Detailed structural elucidation of them is in progress. Compound 1 showed potent BChE-inhibitory activity with $IC_{50}$ values of 11 ng/mL. (omitted)

Micro-algae, one of the biological resources for alternative energy, has been heavily studied. Among various methods to analyze the status of the micro-algae including counting, screening, and flocculation, the flocculation approach has been widely accepted in many critical applications such as red tide removal study or microalgae resource study. To characterize the flocculation status of the micro-alga. A traditional optical modality, i.e., photospectrometry, measuring the optical density of the flocs has been frequently employed. While this traditional optical method needs shorter time than the counting method in flocculation status analysis, it has relatively lower detection accuracy. To address this issue, a novel real-time micro-algae flocculation analysis method based on the lens-free shadow imaging technique (LSIT) is introduced. Both single cell detection and floc detection are simultaneously available with a proposed lens-free shadow image, confirmed by comparing the results with optical microscope images. And three shadow parameters, e.g., number of flocs, effective area of flocs, and maximum size of floc, enabling quantification of the flocculation phenomenon of micro-alga, are firstly demonstrated in this article. The efficacy of each shadow parameter is verified with the real-time flocculation monitoring experiments using custom developed cohesive agents.
This study was evaluated about the seasonal variations of ecosystem in Nammae Reservoir based on the interrelation of physico-chemical characteristics, nutrients, chlorophyll a, b, c and standing crops of phytoplanktons. The amounts of chlorophyll a, b, c were respectively maximum (295 mg/, 9.5mg/l and 48mg/l) at station 1 in June and the standing crop of phytoplanktons was the highest ($1.7{\time}10^5$ cells/1) at stations 3 in July. The range of temperature was $7{\sim}37.4^{\circ}C$. The maxium of pH was $9.9{\sim}10.1$ at all stations in August, the minimum was 7 in September. SS was maximum (308 mg/1) at station 1 in June, while it was minimum (4 mg/l) at the same station in November. The maximal COD and DOC were 33 mg/1 and 16 mg/1 respectively at station 1 in June. As for phytoplanktons, Microcystis aeruginosa, blue-green alga in July${\sim}$August, Scenedesmus acutus, green alga in March${\sim}$May and November${\sim}$January and Cyclotella orientalis, Diatoms in October were dominant species. The amounts of P and Si were generally high in summer, they were low in autumn and winter. Nammae Reservoir assessed by trophic state index was eutrophicated and overtrophicated. These results indicated that Nammae Reservoir was faced with heavy water pollution. As preceding management for the basin of the Reservoir, it will have to be continually studied for an ecosystem reservation.

Cosmopolitan green macroalga, Ulva pertusa is a widely spread species in most coastal waters. This benthic alga is appearing on marine objectives causes significantly economical and social problems. To prevent fouling organisms, new antifouling (AF) agents, such as ziram, diruon, zinc pyrithione, copper pyrithione, DBNPA and triclosan are used. However, the evaluations of new antifouling system of those agents strongly need more information on algae species for considering environment effect. Therefore, the purpose of this study was to evaluate the activity of antifouling system using Ulva spores and Nitzschia pungens. In addition, new AFS were evaluated the toxicity of microalga, Nitzschia pungens. In the motility of Ulva spores, ziram and zinc pyrithione were shown 50% higher inhibition motility than diuron, copper pyritione, DBNPA and triclosan. Ziram was appered the highest AFS of Ulva sore attachment. Copper pyrithione at the concentration of $100{\mu}g/L$ was inhibited 80% growth rate comparison of control. In conclusion, each new AF agents showed their specific AF activities against marine organisms based on this work.
The marine microalga Pavlova viridis can grow fast and has the ability to accumulate essential nutrients for culturing marine animals, such as EPA and DHA, and it has been used as food for raring larval fish and prawn. The symbiotic relationship between the flagellate microalga Pavlova viridis and its associated bacteria was investigated. An axenic culture of P. viridis was obtained by repeated treatment of the microalga with an antibiotic cocktail. The axenic status was confirmed after sub-culturing three times in a sterile f/2 medium without an antibiotic. The axenic alga was then co-inoculated with five bacteria, arbitrarily designated as I1–I5, isolated from the alga to test the growth promotion of the algae. All bacterial strains promoted the growth of P. viridis, and bacterial isolate I3 was the most effective among the five bacteria tested. The cell number of P. viridis in the co-culture with I3 was significantly higher than that of the control culture. A sequence analysis of the 16S rRNA gene isolated from I3 revealed a 97% nucleotide sequence similarity to that of Citrobacter sp. The growth of strain I3 was also significantly enhanced by co-culturing with P. viridis, indicating a symbiotic relationship between the microalga and its associated bacterium. The association between the microalga and bacterium was confirmed by scanning electron microscopy.
The Effect of different temperatures and photon irradiance on the growth of crust and the regeneration of tissue fragments of the commercially important red alga Grateloupia acuminat Okamura were examined in laboratory cultures. The tetraspore developed into basal crusts and produced upright thalli. Crust grew very fast at $25^{\circ}C$ and $80{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ after one week in indoor culture. However, they stopped growing after three weeks. Maximum growth was $275{\mu}m$ in diameter. They required four weeks to get upright thalli at $5^{\circ}C$, while only three weeks were required at $10^{\circ}C$. When different light intensities were compared at $15^{\circ}C$, cells of the crusts were well differentiated $80{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1}$ and consistently divided so that upright thalli were produced. In aeration culture, the upright thalli grew up to 6.5 cm in length within 4 months. Thus, it is possible to produce mass cultures of Grateloupia in the field. In addition, female and male gametophytes developed from the tetraspores and they were fertilized to produce tetrasporohyte thalli. By this procedure, the normal life cycle of the red alga G. acuminata was completed.
Three antimicrobial compounds (SL-l, SL-2 and SL-3) were isolated and identified from the marine red alga, Symphyocladia latiuscula. In addition, their biological functionalities such as cytotoxicity and desmutagenic activity were investigated. From the cryophyllized S. JatiuscuJa, SL-l, SL-2 and SL-3 were purified by solvent extractions and HPLC.SL-2 was crystallized in benzene-diethyl ether solvent. On the EI-MS spectra, it was found that they had three bromines in their structure which showed typical signal strength ratios at $M^+, [M+2]^+, [M+4]^+, [M+6]^+ (13: 38: 37: 12)$. $SL-l$ was identified as 2,3,6-tribromo-4,5-dihydroxybenzyl alcohol ($C_8H_7Br_3O_3, MW=374$) by NMR and MS spectra. SL-2 was assigned as 2,3,6-tribromo-4,5-dihydroxybenzyl methyl ether ($C_8H_7Br_3O_3, MW=388$) and confirmed by X-ray crystallographic analysis. SL-3 was presumed as an isomer of SL-2. Methanol extract of the S. latiuscula showed antimicrobial activities against all strains tested (bacteria, 15 strains; yeasts, 17 strains; fungi, 4 strains), much or less. The strongest inhibition activity of the methanol extract was to the Vibrio mimicus ($50 {\mu}g/ml$) and V. vulnificus ($50 {\mu}g/ml$). The mice injected intraperitoneally with 3 mg of SL-l and 5 mg of 5L-2 showed no acute toxicity response. SL-2 showed higher desmutagenic activity than SL-l against PhIP and MeIQx.
It is important to design photobioreactor by cheap material for economical microalgal biomass production. In this study, two types of marine photobioreactors (MPBR), made by either polyvinyl chloride (MPBR-PVC) or high density poly ethylene (MPBR-HDPE), are used and performance of these were compared. Tetraselmis sp. KCTC 12236BP is a green marine alga that isolated from Ganghwa Island, Korea, and the strain was used for marine cultivations using MPBR-PVC and MPBR-HDPE. The cultivations were performed three times in the spring season of 2012 using MPBR-PVC and of 2013 using MPBR-HDPE in the coastal area of Young Heung Island. As the results, MPBR-PVC shows higher biomass productivities than MPBR-HDPE, due to its high light transmittance. In the cultivations using MPBR-PVC, the average sea water temperature was $11.5^{\circ}C$ during the first experiment and $16.5^{\circ}C$ during the second and third experiments. Average light intensities during three times for experiments were 407.5, 268.1 and $273.0{\mu}{\cdot}E{\cdot}m^{-2}{\cdot}s^{-1}$, respectively. The maximum fresh cell weight and average biomass productivity were $1.2g{\cdot}L^{-1}$ and $0.12g{\cdot}L^{-1}{\cdot}day^{-1}$. These results showed that Tetraselmis sp. KCTC12236BP were adapted well with the environmental conditions from ocean, and grow in the MPBR-PVC and MPBR-HDPE.

Research on the potential biological activity of red alga Symphyocladia spp. has been limited to Symphyocladia latiuscula, which is widely used as a food ingredient in Korea. Here, we examined the biological activity of another species, Symphyocladia linearis, which is found in Korea and was reported as a new species in 2013. The aim of this study was to evaluate the antioxidant, anti-inflammatory, and antibacterial properties of a 70% ethanol extract of S. linearis. Antioxidant activity, which was evaluated using radical scavenging assays, revealed half maximal inhibitory concentration values for 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) of 34.57 and 11.70 ㎍/mL algal extract, respectively. Anti-inflammatory activity of the S. linearis ethanolic extract was evaluated using RAW 264.7 cells by measuring the inhibition of lipopolysaccharide-induced nitric oxide (NO) and prostaglandin E2 (PGE2) production. The potential cytotoxicity of NO and PGE2 was first examined, confirming no toxicity at concentrations ranging from 10-100 ㎍/mL. NO production was inhibited 61.1% and 78.0% at 50 and 100 ㎍/mL S. linearis extract, respectively; and PGE2 production was inhibited 69.1%, 83.2%, and 94.8% at 25, 50, and 100 ㎍/mL S. linearis extract, respectively. Thus, the S. linearis extract showed very strong efficacy against PGE2 production. The cellular production of reactive oxygen species, measured using 2',7'-dichlorofluorescin diacetate fluorescence, was inhibited 48.8% by the addition of 100 ㎍/mL S. linearis extract. Antibacterial activity was evaluated using the disc diffusion method and minimum inhibitory concentration (MIC). S. linearis was effective only against gram-positive bacteria, exhibiting antibacterial activity against Staphylococcus aureus with a MIC of 256 ㎍/mL extract and against Bacillus cereus with a MIC of 1,024 ㎍/mL extract. Based on these results, we infer that a 70% ethanolic extract of S. linearis possesses strong anti-inflammatory properties, and therefore has the potential to be used in the prevention and treatment of inflammatory and immune diseases.
