Molecular Characterization of an Apple cDNA Encoding Cinnamyl Alcohol Dehydrogenase

  • The study of lignin, a major component of secondary cell wall, has been partly focused on its removal from the woody part in the kraft pulping industry. Cinnamyl alcohol dehydrogenase (CAD; EC 1.1.l95) catalyzes the synthesis of cinnamyl alcohols from corresponding cinnamaldehydes. A cDNA clone, MdCADl, encoding putative CAD from apples (Malus domestica Borkh. cv Fuji) was characterized in this study. The clone contains an open reading frame of 325 amino acid residues, which shows a greater than 80% identity with Eucalyptus CADl. MdCADl mRNA was detectable in vegetative tissues and was strongly expressed in the fruit. The expression pattern of MdCADl mRNA in the fruit peel after light exposure was also examined. The mRNA was rapidly increased until 1 day after light exposure and remained stable thereafter, suggesting that MdCADl is light inducible. The inducibility of the MdCADl gene was examined using several environmental stresses. Mechanical wounding of leaves increased the MdCADl mRNA level and the induction was further increased by salicylic acid. Southern blot hybridization showed that there is either one or a few copies of CAD genes in apples. To our knowledge, it is believed that MdCADl is the first CAD clone expressed predominantly in fruit.

Antioxidant Activity and Anti-inflammatory Effects of Raw and Processed Fruits and Vegetables

  • In this study we investigated antioxidant and anti-inflammatory activities of Malus Domestica (apple), Pyrus Communis L. (pear), Daucus carota L. (carrot), Brassica oleracea var. (broccoli), Brassica oleracea var. capitata (cabbage) and Raphanus sativus L. (radish), that were obtained from local market. As these are common fruits and vegetables that are widely consumed, we aimed to investigate their beneficial properties especially the antioxidant and anti-inflammatory. The samples were processed by an indirect heating method and their properties were compared to their raw forms. Based on DPPH and ABTS assay, processed samples showed better antioxidant activity compared to raw samples, and processed pear sample had the best antioxidant activity. The anti-inflammatory activities of the samples were also investigated in LPS-treated RAW 264.7 cells. The mRNA expressions of pro-inflammatory mediators and cytokines (iNOS, COX-2, $TNF-{\alpha}$, $IL-1{\beta}$ and IL-6) were assessed by RT-PCR. Processed samples exhibited better inhibition of iNOS, compared to the raw forms. Processed broccoli and cabbage samples exhibited outstanding anti-inflammatory effects. The samples did not exhibit cytotoxicity against RAW 264.7 cells up to 1mg/ mL as shown in the cell viability assay. Taken together, processed broccoli and cabbage samples exhibited the strongest anti-inflammatory properties.

칼슘급원별 수관살포가 사과 과실의 칼슘축적에 미치는 영향 (Effects of Various Calcium Salt Spray on Calcium Accumulation into Apple Fruits (Malus domestica Borkh.))

  • 사과나무에 대한 칼슘용액의 수관살포 시 칼슘염의 종류에 따른 과실 내 칼슘함량증가효과를 구명하고 자 본 연구를 수행하였다. $CaCl_2$, $CaCO_3$, Ca$(CH_3COO)_2$ 및 Ca$(NO_3)_2$ 등의 칼슘염을 수관살포한 결과 "쓰가루" 품종에는 Ca$(NO_3)_2$, $CaCl_2$$CaCO_3$, 그리고 "후지" 품종에는 $CaCl_2$$CaCO_3$가 과실의 칼슘함량증가에 효과적이었다. 수관살포에 의하여 과실로 흡수된 칼슘은 과피와 과피직하의 과육에 국한되었다. Ca$(CH_3COO)_2$를 살포한 과실은 저장 중에 에틸렌 발생량이 감소하는 경향이었다.

기내 사과나무 조직의 신초 재분화에 미치는 생장조절물질의 영향 (Influence of Growth Regulators on Adventitious Shoot Regeneration from Tissues of Malus domestica cv. 'Gala' in vitro)

  • 'Gala' 잎조직의 신초 재분화에 효과적인 cytokinin 종류와 농도는 BA 4.0 ㎎/L였고, auxin 종류와 농도는 IAA, IBA, 및 NAA 각각 0.1 ㎎/L였다. BA와 NAA를 농도별로 조합 처리하여 'Gala'의 잎, 절간 및 엽병의 신초 재분화 정도를 비교한 결과, 잎조직은 0.5 ㎎/L NAA + 6.0 ㎎/L BA와 0.5㎎/L NAA + 0.8 ㎎/L BA 처리구에서, 절간조직은 0.1 ㎎/L NAA + 0.8 ㎎/L BA와 1.0 ㎎/L NAA + 0.8 ㎎/L BA 처리구에서 신초 재분화율이 가장 높게 나타났으며, 엽병조직은 0.1 ㎎/L NAA + 4.0 ㎎/L BA 처리구에서 가장 높았다.

First Report of an Apple Ring Rot Fungus Botryosphaeria kuwatsukai in Korea

  • Three fungal strains designated as KNUF-23-MG32, KNUF-23-YC8, and KNUF-23-MJ82 were isolated from the abnormal symptomatic apple trees during screening fungal pathogens collected in Jeollanam-do and Gyeongsangbuk-do, Korea. These fungal strains were found to have similar cultural and morphological characteristics close to the genus Botryosphaeria. Morphological characteristics were matched with B. kuwatsukai CBS 135219 but different with B. dothidea KACC 45481 and B. sinensis HMAS 246714T. Pathogenicity tests of strain KNUF-23-MG32 showed that this strain causes rot in Fuji apples. Molecular phylogenetic analysis based on the sequences of the internal transcribed spacer (ITS) regions, translation elongation factor 1-alpha (TEF1α), and partial β-tubulin (TUB2) showed that these strains are B. kuwatsukai. To the best of our knowledge, this is the first report of an apple ring rot fungus Botryosphaeria kuwatsukai in Korea.

Isolation and Identification of an Unreported Fungal Species in Korea and Novel Ice Nucleation Active Fungus: Fusarium diversisporum

  • In this study, the fungal strain KNUF-21-F39 was isolated from a declined apple tree (Malus domestica) in the Chungcheongbuk province in Korea. The strain KNUF-21-F39 presented a slow growth rate and a variety of macroconidia shapes and sizes ranging from ovoid to fusoid and 1- to 5-septate, primarily showing 3- and 4-septate, with "S" -shaped macroconidia rarely observed. The strain was identified based on morphological characteristics along with phylogenetic analysis performed using the internal transcribed spacer region (ITS) and partial sequences of translation elongation factor 1-α (tef1), RNA polymerase largest subunit (rpb1), and calmodulin (cal) genes. The fungal strain KNUF-21-F39 was identified as Fusarium diversisporum, which has not been previously reported in Korea. The ice nucleation activity (INA) of the strain was also evaluated, identifying the strain as positive for INA. This is the first report characterizing F. diversisporum as an IN-active fungal species.

Leaf Thinning and Fruit Quality of 'Hongro'/M.9 Apple Trees by Foliar Application of Cu-EDTA and Fe-EDTA

  • Hand leaf thinning for improvement of fruit coloration and quality is one of the most labor-consuming works in Korean apple cultivation. The study was aimed at investigating whether foliar application of two chelate compounds (Cu-EDTA, Fe-EDTA) could defoliate effectively 'Hongro'/M.9 apple leaves and enhance fruit coloration and quality. At 30 days before harvest, foliar spray with two chelate compounds defoliated significantly the apple leaves. Chelate treatments did not affect the leaf chlorophyll content. High concentration of chelates decreased drastically photosynthetic rate of true leaves, and then it stayed in low levels, whereas low levels of chelates reduced sharply the photosynthetic rate but it was gradually restored over time. Cu-EDTA regardless of its concentration triggered higher defoliation compared to Fe-EDTA. Consequently, higher defoliation improved light interception of fruits and accumulated more anthocyanin. Particularly, Cu-EDTA could target mainly fruit cluster leaves which affect directly light interception of the fruits, i.e. more selective defoliants compared to Fe-EDTA. However, 3% Cu-EDTA rather defoliated excessively, accompanying with reduction of fruit weight and soluble solids. Therefore, our results suggest that 1% Cu-EDTA which defoliates properly the fruit cluster leaves could improve coloration and quality of 'Hongro'/M.9 apple fruits.

Differences in Ethylene and Fruit Quality Attributes during Storage in New Apple Cultivars

  • Physiological characteristics of five new apple cultivars from the Korean apple breeding program were evaluated as a function of harvest time and storage after harvest. Internal ethylene concentration (IEC), flesh firmness, soluble solids concentration (SSC), and titratable acidity (TA) were measured in 'Summer Dream', 'Summer King', 'Green Ball', 'Picnic', and 'Hwangok' apples at harvest, during shelf life at $20^{\circ}C$, and one day after cold storage at $0.5^{\circ}C$ in air. IEC increased during shelf life in 'Summer Dream', 'Summer King', and 'Green Ball' but not in 'Picnic' or 'Hwangok', regardless of harvest time. Flesh firmness decreased towards harvest time and decreased gradually with time in cold storage only in the former three cultivars. In turn, IEC increased during cold storage in the first three cultivars but not for the last two cultivars, irrespective of harvest time. Changes in SSC and TA did not consistently relate to harvest time or storage period but TA tended to decrease as IEC increased. Furthermore, IEC was negatively correlated with flesh firmness except in the 'Green Ball' cultivar but the significance level was much greater in 'Summer Dream' and 'Summer King' (p < 0.0001) than in 'Picnic' (p < 0.01) or 'Hwangok' (p < 0.05) cultivars. Flesh firmness was positively correlated with TA in the first three cultivars but not in the last two cultivars. Overall, the results indicate that cultivars for which IEC increased after harvest had reduced flesh firmness and TA after storage.

충주지역에서의 사과 후지 품종의 만개기 추정 (Estimation of Full Bloom Stage of Apple (Malus domestica Borkh. c. Fugi) in Chungju Area)

  • 충주지역에서 표고가 다른 9개 지점(계명산 5지점, 남산 4지점)에서 측정한 기온 관측자료를 분석하여 기온이 표고가 높아짐에 따라 일정한 비율로 변화하는 단열조건에 의한 기온체감율을 계산하고, 표고별 후지 품종의 만개기를 추정하였다. 충주 계명산 남서사면에서 측정한 시간별 기온자료를 표고와의 관계로부터 표고가 100 m 상승함에 따라 기온은 0.89$^{\circ}C$의 일정한 비율로 체감하였다. 남산 북서사면에서 측정한 시간별 기온자료와 표고와의 상관으로부터는 0.74$^{\circ}C$/100 m의 비율로 기온이 체감함을 알 수 있었다. 충주지역에서 일평균 기온 5$^{\circ}C$ 이상인 날의 발육속도(DVR)를 계산하여 추정한 만개기와 실제로 조사된 만개기는 서로 일치하는 경향이었으며 표고별로 추정된 충주지역의 사과 만개기는 해발고도 100 m 상승함에 따라 4~5일 늦어졌는데 고도에 따라 4월 25~5월 10일의 범위이었다.

추석용(秋夕用) 고품질 사과 '새나라' 육성(育成) ('Saenara', a New Chuseok Season Apple Cultivar)

  • '새나라' 품종은 농촌진흥청 원예연구소에서 1997년에 육성한 중생종으로 착색과 품질이 우수하고 풍산성이다. 이 품종은 1981년도에 'Spur EarliBlaze'에 'Spur Golden Delicious'를 교배하여 육성한 것으로 1992년도에 1차 선발되어 '원교 가-08호'로 명명되었고 1993년부터 1997년까지 5년 동안 7개 지역에서 지역적응성시험을 거쳐 1997년에 '새나라'로 최종 선발 및 명명되었다. 수세는 중정도이고 수자는 개장성이며 스퍼 타입이다. 개화기는 'Spur Golden Delicious'보다 하루 빠르고 'Spur EarliBlaze'보다 3일 늦다. 수원지역에서의 숙기는 만개기로부터 140일 후인 9월 10일경이고 과형은 원추형이며 과피색은 선홍색이다. 과중은 300g 전후이고 당도는 $14.6^{\circ}Brix $이며 점무늬낙엽병에 약하다.
