실험적으로 유발된 직장종양에 대한 치료적 초음파의 효과 (Effects of Therapeutic Ultrasound on Experimental Induced Rectal Sarcoma(CT-26))

  • The use of therapeutic ultrasound(US) in humans with malignant neoplasms has been contra-indicated in physical therapy practice. Some studies have shown the results after application of US inhibited of tumor growth but some studies have shown the results facilitated of tumor growth in mouse. The purpose of this study were to determine the effects of US on rectal sarcoma(CT-26) in mouse and to determine the histological change of tumor. Thirty-five female BALB/C mouse, age 6 to 8 weeks received subcutaneous injection of 0.1 105 tumor cells. When tumors grew to 5 mm in diameters, the mouse were randomly assigned to control group(n=7) and high powered continuous US group(n=7) and low powered continuous US group(n=7) and high powered pulsed US group(n=7) and low powered pulsed US group(n=7). The experimental group (four groups) received 10 treatments over a 10-day period of 3 MHz ultrasound. Tumor dimension were measured on days 1(start of treatment), 5(midtreatment), and 10(end of treatment, preexcision and postexcision). Tumors were weighed after excision and the mouse were observated histological change of tumor. All tumors grew larger over time. Mean tumor weights(in grams) and volumes(in cubic millimeters) were 2.063 g and $2729.313\;mm^3$ for the high powered continuous US group 1.881 g and $2428.002\;mm^3$ for the low powered continuous US group 1.730 g and $2381.002\;mm^3$ for the high powered pulsed US 1.673 g and $2289.562\;mm^3$ for the low powered pulsed US group 1.670 g and $2297.333\;mm^3$ for the control group. Ultrasound increased the weight and volume of subcutaneous tumor in mouse. We urge caution in the use of ultrasound in the areas of tumors.

경골 간부에 발생한 투명세포연골육종: 증례 보고 (Clear Cell Chondrosarcoma of the Tibia Diaphysis: A Case Report)

  • 투명세포연골육종은 매우 드문 골종양으로 장골의 골단이나 골간단에 발생한다. 많은 연구에서 대퇴골의 근위부를 가장 흔하게 발생하는 부위로 보고하였고, 상완골의 근위부가 그 다음으로많이 보고되었다. 조직학적으로 이 종류의 종양 세포들은 투명한 세포질과 뚜렷한 세포질막과 함께 중심성으로 위치하는 둥근 핵소체를 가지고 있다. 일반적으로 투명세포연골육종은 고식적인 연골육종과 혼동되지 않는다. 하지만, 방사선학적으로 장골의 간부에 발생한 경우 진단이 어려우며, 이전 문헌을 통하여 단 3례만이 보고된 바 있다. 저자들은 이전에 보고된 바 없는 42세 남자 환자의 경골 간부에서 발생된 투명세포연골육종을 보고하고자 한다.

  • Burkitt림프종은 중앙아프리카 지역의 어린이 악골에서 흔하게 나타나는 악성 종양으로 림프조직으로부터 발생되며 매우 빠른 성장 양상을 보인다. 임상적으로는 3~8세 아동에 주로 호발하며 악골에 발생하는 경우 안면부 종창이 나타날 수 있으며 유치의 비정상적 동요 및 구치부 조기 맹출등이 나타날 수 있다. 방사선 소견으로는 경계가 불규칙한 방사선투과성 병소로 나타나며 조직학적으로는 종양세포들 사이에 대식세포가 분포되어 "starry-sky" appearance의 특징적인 양상이 관찰된다. 본 증례는 좌측 안면부 종창을 동반한 유구치의 심한 동요도를 주소로 내원한 3세 남환아에서 임상 및 방사선 검사와 조직학적 검사를 통해 Burkitt림프종으로 진단하였고 항암 화학요법에 의해 치료중이며 양호한 치료 결과를 얻고 있다.

  • Bronchial carcinoid tumors are uncommon, constituting approximately 3-5% of all primary lung cancers. Classification of these tumors has evolved substantially as our understanding of the cellular, biologic, and clinical aspects of these neoplasms has improved. Initially, bronchial carcinoids were thought to be benign and therefore were classified as bronchial adenomas. Currently, however, they are well recognized as having the potential for both local invasion and distant metastatic involvement. Consequently, carcinoid tumors are frankly malignant. Thus bronchial adenoma is a misnomer that should no longer be used for bronchial carcinoids. Most investigators currently favor classifying carcinoid tumors as a type of neuroendocrine neoplasm because of their potential to secrete a variety of chemical substances found in both the central nervous system and the epithelial cells of numerous organs. Bronchial carcinoids are usually characterized by a slow growth pattern and a low incidence of metastasis, and histologically conformed by the azurophil staining and the presence of the characteristic neurosecretary granule on electron microscopy. Atypical carcinoid tumor was first defined by Arrigoni et al, who proposed the following criteria for separation of atypical carcinoid from typical carcinoid tumor : 1) increased mitotic activity with 1 mitotic figure per 1-2 high power fields(or 5-10 mitoses /10 HPF), 2) nuclear pleomorphism, hyperchromatism, and an abnormal nuclear-cytoplasmic ratio, 3) areas of increased cellularity with disorganization of the architecture, and 4) tumor necrosis. In contrast, typical carcinoid tumor may have focal cytologic pleomorphism, but necrosis is absent and mitotic figures are rare. Recently we experienced a case of atypical bronchial carcinoid with multiple distant metastasis, so we report this case with a review of the literature.

Mutational Analysis of Extranodal NK/T-Cell Lymphoma Using Targeted Sequencing with a Comprehensive Cancer Panel

  • Extranodal natural killer (NK)/T-cell lymphoma, nasal type (NKTCL), is a malignant disorder of cytotoxic lymphocytes of NK or T cells. It is an aggressive neoplasm with a very poor prognosis. Although extranodal NKTCL reportedly has a strong association with Epstein-Barr virus, the molecular pathogenesis of NKTCL has been unexplored. The recent technological advancements in next-generation sequencing (NGS) have made DNA sequencing cost- and time-effective, with more reliable results. Using the Ion Proton Comprehensive Cancer Panel, we sequenced 409 cancer-related genes to identify somatic mutations in five NKTCL tissue samples. The sequencing analysis detected 25 mutations in 21 genes. Among them, KMT2D, a histone modification-related gene, was the most frequently mutated gene (four of the five cases). This result was consistent with recent NGS studies that have suggested KMT2D as a novel driver gene in NKTCL. Mutations were also found in ARID1A, a chromatin remodeling gene, and TP53, which also recurred in recent NGS studies. We also found mutations in 18 novel candidate genes, with molecular functions that were potentially implicated in cancer development. We suggest that these genes may result in multiple oncogenic events and may be used as potential bio-markers of NKTCL in the future.

TOMM20 as a potential therapeutic target of colorectal cancer

  • Translocase of outer mitochondrial membrane 20 (TOMM20) plays an essential role as a receptor for proteins targeted to mitochondria. TOMM20 was shown to be overexpressed in various cancers. However, the oncological function and therapeutic potential for TOMM20 in cancer remains largely unexplored. The purpose of this study was to elucidate the underlying molecular mechanism of TOMM20's contribution to tumorigenesis and to explore the possibility of its therapeutic potential using colorectal cancer as a model. The results show that TOMM20 overexpression resulted in an increase in cell proliferation, migration, and invasion of colorectal cancer (CRC) cells, while siRNA-mediated inhibition of TOMM20 resulted in significant decreases in cell proliferation, migration, and invasion. TOMM20 expression directly impacted the mitochondrial function including ATP production and maintenance of membrane potential, which contributed to tumorigenic cellular activities including regulation of S phase cell cycle and apoptosis. TOMM20 was overexpressed in CRC compared to the normal tissues and increased expression of TOMM20 to be associated with malignant characteristics including a higher number of lymph nodes and perineural invasion in CRC. Notably, knockdown of TOMM20 in the xenograft mouse model resulted in a significant reduction of tumor growth. This is the first report demonstrating a relationship between TOMM20 and tumorigenesis in colorectal cancer and providing promising evidence for the potential for TOMM20 to serve as a new therapeutic target of colorectal cancer.

Anti-Cancer Effect of Ginsenoside F2 against Glioblastoma Multiforme in Xenograft Model in SD Rats

  • The glioblastoma multiforme (GBM) is the most common malignant brain tumor in adults. Despite combination treatments of radiation and chemotherapy, the survival periods are very short. Therefore, this study was conducted to assess the potential of ginsenoside $F_2$ (F2) to treat GBM. In in vitro experiments with glioblastoma cells U373MG, F2 showed the cytotoxic effect with $IC_{50}$ of 50 ${\mu}g/mL$ through apoptosis, confirmed by DNA condensation and fragmentation. The cell population of cell cycle sub-G1 as indicative of apoptosis was also increased. In xenograft model in SD rats, F2 at dosage of 35 mg/kg weight was intravenously injected every two days. This reduced the tumor growth in magnetic resonance imaging images. The immunohistochemistry revealed that the anticancer activity might be mediated through inhibition of proliferation judged by Ki67 and apoptosis induced by activation of caspase-3 and -8. And the lowered expression of CD31 showed the reduction in blood vessel densities. The expression of matrix metalloproteinase-9 for invasion of cancer was also inhibited. The cell populations with cancer stem cell markers of CD133 and nestin were reduced. The results of this study suggested that F2 could be a new potential chemotherapeutic drug for GBM treatment by inhibiting the growth and invasion of cancer.

Biphasic Effects of Kaempferol on the Estrogenicity in Human Breast Cancer Cells

  • Dietary flavonoids have attracted a great deal of attention as agents for preventing estrogen-related diseases, such as postmenopausal symptoms, and for reducing the risk of estrogen-dependent cancer. Kaempferol is one of the most commonly found dietary phytoestrogen. The aim of this study was to investigate the estrogenic and/or antiestrogenic effect of kaempferol, which can confirm its potency as a preventive agent against estrogen-related diseases. Kaempferol has both estrogenic and antiestrogenic activity, which are biphasic response on estrogen receptor. The estrogenic activity of kaempferol induced via ER-mediated pathway depending on $E_2$ concentration $(\leq\;10^{-12}M)$. Kaempferol $(10^{-5}\;M)$ also caused antiproliferative effect on MCF-7 cell in the presence of $E_2\;(10^{-11}\;M)$ and restored to the addition of excess $E_2\;(10^{-7}\;M)$, which confirms that antiproliferation of kaempferol was induced via ER-dependent pathway. However, at $10^{-4}\;M$, concentration higher than the concentrations at which the estrogenic effects of kaempferol are detected $(10^{-5}\;M)$, kaempferol induced strong antiproliferative effect, but were unaffected by the addition of excess $E_2\;(10^{-7}\;M)$ indicating that kaempferol exerts antiproliferation via ER-independent pathway. In particular, kaempferol blocked the focus formation induced by $E_2$, which confirms that kaempferol might inhibit the malignant transformation caused by estrogens. Therefore, we suggested that kaempferol might regulate a suitable level of estrogenic activity in the body and is expected to have potential beneficial effects in preventing estrogen imbalance diseases (breast cancer, osteoporosis, cardiovascular disease and etc.).

The Processed Radish Extract Melanogenesis in Humans and Induces Anti-Photoaging Effects in Ultraviolet B-Induced Hairless Mouse Model

  • The radish skin and radish greens are an edible part of the radish. But they are removed before eating the radish and used as a byproduct or an animal feed material because of their tough and rough texture. Melanin is a pigment that gives colour to our skin. But increased production of melanin can turn into benign or malignant tumours. These days due to global warming, the amount of Ultra violet (UVB) rays has been extensively increased with sunlight. Due to this, a phenomenon called exogenous photo aging is widely observed for all skin colour and types. As a result of this phenomenon, a set of enzymes called matrix metalloproteinases (MMP's) that serves as degradation enzymes for extracellular matrix proteins mainly collagen is increased, causing depletion in collagen and resulting in early wrinkles formation. Therefore in our study we used the murine melanoma cell line B16/F10 to study the melanogenesis inhibition by Heated radish extract (HRE) in vitro and we used HRM-2 hair less mice exposed to artificial UVB for checking the efficacy of Heated radish extract in vivo. Furthermore, we prepared a 3% Heated radish extract (HRE) cream and checked its effects on human skin. Our results have clearly demonstrated that Heated radish extract (HRE) have potently suppressed the tyrosinase activity and melanin production in B16/F10 cells. It had also reduced the expression of components involved in melanin production pathway both transcriptionally and transitionally. In in vivo studies, HRE had potently suppressed the expression of MMP's and reduced the wrinkle formation and inhibited collagen degradation. Moreover, on human skin, ginseng cream increased the resilience, skin moisture and enhanced the skin tone. Therefore in light of these findings, we conclude that HRE is an excellent skin whitening and antiaging product.
