The Effects of Korean Safflower(Carthamus tinctorious L.) Seed Powder Supplementation Diet on Bone Metabolism Indices in Rats during the Recovery of Rib Fracture (한국산 홍화씨분말 보충식이의 급여가 골절된 흰쥐의 골대사지표에 미치는 영향)

  • This study was carried out to investigate the effects of Korean safflower(Carthamus. tinctorious L.) seed powder supplementation on bone metabolism during the recovery of rib-fracture induced by surgical operation in rats. Male Sprague-Dawley rats of 10 weeks old, weighing 370$\pm$5g, were divided into two groups after arrival : the control group(C group, AIN-76 semifurified diet) and safflower seed group(S group, AIN-76 semifurified diet+10% safflower seed powder). They were fed experimental diets for 12 days before the rib fracture operation and for 30 more days after the operation. A number 9 rib was fractured surgically and sham-operation was also performed. After the rib-fracture operation, rats were sacrificed on the 8th, 11th, 16th, 21st, and 30th day, respectively. The body weights were decreased on the 8th day after operation in both groups but recovered gradually thereafter. The fractured sections of the S group were more strongly adhered and more rapidly repaired than those of C group. Effects of safflower seed powder supplementation on indices of bone metabolism during the repairing of rib-fracture can be summarized as following. There were no differences between C and S group. In the levels of PTH and calcitonin, which regulate the concentration of plasma calcium. However, PTH level on the 8th day was significantly higher than that on the 16th day in control group. There were also no differences between groups or within groups in the levels of calcitonin. Osteocalcin levels, one of the bone-formation indices, were similar in two groups. Activities of total-ALP in plasma were highest at the 8th day in both groups and decreased thereafter, except on the 30th day in control group. The activities of bone-specific ALP that is directly related to bone calcification, showed higher levels in the S group than in the C group throughout the experimental period. They were significantly increased in the S group on the 11th, 16th, and 21st days. In comparisons of plasma calcium and phosphate levels, Ca levels were tended to be lower in the S group than in the C group and P levels showed the opposite trend. It seems that the fractured ribs were more rapidly repaired in rats supplemented with safflower seed powder than in control rats, possibly due to stimulation of bone calcification from increased activity of bone-specific ALP. Fatty Acid Composition of Serum and Liver in Mice and Sancho(Zanthoxylum schinifolium) Seed Oil (산초유의 지방산 조성과 마우스 혈청 및 간장의 지방산 조성변화)

  • The major fatty acids in the Zanthoxylum schinifolium seed oil were eicosenoic acid 30.88%, oleic acid 29.94%, linoleic acid 23.55% and palmitic acid 10.52%. Fatty acid profiles in the each lipid fractions by TLC of the Z. schinifolium seed oil showed the highest composition of eicosenoic acid in triglyceride fraction and oleic acid in other fractions. Mice (ddY male strain) being starved for 24 hr were injected into gastric directly 500 mg of the Z. schinifolium seed oil, and then blood samples were obtained 0, 3 and 6 hr after dosing. In our results, eicosenoic acid appeared to be significantly increased in the serum obtained from 3 and 6 hr after injection of the Z. schinifolium seed oil. In the control mice, however, the serum samples did not exhibited any change of the Z. schinifolium seed oil. Interestingly, eicosenoic acid was significantly increased in the liver of 6 hr mice after injection. In conclusion, eicosenoic acid was the major fatty acid in the Z. schinifolium seed oil, and this fatty acid was significantly increased in the serum obtained 3 and 6 hr after injection in mice.

  • Two types of sperm, apyrene and eupyrene, are identified in A. mylitta. The sperm in the adult moth are motionless in seminal vesicles. At the time of ejaculation they received a secretion from male ejaculatory duct that renders them motile. The dissociation of eupyrene bundles, eupyrene sperm motility and the sequence of events of sperm migration in both sexes are described in the present paper.

Effect of Planting Ratio of Male Sterility (MS) and Restorer Line (RL) on Fatty Acid Content and Composition during Seed Filling Period in $F_1$ Seed of Brassica napus L. (유채의 $F_1$ 종자 생산시 종자친과 화분친의 재식비 및 등숙시기별 종자의 지방산 분석)

  • The objective of this experiment was determined to investigate the effect of the planting ratio of Male Sterility (MS) to Restorer Line (RL) and harvesting time on fatty acid compositions under $F_1$ seed production of Brassica napus L. For rapeseed seed production, two experiments were conducted in the open fields. One experiment studied planting ratios of MS to RL (4:2, 10:2, or 10:1) were planted and investigated fatty acid composition at 40, 45, 50, 55, and 60 days after flowering, the other $F_2$ seeds were analyzed on fatty acid compositions of harvested seeds at five sequential stages. The results showed that fatty acid compositions of developing seeds were influenced by MS:RL planting ratios and $F_2$ hybrid treatments and contaminated level of fatty acid compositions, erucic acid, were unaffected by planting ratio of MS to RL. Fatty acid compositions such as palmitic acid (C16:0), stearic acid (C18:0) and linoleic acid (C18:2) contents decreased during seed maturation period in $1^{st}$ and $2^{nd}$ experiments. In contrast, oleic acid (C18:1) content relatively increased up to 55days after flowering. At day 60 after flowering, oleic acid content was unaffected by MS:RL planting ratios and $F_2$ seeds treatments. Aspects of related gene expression of fatty acid synthesis such as SAD, FAD1 and FAD2 were followed exactly to changes of fatty acid composition during seed maturation. These results suggest that MS ratio may be enlarged and RL may be reduced, indicating this ratio will be useful for rape seed production.

  • Co-segregation of male fertility with DNA markers selected by RAPD analysis as being potentially linked to the restorer gene (Rf) for Cytoplasmic male sterility (CMS) was analyzed using segregating F2 population. One RAPD marker directly linked to the Rf locus was identified. Amplification of OPT-02/570 using the STS primers generated a monomorphic band of each fertile plants randomly selected F2 progenies. From these results, this specific marker would be strongly linked to be restoring gene. The use of STS marker is effective in overcoming the reliability of the RAPD phenotype and improving their utility for MAS, co-dominant STS markers are especially very useful.

  • Callosobruchus chinensis (Coleoptera: Bruchidae) is the main pest of mungbean (Vigna radiata) especially during seed storage. To provide background information for chemical ecology studies, we examined the externalmorphology of the antennal, maxillary palp and labial palp sensilla of this insect with scanning electron microscopy. The antennae of adult C. chinensis exhibited sexual dimorphism. The female have serrated antenna and the male have pectinate morphology. Eight sensillum types have been observed on the antennae of both sexes: $B\ddot{o}hm$ bristles, sensilla chaetica, sensilla trichodea, sensilla basiconica, sensilla styloconicum, sensilla coeloconica, sensillum campaniformia, sensilla cavity. There are no differences on the morphology of maxillary palp and labial palp between female and male. Four different types of sensilla are present on the labial and maxillary palps of both sexes of C. chinensis: sensilla chaetica, sensilla twig basiconica, sensilla basiconica, sensilla digitiformia.

Seed Production by Induction of Male Flowers on Female Plants of Hemp(Cannabis saliva L.) (대마 자주의 웅화 유도에 의한 자성종자 생산에 관한 연구)

  • This study was conducted to overcome the deliciousness by sex reversal in hemp plants. When female plants grown under long day length condition were transferred to short day length condition in 3th and 6th leaf stage, a few male flowers were formed and the gap of flowering days between sex reversed male and normal female flowers were around 10 days and 3 days, respectively. Although the flowering amounts were small, this short gaps of flowering days were enough for artificial pollination. When female plants grown under long day length condition were transferred to short day length condition just after treatment of silver nitrate in the vegetative stage, a great amount of male flower was formed and the gap of flowering days between sex reversed male and normal female flowers were only 1 to 3 days. The increased sex reversed male flowers and induced short gap of flowering days were adequate for natural wind pollination. When female seeds were cultivated, seed yield was 141 kg/10a, while it was 96 kg/10a when normal seeds cultivated.
