Functional Silk Proteins: Molecular Structure and Application to Biomaterials

  • Silk proteins consist of two major proteins, fibroin and sericin. There is currently an enormous reawakening of interest in these silk proteins as a biomaterial due to their mechanical and biological properties based on the detailed findings. Novel method for determination of the crystalline structure of silk proteins in an atomic level using nuclear magnetic resonance (NMR) was reviewed. Recent application of silks to biomaterials and prospects for future were discussed.

  • Morphological observation of a unialgal culture of Lingulodinium polyedrum was conducted under light- and scanning electron microscopes. Lingulodinium polyedrum was redefined as having the thecal formula of APC, Q, 5', 6", 6c, 6s, 5'", 2"" using the reinterpreted tabulation system. In our tabulation system, the Q, which is equivalent to the 3' in the strict Kofoidean system, was treated as an auxiliary platelet because of its considerable morphological variations in culture specimens. Subsequently, apical plates were redefined as plates directly connected to the APC and/or the Q. The 1"' and the 1p in the conventional Kofoidean system were treated as a component of the sulcus (the Ssa) and as an antapical plate (the 1""), respectively. Our reinterpreted tabulation system clearly differentiated extant gonyaulacalean taxa.

N-Acetylglycine Side Chain is Critical for the Antimicrobial Activity of Xanthostatin

  • This study was carried out to elucidate the mode of bacteriostatic property of xanthostatin (XS), a novel depsipeptide antibiotic with an N-acetylglycine side chain and selective antimicrobial activity against Xanthomonas spp. Two biotransformed XSs were isolated by the treatment of XS with the cell lysate of Xanthomonas campestris pv. citri, a solvent partition, preparative TLC, and HPLC. Structure determination of those two biotransformed XSs demonstrated deletion of the N-acetylglycine side chain. Noteworthily, they showed no antimicrobial activity against Xanthomonas spp. This result suggests that the N-acetylglycine side chain plays a critical role in the antimicrobial activity of XS, and that the bacteriostatic property of XS is due to susceptibility of the ester bond between the hexadepsipeptide nucleus and the N-acetylglycine side chain to hydrolytic enzyme(s) produced by Xanthomonas spp.

  • Initial imperfections, such as initial deflection or remaining stress, cause deterioration of buckling strength of structures. The Koiter imperfection sensitivity law has been extended to describe the mechanism of reduction for structures. The extension is twofold: (1) a number of imperfections are considered, and (2) the second order (minor) imperfections are implemented, in addition to the first order (major) imperfections considered in the Koiter law. Yet, in reality, the variation of external loads is dominant over that of imperfection. In this research, probabilistic evaluation of buckling loads against external loads subjected to probabilistic variation is conducted by extending the concept of imperfection sensitivity. A truss arch subjected to dead and live loads is considered as a numerical example. The mechanism of probabilistic variation of buckling strength of this arch is described by the proposed method, and its reliability is evaluated.

  • The effects on birch (Betula spp.) of elevated carbon dioxide ($CO_2$) and ozone ($O_3$), which are both increasing in the troposphere, are surveyed in detail based on the literature. Birches establish themselves in the open field after disturbances, and then become dominant trees in temperate or boreal forests. Ecophysiological approaches include the measurement of photosynthesis, biomass, growth, and survival of seedlings and trees. Elevated $CO_2$ levels give rise to a net enhancement of the growth of birch trees, whereas high $O_3$ generally reduces growth. Although the effects of the two are opposed, there is also an interactive effect. Basic physiological responses of the single genus Betula to $CO_2$ and $O_3$ are set out, and some data are summarized regarding ecological interactions between trees, or between trees and other organisms.

  • A problem of the surface friction is inevitable in the micro machines. However, the levitation by Meissner effect of high Tc superconductor gives a complete solution to the problem. The repulsive force between permanent magnets and superconductors is utilized to levitate and actuate micro structures without the surface friction. In this paper, a micro actuator using; Meissner effect of high Tc superconducting film (Meissnac) is proposed ; Meissnac is drived by the control of (Sugerconducting/ Normal) states of superconductor. The levitating force and the driving force are analyzed by the numerical method.

  • City gas is one of the most important necessities of daily city life and social infrastructures. City gas is delivered to every user through a pipeline network. The gas pressure in the pipeline is regulated by gas regulator. In the pressure control system, characteristics of gas pipeline is as important as characteristics of regulator. There are many reports about the transfer function model of the fluid pipeline. But suitable model about the gas transmission pipeline is not known. In this paper, as the transfer function model of the gas pipeline, new model considering the heat transfer between pipe wall and gas and temperature change of gas is proposed. To evaluate this model, frequency response tests are used. As the result, the proposed model shows a better agreement when compared with the experimental result than conventional models. The results show the effectiveness of the model.

  • In this paper, we fabricated drag force type and pressure difference type gas flow sensor with dry etching technology which used Deep RIE(reactive ion etching) and etching stop technology which used SOI(silicon-on-insulator). we fabricated four kinds of sensor, which are cantilever, paddle type, diaphragm, and diaphragm with orifice type. Both cantilever and paddle type flow sensors have similar sensitivity as 0.03mV/V kPa. Sensitivity of the fabricated diaphragm and diaphragm with orifice type sensor were relatively high as about 3.5mV/V kPa, 1.5mV/V kPa respectively.
