담배나방 (Helicoverpa assulta)의 발생중 Major Haemolymph Protein(MHP)의 변화 및 특성 (Ontogeny and Characterization of Major Haemolymph Protein(MHP) in Helicoverpa assulta)

  • 담배나방 (Helicoverpa assulta)의 발생기간 중 존재하는 major haemolyruph protein (MHP)을 확인하였으며, 그 발생에 따른 변화상 및 물리화학적 특성을 조사하였다. MHP는 유충-용-성충의 발생단계에서 확인되었으며, 그 함량은 유충의 성장에 따라 증가하여 용기와 성충기 동안에도 높게 유지되고 용초기와 성충 초기에서 일시적인 감소를 보여, 총 혈림프 단백질의 농도 변화와 유사한 변화양상을 보였다. ammonium sulfate precipitation, gel filtration 그리고 ion exchange chromatography를 통해 정제한 MHP의 전기영동 분석 결과, 단일 구성소 단위 (69 kDa)가 hexamer를 이룬 분자량 414kDa의 glycolipoprotein (pI 5.9)인 것으로 확인되었다. MHP의 아미노산 조성에 있어서 18.27 mole % 의 tryptophan, 7.47 mole % 의 tyrosine and 6.51 mole % 의 phenylalanine이 검출되어 다른 곤충류의 저단백질들에 비해 비교적 높은 aromatic amino acid의함량을 보였다. 지방체, 장, 말피기관 단백질의 전기영동 및 이들 단백질과 anti-MHP간의 immunodiffusion test 결과 MHP는 지방체에 존재하고 있음이 확인되었다.

  • HVDC is better economic method than HVAC in case of long distance transmission and it is possible to interconnect transmission lines regardless of difference of power frequency. The electrical environment problems of HVDC overhead transmission line are electric field, charged voltage, ion current and so on. For biopolar HVDC lines, most of the ions are directed toward the opposite polarity conductor, but a significant fraction is also directed toward the ground. These problems are major factor to design configuration of HVDC overhead transmission line. Therefore, It is necessary to test an environmental characteristics of HVDC overhead transmission line. In this paper, to assess the ion characteristic of HVDC transmission line, continuous measurements have been done on the biopolar single circuit line with ACSR 480mm2-6 bundle conductors of Gochang HVDC Test line. And then the ion characteristics were analyzed.

Depolarizing Effectors of Bradykinin Signaling in Nociceptor Excitation in Pain Perception

  • Inflammation is one of the main causes of pathologic pain. Knowledge of the molecular links between inflammatory signals and pain-mediating neuronal signals is essential for understanding the mechanisms behind pain exacerbation. Some inflammatory mediators directly modulate the excitability of pain-mediating neurons by contacting the receptor molecules expressed in those neurons. For decades, many discoveries have accumulated regarding intraneuronal signals from receptor activation through electrical depolarization for bradykinin, a major inflammatory mediator that is able to both excite and sensitize pain-mediating nociceptor neurons. Here, we focus on the final effectors of depolarization, the neuronal ion channels, whose functionalities are specifically affected by bradykinin stimulation. Particular G-protein coupled signaling cascades specialized for each specific depolarizer ion channels are summarized. Some of these ion channels not only serve as downstream effectors but also play critical roles in relaying specific pain modalities such as thermal or mechanical pain. Accordingly, specific pain phenotypes altered by bradykinin stimulation are also discussed. Some members of the effector ion channels are both activated and sensitized by bradykinin-induced neuronal signaling, while others only sensitized or inhibited, which are also introduced. The present overview of the effect of bradykinin on nociceptor neuronal excitability at the molecular level may contribute to better understanding of an important aspect of inflammatory pain and help future design of further research on the components involved and pain modulating strategies.

Experimental Study on the Interference of Water Vapor on the Chemical Ionization of OH by Sulfur Hexafluoride Ion

  • The interference of water vapor on the chemical ionization (CI) of hydroxyl radicals (OH) by sulfur hexafluoride ion ($SF_6{^-}$) was investigated using a flow tube system coupled to a high-pressure CI mass spectrometer. Water vapor, which is required to study heterogeneous reactions of OH under real tropospheric conditions, transforms the reagent ion $SF_6{^-}$ into $SF_4O^-$ and $F^-(HF)_n$, resulting in a substantial loss in CI sensitivity. Therefore, under humid conditions, peaks corresponding to OH are drastically diminished, while those corresponding to OH-water complex ions ($[OH(H_2O)_n]^-$) are enhanced. $[OH(H_2O)_3]^-$ was observed as the major OH species. The obsercation of $[OH(H_2O)_n]^-$ by isolating humid conditions to the CI region and preliminary ab initio calculations suggested that $[OH(H_2O)_n]^-$ ions were produced from reactions between OH ions ($OH^-$) and water molecules. An additional helium buffer flow introduced into the CI region reduced loss of the reagent ion and resulted in a partial recovery of OH peak intensities under humid conditions.

Research on Preparation of Sheath-Core Bicomponent Composite Ion Exchange Fibers and Absorption Properties to Metal Ion

  • Based on the sheath-core bicomponent composite fibers with modified polystyrene (PS) and the modified polypropylene (PP), composite fibers obtained were further cross-linked and sulphonated with chlorosulphonic acid to produce strong acidic cation ion exchange fibers. The structures of the fibers obtained were characterized using Fourier transform infrared (FT-IR) spectroscopy, differential scanning calorimetry (DSC) etc. The optimal technology of the fibers obtained is discussed. The static absorption capacity of the sheath-core bicomponent composite cation exchange fibers for $Zn^{2+}$, $Cu^{2+}$ was determined. The absorption kinetics and major factors affecting the absorption capacities of $Zn^{2+}$, $Cu^{2+}$ were studied, and its chemical stability and regenerating properties were probed. The results suggest that cation exchange fibers with better mechanical properties and higher exchange capability were obtained. Moreover, this type of ion exchange fiber has good absorption properties and working stability to various metal ions. Hence, they have higher practicability.

적조생물, Cochlodinium polykrikoides와 Gymnodinium sanguieum의 사멸에 있어 암모니아염의 효과 (The Effects of Ammonium Ion and Salts on the Killing of Red Tides Organism; Cochlodinium polykrikoides and Gymnodinium sanguieum)

  • Cell-free culture broth of marine halophilic bacterium, Kordia algicida was shown to possess specific algicidal ability against red tide organism, Cochlodinium polykrikides. Physiochemical characteristics of algicidal material originated in the bacterial culture broth were analyzed that its molecular weight was estimated to a 3,000 dalton and it was stable in heat and pH treatment. The algicidal fraction against C. polykrikoides obtained from gel permeable chromatography contained high concentration of ammonium ion as analyzed by ICP/Mass spectrum. C. polykrikoides by the fraction was quickly lysed within 1 min. It was shown that the effective concentration for algicide against C. polykrikoides was over 1mM of ammonium chloride. On the other hand, other metal ions presented in the algicidal fraction showed no algicidal effect against C. polykrikoides. In additon, ammonium ion exhibited species-specific killing spectrum for two species of red tide organisms, C. polykrikoides and Gymnodinium sanguieum. Therefore, further researches on the killing mechanism against C. polykrikoides exerted by ammonium ion, and subsequent development of replaceable algicidal materials will perform to provide useful tools for the control of red tide.

제주도 동부지역 지하수의 염수화에 관한 연구 (Study on The Salinization in Groundwater of the Eastern Area of Cheju Island)

  • According to the results of the groundwater quality investigation about 230 holes all over the country, the groundwater which was in excess of standard grows larger every year and closed holes increased to 23,457 holes in 1997 from 15,724 holes in 1996. This is the major reasons that water quality contamination, shortage of water quantity, increasing of salinity and so on. There are 7 groundwater salinization sources which are condisered as most important on a regional level. And among theses the Cheju Island groundwater salinization sources are (1) halite solution, (2) natural saline groundwater, (3) sea-water intrusion. The method of taking an isotopes is one of research methods of the origin of groundwater salinization and is used in so many studies because it has very high confidence. $^{18O}O, ^2H, ^3H, ^{14}C$ and so on in an isotopes are frequently used in the method of them. Consequently on this study we analyzed major ions and $^3H$ in groundwater, sea-water and rain of the eastern part of cheju island known as contaminated site from long time ago to examine the origin of groundwater salinization. Relation ratios of the major ions versus chloride ion shows similar tendency to sea-water. This indicates that sea-water entered the groundwater layer. And amount of $^3H$ in holes of the land side is higher than of the sea side. Relation of chloride ion versus $^3$H indicates negative character. Therefore we can think that the reason of groundwater salinization of this part is natural saline groundwater and halite solution by relation.

Qualitative Analysis of the Major Constituents in Traditional Oriental Prescription Bang-poong-tong-sung-san by Liquid Chromatography/Ultraviolet Detector/Ion-Trap Time-of-Flight Mass Spectrometry

  • An advanced and reliable high performance liquid chromatography (HPLC)/ultraviolet detector (UV)/ion-trap time-of-flight (IT-TOF) mass spectrometry was developed for the simultaneous quantification of 19 marker compounds in Bang-poong-tong-sung-san (BPTS), a traditional oriental prescription. Various parameters affecting HPLC separation and IT-TOF detection were investigated, and optimized conditions were identified. The separation was achieved on a Capcell PAK C18 column ($1.5mm{\times}250mm$, $5{\mu}m$ particle size) using a gradient elution of acetonitrile and water containing 0.1% formic acid at a flow rate of 0.1 mL/min. The column temperature was maintained at $40^{\circ}C$ and the injection volume was $2{\mu}L$. IT-TOF system was equipped with an electrospray ion source (ESI) operating in positive or negative ion mode. The optimized electrospray ionization parameters were as follows: ion spray voltage, +4.5 kV (positive ion mode), or -3.5 kV (negative ion mode); drying gas ($N_2$), 1.5 L/min; heat block temperature, $200^{\circ}C$. Automatic $MS^n$ (n = 1~3) analyses were carried out to obtain structural information of analytes. Elemental compositions and their mass errors were calculated based on their accurate masses obtained from a formula predictor software. The marker compounds in BPTS were identified by comparisons between $MS^n$ spectra from standards and those from extracts. Moreover, the libraries of $MS^2$ and $MS^3$ spectra and accurate masses of parent and fragment ions for marker compounds were constructed. The developed method was successfully applied to the BPTS extracts and identified 17 out of 19 marker compounds in the BPTS extracts.

  • The classical concept for iron uptake into mammalian cells has been the endocytosis of transferrin( $T_{f}$ )-bound F $e^{3+}$ via the $T_{f}$ - $T_{f}$ receptor cycle. In this case, we could not explain the uptake of F $e^{2+}$ ion and the export of iron from endosome. Studies on iron transport revealed that other transport system exists in epithelial cells of the intestine. One of non- $T_{f}$ -receptor-mediated transport systems is Nramp2/DMT1/DCT1 which transports M $n^{++}$, $Mg^{++}$, Z $n^{++}$, $Co^{++}$, N $i^{++}$ or C $u^{++}$ ion as well as F $e^{+2}$ ion. DMT1 was cloned from intestines of iron-deficient rats and shown to be a hydrogen ion-coupled iron transporter and a protein regulated by absorbed dietary iron. DMT1 is founded in other cells such as cortical and hippocampal glial cells as well as endothelial cells in duodenum. Two F $e^{3+}$ ion bound to transferrin( $T_{f}$ ) are taken up via the $T_{f}$ - $T_{f}$ receptor cycle in the intestinal epithelial cell. F $e^{3+}$ in endosome was converted to F $e^{2+}$ ion, and then exported to cytosol via DMT1. F $e^{2+}$ ion is taken up into cytosol via DMT1. Several other transporters such as FET, FRE, CCC2, AFT1, SMF, FTR, ZER, ZIP, ZnT and CTR have been reported recently and dysfunction of the transporters are related with diseases containing Wilson's disease, Menkes disease and hemochromatosis. Evidences from several studies strongly suggest that DMT1 is the major transporter of iron in the intestine and functions critically in transport of other metal ions.

이온성분의 환경거동과 기상인자와의 관계: 제주지역을 중심으로 한 유.무기성 이온성분의 대기-해양지화학 (The Behavior of Particulate-Bound logic Components and Their Relationships with Meteorological Parameters: Air-Sea Geochemistry of Inorganic and Organic tons in Cheiu Island)

  • The concentrations of ten inorganic (sodium, chloride, sulfate, ammonia, etc.) and three organic (acetate, formate, and MSA) ions associated with airborne particulate matter were measured from Cheju Island, Korea during the three field intensive campaigns conducted in (1) Sept./oct. 1997 (fall), (2) Dec. 1997 (winter), and (3) April 1998 (spring). The results of our measurements indicated that the concentration levels of most ionic species were decreasing significantly across the three experimental periods. The patterns of concentration reduction were clear as the sum of all cation and anion species changed dramatically across those periods such as 294> 144 > 65 and 193 >96>74 nequiv/m3, respectively. The changes were best explained in terms of the wind rose patterns of the study site. Since our sampling spot is located on the western-end point of Cheju Island, it is likely to reflect the effects of diverse sources such as natural, marine processes during NW and local non-maritime ones during SE winds. .Hence, the periodical changes in ionic concentrations may be accounted for by the comparable changes in wind direction. To further investigate environmental characteristics of these ionic components, correlation analysis was conducted not only between meteorological and ion data but between different ion-pairs. The results of these analyses confirm that the concentration levels of ionic species are strongly affected by wind speed and temperature and that there are certain patterns between ion species to which such effects apply. In light of the significance of the wind rose patterns in the area, we further extended these analyses into four data groups that were divided on the basis of wind direction. The results of these analyses showed that the strength of correlations between important pairs (e.g.:. between windspeed and most of major inorganic species including sodium and chloride) can be ranked on the distribution of major ions are very diverse, depending on data grouping scheme for such analysis. The results of this study thus suggest that environmental behavior of chemical components be analyzed in various respects, rather than simple standard, especially if measurements are made in complex environmental condition under which both natural and anthropogenic effects are competing each other.
