The effect of Saururus chinensis Baill against oxidative damage and inflammation

  • Objectives : ROS are involved in a wide spectrum of diseases including chronic inflammation and cancer. S.chinensis Baill, a perennial herb commonly called Chinese lizard's tail or Sam-baek-cho in Korea, is used for the treatment of edema and inflammatory diseases in the Oriental folk medicine. In this study, we investigated the antioxidant activities and anti-inflammatory effects of the two extracts, water(WE) and ethyl acetate(EAE) from S.chinensis Baill. Methods : Anti-oxidant activity was evaluated using Fe2+ chelating and hydroxyl radical scavenging assay. DNA cleavage assay, and western blot and immunostaining for phospho-p65 were performed to evaluate anti-oxidative effect. Anti-inflammatory effect was performed using NO generation assay and western blot in LPS-stimulated RAW264.7 cell. Results : In Fe2+ chelating activity and hydroxyl radical scavenging activity, WE showed more strong scavenging activity for hydroxyl radical than EAE. WE scavenged hydroxyl radical by 12% at 3.2 ${\mu}g/ml$, 21% at 16 ${\mu}g/ml$, 32% at 80 ${\mu}g/ml$, 66% at 400 ${\mu}g/ml$ and 82% at 2000 ${\mu}g/ml$, respectively. In addition, WE showed more strong chelating activity than EAE. WE chelated Fe2+ ion by 1.1% at 3.2 ${\mu}g/ml$, 8.2% at 16 ${\mu}g/ml$, 26.3% at 80 ${\mu}g/ml$, 72% at 400 ${\mu}g/ml$ and 89% at 2000 ${\mu}g/ml$, respectively. Also, WE inhibited oxidative damage via its anti-oxidant activity. In anti-inflammatory effect, EAE inhibited NO production and iNOS expression. In addition EAE suppressed the NF-${\kappa}B$ and MAPK signaling pathway in LPS-stimulated RAW 264.7 cells. Conclusions : Together, these data indicate that S. chinensis Baill, shows anti-oxidant activity and anti-inflammatory effect.

Comparative Phenolic Composition and Antioxidant Properties of Honey and Honeycomb Extracts (벌꿀과 벌집채꿀의 페놀성분 및 항산화 효과 비교)

  • Although many studies have described the physiological effects of bee products, such as honey, propolis, pollen, and royal jelly, the health benefits of honeycomb remain incompletely characterized. We performed a comparative study of the antioxidant properties of honey and honeycomb extracts using two different solvents (water and 95% ethanol). The results showed that the total phenolic and flavonoid content of the honeycomb extract was higher than that of the honey extract. They also demonstrated that water was more effective than ethanol in extracting total phenols. The in vitro antioxidant properties of the water honeycomb extract were evaluated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) and nitric oxide (NO) radical scavenging assays and ferrous ion chelating and reducing power assays. The antioxidant activity of the honeycomb extract exhibited was higher than that of the honey extract. The 50% effective concentrations (EC50) of the honeycomb extract were 7.3±0.26 mg/ml for scavenging DPPH radicals, 6.1±0.22 mg/ml for scavenging NO radicals, 6.9±0.44 mg/ml for chelating ferrous ions, and 8.2±0.11 mg/ml for reducing power. A correlation analysis revealed that the total phenolics and flavonoids of the honeycomb extract were the major contributors to the radical scavenging activity, ferrous ion chelating, and reducing power. The honeycomb extract was effective in protecting biological systems against various oxidative stresses in vitro. This is the first report on the antioxidant properties of honeycomb.

Changes in Phenolic Composition, Antioxidant and Antidiabetic Properties of Jeju Citrus sudachi as Influenced by Maturity (수확시기별 제주산 영귤의 항산화 및 항당뇨 활성 비교)

  • The effects of fruit maturation on changes in the total phenolics, flavonoids, and carotenoids of methanolic extracts of Citrus sudachi, in addition to its antioxidant and antidiabetic activities, were determined. Generally, the concentration of these chemical constituents increased as C. sudachi reached maturity. C. sudachi contained high levels of total phenolics, flavonoids, and carotenoids at maturity, contributing 6339.5 mg of gallic acid equivalent per 100 g, 2364.2 mg of rutin equivalent per 100 g, and 678.7 mg/ml, respectively. The scavenging activities of 1,1-diphenyl-2-picryl hydrazyl (DPPH), hydrogen peroxide and nitric oxide radicals and the reducing power of mature fruits were significantly higher at all data points than those of immature fruits (p<0.05). In contrast, the ferrous ion chelating activity of mature and immature C. sudachi fruits was similar. The 50% effective concentrations (EC50) of mature fruits were 4.1±0.10 mg/ml for scavenging DPPH radicals, 3.1±0.02 mg/ml for scavenging hydrogen peroxide, 3.9±0.01 mg/ml for scavenging nitric oxide, and 3.8±0.02 mg/ml for chelating ferrous ion. The antidiabetic activity of C. sudachi was studied in vitro using the α-glucosidase inhibitory method. The inhibitory activity of mature C. sudachi fruits on α-glucosidase was higher than that of immature fruits. These results suggest that the content of bioactive compounds and the antioxidant and antidiabetic activities of C. sudachi change during maturation. These findings can be further extended to exploit them for their possible application for the preservation of food products, as well as their use as health supplements and nutraceuticals.

Characterization and Purification of Anti-Complement Polysaccharide from Spirodela polyrhiza

  • We purified and characterized a crude polysaccharide from Spirodela polyrhiza with anti-complement activities. The crude polysaccharide fraction (SP-0) which had potential anti-complement activity was extracted in hot water for 4 hrs at 10$0^{\circ}C$. The ethanol-precipitate, the crude polysaccharide traction (SP-1), showed a potent anti-complement activity. Further purification of the crude polysaccharide (SP-1) was carried out by cetavlon, ion exchange chromatography and gel column chromatography. Among cetavlon fractions, SP-4 showed the most potent anti-complement activity. When 100 $\mu\textrm{g}$/mL of SP-4 was incubated with an equal volume of normal human serum (NHS), the TCH$_{50}$ was reduced by about 78%. When the SP-4 fraction was further purified by DEAE-Sepharose (Cl$^{[-10]}$ ), the SP-4IIa, SP-4IIb and SP-4IIc, absorbed fractions, were almost the same as the anti-complement activities of SP-4. SP-4IIc, having the greatest potential activation and the highest yield by ion exchange chromatography, was further purified by gel column chromatography on a Sepharose CL-6B column. Four polysaccharide fractions of SP-4IIc-1, SP-4IIc-2, SP-4IIc-3 and SP-4IIc-4 were obtained, consisting mainly of arabinose, rhamnose, galactose and glucose, with approximate molecular weights of about 305,000, 132,000, 64,000 and 12,000, respectively. Among these subfractions, SP-4IIc-1 had the most potent anti-complement activity. When the SP-4IIc-1 aggregate was applied to a gel column chromatography in 10 mM and 50 mM NaCl solution, the position of the peak fractions shifted to a low molecular weight region, and the molecular weight of SP-4IIc-1 decreased with increased NaCl concentration in the gel column chromatography. It was found that the self-aggregation formed spontaneously in void volume by gel column chromatography using Sepharose CL-6B in water and the self-aggregation significantly affected the anti-complement function.

  • From results obtained by adjusting experimental variables based on the kinetic, the nucleation rate for formation of precipitated calcium carbonate (PCC) was investigated. Formation behavior of PCC was investigated for various concentrations of NaOH solution and $Na_2CO_3$ addition methods in the $Ca(OH)_2$ slurry. The range of nucleation rate was investigated for dissolution rates of major ion concentrations, $Ca^{2+}$ and $CO{_3}^{2-}$. In case of high concentration of major ions, vaterite and calcite were synthesized. The high nucleation rate was achieved for lower either $Ca^{2+}$ or $CO{_3}^{2-}$ ion concentration, calcite was mainly synthesized and when concentration of major ions was low, aragonite was synthesized. Furthermore, the formation of calcite was decreased with increasing concentration of NaOH. homogeneous aragonite could be obtained by addition 5 M NaOH. Therefore, in this study, specific shape of polymorphs could be prepared through controlling supersaturation.

Urban Nonpoint Source Pollution Assessment Using A Geographical Information System (GIS를 이용한 도심지 Nonpoint Source 오염 물질의 평가연구)

  • A geographical information systems(GIS) was a useful aid in the assessment of urban nonpoint source pollution and the development of a pollution control strategy. The GIS was used for data integration and display, and to provide data for a nonpoint source model. An empirical nonpoint source loading model driven by land use was used to estimate pollutant loadings of priority pollutant. Pollutant loadings were estimated at fine spatial resolution and aggregated to storm sewer drainage basins(sewershedsl. Eleven sewersheds were generated from digital versions of sewer maps. The pollutant loadings of individual land use polygons, derived as the unit of analysis from street blocks, were aggregated to get total pollutant loading within each sewershed. Based on the model output, a critical sewershed was located. Pollutant loadings at major sewer junctions within the critical sewershed were estimated to develop a mi t igat ion strategy. Two approaches based on the installat ion of wet ponds were investigated -- a regional approach using one large wet pond at the major sewer outfall and a multi-site approach using a number of smaller sites for each major sewer junction. Cost analysis showed that the regional approach would be more cost effective, though it would provide less pollution control.

Development of Chemical and Biological Decontamination Technology for Radioactive Liquid Wastes and Feasibility Study for Application to Liquid Waste Management System in APR1400 (액체방사성폐기물에 대한 화학적, 생물학적 제염기술 개발 및 APR1400 액체폐기물관리계통 적용을 위한 타당성 연구)

  • A decontamination technology for radioactive liquid wastes was newly developed and hypothetically applied to the liquid waste management system (LWMS) of the nuclear power plant (NPP) to evaluate its decontamination efficacy for the purpose of the fundamental reduction of spent resins. The basic principle of the developed technology is to convert major radionuclide ions in the liquid wastes into inorganic crystal minerals via chemical or biological techniques. In a laboratory batch experiment, the biological method selectively removed more than 80% of cesium within 24 hours, and the chemical method removed more than 95% of cesium. Other major nuclides (Co, Ni, Fe, Cr, Mn, Eu), which are commonly present in nuclear radioactive liquid wastes, were effectively scavenged by more than 99%. We have designed a module including the new technology that could be hypothetically installed between the reverse osmosis (R/O) package and the organic ion-exchange resin in the LWMS of the APR1400 reactor. From a technical evaluation for the virtual installation, we found that more than 90% of major radionuclides in the radioactive liquid wastes were selectively removed, resulting in a large volume reduction of spent resins. This means that if the new technology is commercialized in the future, it could possibly provide drastic cost reduction and significant extension of the life of resins in the management of spent resins, consequently leading to delay the saturation time of the Wolsong repository.

Effect of Inhibitors of Ethylene Production on Growth and Gravitropism Inhibited by Oryzalin in Arabidopsis Roots (애기장대 뿌리에서 ethylene 생성 억제제가 oryzalin에 의해 억제된 뿌리 생장과 굴중성 반응에 미치는 영향)

  • Oryzalin is a herbicide that disrupts the arrangement of microtubules by binding to tubulin, thereby blocking the anisotropic growth of plant cells. Microtubules and microfilaments are cytoskeleton components that have been implicated in plant growth through their influence on the formation of cell walls. Microtubules also play roles in the sedimentation of amyloplasts in the root tip columella cells; this sedimentation is related to gravity sensing and results in downward root growth in the soil for absorption of water and minerals. However, the orientation of microtubules changes depending on the level of ethylene in plant cells. A recent study reported that oryzalin stimulated ethylene production via 1-aminocyclopropane-1-carboxylic acid (ACC) synthase and ACC oxidase and caused a concentration-dependent inhibition of root growth and gravitropic responses. The aim of the present study was to investigate the possibility that oryzalin-induced inhibition might be recovered by the application of inhibitors of ethylene production, such as 10-4 M cobalt ions and 10-8 M aminoethoxyvinylglycine (AVG). The inhibition of root growth and gravitropic response was overcome by 10-20% by an 8 hr treatment with cobalt ions or AVG. These results suggest that ethylene levels could regulate root growth and gravitropic responses in Arabidopsis.

A Study on the Chemical Components of Acid Deposition (산성 강하물질의 화학적 성분에 관한 연구)

  • Dry and wet deposition samples were continuously collected by deposit gauge. In Bulkwang area of Seoul and Kanghwa in west coastal area of Korea. In order to evaluate the level of air pollution and its chemical composition, Bulkwang area located in Seoul and Kanghwa in west coastal area were chosen for sampling site. dry deposition concentrations, pH, electric conductivity and water soluble ion concentrations of deposit gauge were analysed. The results of comparison between urban area and coastal area were summarized as follows. Mean concentrations of dry deposition in Bulkwang was 2.807 ton/km$^2$/month (range: 5.171~1.128 ton/km$^2$/month) while that in Kanghwa was 1.990 ton/km$^2$/month (range: 3.358 ~ 1.084 ton/km$^2$/month), which showed a significant difference between two areas. The rainfall during the period from June to September in 1990 recorded 1859.7 mm which was 78.8% of its mean amount in Seoul, and 1846.9 mm which was 81.6% that of Kanghwa. In Bulkwang area, correlation coefficients of deposit chemical composition were 0.95 for SO$_4^{-2}$ and Na$^+$, 0.94 for SO$_4^{-2}$ and NH$_4^+$, 0.93 for CI$^-$ and NH$_4^+$ and 0.85 for Cl$^-$ and Ca$^{2+}$, respectively. Then, the results indicate that sulfates such as $Na_2SO_4$, $(NH_4)_2SO_4$, and CaSO$_4$ were the major chemical state of deposit. In Kanghwa area, it was considered that NaCl, $NH_4NO_3$, NaNO$_3$, and $Ca(NO_3)_2$ were the major chemical state of deposit.

Minor Thermostable Alkaline Protease Produced by Thermoactinomyces sp. E79

  • Thermoactinomyces sp. E79 produced two types of thermostable alkaline proteases extracellularly. A minor protease was separated from a major protease by using DEAE-column chromatography. This enzyme was purified to homogeneity by ammonium sulfate and DEAE-Sepharose ion-exchange chromatography. The purified minor protease showed different biochemical properties compared to the major protease. The molecular mass of the purified enzyme was estimated by SDS-PAGE to be 36 kDa. Its optimum temperature and pH for proteolytic activity against Hammarsten casein were $70^{\circ}C$ and 9.0, respectively. The enzyme was stable up to$75^{\circ}C$ and in an alkaline pH range of 9.0-11.0. The enzyme was inhibited by phenylmethylsulfonyl fluoride (PMSF) and $Hg^{2+}, indicating that the enzyme may be a cysteine-dependent serine protease. In addition, the enzyme cleaved the endoproteinase substrate, succinyl-Ala-Ala-Pro-Phe-p- nitroanilide, and the $K_m$ value for the substrate was 1.2 mM.
