A new, improved maize varieties are greatly needed to meet the national demand for the maize, and a new germplasm should be investigated to supply necessary breeding materials for the required maize breeding. The Korean local maize lines, which has never been wholly studied for the purpose of breeding should be enough to supply such breeding materials at least in Korea. The objectives of this study were to maintain useful Korean local lines, to investigate good plant characters and to analyze the Korean local maize lines in the respect of genetic make-up. A total of 820 lines and 200 lines were used for the present study. Lines to maintain were planted at experimental farm of the Crop Experiment Station, Office of Rural Development, Suwon and the lines to study genetic make-up were planted in Dae-jon. The summarized results are as follows : 1. A total of 1200 lines from 820 collections were sibbed for future use. 2. Two hundred collection were sibbed, selfed and testcrossed for genetic study in 1980. 3. Ten lines were surely confirmed as resistant lines to virus disease epidemic in most introduced maize varieties. 4. A dozen Lines were having three to four ears per plant. 5. Five lines were showing three to four tillers per plant. 6. One of the most significant and important finding through the study was to obtain the lines with three to four tillers bearing all together eight to nine ears. The lines of multi-ears and multi-tillers must have great breeding potential for the future use. 7. A great variation was observed in the tasselling days, 100 kernel weight and protein content. 8. From the genetic study of local maize lines for plant height and 100 kernel weight, the following findings were obtained. a. On the average six percent of inbreeding depression expressed in percent decrease over sibbed counterpart were found for the plant height. b. Fourty percent of heterosis expressed in percent increase over sibbed counterpart were observed in the plant height. c. The homogeneity or degree of homozygosity calculated from heterosis minus inbreeding depression was on the average 34 percent for plant height. d. The average inbreeding depression expressed in percent decrease over sibbed counterpart was 15 percent for 100 kernel weight. e. The average heterosis expressed in percent increase over sibbed counterpart was 43 percent for 100 kernel weight. f. The degree of homozygosity calculated from the heterosis minus inbreeding depression was 28 percent for 100 kernel weight.

Four lactating Holstein cows were used in a $4{\times}4$ Latin-square design to determine the effects of various levels of whole cottonseed (WCS) in diets on parameters including milk (yield and fat content), rumen fluid (pH, ammonia and TVFA) and blood (${\beta}$-carotene, vit. A, vit. E, urea, $NH_3$, Ca, P and Mg levels). Cows consumed 0, 1, 2 or 3 kg WCS per day. No significant differences were observed among the groups on analysed parameters except plasma vitamin E concentration. In addition, when the amount of cottonseed was increased, milk yield and milk fat content also tended to increase but this increase was not statistically significant. In conclusion, feeding of WCS up to 3 kg per day with ad libitum maize silage did not cause negative effects on milk yield, milk fat and blood vitamin levels in the short term in dairy cows.

Northern corn leaf blight caused by Exserohilum turcicum Pass is considered the most important disease infecting corn (Zea mays L.) in the Peoples' Republic of Korea (North Korea). It contributes to the food shortage in North Korea. The objectives of the current research were to study resistance expression and responses of corn crosses made between ten hybrids from North Korea and inbreeding lines ($S_{3-4}$ stage) from the Republic of Korea (South Korea). The experiments were conducted in six trials with a total of 184 crosses including two commercial hybrids in each trial. The trials were conducted at two locations in North Korea (Mirim and Eunsan) and one location in South Korea (Gunwi) under natural infestation of E. turcicum. Host plant responses were rated on a scale of 1 (highly tolerant) to 9 (highly susceptible). A total of 111 crosses (62.4%) showed significant tolerant or susceptible response variations among three locations; 42 crosses (22.8%) at two locations and 69 crosses (39.0%) at one location, respectively. At least 8 crosses of high level of tolerance and 12 crosses of high level of susceptibility showed significantly different biotic responses (P = 0.05). The results of the current study and historical reviews of E. turcicum epidemics in both North and South Korea suggest that breeding of tolerance with quantitatively inherited genes should be carried out for a sustainable corn production in North Korea.
Accurate and rapid detection of bacterial plant pathogen is the first step toward disease management and prevention of pathogen spread. Bacterial plant pathogens Clavibacter michiganensis subsp. nebraskensis (Cmn), Pantoea stewartii subsp. stewartii (Pss), and Rathayibacter tritici (Rt) cause Goss's bacterial wilt and blight of maize, Stewart's wilt of maize and spike blight of wheat and barley, respectively. The bacterial diseases are not globally distributed and not present in Korea. This study adopted comparative genomics approach and aimed to develop specific primer pairs to detect these three bacterial pathogens. Genome comparison among target pathogens and their closely related bacterial species generated 15-20 candidate primer pairs per bacterial pathogen. The primer pairs were assessed by a conventional PCR for specificity against 33 species of Clavibacter, Pantoea, Rathayibacter, Pectobacterium, Curtobacterium. The investigation for specificity and sensitivity of the primer pairs allowed final selection of one or two primer pairs per bacterial pathogens. In our assay condition, a detection limit of Pss and Cmn was $2pg/{\mu}l$ of genomic DNA per PCR reaction, while the detection limit for Rt primers was higher. The selected primers could also detect bacterial cells up to $8.8{\times}10^3cfu$ to $7.84{\times}10^4cfu$ per gram of grain seeds artificially infected with corresponding bacterial pathogens. The primer pairs and PCR assay developed in this study provide an accurate and rapid detection method for three bacterial pathogens of grains, which can be used to investigate bacteria contamination in grain seeds and to ultimately prevent pathogen dissemination over countries.
A cDNA (Oslls1) encoding Lls1-homologue of maize was isolated from cDNA library of rice (Oryza sativa cv. Ilpum). The 2,138 bp of full length Oslls1 clone contains an open reading frame of 1,623 nucleotides encoding 575 amino acid residues. The deduced amino acid sequence of Oslls1 has a high level of homology with chlorophyll a oxygenases of Arabidopsis thaliana (67%) and Marchantia polymorpha (65%). Southern blot analysis of genomic DNA indicates the existence of a small gene family for Oslls1 in the rice genome. The expression of Oslls1 mRNA was induced in leaves and germinating seeds. Treatment of $H_2O$$_2$significantly down-regulated Oslls1 expression. The expression of Oslls1 mRNA was consititutively down-regulated in the blm, a rice mutant exhibiting spontaneous necrotic lesions. These results suggest that this Oslls1 gene may be involved incell death mechanisms in the blm mutant of rice.
This study was carried out to identify the Rice black-streaked dwarf virus that infected maize plants collected from Gochang-gun in Jeollabukdo in 2005. The genomic dsRNA from infected plants was extracted and the genome pattern was analyzed by polyacrylamide gel electrophoresis. Results of the electrophoresis revealed the already known to-segment genome and the difference of mobility was confirmed in isolates by collected areas. The RBSDV was identified from the result of RT-PCR using the template of extracted dsRNA and specific primer. The results of S10 cloned to pGEM-T vector and the conducted in sequence analysis consisted of 1,801nt and 559aa. This was of the same size as the RBSDV S10 identified in rice, and the change was confirmed in 18 base and displayed homology of 99%.

The studies were conducted to test for combining ability and to evaluate resistance to maize dwarf mosaic virus (MDMV) by diallel crosses of corn inbreds. For the genetic analysis of the resistance, a diallel set of crosses without reciprocals was made using the eight corn inbreds which had different degrees of resistance to MDMV. Twenty eight $F_1$ hybrids showed different symtom severity. the highest value is 3.63 and the lowest value is 1.87 in disease ratings (1-4). General combining ability (GCA) for resistance to MDMV was highly significant, but specific combining ability (SCA) was not significant. Two inbreds, A632 and KS15 showed negative GCA effects, indicating that these parents were good general combiners and that resistance to MDMV increased in hybrid combinations. Hyrid A632 x KS5, showed the highest negative SCA effect and several combinations showed negative SCA effects. The analysis of parent-offspring covariance (Wr) and array variance (Vr), suggest that there may be many dominant genes in the resistant inbreds and many recessive genes in the susceptible inbreds.
Five parents and their 10 $F_1$ crosses of maize (Zea mays L.) were tested by means of diallel analysis for the inheritance of peroxidase activity following Puccinia sorghi rust infection. Peroxidase activity was measured at day zero and at 2 days and 8 days after inoculation. Peroxidase activity was increased significantly by P. sorghi infection in all 15 genotypes after 8 days, but not after 2 days. Highly significant differences in peroxidase activity were detected among the 15 genotypes. The highly genera] resistant inbred, CM105, and its hybrids showed exceptionally high peroxidase activity in both healthy and infected plants. However, another highly resistant inbred, Oh545, showed exceptionally low peroxidase activity. Significant general combining ability (GCA) and specific combining ability (SCA) means quares were detected for peroxidase activity independent of disease. GCA mean spuares, however, were consistently a major contiribution to the inheritance of peroxidase activity in the infected plants whereas SCA men square contributions were minor. Rust resistant maize plants controlling mongenic dominant $Rp_1^d$gene showed stronger peroxidase reroxidase responses than their susceptible counterparts in the gel electrophoresis and densiometric tracings. The increased peroxidase activity occurred in both major leaf peroxidases, $Px_3\;and\; Px_7$.
The Rice black-streaked dwarf virus (RBSDV) infected 99-100% of a $1000\;m^2$-maize field in Mungyeong City in 2007. Adjacent to the area is a Persimmon orchard where barley crops were grown under the trees as green manure crops and for soil amendments. The barley acted as winter host to the small brown plant hoppers (SBPH) enabling the insects to survive and pass the winter season. The existence of RBSDV was detected and confirmed by RT-PCR using S9 specific primer. Samples of the insect vector SBPH were collected in the area on May 3, June 7 and, August 4 and the results of the RT-PCR analysis revealed viruliferous insect rates of 2.9, 4.8, and 4.4%, respectively. These observed viruliferous insect rates were similar with those detected in RBSDV infected rice fields.
A single cross hybrid, Heukjinjuchal, is a black color waxy corn (Zea mays L.) developed by the maize breeding team at NICS in 2008. Heukjinjuchal was produced by crossing two inbred lines, KBW24 as seed parent and KBW2 as pollen parent. Ear length and ear diameter of Heukjinjuchal was 16.0 cm and 4.4 cm, respectively. The ratio of kernel set length/ear length was 89%, similar with that of a check hybrid, Chalok1. It is resistant to Northern corn leaf blight disease and also lodging tolerant. Fresh ear yield of Heukjinjuchal comparable with those of a check hybrid, Chalok1 in regional yield trials for three years. Seed production of this hybrid is relatively easy because of well overlapping period of silking and pollen dispersal between the two parents. This cultivar would be well adaptable to the whole country.
