C-banding Pattern of Mitotic Chromosome in Korean Indigenous Maize (한국 재래종 옥수수 체세포 염색체의 C-분염패턴)

  • A Giemsa C-banding method was used for the identification of somatic chromosomes of Korean indigenous maize(Zea mays L.). Two Korean indigenous maize stocks and an American hybrid maize for comparison were examined. Ten deeply stained heterochromatic knobs whose position and size were different between the genotypes, two satellites and interstitial bands were observed. The length of homologous chromosomes compared by the relative lengths of chromosomes presented as a percentage of the length of chromosome 10 were different between the genotypes. The Giemsa method proved to be useful for the identification of somatic chromosomes and for the characterization of different stocks of Korean indigenous maize.

Co-inoculation of Burkholderia cepacia and Alcaligenes aquatilis enhances plant growth of maize (Zea mays) under green house and field condition

  • The synergistic effect on phosphate solubilization of single- and co-inoculation of two phosphate solubilizing bacteria, Burkholderia cepacia (C1) and Alcaligenes aquatilis (H6), was assessed in liquid medium and maize plants. Co-inoculation of two strains was found to release the highest content of soluble phosphorus (309.66 ?g/mL) into the medium, followed by single inoculation of B. cepacia (305.49 ?g/mL) and A. aquatilis strain (282.38 ?g/mL). Based on a plant growth promotion bioassay, co-inoculated maize seedlings showed significant increases in shoot height (75%), shoot fresh weight (93.10%), shoot dry weight (84.99%), root maximum length (55.95%), root fresh weight (66.66%), root dry weight (275%), and maximum leaf length (81.53%), compared to the uninoculated control. In a field experiment, co-inoculated maize seedlings showed significant increases in cob length (136.92%), number of grain/cob (46.68%), and grain weight (67.46%) over control. In addition, single inoculation of maize seedlings also showed improved result over control. However, there was no significant difference between single inoculation of either bacterial strains and co-inoculation of these two bacterial strains in terms of phosphate solubilization index, phosphorous release, pH of the media, and plant growth parameters. Thus, single inoculation and co-inoculation of these bacteria could be used as biofertilizer for improving maize growth and yield.

The Effect of Taxol and Ethyl-N-phenylcarbamate (EPC) on Growth and Gravitropism in Zea mays L

  • The effect of taxol and ethyl-N-phenylcarbamate (EPC) on the growth and gravitropism of maize roots and coleoptiles was studied. Taxol is known to promote the assembly of microtubules (MTs) and stabilizes MTs by preventing depolymerization. EPC, on the contrary, is an anti-microtubule drug that promotes disassembly of MTs. Taxol, at 1 $\mu$M, inhibited gravitropic response of maize roots to about 40%, but did not inhibit growth; at 10 $\mu$M, it inhibited the gravitropic response of coleoptile segments of maize by approximately 50%, but did not inhibit growth, while 0.5 mM EPC inhibited both the gravitropic response and growth of maize roots by approximately 50%. Taxol, which inhibited the gravitropic response of maize roots and coleoptile segments, had no effect on either the polar or the bilateral transport of auxin. These results indicated that MT polymerization could not occur normally with taxol or EPC, so that if there was any abnormal rearrangement of MT, the gravitropic response was inhibited, which resulted from the inhibition of neither growth nor auxin transport. This results suggested that gravitropic response was related to the MT arrangement, and that both straight growth and the differential growth in gravitropic response could be regulated by different mechanisms.

Radical Scavenging Activity and Cytotoxicity of Maysin(C-glycosylflavone) isolated from Silks of Zea mays L.

  • Maysin, a C-glycosylflavone, was isolated from the silks of maize, Zea mays L. The ESI mass spectrum indicates that molecular weight of maysin is $577\textrm{M}^+$m/z, and the ether-linked sugar is rhamnose, $431\textrm{M}^+$m/z (MW$^{+}$-146). The DPPH (1,1-Diphenyl-2-picrylhydrazyl) radical scavenging activity of maysin was higher than that of rutin. However, as compared with its aglycon luteolin, maysin showed the relatively moderate DPPH scavenging activity mainly due to the glycosylation of two sugars moieties, keto-fucose and rhamnose. In the in vitro cytotoxicity test against the five human tumor cell lines such as lung (A549), ovarian (SK-OV-3), melanoma (SK-MEL-2), central nerve system (XF-489), and colon (HCT-15), maysin exhibited the relatively weaker activities than cisplatin. The $\textrm{ED}_{50}$ values of maysin were 62.24, 43.18, 16.83, 37.22, and 32.09/$m\ell$, respectively. Result suggests that maysin is a potential cytotoxicity compound, particularly for human colon, central nerve system, and melanoma tumors.s.

Detection and environmental unintentional release monitoring of living modified maize (Zea mays L.) in Gyeonggi-do of South Korea in 2014 (2014년 경기지역 유전자변형 옥수수 모니터링 및 발견현황)

  • In South Korea, LM crops are not allowed to grow locally, but have been allowed to be imported as food and feed purposes. Currently, the typical LMO imports are continuously increasing in the region of South Korea. In 2014, we carried out a review of the environmental release monitoring of LM maize (Zea mays L.) in Gyeonggi-do of South Korea, and analyzed volunteer samples using strip test kits and polymerase chain reaction (PCR) methods. We thereby collected 44 volunteers of released LM maize in 169 locations around ports, from roadsides, feed factories and stockbreeding farmhouses. We found 4 positive samples at 3 sites using strip test kits. Based on the PCR analysis, the LM maize plants were found using event-specific primers. These results suggested that our monitoring is necessary to detect the presence of released LM maize in the natural environment of South Korea.

Characterization of CH4-oxidizing and N2O-reducing Bacterial Consortia Enriched from the Rhizospheres of Maize and Tall Fescue (옥수수와 톨페스큐 근권 유래의 메탄 산화 및 아산화질소 환원 세균 컨소시움 특성)

  • CH4-oxidizing and N2O-reducing bacterial consortia were enriched from the rhizosphere soils of maize (Zea mays) and tall fescue (Festuca arundinacea). Illumina MiSeq sequencing analysis was performed to comparatively analyze the bacterial communities of the consortia with those of the rhizosphere soils. Additionally, the effect of root exudate on CH4 oxidation and N2O reduction activities of the microbes was evaluated. Although the inoculum sources varied, the CH4-oxidizing and N2O-reducing consortia derived from maize and tall fescue were similar. The predominant methanotrophs in the CH4-oxidizing consortia were Methylosarcina, Methylococcus, and Methylocystis. Among the N2O-reducing consortia, the representative N2O-reducing bacteria were Cloacibacterium, Azonexus, and Klebsiella. The N2O reduction rate of the N2O-reducing consortium from maize rhizosphere and tall fescue rhizosphere increased by 1.6 and 2.7 times with the addition of maize and tall fescue root exudates, respectively. The CH4 oxidization activity of the CH4-oxidizing consortia did not increase with the addition of root exudates. The CH4-oxidizing and N2O-reducing consortia can be used as promising bioresources to mitigate non-CO2 greenhouse gas emissions during remediation of oil-contaminated soils.

Distinctive response of maize (Zea mays L.) genotypes in vitro with the acceleration of phytohormones

  • In maize, immature embryos (IEs) are highly regenerative explants most suitable for producing high frequencies of plantlet regeneration in vitro. Apart from media, explants, and hormones, genotypic variation also influences in vitro characters to a great extent. In the present study, IEs were used to study the distinctive effect of variation of size/stage and hormones in different genotypes on five in vitro characters viz., frequency of callus induction, growth rate of total callus, frequency of E. callus induction, and volume and number of regenerated plantlets. LS medium with different concentrations of 2,4-D (0.5, 1.5, 2.5, 4.0 and 5.0 mg/L) were used to study the former four in vitro characters, and medium with 6-benzylaminopurine and kinetin (0.5 mg/L, each) was used for plantlet regeneration. IEs of 1.0, 1.5, 2.0, 2.5 and 3.0 mm in size were isolated from four inbred lines viz., NM74C, NM81A, NM5883 and NM5884. Two-way ANOVA revealed that explant size and genotypes, as well as hormonal concentrations showed significant effects on in vitro characters. Two millimeter IEs were found to be suitable for in vitro cultures. LS medium with 1.5 mg/L 2,4-D and LS with BAP and Kn (0.5 mg/L, each) were found to be the best hormonal concentrations for callus induction, maintenance, and regeneration, respectively. Among the four genotypes, NM81A and NM5883 yielded more non-embryogenic and Type I E. calli. In contrast, NM74C and NM5884 yielded more highly regenerative Type II calli. Inbred line NM5884 was found to be the best among these four genotypes.

Quantitative Change in rbcL mRNA of Maize by Phytohormones (식물 호르몬에 의한 옥수수 rbcL mRNA의 양적 변화)

  • In order to investigate the effects of plant hormones on the quantitative changes in mRNA of maize (Zea mays L.) rbcL, we used GA3, IAA, ABA and BAP. GA3 at the concentration of 10-4M resulted in decrease in rbcL gene transcript to 62%. IAA decreased the amount of rbcL transcript to about 70% at all the hormone concentrations tested. ABA did not cause a noticeable change in the amount of rbcL transcript, but BAP increased the amount of rbcL transcript to 153% at 10-8M and 123% at 10-5M, respectively. Thus, it appears that BAP is related to the increase in the amount of rbcL transcript by light.

Characteristics of Waesungri Maize (Zea mays L.) Inbred with Multi Tillers and Ears for Crude Forage Use

  • Major characteristics of new Waesungri maize inbred line has multi-tiller and ears: five to six tillers and seven to eight ears per plant and flowering date of Waesungri was delayed about 18 and 24 days compared to Mo17 U.S line and IK$_4$ Korean local lines, respectively. Number of ears, fresh and dry weight per plant were significantly different among all tested hybrids including Waesungri/Sinkihong hybrid under different planting times and densities. Especially, both fresh and dry weight of IK$_1$/FR140//Waesungri F$_1$hybrid were significantly higher at high planting density. In kernel weight per unit area, Wnesungri/Sinkihong hybrid was high at high density and IK$_1$/FR140//Waesungri hybrid was high at low planting density. As results of analysis of variance, flowering date was shown a significantly different both planting times and varieties, while other characters including stem height were shown very variable in interactions with enviromental factors.

Photochemical Response Analysis on Different Seeding Date and Nitrogen (N) level for Maize (Zea mays L.) (옥수수의 파종시기 및 질소수준별 광화학적 반응 해석)

  • The photochemical characteristics were analyzed in the context of sowing time and different levels of fertilized nitrogen during the maize (Zea mays L.) growth. When maize was early sawn, the fluorescence parameters related with electron-transport, in photosystem II (PSII) and PSI, were effectively enhanced with the higher level of fertilized nitrogen. Highest values were observed in maize leaves grown in double N-fertilized plot. The photochemical parameters were declined in the progress of growth stage. In early growth stage, the fluorescence parameters were highest, and then reduced to about half of the parameters related with electron transport on PSII and PSI at middle and late growth stages. In 1/2 N plot, the photochemical energy dissipation was measured to 13% in term of active reaction center per absorbed photon resulting in decrease in performance index and driving force of electron. This decrease induced to lower the photochemical effectiveness. In 2 N plots, the electron transport flux from $Q_A$ to $Q_B$ per cross section and the number of active PSII RCs per cross section were considerably enhanced. It was clearly indicated that the connectivity between photosynthetic PSII and PSI, i.e. electron transport, was far effective.
