Study on the Applications of prescriptions including Magnoliae Cortex as a main component in Dongeuibogam (東醫寶鑑 中 厚朴이 主藥으로 配伍된 方劑의 活用에 대한 考察)

  • This report describes the remedial fields, symptoms, pathology, dosage, prescriptional constitution of 40 prescriptions related to the use of Magnoliae Cortex main blended prescriptions from Dongeuibogam. The following conclusions were reached through investigations on the prescriptions that use Magnoliae Cortex as a key component. Prescriptions that Magnoliae Cortex was taken as a monarch drug are utilized for 13 therapeutic purposes, for example, abdominal dropsy, abdominal lump. In particular, 27.5% of the prescriptions appear in the chapter of abdominal dropsy. Prescriptions that utilize Magnoliae Cortex as the main ingredient are used in the treatment of abdominal lump, abdominal dropsy. Magnoliae Cortex is used in pathogenic factors such as cold, congestion of Qi and used in pathology related to digestive system. The dosage of Magnoliae Cortex is 2pun(about 0.75g) to 3don(about 11.25 g), however 1don(about 3.75g) has been taken the most for clinical application. Pyeongwisan is the most useful base prescription which use the Magnoliae Cortex as the main component.

Inhibitory Effect of Immediate-Type Allergic Reaction by Magnoliac flos (辛夷花에 의한 卽時型 알레르기의 反應 抑制 效果)

  • The mast cell plays a pivotal role in initiating allergic response by secreting intracytoplasmic granular mediators such as histamine. Magnoliae flos has been used for treatment of allergic disease in Korea. However, its effect in experimental models remains unknown. The present report describes an inhibitory effect of Magnoliae flos on mast cell-mediated immediate-type allergic reactions. Topical application of compound 48/80 can induce an ear swelling response in normal($WBB6F_1-+/+$) mice but not in the congenic mast cell-deficient $WBB6F_1-W/W^v$ mice. Magnoliae flos inhibited concentration-dependently mast cell-dependent ear swelling response induced by compound 48/80 by topical application. Magnoliae flos inhibited concentration-dependently passive cutaneous anaphylaxis induced by anti-dinitrophenyl (DNP) IgE in rats by topical application. Magnoliae flos also inhibited concentration-dependently the histamine release from the rat peritoneal mast cells by compound 48/80 or anti-DNP IgE. Moreover, Magnoliae flos had a significant inhibitory effect on compound 48/80-induced systemic anaphylactic reaction. These results indicate that Magnoliae flos inhibits immediate-type allergic reactions by inhibition of mast cell degranulation in vivo and in vitro.

Synergistic Antifungal Activity of Magnoliae Cortex and Syzyii Flos against Candida albicans

  • Candida albicans is a dermal fungus of the human body that is known to cause oral candidiasis, vaginal candidiasis, and bloodstream infections in immunocompromised people or in certain environmental conditions. As cases of strains resistant to antifungal agents in C. albicans have been reported, studies using plant materials as safe antifungal agents are being actively conducted. In this study, a total of 17 edible plant extracts showed antifungal activity against C. albicans as a result of evaluating a 280-plant extract library using paper disk diffusion method. Among them, the four extracts with the strongest antifungal activity (Cinnamomi Cortex, Cinnamomi Ramulus, Magnoliae Cortex, and Syzygii Flos) were selected and evaluated for synergistic antifungal activity against C. albicans. The combination of Magnoliae Cortex and Syzygii Flos showed a synergistic activity. The antifungal activity was evaluated based on the concentrations of magnolol and eugenol, the respective components of Magnoliae Cortex and Syzygii Flos. Magnolol and eugenol showed synergistic antifungal activities at the concentration ratio of 1:25 - 1:61. The antifungal activity of these two compounds contributes 28 to 48% to the synergistic antifungal activity of the combination of Magnoliae Cortex and Syzygii Flos extract. In this study, we propose that a combination of Magnoliae Cortex and Syzygii Flos can effectively inhibit the growth of C. albicans and that magnolol and eugenol are the responsible inhibitory compounds.

Antimicorbial effect of Zea Mays L. and Magnoliae cortex extract mixtures on periodontal pathogen and effect on human gingival fibroblast cellular activity (옥수수 불검화 추출물(Zea Mays L.)과 후박(Magnoliae cortex) 추출물 혼합물의 치주질환원인균에 대한 항균작용 및 치은섬유아세포 활성도에 미치는 영향)

  • Zea Mays L. has been known to be effective for improving tissue health and Magnoliae cortex to have effective antibacterial and antimicrobial activity against pathogenic microbes. The purpose of this study was to examine the antimicrobial effects of Zea Mays L. and Magnoliae cortex extract mixtures on periodontal pathogens(Prevotella intermedia, Actinobacillus actinomycetemcomitans, Porphyromonas gingivalis, Streptococcus mutans )and to examine the effects on human gingival fibroblast cellular activity. Zea Mays L. and Magnoliae cortex extracts and their mixtures were prepared with various mixing ratios (0.5:1, 1:1, 1.5:1, 2:1). These extracts were loaded to periodontal pathogen cultured petri dish for antimicrobial test and also loaded to cultured human gingival fibroblast for cellular activity test. Each test was repeated 3 times and data were analyzed by one-way ANOVA with 95% confidence level. Mixture of these two extracts showed greater amount of inhibition area on periodontal pathogen and more improved gingival fibroblast activity as Zea Mays L. ratio reduced. So, mixture ratio 0.5:1 (Zea Mays L. : Magnoliae cortex) group showed statistical significance in antimicrobial activity and cellular activity among various mixtures(p < 0.05). In conclusion, 0.5:1 (Zea Mays L. : Magnoliae cortex) mixture possessed best gingival fibroblast cellular activity and antimicrobial activity toward periodontal pathogens.

  • Magnoliae Flos, Xanthii Fructus and Menthae Herba have long been used for treatment of nasal disease in Korea. The 70%-ethanol extracts of Magnoliae Flos, Xanthii Fructus and Menthae Herba were used externally in order to investigate the effects on a few strains of microbe, nasal alergy and inflammation, rhinitis and sinusitis. When the proportion of Magnoliae Flos : Xanthii Fructus : Menthae Herba was 2 : 2 : 1, the results showed great significance compared to the control group(non-extracts) in treatment of various nasal disorder.

  • In order to prove the anticonvulsive effect of the fragrance of Magnoliae Flos in convulsion-induced mice, experiments were performed on anticonvulsive effect, GABA level, glutamic acid level, GABA-T activity and GAD activity. The results were obtained as follows: 1. As far as anticonvulsive effect was concerned, on the convulsion induced by such as maximal electric seizure, strychnine, bicuculline, or picrotoxin it was not significant, but the convulsion induced by pentylenetetrazole it was significant comparing to the control group. 2. GABA level was increased significantly in mice. 3. Glutamic acid level was decreased significantly in mice. 4. GABA-T activity was decreased by the fragrance of Magnoliae Flos. 5. The fragrance of Magnoliae Flos was not effective in GAD activity. From above result, the fragrance of Magnoliae Flos had significant effects on convulsion induced by pentylenetetrazole, so it is expected to clinical application on convulsive diseases such as epilepsy.

Cloning of the Transketolase Gene from Erythritol-Producing Yeast Candida magnoliae

  • The entire nucleotide sequence of the TKL1 gene encoding transketolase (TKL) in an erythritol-producing yeast of Candida magnoliae was determined by degenerate polymerase chain reaction and genome walking. Sequence analysis revealed an open reading frame of C. magnoliae TKL1 (CmTKL1) that spans 2,088 bp and encodes 696 amino acids, sharing 61.7% amino acid identity to Kluyveromyces lactis TKL. Functional analysis showed that CmTKL1 complemented a Saccharomyces cerevisiae tkl1 tkl2 double mutant for growth in the absence of aromatic amino acids and restored transketolase activity in this mutant. An enzyme activity assay and RT-PCR revealed that the expression of CmTKL1 is induced by fructose, $H_2O_2$, and KCl. The GenBank accession number for C. magnoliae TKL1 is KF751756.

The Effects of the Bojungikgi-tang added Magnoliae Flos on the Pulmonary Thromboembolism and Delayed Allergy Response (보중익기탕(補中益氣湯) 가(加) 신이(辛夷)가 폐혈전색전(肺血栓寒栓) 및 지연형(遲延型) allergy반응(反應)에 미치는 영향(影響))

  • Objective : This experimental studies were done to research the effects of Bojungikgi-tang added Magnoliae Flos on the pulmonary thromboembolism and delayed allergy response. Methods : Healthy rats were devided into each group. This test is to inject sodium arachidonic acid to one group, and ADP(adenocine diphophate) to the other one afterward to compare the survival rate between each other. Delayed allergy response test is to inject PC(picryl chloride) to one group and measure the thickness of their ears, to inject SRBC(sheep red blood cell) allergen to the other, and measure their foot edema, and compare the each other rates Results : The extract of the Bojungikgi-tang added Magnoliae Flos revealed significant effect on the pulmonary thromboembolism induced by sodium arachidonate, or adenocine diphophate(ADP). And the extract of the Bojungikgi-tang added Magnoliae Flos revealed significant effect on delayed type hypersensitivity induced by picryl chloride(PC), or sheep red blood cell(SRBC). Conclusions : According to the above findings it is suggested that the extract of the Bojungikgi-tang added Magnoliae Flos revealed significant effect on the delayed type hypersensitivity and effect on the pulmonary thromboembolism.

Cloning and Characterization of the Orotidine-5'-Phosphate Decarboxylase Gene (URA3) from the Osmotolerant Yeast Candida magnoliae

  • We determined the nucleotide sequence of the URA3 gene encoding orotidine-5'-phosphate decarboxylase (OMPDCase) of the erythritol-producing osmotolerant yeast Candida magnoliae by degenerate polymerase chain reaction and genome walking. Sequence analysis revealed the presence of an uninterrupted open-reading frame of 795 bp, encoding a 264 amino acid residue protein with the highest identity to the OMPDCase of the yeast Kluyveromyces marxianus. Phylogenetic analysis of the deduced amino acid sequence revealed that it shared a high degree of identity with other yeast OMPDCase homologs. The cloned URA3 gene successfully complemented the ura3 null mutation in Saccharomyces cerevisiae, revealing that it encodes a functional OMPDCase in C. magnoliae. An enzyme activity assay and reverse transcription polymerase chain reaction indicated that the expression level of the C. magnoliae URA3 gene in S. cerevisiae was not as high as that of the S. cerevisiae URA3 gene. Quality Evaluation on Magnoliae Cortex (후박의 품질평가)

  • Magnolol and honokiol, the main components of Magnoliae Cortex, were isolated and used as the standard substances for the analysis. In order to determine the contents of magnolol and honokiol in Magnoliae Cortex originated from Korea, China and Japan, both HPLC and HPTLC methods are applied and compared with each other. The components were separated on C8 column with acetonitrile-water-acetic acid (50:50:1) in HPLC and detected at UV 294nm. The components separated on HPTLC precoated silica gel plate with chloroform-methanol (9:1) were detected directly on the plate at 254nm. The contents of magnolol and honokiol in Magnoliae Cortex were in the wide range of 0.01~2.8% and 0.005~0.8%, respectively, according to their purchase places. It is also applicable to the quality control of various preparation from Magnoliae Cortex.
