형태특징 및 RAPD분석에 의한 후박(厚朴) 약재의 진위감별 (Discrimination of Genuines and Misuses of Hu-Bak medicines by Morphological Characters and Random Amplified Polymorphic DNA Analysis)

  • For identification of genuine and misuse samples of Hu-Bak medicines, an morphological characters, and RAPD (random amplified polymorphic DNA) were performed. In this study, three samples were collected, of which two were genuine and one was misuse sample. Genuine samples of 'Hu-Bak' were Magnolia obovata Thunberg and Magnolia officinalis Rehder et Wilson. Misuse samples was Machilus thunbergii S. et Z. In the morphological characters, M. obovata Thunberg and M. officinalis Rehder et Wilson were similar, but M. thunbergii S. et Z. was different with cortex, cambium, and fibrous layer. The result of RAPD analysis, seven primers can distinguish between genuines and counterfeit samples of Hu-Bak medicines.

Cytotoxic Compounds from the Stem Bark of Magnolia obovata

  • Two sesquiterpenes (1 - 2), a tetralone (3), and two phenylpropanoids (4 - 5) were isolated from the stem bark of Magnolia obovata Thunberg (Magnoliaceae) through repeated column chromatography. Their structures were identified as ${\beta}-eudesmol$ (1), cryptomeridiol (2), 4R-4,8-dihydroxy-${\beta}-tetralone$ (3), trans-pcoumaryl aldehyde (4), and p-coumaric acid (5) on the basis of spectroscopic analysis including two dimensional NMR and mass. Compounds 1 - 3 were tested in vitro for their cytotoxic activity against the K562, HeLa, A549, and HCT116 cancer cell lines. However, compounds 1 - 3 were inactive in this assay system.

Anti-complement Activity of Phenolic Compounds from the Stem Bark of Magnolia obovata

  • Five neolignans (1 - 4, 8), two sesquiterpene-lignans (5 - 6), and two phenylpropanoids (7, 9) were isolated from the stem bark of Magnolia obovata Thunberg (Magnoliaceae) by repeated column chromatography. The structures of isolated compounds were identified as 4-methoxyhonokiol (1), obovatol (2), magnolol (3), honokiol (4), eudeshonokiol B (5), eudesobovatol B (6), coumaric acid (7), magnaldehyde B (8), and ${\rho}-coumaric$ acid (9) on the basis of spectroscopic analysis including 2D-NMR and MS data. Compounds 1 - 9 were evaluated for their anti-complement activities against the classical pathway of the complement system. Of them, compound 8 showed significant anti-complement activity on the classical pathway with $IC_{50}$ value of 102.7 ${\mu}M$, whereas compounds 1 - 7 and 9 were inactive. This result indicated that an aldehyde group in the neolignan is important for the anti-complement activity against the classical pathway.

한국산 생약제들의 혈압강하작용에 대한 연구 II. 한국산 후박수피의 혈압강하 작용 (Pharmacological Action of Machilus Thunbergii Siebold Zuccarini)

  • With a view to searching after a new antihypertensive or hypotensive agents in the botanical crude plants, authors intended to reevaluate several natural products caltivated in Korea. This experiment was undertaken to compare pharmacogical actions of Machilus thunbergii Siebold et Zuccarini with those of Magnolia obovata Thunberg in anesthetized rats and in normal mice. Machilus thunbergii Sieb. et Zucc., a tree belonging to the Lauraceae family, is caltivated at Ull-ung Do, and their cortecies have been used as folk medicine mingled with those of Magnolia obovata Thunberg. These two cortecies have teen also applied in chinese medicine, it was advocated that these cortecies exerted good therapeutic effects on gastritis, convulsive abdominal pain, nausea, vomiting and urinary tract disorders. Therefore, we intended to determine the pharmacological action of two palnt of different family each other, especially their effects on blood pressure and heart rate, and also their mechanism of action were observed. We studied their action with extracts of hexane(MTHE), ether(MTEE), methanol(MTME) and water(MTWE) from Machilus thunhergii Sieb. et Zucc., and also fractionations of methanol(MOME), chloroform(MOCE) and water(MOWE) from Mapolia obovata Thunberg. The results of this experiment were as follows; 1) MTME, when intravenously administered to rats, elicited the significant hypotensive responses dependent on the administered dosage. 2) MOWE was also exhibited the hypotensive effect dependent on the treated dose. 3) Depressor effect of MTME was blocked by pretreatment with hexamethonium. 4) The hypotensive response of MOWE was blocked by pretreatment with hexamethonium or hrdralazine. 5) HTME and MOWE were also observed the anticonvulsive effect and sedative effect. These results suggested that MTME may induce the hypotensive response via central sympathetic effect, but the site of action in brain are not clarified, and the hypotensive effect of MOWE may be due to dual mechanism of central sympathetic action and direct vasodilation of blood vessel.

$\beta$-EUDESMOL CAUSES VASODILATORY EFFECT IN THE NORMOTENSIVE RAT

  • $\beta$-Eudesmol is one of various compounds derived from the bark of Magnolia obovata Thunberg, a medicinal plant. It has been shown that $\beta$-eudesmol also markedly alleviated muscle fasciculation, tremor and convulsion induced by diisopropylfluorophosphate and prolonged the time to death in mice (Chiou et al., 1995). Actually, the extract of magnolia bark has been shown to have depressant actions on the cental nervous system (Watanabe et al., 1973). (omitted)

Influence of ${\beta}-Eudesmol$ on Blood Pressure

  • The present study was undertaken to investigate the effects of ${\beta}-eudesmol$, one of various ingredients isolated and identified from the bark of Magnolia obovata Thunberg, on arterial blood pressure and vascular contractile responses in the normotensve rats and to establish its mechanism of action. ${\beta}-Eudesmol\;(30{\sim}300\;{\mu}g/kg)$ given into a femoral vein of the normotensive rat produced a dose-dependent depressor response. These ${\beta}-eudesmol-induced$ hypotensive responses were markedly inhibited in the presence of chlorisondamine (1.0 mg/kg, i.v.) or phentolamine (2.0 mg/kg, i.v.). Interestingly, the infusion of ${\beta}-eudesmol$ (1.0 mg/kg/30min) into a femoral vein made a significant reduction in pressor responses induced by intravenous norepinephrine. Furthermore, the phenylephrine $(10^{-5}\;M)-induced$ contractile responses were depressed in the presence of high concentrations of ${\beta}-eudesmol\;(10{\sim}40\;{\mu}g/ml)$, but not affected in low concentration of ${\beta}-eudesmol\;(2.5{\sim}5\;{\mu}g/ml)$. Also, high potassium $(5.6{\times}10^{-2}\;M)-induced$ contractile responses were greatly inhibited in the presence of ${\beta}-eudesmol\;(10{\sim}40\;{\mu}g/ml)$ in a dose-dependent fashion. Taken together, these results obtained from the present study demonstrate that intravenous ${\beta}-eudesmol$ causes a dose-dependent depressor action in the anesthetized rat at least partly through the blockade of vascular adrenergic ${\alpha}_1-receptors$, in addition to the some unknown mechanism of direct vasorelaxation.

제주 자생 식물들에 대한 항비만 효능에 대한 연구 (The Study on Anti-obesity Activity of the Wild Plants of Jeju Island)

  • 본 연구에서는 제주도에서 자생하는 30여 종의 식물추출물에 대한 항비만 효능을 조사하였다. 항비만 효능은 in vitro oil red-O staining 방법을 이용하여 지방전구세포 3T3-L1에서 분화억제력을 측정하였다. 그 결과, 30여 종의 식물 중에서 약도라지, 호장근, 유근피 등을 포함한 6종의 식물이 지방전구세포 3T3-L1의 분화를 억제하였다. 지방전구세포의 분화를 억제하는 물질 중 유근피, 약쑥, 호장근, 후박 4종은 우수한 항산화 효능도 동시에 가지고 있음을 확인하였다. 이러한 결과를 통해, 본 실험에서 확보된 추출물이 항비만 물질로 사용될 수 있는 가능성을 확인하였다.
