A Comparison between Extract Products of Magnolia officinalis on Memory Impairment and Amyloidogenesis in a Transgenic Mouse Model of Alzheimer's Disease

  • The components of Magnolia officinalis have well known to act anti-inflammatory, anti-oxidative and neuroprotective activities. These efficacies have been sold many products as nutritional supplement extracted from bark of Magnolia officinalis. Thus, to assess and compare neuroprotective effect in the nutritional supplement (Magnolia $Extract^{TM}$, Health Freedom Nutrition LLC, USA) and our ethanol extract of Magnolia officinalis (BioLand LTD, Korea), we investigated memorial improving and anti-Alzheimer's disease effects of extract products of Magnolia officinalis in a transgenic AD mice model. Oral pretreatment of two extract products of Magnolia officinalis (10 mg/kg/day in 0.05% ethanol) into drinking water for 3 months ameliorated memorial dysfunction and prevented $A{\beta}$ accumulation in the brain of Tg2576 mice. In addition, extract products of Magnolia officinalis also decreased expression of ${\beta}$-site APP cleaving enzyme 1 (BACE1), amyloid precursor protein (APP) and its product, C99. Although both two extract products of Magnolia officinalis could show preventive effect of memorial dysfunction and $A{\beta}$ accumulation, our ethanol extract of Magnolia officinalis (BioLand LTD, Korea) could be more effective than Magnolia $Extract^{TM}$ (Health Freedom Nutrition LLC, USA). Therefore, our results showed that extract products of Magnolia officinalis were effective for prevention and treatment of AD through memorial improving and anti-amyloidogenic effects via down-regulating ${\beta}$-secretase activity, and neuroprotective efficacy of Magnolia extracts could be differed by cultivating area and manufacturing methods.

  • It has been known that Magnolia officinalis, Magnolia and Machilus Barks are reputed to be effective in 'Hagi', 'Sanman' and to be analgesic but there was differences of efficacy of these crude drugs against the kinds and habitates. For this reason, this study was conducted to compare the acute toxicities, analgesic effects and motilities of isolated intestine. This results obtained in this work were summarized as follow: 1. In the test of acute toxicies, Machilus bark was found to be more toxic than the others. 2. Both these was found to have analgesic effect. 3. In the effects against the motility of isolated intestine, Magnolia officinalis and Magnolia barks were found to be relaxant, Machilus bark was found to be contractive. From these results, Magnolia officinalis and Magnolia barks might have analgesic effect and relaxant effect of intestine.

Experimental Study on the Antidepressant Effects of Magnolia Officinalis Extracts (후박의 항우울 효과에 대한 실험적 연구)

  • Objectives : The purpose of this study was to investigate the protective effects of Magnolia Officinalis extracts on the animal model of depression induced by immobilization stress. Methods : The subjects were divided into 4 groups : normal, saline solution-administered during immobilization stress, 200 mg/kg of magnolia extracts-administered (magnolia extract 200), and 400 mg/kg of magnolia extracts- administered (magnolia extract 400). During 2 days of immobilization stress treatment, they underwent forced swimming test (FST) and tail suspension test (TST). The number of serotonin (5-HT) immunostained nuclei in the dorsal raphe nucleus regions was measured by immunohistochemistry. Superoxide dismutase (SOD) and glutathione peroxidase (GPX) in blood were measured. Results : In FST, magnolia-administered groups showed significantly decreased immobilization. In TST, the magnolia extract 400 group showed decreased immobilization. The stress group showed significantly decreased number of 5-HT immunostained nuclei in the dorsal raphe nucleus regions, while magnolia extract 400 group showed increased number of 5-HT immunostained nuclei. Stress group showed decrease in serum level of SOD and GPX, while the magnolia extract 200 group showed increase in serum level of SOD and GPX. Conclusions : These results suggest potent effectiveness of magnolia extracts in the treatment of depression.

Effect of Magnolia Obovata Water-extracts on the Atherosclerosis Treated with High-cholesterol Diet in Rat (고(高)콜레스테롤 식이로 유발된 흰쥐 동맥경화에 Magnolia obovata가 미치는 영향)

  • Magnolia obovata exhibit several beneficial effects including anti-oxidant effects. Magnolia obovata is used as a therapeutic agent to stop hemorrhages and a tonic to promoted health in Korean and Chinese medicine. The pharmacokinetic profiles of the main Magnolia obovata is still not accurately investigated. In present study, I examined the effects of water extracts of Magnolia obovata on atherosclerosis induced high cholesterol diet in rats in serum and abdominal aorta. A total of 3-week old 9 male rats of Sprague-Dawley were divided into 3 groups and fed with the basal diet (normal group), high cholesterol diet (atherosclerosis induced group) for 8 weeks, high cholesterol diet supplemented with water extracts of Magnolia obovata (Magnolia obovata group) for 2 weeks. And rats were sacrificed, serum lipid level, abodominal aortic anti-oxidant activities and lipid peroxide were measured. These results indicated that serum total cholesterol, LDL-cholesterol, triglycerides concentration significantly lower in Magnolia obovata group than high cholesterol diet group. But HDL-cholesterol concentration had significantly higher in Magnolia obovata group than high cholesterol diet group. In conclusion, Magnolia obovata controls of lipid level of hyperlipidemia and hypercholesterolemia, thus suppresses the proliferation of intima in endothelial cells, which is risk factors of atherosclerosis induced high cholesterol diet in rats. I consider as Magnolia obovata is worth to using as atherosclerosis treatment.

  • This experiment was carrie out to investigate the amount of essential of leaves and fruits of Magnolia sieboldii, Magnolia hypoleuca, Evodia danillii male and female, Phellodendron amurense male and female and Zanthoxylum schinifolium male and emale by Karlsrube set on August, August, September and October in 1994. Essential oil yield was highest in the fruits of Zanthoxylum schinifollium leaves on Augus the amount of essential oil of Magnolia sieboldii and Magnolia hypoleuca leaves was more than the one of Evodia danillii, Phellodendron amurense and Zanthoxylum schnifolium leaves. The amount of essential oil were not diferent between male and female leaves.

The Effect of Magnolia Bark on the Metabolic Inflammation and Insulin Resistance of ob/ob Mice (후박(厚朴)이 ob/ob 마우스의 대사성 염증과 인슐린 저항성에 미치는 영향 및 관련기전에 대한 연구)

  • Objective: This study was undertaken to investigate how magnolia bark extract affects ob/ob mouse in terms of metabolic inflammation and insulin resistance. Methods: Leptin-deficient ob/ob mice were divided into 2 groups (n=5): a normal saline treatment (=control) and magnolia bark treatment. Wild type mice were the lean group (n=5). After 5 weeks, we measured fasting blood sugar (FBS) and conducted oral glucose tolerance tests (OGTTs) in each group. After 6 weeks, we measured body weight, epididymal fat pad weight, liver weight, serum glucose, serum insulin, and gene expression of tumor necrosis factor-${\alpha}$, interferon-${\gamma}$, and interleukin-6. We characterized the phenotype of adipose tissue macrophages (ATMs) and analyzed fractions of the phenotype in each group by flow cytometry. Results: In the magnolia bark group, fasting blood sugar, oral glucose tolerance levels, and insulin resistance (HOMA-IR) were significantly decreased. The population and proportion of ATMs among leukocytes in adipose tissue were significantly decreased in the magnolia bark group. The population and proportion of M1 type ATMs among ATMs were significantly decreased in the magnolia bark group. Gene expression of tumor necrosis factor-${\alpha}$ was significantly decreased in the magnolia bark group. Conclusions: These results support a positive effect of magnolia bark on metabolic diseases such as insulin resistance and metabolic inflammation in leptin-deficient ob/ob mice.

  • This study is how to effectively abstrcact and transform medicine manufacture from Magnolia biondii to treat nasal allergy. The results are as follows : 1. The original treatment of Magnolia biondii was how to exclude wool and flower center, but form now it have to treat to be crushed, because of effectively abstraction from Magnolia biondii. 2. The original medicine manufacturing form of Magnolia biondii was a boiling, a plaster, a powder, and a insertion, but nowadays in china there are so many kinds of medicine manufactuhng forms. including an injection. 3. Magnolia biondii was recorded on ${\ll}Shinnong-Bonchogyung{\gg}$ at first, but there were no description about nasal allergy. It was described on ${\ll}Mengyebelrok{\gg}$ about nasal allergy at first. 4. A nebulizer is necessary for nasal allergy, western medicine already had it, but on the other hand korea medicine does't have it. 5. We developed new construction for effectively abstrcaction from Magnolia biondiiand, and we succeeded it.

The Dispersal and Plant Community Characteristics of Magnolia obovata - Focused on Case of Korea UNESCO Peace Center area in Gyeonggi-do - (일본목련의 분산 및 식물군집 특성에 관한 연구 -한국유네스코평화센터 주변을 대상으로-)

  • This research had been executed to find out the structure of Magnolia obovata community, a dispersal range and characteristics of scatter investigation, and to prepare management measure of it. In the result of this study, Magnolia obovata was distributed in a native plant community from planting trees of UNESCO Peace Center. The maximum numbers of Magnolia obovata were found in $20{\sim}100m$ from trees of mother. Seed of Magnolia obovata has a characteristics of gravity dispersal. But some individuals was found in 420m from its tree of mother. Seeds of Magnolia obovata has a characteristics of animal dispersal, too. Major species of animal dispersal are Garrulus glandarius, Ciurus vulgaris coreae, and Tamias sibiricus asiaticus in the study site. The importance value between native plants and Magnolia obovata had a negative relation. Because Magnolia obovata is more shade tolerant than other trees. Also, the growth of Magnolia obovata is faster than other trees. And so if the influence of Magnolia obovata reduce, it is necessary to expand native plant community, and to cut Magnolia obovata over 10m of the height and over 10cm of the DBH, for preventing influence of Magnolia obovata.

Effects of Chinese magnolia vine Extract on the Renal Function in Rats (오미자(五味子)의 백서(白鼠) 신장기능(腎臟機能) 및 혈장(血漿) 호르몬 농도(濃度)에 미치는 영향(影響))

  • The aim of this experiments was to investigate the effects of Chinese magnolia vine water extract on the renal function, plasma renin activity, plasma levels of aldosterone and atrial natriuretic peptide in rats. The results of this study were as follows; 1. Water balance decreased significantly after administration with Chinese magnolia vine extracts (0.2 ml/200 g of body weight). 2. Urine volume increased significantly after administration with Chinese magnolia vine extracts. 3. Urinary excretion of sodium increased significantly after administration with Chinese magnolia vine extracts. 4. Urinary excretion of creatinine increased significantly after administration with Chinese magnolia vine extracts (0.2 ml/200 g of body weight). 5. Plasma levels of atrial natriuretic peptide decreased significantly after administration with Chinese magnolia vine extracts (0.1 ml/200 g of body weight). 6. Plasma levels of aldosterone decreased significantly after administration with Chinese magnolia vine extracts (0.2 ml/200 g of body weight).

  • Advanced glycation end products(AGEs) are largly involved in the pathogenesis of diabetic nephropathy. It is obvious that inhibition of AGEs formation is important in preventing the occurrence and progression of diabetic nephropathy. In diabetes, this reaction is greatly accerated and is important in the pathogenesis of diabetic complications, especially diabetic nephropathy. Therefore, to seek possible AGEs inhibitors in herbal medicines, unprocessed - and processed Magnolia Bark were examined in vitro as basic data for aniaml experiment. The content of magnolol in unprocessed Magnolia Bark was $0.796{\pm}0.072%$, and after processing was decreased to $0.586{\pm}0.101%(p<0.01)$. The content of AGEs was measured by their intrinsic fluorescence. The $IC_{50}({\mu}g/ml)$ values of aminoguanidine, unprocessed- and procesled Magnolia Bark are $38.845{\pm}8.36{\mu}g/ml$, $54.264{\pm}3.153{\mu}g/ml$ and $27.882{\pm}1.836{\mu}g/ml$, respectively. This result means that prcessed Magnolia Bark was more effective than aminoguanidine, as positive control.
