다층구조의 미세유체채널을 이용한 자성입자 분리 (Magnetic beads separation using a multi-layered microfluidic channel)

  • This paper presents the design and experiment results of a multi-layered microsystem for magnetic bead applications. The magneto-microfluidic device is designed for capable of separating magnetic beads. In the presence of the magnetic field, magnetic beads are attracted and moved to high gradient magnetic fields. A multi-layered microfluidic channel consists of top and bottom layers in order to separate magnetic beads in the vertical direction. Our channel is easily integrated magnetic cell sorter, especially on-chip microelectromagnet or permanent magnet device. Fast separation of magnetic beads in top and bottom channels can be used in high throughput screening to monitor the efficiency of blood and drug compounds.

  • An automatic control system is proposed and implemented for a miniaturized DNA extraction system using magnetic bead. A host-local system is employed for the accommodation of the graphical user interface and the basic control function. The functional partitioning into the local and the host system is discussed. The control functions are classified and formalized for the flexible control scenario, which is the input of the proposed system. As the proposed scenario is consists of the sequence of the user-centric actions, the user goal can be easily programmed and modified. The DNA extraction performance of the implemented system was compared with the existing silica-membrane-based method, resulting in the comparable concentration and purity of the extracted DNA. The proposed system is currently being utilized for the development of the DNA extraction system only changing scenario, without any alteration of the system.

Immobilized Saccharomyces cerevisia의 반응특성에 관한 연구 (Studies on the Reactive Characteristics of Immobilized Saccharomyces cerevisia in Ethanol Production)

  • In an attempt to develop the immobilized biocatalysts based on immobilized Saccharomyces cerevisiae, immobilized yeast was investigated with respect to the conditions affected to ethanol productivities. Saccharomyces cerevisiae was immobilized in the form of the beads by magnetic-calcium alginate, non magnetic-calcium alginate and acrylamide polymerization. Magnetic immobilized yeast, nonmagnetic immobilized yeast and polyacrylamide immobilized yeast were compared in respect of their pH stability, thermostability, heat tolerance, the relation between the concetration of native yeast and retained activity of immobilized yeast, the activity depending on bead size of immobilized yeast, and the effects of magnesium and cobalt on the activities. The more small bead had retained the higher activity for the three kinds of immobilized yeast. In case of 1.0mm diameter of beads, the retained activity was 40~50% for the all groups. The pH stability profile for the immobilized yeast showed a broad range of optimun activity while the native yeast gave a sharp pick for its optimun pH value. The thermostability was at the range of 25~55$^{\circ}$C for the immobilized yeast groups. It was investigated that the influent magnesium and cobalt concentration, and the relative activity have an influent on heat tolerance at steady state. Both protein content released from immobilized yeast and activity of immobilized yeast were changed after activation of immobilized yeast cell.

  • We developed a microfluidic platform able to control the trap and release of magnetic beads used for separation of a specific biomolecules. The magnetic beads can be trapped and released conditionally by controlling the difference between the Stokes force induced by the fluid flow and magnetic force resulting from a permanent magnet. The permanent magnet of CoNiP alloy is electroplated. It is characterized to have the 1369 Oe of coercivity, 1762 Gauss of remanence, and 0.603MGOe of (BH)max. Through the experimental and numerical investigation, the magnetic beads are trapped under the flow velocity of 17 ${\mu}m/s$ and are released perfectly above the velocity of 174 ${\mu}m/s$.

Direct Detection of Escherichia coli, Staphylococcus aureus, and Salmonella spp. in Animal-derived Foods Using a Magnetic Bead-based Immunoassay

  • In this study, an immuno-magnetic bead (IMB)-based assay was developed to simultaneously detect Escherichia coli, Staphylococcus aureus, and Salmonella spp. and was tested in four animal-derived foods: beef, ham, egg, and ricotta cheese. The IMB-based assay exhibited good specificity by binding to five E. coli serotypes [capture efficiency (CE) average (avg.) 90.4%], five S. aureus strains (CE avg. 91.4%), and five Salmonella serotypes (CE avg. 95.4%) but not binding to non-target bacteria (CE<10%). Furthermore, the assay detected all three pathogens with a detection limit of 10 CFU/g without the need for enrichment or additional platforms. Since the results demonstrated that the IMB-based assay can effectively separate and enrich target bacteria from a variety of animal-derived food matrixes, the assay exhibits good specificity for potential use in providing rapid, immunological, presumptive identification of pathogenic bacteria.

적혈구 포획용 미크론 크기 코일에 흐르는 전류의 크기에 따른 자기장 분포 특성 (Distribution of Magnetic Field Depending on the Current in the μ-turn Coil to Capture Red Blood Cells)

  • 광 리소그래피 공정을 이용하여 반강자성체 IrMn 층을 기반으로 하는 GMR-SV(giant magnetoresistance-spin valve) 소자 위에 ${\mu}m$ 선폭 크기의 코일을 적층하여 미크론 크기 코일을 제작하였다. GMR-SV 박막일 때와 소자 제작 후의 자기저항비와 자장감응도는 각각 4.4 %, 2.0 %/Oe와 1.6 %, 0.1 %/Oe로 나타났다. 여러 개의 $1{\mu}m$ 크기인 자성비드가 붙은 적혈구 바로 아래 위치에 놓일 소자와 10번 감은 미크론 크기 코일에 흐르는 AC 및 DC 전류 크기에 따른 자기장 분포를 유한요소법 전산모사로 분석하였다. 코일 중심인 $z=0{\mu}m$에서 주파수 20 kHz인 AC 전류가 0.1 mA에서 10.0 mA로 증가하면 이 값에 비례하여 자기장의 크기는 $30{\mu}T$에서 $3060{\mu}T$로 증가하였다. 그리고 코일 중심에서부터 $z=10{\mu}m$에서 자기장 크기는 $z=0{\mu}m$인 중심에서 보다 1/6배로 줄어들었다. 미크론 크기 코일에 흐르는 전류에 의해 생성되는 자기장은 적혈구 포획용으로 충분한 크기를 갖고 있어서 검출용 바이오센서로 활용 가능함을 확인 할 수 있었다.

  • Effects of electromagnetic force which is one of the most important factor of metal transfer that affects bead geometry and microstructure of weld metal in GMAW(gas metal arc welding). In this paper, different ways of external electromagnetic forces were applied on GMAW process and their effects on the welding were studied. On certain conditions, better bead geometry, better influence on the arc and metal transfer mode and higher welding efficiency could be obtained. Experimental methods and their results will be presented.

Multiplexed single-molecule flow-stretching bead assay for DNA enzymology

  • Single-molecule techniques have been used successfully to visualize real-time enzymatic activities, revealing transient complex properties and heterogeneity of various biological events. Especially, conventional force spectroscopy including optical tweezers and magnetic tweezers has been widely used to monitor change in DNA length by enzymes with high spatiotemporal resolutions of ~nanometers and ~milliseconds. However, DNA metabolism results from coordination of a number of components during the processes, requiring efficient monitoring of a complex of proteins catalyzing DNA substrates. In this min-review, we will introduce a simple and multiplexed single-molecule assay to detect DNA substrates catalyzed by enzymes with high-throughput data collection. We conclude with a perspective of possible directions that enhance capability of the assay to reveal complex biological events with higher resolution.
