Oleanolic acid regulates NF-κB signaling by suppressing MafK expression in RAW 264.7 cells

  • Oxidative stress and inflammation are common to many pathological conditions. Defense mechanisms protect cells from oxidative stress, but can become over-activated following injury and inflammation. NF-${\kappa}B$ and Nrf2 transcription factors regulate proinflammatory and antioxidant gene expression, respectively. Studies have shown that many natural dietary compounds regulate NF-${\kappa}B$ and Nrf2, preventing inflammation and oxidative stress. Here, we report major compounds of Prunella vulgaris var. lilacina such as rosmarinic acid, oleanolic acid, ursolic acid and caffeic acid as a potential therapeutic for oxidative stress and inflammation. The major compounds exhibited high anti-inflammatory activity, inhibiting NO, PGE2 production, NF-${\kappa}B$ expression and activating Nrf2 expression. In addition, we examined the effect of major compounds on MafK expression. Among the compounds, oleanolic acid significantly decreased MafK expression and MafK-mediated p65 acetylation. These findings suggest that oleanolic acid as NF-${\kappa}B$ inhibitors can potentially be used in therapeutic applications for the treatment of oxidative stress-induced diseases.

Negative regulators in RANKL-induced osteoclastogenesis

  • Receptor activator of nuclear factor ${\kappa}B$ ligand (RANKL) induces osteoclast formation from hematopoietic cells via up-regulation of positive regulators, including $NF-{\kappa}B$, c-Fos, microphthalmia transcription factor (Mitf), PU.1, and nuclear factor of activated T cells (NFAT) c1. In addition to the positive regulation by these transcription factors, RANKL appears to regulate negative regulators such as MafB and inhibitors of differentiation (Ids). Ids and MafB are abundantly expressed in osteoclast precursors, bone marrowderived monocyte/macrophage lineage cells (BMMs). Expression levels of these genes are significantly reduced by RANKL during osteoclastogenesis. Overexpression of these genes in BMMs inhibits the formation of tartarate-resistant acid phosphatase (TRAP)-positive multinuclear osteoclasts by down-regulation of NFATc1 and osteoclast-associated receptor (OSCAR), which are important for osteoclast differentiation. Furthermore, reduced expression of these genes enhances osteoclastogenesis and increases expression of NFATc1 and OSCAR. Taken together, RANKL induces osteoclastogenesis via up-regulation of positive regulators as well as down-regulation of negative regulators.

PCR 기법을 이용한 인공감염토양 및 감염동물 장기로 부터 Bacillus anthracis의 검출 (Establishment of PCR to detect Bacillus anthracis in the experimentally infected soil and mice)

  • Anthrax caused by Bacillus anthracis is one of the most important zoonotic diseases in the worldwide. To control and prevent the disease effectively, several methods such as development of a fast and specific diagnostic method and vaccine, education etc, have been carried out. However, it still has a problem in the control and prevention. To control, the most important method is the prevention of direct or indirect contact of the causative agent with susceptible host. Therefore, we developed a fast and specific detection method, polymerase chain reaction, of B anthracis from soil and infected animals because the organism could survive long time in the environment including soil due to formation of spore. With the method, virulence genes of B anthracis were successfully amplified from experimentally infected soil and mice. Up to $4.2{\times}10$ of the organisms per gram could be detected with the PCR method from experimentally infected soil. These results suggested that this PCR method could be effectively used not only to detect B anthracis in soil and infected animal but also to provide the information to prevent the disease.

Enhancement of In Vitro Regeneration of Several Ocimum Species and Varieties

  • Tissue culture systems to optimize regeneration plant species of Ocimum spp were evaluated as a method to micropropagate individual plants and to better study their biology in vitro. Ocimum species were also evaluated for the production of natural plant products during and following the regeneration process. The primary goal of this project was to enhance the regeneration efficiency of basil. Several factors were examined using different Ocimum species and commercial varieties. The effect of cytokinin combination, activated charcoal, gelling agents, and different carbon sources were investigated. Anthocyanin callus spots were produced only in four varieties among six tested. 'Sweet Dani' showed the best results on anthocyanin accumulation, while 'African beauty', 'Tree basil' and 'Methylcinnamate' produced only a few spots. Shoot regeneration was only achieved from 'Sweet Dani' explants. As the activated charcoal concentration increased, callus formation rate decreased respectively compare to the controls for all varieties. There was a decrease in callus growth with increasing concentration of agar and phytagel.

Brucella canis에 감염된 개의 항생제 치료효과 (Therapeutic value of antibiotics in dogs infected with Brucella canis)

  • In a kenel of 62 dogs, 33 were diagnosed as infected dogs Brucella canis by serological test and blood cultures in a large kennel in Chonnam area. Twenty eight of 33 dogs were treated with combined antibiotics therapy consisting of tetracycline and dihydrostregtomycin. After the first treatment, all dogs became abacteremic and serologic titers declined. Abortion due to B canis infection could be prevented by antibiotic therapy during pregnancy. However, sequential antibiotics therapy for 4 weeks did not eradicate B canis from affected bitch. According to results of serological and blood culture tests, effect of antibiotics treatment respectively revealed that 17 of the originally infected dogs were cure with second therapy schedule at 2 months and 27 of that with 6 months after second therapy.

천마가 RANKL에 의해 유도된 파골세포의 분화에 미치는 효과 (Effects of Gastrodia elata Blune Water Extract on RANKL-induced Osteoclast Differentiation)

  • Impairment of balance between bone-resorbing osteoclasts and bone-forming osteoblasts result in bone disease. Especially, increased osteoclast formation and activity are responsible for bone diseases such as osteoporosis, rheumatoid arthritis, periodontal disease. Natural metabolites of plants have recently received much attention as an alternative tools for the development of novel therapeutic strategy. The aim of this study was to search the natural products to inhibit osteoclast differentiation and was to evaluate of its mechanism. Water extract of Gastrodia elata Blune significantly inhibited receptor activator of nuclear factor-κB ligand (RANKL)-induced osteoclast differentiation in bone marrow macrophages (BMMs) in a dose dependent manner. However, water extract of Gastrodia elata Blune did not affect cytotoxicity when compared with control. The mRNA expression of c-Fos, NFATc1, and TRAP induced by RANKL was inhibited by water extract of Gastrodia elata Blune treatment. Also, water extract of Gastrodia elata Blune inhibited the protein expression of c-Fos and NFATc1 expression in BMMs treated with RANKL. Water extract of Gastrodia elata Blune suppressed the phosphorylation of p38 induced by RANKL. In general, RANKL considerably inhibited the expression level of Id2 and MafB known as negative regulators of osteoclastogenesis, but RANKL did not inhibit Id2 and MafB expression in BMMs when it was co-treated with Gastrodia elata Blune. Taken together, these results suggest that Gastrodia elata Blune may be a useful drug in the treatment of bone-related disease.

전남지방의 소형견 번식장으로부터 발생한 canine brucellosis (Occurance of canine brucellosis in a large kennel in Chonnam area)

  • Reproductive failures, such as abortions were occured among dogs in a large kennel in Chonnam area April in 1994. After an initial positive result by the 2 mercaptoethal rapid slide agglutination test(2ME-RSAT) to Brucella canis on five sera, additional specimens from all dogs in the population were tested. The blood cultures and 3 following serological tests (2ME-RSAT, TAT, AGID) performed on all samples on the basis of surveys, the following results were obtained. 1. Thirty three of 62 dogs were seropositive. 2. Twenty blood samples from 33 dogs were cultured, all of the isolates were identified as B canis. 3. Although there was not siginificant difference in sex, age and breed of the cause brucellosis dectected, abortions was occured late in gestation stage.

In vitro에서 Levamisole이 양식뱀장어의 면역조절작용에 미치는 영향 (Effect of Levamisole on Immunomodulation of Eels (Anguilla japonica) In Vitro)

  • Levamisloe(LMS)이 뱀장어의 면역반응에 미치는 영향을 측정하기 위해서 뱀장어의 혈액 및 신장으로부터 백혈구를 분리하여, 림프구의 증식능, MIF 생산능, 자연살해세포활성능, 탐식세포의 탐식능, superoxide anion, hydrogen peroxide 및 lysozyme 활성능 등을 측정하였다. LMS는 T-cell 마이토젠인 Con A 및 PHA 처리시에는 LMS 농도에 비례감소를 보인 반면에 B-cell 마이토젠인 LPS에는 LMS 처리와는 무관하게 큰 차이를 보이지 않았다. 한편 MIF 및 MAF의 생성에는 LMS 처리 농도에 비례 증가되었다. NK 세포의 활성은 LMS 첨가농도에 비례 증가하였는데, 이는 표적세포에 대한 결합율을 증가시킨 결과 NK 세포의 활성이 증가된 것으로 보인다. LMS는 백혈구의 탐식능, superoxide anion의 생성능, hydrogen peroxide 생성능 및 Iysozyme의 활성을 증가시켰다. 이러한 결과로부터 LMS가 어체의 면역작용에 관여하는 작용기전은 면역 작동세포수의 증가를 촉진시키기보다는 분화를 촉진시켜서 cytokine의 방출증가 및 세포의 기능활성을 증강시킴을 알수 있다.
