STUDIES ON ANTITUMOR AGENTS PRODUCED BY STREPTOMYCES spp. ISOLATED IN KOREA

  • 1993년 국내토양으로부터 분리동정된 Streptomyces속 균주중 항종양성이 우수한 균주 3주(DKM104, DKM117, DKM409)를 선정하고 이들을 대량 배양하여 종양억제인자를 분리하고 시험관내 종양세포 독성능 및 생체내 항암활성능과 이들 물질생산과 PLASMID DNA와의 관련성, $LD_{50}$등을 시험하여 새로운 항종양물질의 개발을 목적으로 하였다. 분리선별균주의 배양조건을 확립하였으며 배양여액을 국성이 적은 유기용매로부터 큰쪽으로 단계적으로 유효성분을 추출하여 Gel Chromatography를 이용하여 유효성분을 분획하였다. 시험관내 항종양능 시험은 MTT colorimetric검정법을 이용하여 $IC_{50}$값을 산정하였으며 생체내 항종양능은 마국의 NIC에서 권장하는 tumor panel system에 따랐다. 선정된 균주의 plasmid 분리는 alkalin lysis법을 채택하였으며 agarose gel electrophoresis로 plasmid profile을 시험하였다. Novobiocin등을 이용하여 Curing test를 시행하였고 독성실험은 $LD_{50}$량을 구해 항암효과 측정시에 Maximum dost로 투여하였고 최고용량을 기준으로 일정한 공비를 적응하여 3단계의 투여량을 설정하였다.(중략)

Cytotoxic Anthraquinones and Stilbenes from Reynoutria sachalinensis (Fr. Schm.) Nakai

  • Five known anthraquinones, physcion (1), I-O-methylemodin (2), emodin (3), $physcion-8-O-{\beta},-D-glucopyranoside$ (5), $emodin-8-O-{\beta},-D-glucopyranoside$ (6) and two known stilbenes, trans-resveratrol (4), $trans-resveratrol-3-O-{\beta},-D-glucopyranoside$ (7) were isolated from MeOH extract of Reynoutria sachalinensis (Polygonaceae). All structures were unambiguously established by 1D and 2D NMR and MS data and the compounds were evaluated for their cytotoxicity against L1210, HL-60, BI6F10 tumor cell lines in MTT assay. Among the compounds, trans-resveratrol (4) exhibited significant cytotoxic activity with $IC_{50}$ values of 9.2, 6.7 and $9.8\;{\mu}g/ml$, against the test cell lines respectively, but compounds 1-3 exhibited the moderate cytotoxic activity.

Biological Activities of Larix kaempferi Needles

  • The needles of L. kaempferi was extracted with 95% ethanol and successively partitioned with n-hexane, $CH_2Cl_2$ and EtOAc. Repeated column chromatography on the EtOAc and $H_2O$ soluble fractions gave three flavan-3-ols, one flavone glycoside, six flavonol glycosides and one lignan derivative. Their structures were elucidated on the basis of chemical and spectroscopic evidences. The antioxidant activities of the isolated compounds were evaluated by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging method. Flavan compounds indicated good antioxidative potentials compared with BHT (butylated hydroxytoluene) and ${\alpha}$-tocopherol as controls. In the anti-inflammatory test on most of the isolated compounds, NO (nitric oxide) assay against the RAW 264.7 (Mouse Macrophage) showed similar inhibitory potentials to NO production of the control. The cytotoxicity was determined by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay and most of the isolated compounds indicated no toxicity in various concentration.

  • 23-Dihydroganoderic acid N, a triterpenoid compound, was investigated whether it may show cytotoxic activity against U937, HeLa, NCI-H460 and MCF-7 cancer cells by MTT test. As a result, 23-dihydroganoderic acid N show sensitive to MCF-7 cells among NCI-H460, HeLa, U937, and MCF-7 cells.

  • Ecklonia cava is a brown alga(Laminariaceae) that is abundant in the subtidal regions of Jeju island in Korea. Phlorotannins were identified to be responsible for the biological activities in Ecklonia species. In the present study, triphlorethol-A, a phlorotannin, was isolated from Ecklonia cava and its anticancer properties were investigated. Triphlorethol-A was investigated whether it may show cytotoxicity effects against U937, HeLa, NCI-H460 and MCF-7 cancer cells by MTT test. As a result, triphlorethol-A did not show cytotoxic effects against tested four cell lines.

  • The action of epigallocatechin-3-gi:lllale (EGCG). polyphenol compound from green lea, on the release pattern of glycosylphosphatidylinositol (GPI)-anchored renal dipeptidase (RDPase) from renal proximal tubules (PTs) was examined. EGCG had a stronger inhibitory effect on the release of RDPase than alkaline phosphatase (APase), another GPI-anchored ectoenzyme used as a reference protein. The effect of EGCG on cell viability as assessed by MTT test was found to be intact, and moreover, was indicative of potent cell activation or proliferation. (omitted)

  • Objectives: The purpose of this study is to review experimental studies of Parkinson's Disease in Korean medical journals, which have been published since 1990. Methods: Experimental studies related to Parkinson's disease published in Korean medical journals, registered with Korea research foundation, were searched. Articles were grouped into journal of publication, subject, and publication year. Experimental study subjects and models were analysed. Results and Conclusions: Twenty-two articles about experimental studies of Parkinson's Disease in Korean medical journals were included in our review. There were 13 articles in which herbs were used for treatment, 4 articles in which bee venom was used for treatment, 4 articles in which acupuncture was used for treatment, and 1 article in which pharmacopuncture was used for treatment. Most of the studies used experimental models of MPTP-induced Parkinson's disease and MTT assay was used to analyse cell toxicity and cell viability. Pole test, rotarod test, Morris water maze test, and forced swimming test were commonly used to analyse MPTP-induced behavioral deficits in animals. Analysis on experimental studies of Parkinson's disease was categorized into the subjects of dopaminergic cell protective effect, cell-apoptosis protectiveness, anti-inflammatory effect, and anti-oxidative effect.

Effect of Steamed Codonopsis lanceolata on Spatial Learning and Memory in Mice (증숙 더덕 추출물의 인지능력 개선 효과)

  • Alzheimer's disease is progressive neurodegenerative disorder by the loss of memory and learning abilities. Codonopsis lanceolata (C. lanceolata) is traditional medicinal plant used for the treatment of inflammatory diseases. The aim of study was to evaluate the effect of steamed C. lanceolata on scopolamine-induced memory impairment in the Morris water maze test and passive avoidance test. In addition, this study investigated the neuroprotective effects of steamed C. lanceolata on glutamate-induced cell death in HT22 cells using MTT assay. The results showed that steamed C. lanceolata (500 mg/kg body weight, p.o.) reversed spatial memory impairment by scopolamine in Morris water maze test and passive avoidance test. Steamed C. lanceolata attenuated memory impairment by scopolamine compared with common C. lanceolata. In addition, administration of steamed C. lanceolata significantly also reduced cell death. We suggest that steaming process more improve cognitive enhancing and neuroprotective effect of C. lanceolata than common C. lanceolata.

A Study on Oxidation Behavior and Cytotoxicity Test of Ti-10Ta-10Nb Alloy (생체용 타이타늄 합금의 산화거동 및 세포독성에 관한 연구)

  • A new Ti-10Ta-10Nb alloy has designed and examined some possibility of forming more passive oxide film by oxidation treatment which is closely related to corrosion resistance and biocompatibility. Ti-6Al-4V and Ti-10Ta-10Nb alloys were prepared by consumable vacuum arc melting and homogenized at 1050$^{\circ}C$ for 24hours. Alloy specimens were oxidized at the temperature range of 400 to 750$^{\circ}C$ for 30minutes, and the oxide films on Ti alloys were analysed by optical microscope, SEM, XPS and TGA. Cytotoxicity test was performed in MTT assay treated L929 fibroblast cell culture by indirect method. It is found out that the oxide film on Ti-10Ta-10Nb alloy is denser and thinner compared to Ti-6Al-4V alloy. The weight gain during the oxidation was increased rapidly at the temperature above 650$^{\circ}C$ for Ti-6Al-4V alloy and above 700$^{\circ}C$ for Ti-10Ta-10Nb alloy respectively. It was analysed that the passive film of the Ti alloys consisted of TiO2 through X-ray photoelectron spectroscopy (XPS) analysis. It is found out by cytotoxicity test that moderate oxidation treatment lowers cell toxicity, and Ti-10Ta-10Nb alloy showed better result compared to Ti-6Al-4V alloy.

Cytotoxic effects of different self-adhesive resin cements: Cell viability and induction of apoptosis

  • PURPOSE. The effects of four different self-adhesive resin cement materials on cell viability and apoptosis after direct and indirect exposure were evaluated using different cell culture techniques. MATERIALS AND METHODS. Self-adhesive cements were applied to NIH/3T3 mouse fibroblasts by the extract test method, cell culture inserts, and dentin barrier test method. After exposure periods of 24 h and 72 h, the cytotoxicity of these self-adhesive materials was evaluated using the MTT assay (viability) and the Annexin-V-FITC/PI staining (apoptosis). RESULTS. The lowest cell viability was found in cells exposed to BeautiCem SA for 24 h in the extract test method. Cell viability was reduced to 70.6% compared to negative controls. After the 72 h exposure period, viability rate of cell cultures exposed to BeautiCem SA decreased more than 2- fold (29.5%) while cells exposed to RelyX U200 showed the highest viability rate of 71.4%. In the dentin barrier test method, BeautiCem SA induced the highest number of cells in apoptosis after a 24 h exposure (4.1%). Panavia SA Cement Plus was the material that caused the lowest number of cells in apoptosis (1.5%). CONCLUSION. The used self-adhesive cements have showed different cytotoxic effects based on the evaluation method. As exposure time increased, the materials showed more cytotoxic and apoptotic effects. BeautiCem SA caused significantly more severe cytotoxic and apoptotic effects than other cements tested. Moreover, cements other than BeautiCem SA have caused necrotic cell death rather than apoptotic cell death.
