Cytotoxic activity of 1-phenyl-2-alkylsulfonylamido propanol derivatives

  • The 20 alkylsulfonylamido propanol derivatives had been investigated for their cytotoxic activity against HT-29 colon cancer, Caki-2 renal cancer, A549 lung cancer, PC-3 prostate cancer, HL-60 leukemia cell using MTT assay. Cytotoxic activity was strongly influenced by the substituted alkyl chain length and the optimal alkyl chain length for cytotoxicity was C11. Some of alkylsulfonylamido porpanol derivatives showed stronger activity than reference compound, B13.

Cytotoxic Effects of Chloroform Extracts and Fraction from Cornis fructus on Cancer Cell Lines

  • Cornis fructus were extracted by successive extractions and then fractionated with chloroform extract to get active fractions. This study was performed to determine the cytotoxic effect of chloroform extract from Cornis fructus on NIH 3T3 fibroblasts and cancer cell lines using MTT assay. All extracts did not exhibit cytotoxicity in HIH 3T3 fibroblasts. Chloroform extract exhibited antitumor activity in A549, MDA-MB-123, B16 melanoma and SNU-C4 cells. Futher fractionation with chloroform extract was performed to obtain effective fractions. (omitted)

  • The 20 thiourea derivatives had been investigated for their cytotoxic activity using MTT assay. Cytotoxic activity was strongly influenced by the substituted alkyl chain length. but not by configuration of Cl and C2. The optimal alkyl chain length for cytotoxicity was C12. The 9 thiourea derivatives showed stronger activity than reference compound. B13. some of them gave 2-3 times stronger activity than B13. (omitted)

만병초 잎 추출물의 유전 독성과 사람의 암세포주 등에 대한 세포독성 (Genotoxicity and Cytotoxicity in Human Cancer and Normal Cell Lines of the Extracts of Rhododendron brachycarpum D. Don leaves)

  • 강원도 산지에서 채취한 만병초 잎의 70% ethanol 추출물과 다른 유기용매 분획 과정을 통한 3가지의 분획물에 대한 in vitro에서의 정상세포에 대한 세포독성과 유전독성, 여러 암세포에 대한 항암 기능, 면역세포에 대한 면역기능 등을 조사하여 다음과 같은 결과를 얻었다 1. 만병초 잎의 EtOH 추출물과 각 용매 분획물 들의 사람의 정상 세포인 HEL 299와 Chang에 대한 세포 독성을 SRB assay로 측정한 결과, 대부분의 시료 0.5 mg/ml 이하의 농도에서 35% 이하의 세포 독성이 나타났다. 2. 만병초 잎의 70% EtOH 추출물 및 각 다른 용매 분획물만 처리한 정상 세포에 대한 유전독성을 comet assay에서 측정한 결과, 양성 대조구 $(10^{-2}M,\;H_2O_2)$과 비교시, 가장 높은 농도인 1 mg/ml의 농도에서 1/3 이하의 낮은 DNA damage가 관찰되었다. 이것은 세포독성과 관련하여 대부분의 각 시료가 세포 DNA에 대하여 매우 약한 손상만 일으키는 것이 확인되었다. 3. 사람의 암세포에 대한 SRB 및 MTT assay를 이용한 만병초의 잎의 모든 시료의 1 mg/ml의 농도의 70% EtOH의 추출물은 실험대상인 모든 사람의 암세포에 대하여 80% 이상의 항암 기능을, EtOAc fr.는 모든 암세포에 대하여 가장 높은 농도인 1 mg/ml에서 70% 이상의 높은 억제 활성이 보인 결과로 부터 유효한 성분이 역시 EtOAc fr.에 함유되어 있을 것을 시사하였다. 4. 만병초 잎 추출물의 면역세포 생육에 대한 조사에서 사람의 B-cell과 T-cell에 대하여 0.5 mg/ml의 농도에서 배양 3일째에 BuOH fr.에서 대조구와 비교시 1.6배 정도의 생육 촉진활성을 보였다. 5. SCGE를 통해 $H_2O_2$로 유도된 산화적 손상에 대한 만병초 잎 추출물과 분획물들의 유전독성 억제 기능은 EtOAc fr.이 $100\;{\mu}g/ml$ 농도에서 대조구 대비 68.5%의 가장 높은 산화손상 억제율을 보였으며 BuOH fr.에서도 강한 유전독성 억제기능이 있음이 확인되었다. 그러나 수용액 분획은 매우 약한 유전독성 억제 활성을 보여 각 분획의 특성이 다양함을 알 수 있었다.

In Vitro Chemosensitivity Test of SK-302B on Human Colon Carcinoma Cell Lines

  • SK-302B, an antibiotic purified from soil Streptomyces sp. 302, was structurally identified as echinomycin (C/sub 50/H/sub 66/N/sub 11/S/sub 2/). In the present experiment, the possibility of SK-302B as an anticolon cancer agent was investigated by using chemosensitivity system (MTT assay, clonogenic assay). Treatment of SK-302B on various colon cancer cell lines resulted in a significant cytotoxicity and tumor colony formation inhibition. These studies showed that SK-302B had a potent inhibition on colon cancer cells.

마우스 대식세포(Raw 264.7)에 대한 한약조성물 KOCO-P1의 세포활성 연구 (Study on Biological Effect of Multi-Herbal Drug KOCO-Pl on Mouse Macrophage Raw 264.7 Cells)

  • Objectives : The purpose of this study is to investigate the biological Effect of multi-herbal drug 'KOCO-P1' on mouse macrophage Raw 264.7 cells. Methods : Multi-herbal drug 'KOCO-P1' was composed of Ginseng Radix, Astragali Radix, Polygonati Rhizoma, Liriopis Tuber, and Scrophulariae Radix. Cytotoxicity and cytoprotective activity of K0C0-P1 was verificated by MTT assay. And antioxidative effect of K0C0-P1 against EtOH, Nicotine was inspected by Hydroperoxide assay. Results : K0C0-P1 showed no cytotoxicity on RAW 264.7 cells for 24, 48, 72 hours. KOCO-P1 at 200, 100, and 50 ug/mL reduced the production of H202 in Raw 264.7 cells by EtOH. KOCO-P1 at 50 ug/mL reduced the production of H202 in Raw 264.7 cells by Nicotine. Conclusions : KOCO-P1 could be supposed to have antioxidative effect on macrophage with no cytotoxicity.

A novel cis/trans-diaminocyclohexane platinum coordination complexes possessing in vitro and in vivo antitumor activity

  • As part of a drug discovery program to develope more effective platinum-based anticancer drugs, a series of platinum complexes trans -diaminocyclohexane platinum bi sdiphenylphosphino -ethane (KHPC-002) cis-diaminocyclohexane platinum bisdiphenylphosphino-ethane (KHPC-006) has been evaluated in vitro against 4 human carcinoma cell lines with those of cisplatin using a tetrazolium-based colorimetric assay (MTT assay). The cell lines were two human bladder carcinoma cell lines, HT-1197 and HT-1376, human colon carcinoma cell line, HCT-116, and prostate cancer cell line DU-145.

  • Zinc is known to have an inhibitory effect on apoptosis and an antioxidative effect scavenging reactive oxygen species (ROS) under oxidative stress. We studied the influence of zinc on cadmium-induced apoptosis especially associated with ROS in MCF-7 human breast carcinoma cell line. For the determination of appropriate experimental concentration and time, we excecuted MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide] assay and DNA fragmentation assay. 인진쑥 추출물의 항산화 및 항암 활성 (Antioxidant and Anticancer Activities of Extract from Artemisia capillaries)

  • Artemisia capillaries is a major important food and medicinal resource in Korea. In order to confirm the biological activities of Artemisia capillaries, we investigated antioxidant and anticancer activities from in vitro assays. The Artemisia capillaries methanol (MeOH) extracts was used for the evaluation of DPPH scavenging, total phenolic content, total flavonoid content, hydroxyl radical (${\bullet}OH$) scavenging, reducing power assay as antioxidant activity, as well as anticancer activities as MTT assay. As a result, the Artemisia capillaries MeOH extracts showed potent antioxidative activity and anticancer activity in vitro. These results suggest that the Artemisia capillaries MeOH extracts have a potential alleviated oxidation process, cell motility activity, and tumorigenesis.
