Keratanase II Digestion Accompanied with a Liquid Chromatography/Tandem Mass Spectrometry for Urinary Keratan Sulfate Quantitative Analysis

  • Purpose: Mucopolysaccharidosis IV (MPS IV) is a disease characterized by deficient activity of N-acetylgalactosamine-6-sulfatase (GALNS) causing excessive lysosomal storage of keratan sulfate (KS). The identification of the relevant disaccharide units of KS after keratanase II digestion followed by liquid chromatography/tandem mass spectrometry detection (LC-MS/MS) is validated and applicable for the preliminary diagnosis of MPS IV. Methods: A total of 67 urine samples were collected and analyzed from 11 MPS IV patients comprising 10 MPS IVA and one MPS IVB patients, and 56 normal controls. Urinary glycosaminoglycan was first precipitated by the Alcian blue method followed by a digestion of keratanase II. The protonated species of the digested disaccharide products were detected by using multiple reaction monitoring experiment. Results: One particular disaccharide of KS was selected. The transition mass-to-charge (m/z) of the parent ion and its daughter ion after collision was $462.0{\rightarrow}97.0$, whereas the chondrosine used as an internal standard in this assay was m/z $353.9{\rightarrow}73.0$. The results corresponded well with the two-dimensional electrophoresis method. The quantities of urinary KS were significantly raised in confirmed MPS IV patients when comparing with those of normal controls ($170.2{\pm}81.1$ vs. $4.06{\pm}1.92{\mu}g/mL$). Conclusion: The LC-MS/MS method for MPS IVA determination is specific, sensitive, validated, and applicable for urinary KS quantification. This method can be used not only as a first-line biochemistry examination of MPS IVA, but also as an outcome survey after enzyme replacement therapy.

  • The violent free-surface motions and the corresponding impact loads are numerically simulated by using the Moving Particle Semi-implicit (MPS) method, which was originally proposed by Koshizuka and Oka (1996) for incompressible flows. In the original MPS method, there were several shortcoming including non-optimal source term, gradient and collision models, and search of free-surface particles, which led to less-accurate fluid motions and non-physical pressure fluctuations. In the present study, how those defects can be remedied is illustrated by step-by-step improvements in respective processes of the revised MPS method. The improvement of each step is explained and numerically demonstrated. The numerical results are also compared with the experimental results of Martin and Moyce (1952) for dam-breaking problem. The current numerical results for violent free-surface motions and impact pressures are in good agreement with their experimental data.

Using a Lagrangian-Lagrangian approach for studying flow behavior inside a bubble column

  • Bubble columns are widely encountered in several industries, especially in the field of nuclear safety. The Eulerian-Eulerian and the Eulerian-Lagrangian methods are commonly used to investigate bubble columns. Eulerian approaches require additional tasks such as strict volume conservation at the interface and a predefined well-structured grid. In contrast, the Lagrangian approach can be easily implemented. Hence, we introduce a fully Lagrangian approach for the simulation of bubble columns using the discrete bubble model (DBM) and moving particle semi-implicit (MPS) methods. Additionally, we propose a rigorous method to estimate the volume fraction accurately, and verified it through experimental data and analytical results. The MPS method was compared with the experimental data of Dambreak. The DBM was verified by analyzing the terminal velocity of a single bubble for each bubble size. It agreed with the analytical results for each of the four drag correlations. Additionally, the improved method for calculating the volume fraction showed agreement with the Ergun equation for the pressure drop in a packed bed. The implemented MPS-DBM was used to simulate the bubble column, and the results were compared with the experimental results. We demonstrated that the MPS-DBM was in quantitative agreement with the experimental data.

  • PDA와 스마트폰 등의 모바일 단말기 사용이 증가됨에 따라 새로운 결제수단으로서 모바일결제시스템(Mobile Payment System, MPS)이 많이 활용되고 있다. 이에 따라, 국내외 MPS 사용자들은 시공간의 제약에서 벗어나 본인이 원하는 제품 및 서비스를 편리하게 구매 혹은 이용할 수 있게 되었다. 이러한 상황에서, 본 연구는 MPS 사용자들의 특성(인구통계적 특성, MPS 이용 특성)에 따라 MPS의 가치에 대한 인식도가 어떻게 달라지는가를 파악하고자 하였으며, 이에 더하여 MPS의 가치에 대한 인식도가 궁극으로 사용자 만족도에 유의미한 영향을 미치는가를 파악하고자 하였다. 연구목적을 달성하기 위해서, 본 연구에서는 국내에서 MPS 이용 경험자들을 대상으로 배포한 설문지를 이용하여 다양한 통계분석을 수행하였다. 본 연구에서 제시된 실증적 분석결과는 향후 국내외에서 MPS의 성공적 확산 및 사용자 만족도 제고방안을 모색하는데 있어서 유용하게 활용될 수 있다고 판단된다.

MPS eutectic reaction model development for severe accident phenomenon simulation

  • During the postulated severe accident of nuclear reactor, eutectic reaction leads to low-temperature melting of fuel cladding and early failure of core structure. In order to model eutectic melting with the moving particle semi-implicit (MPS) method, the eutectic reaction model is developed to simulate the eutectic reaction phenomenon. The coupling of mass diffusion and phase diagram is applied to calculate the eutectic reaction with the uniform temperature. A heat transfer formula is proposed based on the phase diagram to handle the heat release or absorption during the process of eutectic reaction, and it can combine with mass diffusion and phase diagram to describe the eutectic reaction with temperature variation. The heat transfer formula is verified by the one-dimensional melting simulations and the predicted interface position agrees well with the theoretical solution. In order to verify the eutectic reaction models, the eutectic reaction of uranium and iron in two semi-infinite domains is simulated, and the profile of solid thickness decrease over time follows the parabolic law. The modified MPS method is applied to calculate Transient Reactor Test Facility (TREAT) experiment, the penetration rate in the simulations are agreeable with the experiment results. In addition, a hypothetical case based on the TREAT experiment is also conducted to validate the eutectic reaction with temperature variation, the results present continuity with the simulations of TREAT experiment. Thus the improved method is proved to be capable of simulating the eutectic reaction in the severe accident.

Cochlear Implantation via the Transmeatal Approach in an Adolescent with Hunter Syndrome-Type II Mucopolysaccharidosis

  • Type II mucopolysaccharidosis (MPS II) commonly known as Hunter syndrome, is a rare X-linked lysosomal storage disorder caused by iduronate-2-sulfatase deficiency, which in turn causes otorhinolaryngological manifestations, including sensorineural hearing loss (SNHL). Previously, the median survival age of patients with MPS was approximately 13.4 years. However, in the era of enzyme replacement therapy and other multidisciplinary care modalities, the life expectancy has increased. Herein, we report a rare case of an adolescent with MPS II who underwent SNHL treatment with cochlear implantation (CI). Based on unexpected findings of mastoid emissary veins and overgrowth of the vessels around the temporal bone, CI was performed using the transmeatal approach instead of the conventional transmastoid method, to avoid damage to the vessels. The average hearing threshold after CI was 35 dB and no surgical complications were encountered. Adolescent MPS II may present vessel abnormalities, which can reduce the success rate of surgery. In patients with MPS II with SNHL, CI should be performed under careful monitoring of vessel overgrowth. Moreover, with regard to feasibility of CI in adolescent patients with MPS II with SNHL, surgical techniques such as the transmeatal approach should be selected based on adequate assessment of the case.

크기가 제어된 실리카 나노입자 합성과 제조된 입자의 고분자계 복합재 응용 (Synthesis of Silica Nanoparticles Having the Controlled Size and their Application for the Preparation of Polymeric Composites)

  • 고분자계 치아수복용 복합재료에 사용되는 다양한 크기의 나노실리카 입자를 Stober method를 이용하여 제조하고, 3-methacrylofpropyltrimethoxysilane $(\gamma-MPS)$로 제조된 입자 표면 처리 여부에 따른 유기 수지 내에서의 실리카 입자의 분산도 변화를 조사하였다 반응에 사용된 반응물인 tetraethylorthosilicate(TEOS)와 물의 양, 촉매인 암모니아의 양, 용매의 종류와 양을 조절하여 다양한 크기의 나노실리카 입자를 제조하였다. 용매로 에탄올보다 메탄올을 사용할 경우 더 작은 입자가 생성되었다. 또, 물의 함량이 증가할수록 작은 입자가 형성되는 반면, 촉매와 전구체의 경우는 함량이 증가함에 따라 형성된 입자의 크기도 증가하였다. 유기 소재들과의 혼합시 계면 특성 향상을 위해 제조된 친수성의 나노실리카를 $\gamma-MPS$와 반응시켜 소수성의 나노실리카 입자를 제조하였다 실리카 입자 크기가 작을수록 단위 질량당 존재하는 $\gamma-MPS$의 함량은 많았지만, 단위 표면적당 존재하는 $\gamma-MPS$의 양은 실리카 입자의 크기에 영향을 받지 않았다. $\gamma-MPS$로 표면 처리된 실리카 입자의 고분자계 치아수복용 레진 내에서의 분산성은 표면 처리되지 않은 실리카에 비해 크게 향상되었다.

Thermosensitive Chitosan as an Injectable Carrier for Local Drug Delivery

  • Two types of injectable system using thermosensitive chitosan (chitosan-g-NIPAAm), hydrogel and microparticles (MPs)-embedded hydrogel were developed as drug carriers for controlled release and their pharmaceutical potentials were investigated. 5-Fluorouracil (5-FU)-loaded, biodegradable PLGA MPs were prepared by a double emulsion method and then simply mixed with an aqueous solution of thermosensitive chitosan at room temperature. All 5-FU release rates from the hydrogel matrix were faster than bovine serum albumin (BSA), possibly due to the difference in the molecular weight of the drugs. The 5-FU release profile from MPs-embedded hydrogel was shown to reduce the burst effect and exhibit nearly zero-order release behavior from the beginning of each initial stage. Thus, these MPs-embedded hydrogels, as well as thermosensitive chitosan hydrogel, have promising potential as an injectable drug carrier for pharmaceutical applications.

슬로싱 해석을 위한 CCUP 기반 시뮬레이션 기술 개발 (DEVELOPMENT OF A NUMERICAL SIMULATION METHOD FOR THE ANALYSIS OF SLOSHING PROBLEMS BASED ON CCUP SCHEME)

  • A new computational program, which is based on the CIP/CCUP(Constraint Interpolation Profile/CIP Combined Unified Procedure) method, has been developed to numerically analyse sloshing phenomena dealt as multiphase-flow problems. For the convection terms of Navier-Stokes equations, the RCIP(Rational function CIP) method was adopted and the THINC-WLIC(Tangent of Hyperbola for Interface Capturing-Weighted Line Interface Calculation) method was used to capture the air/water interface. To validate the present numerical method, two-dimensional dam-breaking and sloshing problems in a rectangular tank were solved by the developed method in a stationary Cartesian grid system. In the case of sloshing problems, simulations by using a improved MPS(Moving Particle Simulation) method, which is named as PNU-MPS(Pusan National University-MPS), were also carried out. The computational results are compared with those of experiments and most of the comparisons are reasonably good.
