Overexpression of tumor necrosis factor receptor-associated protein 1 (TRAP1), leads to mitochondrial aberrations in mouse fibroblast NIH/3T3 cells

  • Cancer cells undergo uncontrolled proliferation, and aberrant mitochondrial alterations. Tumor necrosis factor receptor-associated protein 1 (TRAP1) is a mitochondrial heat shock protein. TRAP1 mRNA is highly expressed in some cancer cell lines and tumor tissues. However, the effects of its overexpression on mitochondria are unclear. In this study, we assessed mitochondrial changes accompanying TRAP1 overexpression, in a mouse cell line, NIH/3T3. We found that overexpression of TRAP1 leads to a series of mitochondrial aberrations, including increase in basal ROS levels, and decrease in mitochondrial biogenesis, together with a decrease in peroxisome proliferator-activated receptor gamma coactivator-$1{\alpha}$ (PGC-$1{\alpha}$) mRNA levels. We also observed increased extracellular signal-regulated kinase (ERK) phosphorylation, and enhanced proliferation of TRAP1 overexpressing cells. This study suggests that overexpression of TRAP1 might be a critical link between mitochondrial disturbances and carcinogenesis.

MOUSE TRAP 모델을 이용한 하수관거내 수질변화 예측 (Estimation of Water Quality Variation in Sewer Network using MOUSE TRAP Model)

  • One of the major problems associated with operation of domestic sewer lines involves hydraulic problems such as insufficient conveyance capacity, exceeding maximum velocity, and deficiency of minimum velocity. It has also been pointed out that influent concentration lower than design concentration of pollutants, which is mainly caused by unidentified inflow and infiltration, degrades the operational efficiency of many sewage treatment plants (STPs). A computer-added analysis method supporting a coupled simulation of sewage quality and quantity is essentially required to evaluate the status of existing STPs and to improve their efficiency by a proper sewer rehabilitation work. In this study, dynamic water quality simulations were conducted using MOUSE TRAP to investigate the principal parameters that governs the changes of BOD, ${NH_4}^+$, and ${PO_4}^{3-}$3- concentrations within the sewer networks based on data acquired through on-site and laboratory measurements. The BOD, ${NH_4}^+$ and ${PO_4}^{3-}$3- concentrations estimated by MOUSE TRAP was lower than theoretical pollution loads because of sedimentation and decomposition in the sewer. The results revealed that sedimentation is a most important factor than other biological reactions in decreasing pollutant load in the sewers of C-city. The sensitivity analysis of parameters pertaining to water quality changes indicated that the effect of the BOD decay rate, the initial DO concentration, the half-saturation coefficient of dissolved BOD, and the initial sediment depth is marginal. However, the influence of settling rate and temperature is relatively high because sedimentation and precipitation, rather than biological degradation, are dominant processes that affect water quality in the study sewer systems.

  • RIKEN BioResource Center (BRC) has collected, preserved, conducted quality control of, and distributed mouse resources since 2002 as the core facility of the National BioResource Project by the Ministry of Education, Culture, Sports, Science and Technology (MEXT), Japan. Our mouse resources include over 5,000 strains such as humanized disease models, fluorescent reporters, and knockout mice. We have developed novel mouse strains such as tissue-specific Cre-drivers and optogenetic strains that are in high demand by the research community. We have removed all our specified pathogens from the deposited mice and used our quality control tests to examine their genetic modifications and backgrounds. RIKEN BRC is a founding member of the Federation of International Mouse Resources and the Asian Mouse Mutagenesis and Resource Association, and provides mouse resources to the one-stop International Mouse Strain Resource database. RIKEN BRC also participates in the International Gene Trap Consortium, having registered 713 gene-trap clones and their sequences in a public library, and is an advisory member of the CREATE (Coordination of resources for conditional expression of mutated mouse alleles) consortium which represents major European and international mouse database holders for the integration and dissemination of Cre-driver strains. RIKEN BRC provides training courses in the use of advanced technologies for the quality control and cryopreservation of mouse strains to promote the effective use of mouse resources worldwide.

Effects of Leptin on Osteoclast Generation and Activity

  • Leptin, the product of the obese gene, is a circulating hormone secreted primarily from adipocytes. Several results suggest that leptin is important mediators of bone metabolism. The present study was undertaken to determine the effects of leptin on anti-osteoclastogenesis using murine precursors cultured on Ca-P coated plates and on the production of osteoprotegerin (OPG) in osteoblastic cells. Additionally, this study examined the possible involvement of prostaglandin $E_2\;(PGE_2)$/protein kinase C (PKC)-mediated signals on the effect of leptin on anti-osteoclastogenesis to various culture systems of osteoclast precursors. Osteoclast generation was determined by counting tartrate-resistant acid phosphatase positive [TRAP (+)] multinucleated cells (MNCs). Osteoclastic activity was determined by measuring area of resorption pits formed by osteoclasts on Ca-P coated plate. The number of 1,25-dihydroxycholecalciferol $(1,25[OH]_2D_3)$- or $PGE_2$-induced TRAP (+) MNCs in the mouse bone marrow cell culture decreased significantly after treatment with leptin. The number of receptor activator of NF-kB ligand (RANKL)-induced TRAP (+) MNCs in M-CSF dependent bone marrow macrophage (MDBM) cell or RAW264.7 cell culture decreased significantly with leptin treatment. Indomethacin inhibited osteoclast generation induced by $1,25[OH]_2D_3$ and dexamethasone, however, no significant differences were found in the leptin treated group when compared to the corresponding indomethacin group. Phorbol 12-myristate 13-acetate (PMA), a PKC activator, inhibited osteoclast generation induced by $1,25[OH]_2D_3$. The number of TRAP (+) MNCs decreased significantly with treatment by PMA at concentrations of 0.01 and $0.1{\mu}M$ in culture. Leptin inhibited PMA-mediated osteoclast generation. Isoquinoline-5-sulfonic 2-methyl-1-piperazide dihydrochloride (H7) had no effect on osteoclast generation induced by $1,25[OH]_2D_3$. Cell culture treatment with leptin resulted in no significant differences in osteoclast generation compared to the corresponding H7 group. Indomethacin showed no significant effect on TRAP (+) MNCs formation from the RAW264.7 cell line. PMA inhibited TRAP (+) MNCs formation induced by RANKL in the RAW264.7 cell culture. H7 had no effect on osteoclast generation from the RAW264.7 cell line. There was no difference compared with the corresponding control group after treatment with leptin. $1,25[OH]_2D_3$- or $PGE_2$-induced osteoclastic activity decreased significantly with leptin treatment at a concentration of 100 ng/ml in mouse bone marrow cell culture. Indomethacin, PMA, and H7 significantly inhibited osteoclastic activity induced by $1,25[OH]_2D_3$ in mouse bone marrow cell culture. No significant differences were found between the leptin treated group and the corresponding control group. The secretion of OPG, a substance known to inhibit osteoclast formation, was detected from the osteoblasts. Treatment by leptin resulted in significant increases in OPG secretion by osteoblastic cells. Taken these results, leptin may be an important regulatory cytokines within the bone marrow microenvironment.

Mouse phenogenomics, toolbox for functional annotation of human genome

  • Mouse models are crucial for the functional annotation of human genome. Gene modification techniques including gene targeting and gene trap in mouse have provided powerful tools in the form of genetically engineered mice (GEM) for understanding the molecular pathogenesis of human diseases. Several international consortium and programs are under way to deliver mutations in every gene in mouse genome. The information from studying these GEM can be shared through international collaboration. However, there are many limitations in utility because not all human genes are knocked out in mouse and they are not yet phenotypically characterized by standardized ways which is required for sharing and evaluating data from GEM. The recent improvement in mouse genetics has now moved the bottleneck in mouse functional genomics from the production of GEM to the systematic mouse phenotype analysis of GEM. Enhanced, reproducible and comprehensive mouse phenotype analysis has thus emerged as a prerequisite for effectively engaging the phenotyping bottleneck. In this review, current information on systematic mouse phenotype analysis and an issue-oriented perspective will be provided.

마우스 수정란의 초기 배 발생단계별 Telomeric DNA의 양적 분석과 Telomerase 활성도 분석 (Analysis of the Amount of Telomeric DNA and Telomerase Activity on Preimplantation Mouse Embryoic Cells)

  • 텔로미어란 염색체 말단부에 (TTAGGG)n의 반복 염기서열이 단백질과 결합된 형태를 말하는데 이의 역할은 염색체의 안정성에 본질적으로 작용하여 세포의 노화, 사멸 및 암의 발생과 관련이 있다고 알려져 있다. 반면 텔로머레이스는 telomeric DNA 합성에 직접 관여하는 ribonucleoprotein이다. 본 연구에서는 마우스 염색체의 텔로미어 분포 양상을 제시하고, 초기 배 발생단계별 수정란의 텔로미어 함량과 이들 수정란의 텔로머레이스 활성도를 분석하고자 하였다. 본 분석에는 마우스의 섬유아세포, 생식세포, 정자, 난자 및 1세포기, 2세포기, 4세포기, 8세포기, 상실배와 배반포배의 각 단계별 수정란을 대상으로 하였다. 텔로미어의 양적 분석은 human telomeric DNA probe를 이용한 Q-FISH 방법을 이용하였고, 텔로머레이스 활성도는 TRAP 방법을 이용하였다. 분석 결과 마우스 염색체의 텔로미어는 성 염색체를 포함한 모든 염색체의 앙 말단부에 분포되어 있고, 염색체별 다소의 양적 차이를 보이나 대부분의 염색체에서 q-arm 말단이 p-arm 말단에 비해 높은 텔로미어의 함량을 나타내었다. Q-FISH를 이용한 마우스 초기 배 발생단계별 수정란의 텔로미어의 양적 분석에서 수정 직후 1세포기에서부터 상실배까지 거의 비슷한 텔로미어 함유율을 나타내고 있으나 배반포기에서 월등히 증가된 양상을 나타내었다. TRAP 분석을 이용한 초기배아의 텔로머레이스 활성도는 초기 배 발생 모든 단계에서 이의 활성도를 나타내었으며, 특히 상실배 및 배반포기에서 점진적으로 강한 활성을 보였다. 이상의 분석 결과로부터 마우스의 초기 배 분열단계의 각 세포들에 있어 텔로미어의 함유율과 텔로머레이스 활성도는 높은 상관관계가 있는 것으로 나타났다. 따라서 포유동물의 초기 배자에 있어 텔로미어의 함유율과 텔로머레이스 활성도는 배 발생 및 배자의 세포 분화와 매우 밀접한 관련이 있는 것으로 사료되어 텔로미어의 양적 분석 및 텔로머레이스 활성도 분석은 발생학적 연구를 위한 또 다른 좋은 자료로 생각된다.

치자 추출물이 RANKL 유도 파골세포 형성 및 골 흡수에 미치는 영향 (Effects of Gardeniae Jasminoides on RANKL-induced Osteoclastogenesis and Bone Resorption)

  • Objectives: This study was performed to investigate the effects of Gardenia jasminoides extract (GJ) on osteoclast differentiation and bone resorption in vitro. Methods: To investigate the effect of GJ on osteoclast differentiation, the mouse leukemic myeloid cell line RAW 264.7 was stimulated by RANKL (receptor activator of nuclear factor kB ligand). Osteoclast differentiation was measured by counting TRAP (+) MNC in the presence of RANKL. To elucidate the mechanism of the inhibitory effect of GJ on osteoclast differentiation, gene expression of TRAP, Cathepsin K, MMP-9, NFATc1, c-Fos, MITF, DC-STAMP, CTR, OC-STAMP and Atp6v0d2 was measured using reverse transcription-PCR (RT-PCR). Bone resorption was measured using the bone pit formation assay. Results: GJ decreased the number of TRAP (+) MNCs in the presence of RANKL. GJ inhibited the expression of cathepsin K, MMP-9, TRAP, MITF, NFATc1, c-Fos, iNON, OC-STAMP, Atp6v0d2, and DC-STAMP in the osteoclast, and inhibited bone pit formation in vitro. Conclusions: The results suggest that GJ has inhibitory effects on bone resorption resulting from inhibition of osteoclast differentiation and gene expression.

애엽 추출물이 골 대사에 미치는 영향 (Effects of Artemisia princeps Extract on Bone Metabolism)

  • 본 연구에서는 마우스 골수유래 파골세포와 난소절제 동물모델을 이용하여 애엽 추출물이 골대사에 미치는 영향을 살펴보았다. 애엽 추출물이 파골세포의 분화 및 활성에 미치는 영향을 알아보기 위하여 마우스 골수유래 전구세포에 M-CSF와 RANKL을 처리하여 파골세포로의 분화를 유도한 후, TRAP 염색법을 실시하고 TRAP 활성도를 측정하였다. 그 결과 애엽 추출물은 TRAP 양성 다핵세포의 형성과 TRAP 활성을 농도 의존적으로 감소시켰으며, 파골세포의 골흡수 또한 애엽 추출물의 최고농도($30\;{\mu}g/mL$) 처리 시약 90%까지 억제되는 것을 확인하였다. 난소절제 동물모델을 이용한 실험에서도 애엽 추출물이 난소절제에 따른 급격한 골밀도 감소를 막아준다는 사실을 확인할 수 있었다. 이상의 결과로 미루어 볼 때, 애엽 추출물이 에스트로겐 분비 감소로 인하여 급격한 골 손실이 나타나는 폐경기 여성의 골다공증 예방에 효과가 있을 것으로 사료된다.

한약재 추출물의 조골세포 분화 및 파골세포 형성에 미치는 영향 (Effects of Medicinal Herb Extracts on Osteoblast Differentiation and Osteoclast Formation)

  • 천연물 유래의 생약에서 조골세포 증식을 높이면서, 파골세포의 분화 억제에 효과가 있는 시료를 검색하고자 15종의 한약재추출물의 효과를 검토해 보았다. Mouse calvaria 유래의 osteoblastic cells를 이용하여 세포 생존률 및 ALP 활성을 측정하였으며, 또한 마우스 골수 세포를 이용하여 M-CSF와 RANKL을 처리하여 파골세포의 분화를 유도한 후, 세포 생존율과 TRAP효소활성을 측정하였다. 그 결과, 두충, 곽향, 개다래, 형개, 정공등 추출물은 조골세포 증식 및 ALP 활성를 증가시켰으며, 파골세포 활성을 나타내는 TRAP활성이 억제되는 것으로 확인되었다. 따라서 이 추출물들은 조골세포의 기능을 향상시키는 동시에 파골세포의 기능을 억제하여 골 흡수와 관련된 질환과 함께 골 질환 예방에 효과가 있을 것으로 생각되어진다.
