Changes in Microbial Counts, Enzyme Activity and Quality of Foxtail Millet Takju Treated with High Hydrostatic Pressure during Storage (초고압 처리한 좁쌀탁주의 저장 중 미생물수, 효소활성 및 품질변화)

  • Foxtail Millet Takju was treated with heat ($65^{\circ}C/30\;min$) (HT) or high hydrostatic pressure ($27^{\circ}C/400\;MPa/10\;min$) (PT), and changes in microbial count, enzyme activity, and quality were determined during 30-day storage at 10 and $25^{\circ}C$. Total viable cellcount remained constant, while lactic acid bacteria and yeast were not detected in HT and PT Takjus. Relative activities of ${\alpha}-amylase$ in PT Takju significantly increased by 169.7% at 3 days storage, then decreased to 137.7 and 68.7% at 10 and $25^{\circ}C$, respectively, at 30 days. Relative activities of glucoamylase in HT Takju showed reversible change, and were 36.5 and 54.3% at $10\;and\;25^{\circ}C$, respectively, at 30 days storage. Activities in PT Takju increased with storage period, 158.2% at 30 days storage at $10^{\circ}C$. Titratable acidity in untreated Takju increased, while those in HT and PT Takjus remained almost constant during 30 days storage. Reducing sugar content in untreated Takju showed no change, while that in HT Takju increased gradually, reaching 2.9% at 30 days, whereas that in PT increased sharply after 3 days, reaching 4.8% at 30 days. Sensory evaluation showed sourness and bitterness were low, and sweetness and overall acceptance were high in PT Takju after 30 days storage at $10^{\circ}C$.

Regulation of Photosynthesis Genes (puf, puc, puhA, bchC, bchE, bchF, and bchI) in Rhodobacter sphaeroides (Rhodobacter sphaeroides에서의 광합성유전자(puf, puc, puhA, bchC, bchE, bchF와 bchI)의 발현조절)

  • Here we examined the expression patterns and regulation of seven photosynthesis (PS) genes (puf, puc, puhA, bchC, bchE, bchF, and bchI) in the anoxygenic photosynthetic bacterium, Rhodobacter sphaeroides, based on lacZ reporter gene assay. Expression of the tested PS genes, except puhA and bchI, were strongly induced in R. sphaeroides grown under anaerobic conditions relative to that under aerobic conditions. The puhA and bchI genes appear to form the operons together with bchFNBHLM-RSP0290 and crtA, respectively. Expression of the puf, puc, and bchCXYZ operons in R. sphaeroides grown photosynthetically was proportional to the incident light intensity, whereas that of bchFNBHLM(RSP0290-puhA) was inversely related to light intensity. Expression of bchEJG was lowest under medium-light photosynthetic conditions $(10\;W/m^2)$ and highest under high light conditions $(100\;W/m^2)$. The regulation of PS genes by the three major regulatory systems involved in oxygen- and light-sensing in R. sphaeroides is as following: puf and bchC are regulated by both the PpsR repressor and the PrrBA two-component system. The puc operon is under control of PpsR, FnrL, and PrrBA system. Expression of bchE is controlled by FnrL and PrrBA two-component system, whereas bchF is regulated exclusively by PpsR. It was demonstrated that the PpsR repressor is responsible for high-light repression of bchF and that FnrL might be involved in perceiving the cellular redox state in addition to sensing $O_2$ itself.

Acceleration of Mycelial growth of Lentinus edodes in Coniferous Sawdust (침엽수 톱밥에서 표고 균사생장 촉진에 관한 연구)

  • In Lentinus edodes(oak mushroom) cultivation, commonly are logs and sawdusts of oak and some other broadleaved tree species used. Recently oak trees have been substantially diminished due to extensive logging. Thus, to develop comparable synthetic formula using other tree species for the cultivation of Lentinus edodes, we investigated the effect of various nutrients and pretreatment on L. edodes mycelial growth in coniferous sawdusts(i.e., Pine and Larch). We found that 1.5 hr pretreatment of sawdust with hot water and adding 10% rice bran, 3% charcoal, 0.02% $NH_4CI$ and 0.5-1% lignosulfornic acid were effective for the growth of L. edodes in pine sawdust media. In larch sawdust pretreatment with acetone for one hr and adding 20% rice bran, 3% charcoal and 0.02% $NH_4CI$ increased L. edodes mycelial growth. We also analyzed the components of oak and coniferous sawdusts and found oak has higher content of xylose and lower content of lignin, arabinose and mannose than conifers. Rice bran, compared with BITEL(HOKKEN Co.) known for better commercial substitute for rice bran, has lower content of xylose and galactose, but the similar C/N ratio.

Metalloproteinase Plays a Role in Mucin Secretion (Mucin 분비에 영향을 미치는 Metalloproteinase)

  • Background : Mucus hypersecretion in the patients with airway diseases represents poor prognosis as well as discomfort. However, there is no known therapy for its effective control. One important component of mucus is mucin, a glycosylated protein, which endows mucus with viscosity. We studied whether a proteinase has a role in mucin secretion and if so, which. Methods : (1) Inhibition of mucin secretion Group-specific proteinase inhibitors were tested to evaluate whether a proteinase belonging to a group of proteinases plays a role in mucin secretion. Phenylmethylsulfonyl fluoride(PMSF, a serine proteinase inhibitor), E-64(a cysteine proteinase inhibitor), Pepstatin(an aspartic proteinase inhibitor) and 1, 10-Phenanthroline(a metalloproteinase inhibitor) were treated into the Calu-3 cell line for 24 hours. The enzyme linked immunoabsorbant assay(ELISA) for MUC5AC was performed to evaluate the amount of mucin secretion and to compare with a control. (2) Stimulation of mucin secretion Matrix metalloproteinase-9(MMP-9), MMP-12 and TACE(TNF-alpha converting enzyme), which are known to be related with airway diseases, were used to be treated into Calu-3 for 24 hours. ELISA for MUC5AC was performed to evaluate the amount of mucin secretion and to compare with the controls. Results : (1) PMSF($10^{-4}M$), E-64($10^{-4}M$), Pepstatin($10^{-6}M$) and 1, 10-Phenanthroline($10^{-4}M$) reduced the MUC5AC secretion by $1{\pm}4.9%$(mean${\pm}$standard deviation; P=1.0 compared with the control), $-6{\pm}3.9%$(P=0.34), $-13{\pm}9.7%$(P=0.34) and $41{\pm}8.2%$(P=0.03), respectively. (2) The amounts of MUC5AC secretion stimulated by MMP-9(250ng/ml), MMP-12(100ng/ml) and TACE(200ng/ml) were $103{\pm}6%$(P=0.39), $102{\pm}8%$(P=1.0) and $107{\pm}13%$(P=0.39), respectively, compared with the controls. Conclusion : Metalloproteinase(s) is (are) suggested to play a role in mucin secretion. It appears that metalloproteinases, other than MMP-9, MMP-12 or TACE, affect the mucin secretion in this in vitro model.

  • Chronic obstructive pulmonary disease (COPD) is a substantially under-diagnosed disorder, and the diagnosis is usually delayed until the disease is advanced. However, the benefit of early diagnosis is not yet clear, and there are no guidelines in Korea for doing early diagnosis. This review highlights several issues regarding early diagnosis of COPD. On the basis of several lines of evidence, early diagnosis seems quite necessary and beneficial to patients. Early diagnosis can be approached by several methods, but it should be confirmed by quality-controlled spirometry. Compared with its potential benefit, the adverse effects of spirometry or pharmacotherapy appear relatively small. Although it is difficult to evaluate the benefit of early diagnosis by well-designed trials, several lines of evidence suggest that we should try to diagnose and manage patients with COPD at early stages of the disease.

Antibacterial Activities of Trace Elements in Combination with Food Additives (미량원소 강화 식품소재의 항균효과)

  • Antibacterial activities of the trace elements in combination with the food additives were measured against 6 kinds of food-borne microorganisms such as Escherichia coli, Vibrio parahaemolyticus, Staphylococcus aureus, Bacillus cereus, Bacillus subtilis and Pseudomonas fluorescens. The difference of antibacterial activity was not shown among the kinds of food additives, such as dextrin, gelatin and collagen. Zn and Ge in combination with food additives had strong antibacterial effect. Especially, $1\%$ zinc acetate in combination with $1\%$ gelatin was more effective against P. fluorescens and S. aureus than against Bacillus sp., E. coli and V. parahaemolyticus. Minimum inhibitory concentration of zinc acetate in combination with $1\%$ gelatin appeared to be 0.5 mg/mL on S. aureus and P. fluorescens, and 1.0 mg/mL on E. coli, V. parahaemolyticus, B. cereus and B. subtilis. Minimum bactericidal concentration of zinc acetate in combination with $1\%$ gelatin appeared to be 0.5 mg/mL on P. fluorescens and 1.0 mg/mL on E. coli, V. parahaemolyticus, S. aureus, B. cereus and B. subtilis. The zinc acetate in combination with gelatin showed stronger inhibitory effect in acidic range below pH 6.0, and remained active even after heat treatment at $121^{\circ}C$ for 15 min. In comparison with control, the viable cell counts of fish pastes, which were coated with the solution containing both $1\%$ zinc acetate and $3\%$ gelatin, were decreased by more than 100-fold until the storage of 7 days at $10^{\circ}C$. The results indicate that the combined use of zinc acetate and some food additives could prolong the shelf life of fish pastes by 8 days or more at $10^{\circ}C$.

Contents of Heavy Metals in Fishes from the Korean Coasts (한국 연안산 어류의 중금속 함량)

  • We collected 177 fishes representing 53 species of fish from the eastern (Pohang), western (Gunsan), and southern (Tongyeong) coasts of Korea, and measured their heavy metal contents. The mean recoveries of the heavy metals extracted from cod muscle (certified reference material, CRM) were $88.7{\sim}100.6%$. The mean levels of the heavy metals in the samples taken from the edible portion of each fish were high in the order of Zn ($8.981{\pm}4.835{\mu}g/g$), Cu ($0.755{\pm}0.507{\mu}g/g$), and Mn ($0.433{\pm}0.699{\mu}g/g$), which are necessary metals in the human body, and then followed by Cr ($0.206{\pm}0.181{\mu}g/g$), Ni ($0.081{\pm}0.110{\mu}g/g$), Pb ($0.038{\pm}0.046{\mu}g/g$), Cd ($0.017{\pm}0.023{\mu}g/g$). The average daily intakes of the heavy metals by the fishes were as follows: Cd (0.81 ${\mu}g$), Cr (9.98 ${\mu}g$), Cu (36.63 ${\mu}g$), Mn (21.01 ${\mu}g$), Ni (3.93 ${\mu}g$), Pb (1.84 ${\mu}g$) and Zn (435.58 ${\mu}g$). The average weekly intakes of Cd, Cu, Pb and Zn by the fishes were 1.35%, 0.12%, 0.86%, and 0.73% respectively, as compared with PTWI (Provisional Tolerable Weekly Intakes) established by FAO/WHO Expert Committee for Food Safety Evaluation.

Clinicopathologic Features and Prognostic Factors for Patients with Large Gastric Tumors (대형 위암의 임상병리학적 분석 및 예후)

  • Purpose: Tumor size has been reported to be one of the prognostic factors in the preoperative setting and 8 cm has been confirmed as a cut-off value for large gastric tumors with respect to postoperative complications. The aim of this study was to investigate the clinicopathologic features and the prognosis in patients with tumors larger than 8 cm in diameter. Materials and Methods: We retrospectively studied 2,260 patients with gastric cancer who underwent a gastrectomy from 1983 to 2001 at the Department of Surgery, Korea University College of Medicine. For a comparative analysis we divided the cases into the large and the small groups according to tumor size. The clinicopathological factors associated with large gastric tumors were analyzed by using univariate and multivariate analyses. To determine which variables were independent prognostic factors for overall survival, we applied the Cox proportional hazards model and we used P<0.05 as the cutoff value for statistical significance. Results: Univariate and multivariate analyses disclosed that tumor location (P<0.001), resection type (P<0.001), curability (P<0.001), depth of invasion (P<0.001), number of metastatic lymph nodes (P<0.001), differentiation (P<0.001) and combined resection (P<0.001) were significantly different between the two groups. The independent factors for survival identified by using the Cox proportional hazards model for large gastric tumors were nodal status (P<0.001), curative resection (P<0.001), depth of invasion (P=0.010), type of resection (P=0.018) and age (P=0.033). Conclusion: Large gastric tumors showed more aggressive local findings than their smaller counterparts. In patients with large gastric tumors, a curative resection was the most important factor for the prognosis. Therefore, we suggest that every effort should be made to do a curative gastrectomy and an accurate preoperative examination. (J Korean Gastric Cancer Assoc 2006;6:244-249)

Clinicopathological Features and Prognostic Factors for Patients with Clinical T4 Gastric Cancer that Underwent Combined Resection of Invaded Organs (위암의 주위 장기 침윤으로 합병 절제를 실시한 환자의 병리학적 병기 및 예후)

  • Purpose: The surgical treatment of gastric cancer that invades adjacent organs is a radical gastrectomy with combined resection including the adjacent organs or a palliative operation by performing either a gastrojejunostomy or gastrectomy. However, since it is impossible to determine the exact stage of the cancer, either T or N, in the case of palliative surgery, it is inappropriate to predict patient prognosis. This study analyzes the prognoses for patients whose final TNM stages are determined by a combined resection performed due to macroscopical infiltration into the adjacent organs. Materials and Methods: Of 2,452 patients that underwent surgery for gastric cancer at our hospital from 1983 to 2002, we evaluated 102 patients where a combined resection was performed because direct infiltration into the adjacent organs was discovered. Results: Univariate analysis showed that the survival rate differed by the depth of invasion into the gastric walls, the degree of lymph node metastasis, distant metastasis, pathological TNM stage, surgical curability, the location of tumor, and histological differentiation. By multivariate analysis, it was found that the surgical curability, the location of the tumor and the degree of lymph node metastasis were independent prognostic factors. Conclusion: It is suggested that even when infiltration into adjacent organs is suspected, radical surgery should be performed as to allow a prediction of prognosis through an exact determination of disease stage, and to improve the survival rate.

Synthesis of Ceramic Support for Immobilization of Microorganisms Using Fly Ash (석탄회를 이용한 미생물 고정화 세라믹 담체 제조)

  • Porous ceramic supports with immobilized microorganisms for the water purifier were synthesized by firing green compacts of mixed powder comprising of fly ash, bentonite and an additive of yeast powder at 800∼1,000$^{\circ}C$ for 1h and the pore and mechanical properties of specimens were investigated. The compressive strength was increased in FB (Fly Ash + Bentonite) specimens while pore properties was decreased with increasing the bentonite content and sintering temperature. The compressive strength, bulk density, apparent density, porosity, mean pore size, pore volume and specific surface area of FB specimens at 800∼1,000$^{\circ}C$ were 89.6∼128.9 kgf/$cm^2$, 1.25∼1.43, 1.61∼1.78, 27.2∼62.2%, 7.9∼25.6 ${\mu}m$, 8.9∼$22.2{\times}10^{-5}\;cm^3/g$ and 35.2∼134.3 $m^2/g$, respectively. The pore properties of FBY (FB+yeast powder) specimens were superior to that of FB specimens, however compressive strength was decreased with increasing yeast powder content. The overall properties of 9F1B1Y (9F1B+10% of yeast powder) specimens at 900$^{\circ}C$ for 1 h were 98.7 kgf/$cm^2$, 1.20, 1.67, 68.1%, 48.9 ${\mu}m$, $29.5{\times}10^{-5}\;cm^3/g$ and 152.2 $m^2/g$, respectively. In this study, it was revealed that 9F1B1Y specimen demonstrated better S. saprophyticus adherence properties n their surface pores. Consequently, the microorganisms immobilized on porous ceramic supports showed better water purifying performance with many pores and adequate strength.
