A Case study on the Effects of Mathematically Gifted Creative Problem Solving Model in Mathematics Learnings for Ordinary students (수학 영재의 창의적 문제해결 모델(MG-CPS)을 일반학생의 수학 학습에 적용한 사례연구)

  • This research is a case study of the changes of students's problem solving ability and affective characteristics when we apply to general students MG-CPS model which is creative problem solving model for gifted students. MG-CPS model which was developed by Kim and Lee(2008) is a problem solving model with 7-steps. For this study, we selected 7 first grade students from girl's high school in Seoul. They consisted of three high level students, two middle level students, and two low level students and then we applied MG-CPS model to these 7 students for 5 weeks. From the study results, we found that most students's describing ability in problem understanding and problem solving process were improved. Also we observed that high level students had improvements in overall problem solving ability, middle level students in problem understanding ability and guideline planning ability, and that low level students had improvements in the problem understanding ability. In affective characteristics, there were no significant changes in high and middle level classes but in low level class students showed some progress in all 6 factors of affective characteristics. In particular, we knew that the cause of such positive changes comes from the effects of information collection step and presenting step of MG-CPS model.

  • 황색포도상구균이 Capsular polysaccharide (CPS)는 백혈구의 탐식에 저항하기 때문에 젖소의 유방염 발생의 중요한 병원상 인자로 오랫동안 연구자들의 관심이 집중되어 왔다. 따라서 CPS는 젖소의 유방염을 최소화시키기 위한 유방염 백신개발에 있어 잠재적인 백신 항원물질로 알려져 왔다. 본 연구의 복적은 황색포도상구균의 CPS 항원물질을 좀더 효과적으로 발현시키고 향후 백신항원으로 사용시 좀 더 간편하게 정제하는 방법을 찾아보고자 본 연구를 수행하였다. CPS는 락토스와 덱스트로스를 첨가한 brain heart infusion media 에서도 부분적으로 발현되었지만 CPS가 가장 잘 발현되는 배지 조건은 젖소 유래의 유청을 10% 첨가한 조건에서 가장 발현율이 높았다. 또한 CPS발현은 salt agglutination test, india ink법 및 투과전자현미경을 이용하여 확인하였으며 균체표면의 hydrophobicity에서는 락토스, 덱스트로스 및 젖소 유래의 유청을 첨가한 배지조건에서 다른 조건에 비해 높았다. CPS의 가장 손쉬운 검사법은 균체를 절편하지 않고 직접 투과전자현미경상에서 염색없이 관찰하는 것이 가장 간편한 방법이었다. CPS정제는 상층액을 농축시킨후 이온교환크로마토그라피와 젤 여과법을 이용하여 정제하였으며 CPS의 분자량은 대부분 97kDa이상이었다. CPS 회수율은 배지와 균체 1리터당 1.5mg이었다. 결론적으로 CPS를 백신항원으로 포함하고자 할 경우, CPS의 최적 발현조건은 젖소 유래의 유청을 10% 첨가한 Brain heart infusion 배지에서 배양하여 사용하는 것이 가장 좋은 방법이라고 생각된다.

  • Carbamoyl phosphate synthetase 1 (CPS1) deficiency is an autosomal recessive urea cycle disorder which causes hyperammonemia. CPS1 is the first enzyme step in the urea cycle and almost patients present their symptoms during neonatal period. We report a case of CPS1 deficiency in a boy who developed symptoms including lethargy and seizure at 3 days of age. The ammonia level was up to $2,325{\mu}mol/L$, sodium benzoate (250 mg/kg/d) and high calories of both dextrose and lipid was promptly administered. Central access by experienced pediatric surgeon and emergent continuous hemodialysis by pediatric nephrologist was performed within 3 hours and ammonia was less than $100{\mu}mol/L$ at 5 days of age. Currently, he has showed excellent response to treatments including scavenging drugs and a low-protein diet. Despite of diffuse increasing signal intensity on cerebral white matters and basal ganglia on brain MRI, his development and weight gain were good at the last follow-up at 11 months of age. Molecular assay of the CPS1 gene demonstrated that patient had compound heterozygous for c.1529del ($p.Gly510Alafs^*5$) in exon 14 and c.3142-1G>C (IVS25(-1)G>C) in intron 25 and exon 26 boundary. The splicing mutation was novel mutation and inherited from patient's mother. Here, we report a neonatal lethal type CPS1 deficiency patient having novel mutation.

Optimization of Capsular Polysaccharide Production by Streptococcus pneumoniae Type 3

  • Response surface methodology (RSM) examining the effects of five-level-three-factors and their mutual interactions was utilized to optimize the fermentation conditions to enhance capsular polysaccharide (CPS) production of Streptococcus pneumoniae type 3. Twenty experiments conducted in an 8-l lab-scale fermentor were designed to assess fermentation pH, supplemented glucose concentration, and stirring rate. The predicted highest CPS production by the obtained optimization model equation was 256.14 mg/l at optimal conditions [pH 7.5, stirring rate 180 rpm, and supplemented glucose concentration 1% (w/v)]. The validity of the response model was confirmed by the good agreement between the predicted and experimental results. The maximum amount of CPS obtained was $255.03\pm2.23$ mg/l.

Effects of Concurrent Administration of Sopunghwalhyeol-tang and Clopidogrel on Atherosclerosis in the $ApoE^{(-/-)}$ Mouse (동맥경화증이 유발된 $ApoE^{(-/-)}$ mouse에서 소풍활혈탕(疎風活血湯)과 Clopidogrel의 병용투여 효과에 대한 연구)

  • Background and Objective: Atherosclerosis is a diffuse, systemic disease that affects the coronary, cerebral, and peripheral arterial trees. Clopidogrel is widely used antiplatelet agent and its efficacy has been proven in cardiac and extracardiac vascular diseases, but it has several side effects. Therefore we investigated whether Sopunghwalhyeoltang, which is widely used for treating the blood stasis syndrome in traditional medicine, could decrease the side effect of antiplatelets and have a synergic effect. Methods & Materials: Male $ApoE^{(-/-)}$ mice were randomly divided into three different experimental groups, non-treated group (Control group), clopidogrel-treated group (CP group) and clopidogrel with Sopunghwalhyeol-tang treated group (CPS group). The control group was fed with only an atherogenic diet, the CP group an atherogenic diet plus clopidogrel 25mg/kg and the CPS group an atherogenic diet plus clopidogrel 25mg/kg with Sopunghwalhyeol-tang 100 mg/kg. We investigated plasma lipids with liver function test, and performed a histological investigation of liver and abdominal aorta. Results: 1. Photomicrographs of liver and abdominal aorta tissue showed lower histological injury and lipid accumulation in the CP and CPS groups than those in the Control group. 2. In the CPS group, plasma triglyceride level was significantly lower than in the Control and CP groups. 3. In the CPS group, the plasma aspartate aminotransferase (AST) level was significantly lower than in the CP group. Conclusions: The above results shows that a combined treatment of Sopunghwalhyeol-tang and clopidogrel have a synergic effect through inhibiting vessel injury and decrease the side effects of clopidogrel alone.

Dependence of Hardness Change on Microstructure during Isothermal Aging in Mg-Al Alloy (Mg-Al 합금에서 등온 시효 중 경도 변화의 미세조직 의존성)

  • This study is intended to clarify the main microstructural factors that contribute to an increase of hardness during isothermal aging in Mg-Al alloy. For this work, Mg-9.3%Al alloy specimens were solution-treated at 688 K for 24 h followed by water quenching, and then aged at 473 K for up to 24 h. The aging at 473 K yielded nodular discontinuous precipitates (DPs) with (${\alpha}+{\beta}$) lamellar morphology at the grain boundaries, and the volume fraction of DPs increased from 0% to ~30% with increasing aging time up to 12 h. For the aging times longer than 12 h, further formation of DPs was substantially inhibited owing to the occurrence of significant continuous precipitation within the ${\alpha}-(Mg)$ matrix, and the density of continuous precipitates (CPs) becomes greater with increasing aging time. Hardness of the specimen was steadily increased with aging time up to 24 h. Microstructural examination on the aged specimens revealed that the increased overall hardness at the early stage of aging is associated with the increased volume fraction of DPs, but at the later stage of aging, where the amount of DPs was hardly changed, the increased hardness of the ${\alpha}-(Mg)$ matrix in response to the higher density of CPs within the matrix, plays a key role in increasing the overall hardness value.

  • Glass composed of $Li_2$O.MgO. $MgF_2$.$SiO_2$.$B_2O_3$ for the fabrication of green sheet was prepared by melting process, and glass ceramics was prepared by the process of nucleation and grystal growth for the glass of $Li_2$O.MgO. $MgF_2$.$SiO_2$.$B_2O_3$ system with Lithium fluorhectorite and Lithium boron fluorphlogopite crystal phase. Powderization of the glass ceramics was carried out by water swelling. The average particle size at this point was 2.574 $\mu\textrm{m}$. Slurry was prepared for green sheet using high viscous sol fabricated by water swelling, which shows cleavage phenomenon in prepared glass ceramics. The optimum ratio of powder to water for the tape casting was 18:100, and its viscosity was 11,000~14,000 cps.

Effect of Combined Addition of Ca and Y on Aging Behavior of Extruded AZ91 Magnesium Alloy (Ca과 Y 복합 첨가가 AZ91 마그네슘 압출재의 시효 거동에 미치는 영향)

  • The purpose of this study is to investigate the effects of combined addition of Ca and Y on the precipitation and age-hardening behavior of an extruded AZ91 alloy by conducting the aging treatment at 200 ℃ for hot-extruded AZ91 and AZ91-0.3Ca-0.2Y alloys. In the AZ91 alloy, many Mg17Al12 discontinuous precipitate (DP) bands formed during air cooling immediately after extrusion are present, whereas in the AZ91-0.3Ca-0.2Y alloy, a few DP bands and numerous Al2Y, Al8Mn4Y, and Al2Ca phase particles are distributed along the extrusion direction. The peak-aging time of the AZ91-0.3Ca-0.2Y alloy is 16 hours, twice that of the AZ91 alloy. Although both alloys have similar hardness before aging treatment, the hardness after peak-aging treatment (i.e., peak hardness) of the AZ91-0.3Ca-0.2Y alloy is higher than that of the AZ91 alloy, as 93.1 and 88.7 Hv, respectively. The microstructures of both peak-aged alloys comprise DPs and continuous precipitates (CPs). However, the peak-aged AZ91-0.3Ca-0.2Y alloy has a smaller amount of DPs and a larger amount of CPs than the peak-aged AZ91 alloy. Additionally, the inter-particle spacings of DPs and CPs in the former are significantly narrower than those in the latter. These results demonstrate that the addition of small amounts of Ca and Y to a commercial AZ91 alloy considerably affects the formation rate, size, and amount of CPs and DPs during aging and resultant age-hardening behavior.

Changes in Phosphatase Activities of Mouse Epididymal Spermatozoa during Maturation (생쥐 부정소 정자의 성숙과정에서 Phosphatase 활성도 변화)

  • The change of phosphatase activities of the epididymal spermatozoa has been examined during epididymal maturation in mouse. The quantitative analysis of phQsphatase activities have been carried out using the method modified by Emst(1975). The results of experiment were summarized as the followings. Total protein of the caput epididyrnal spermatozoa(CPS) was measured as 59.1 $\pm$8.4(mg/10 9 spermatozoa), and that of the cauda epididymal spermatozoa(CDS) was 14.0$\pm$12.3(mg/10 9 spermatozoa). When phosphatase activities of the CDS in basic reaction medium were 29.2% in alkaline phosphatase, 44.9% in ATPse and 53.8% in acid phosphatase. The activities were eminently decreased in all CDS in contrast to those of CPS. The alkaline phosphatase and ATPase activities of K+ -dependent were decreased in CDS when compared with caput epididymal spermatozoa, and alkaline phosphatase, ATPase and acid phosphatase activities of $Ca^2$+ -dependent were increased in homogenized spermatozoa when compared with intact spermatozoa. From these results, it may be concluded that the decrease of phosphatases activities in spermatozoa during epididymal maturation may play some significant roles in acquiring fertilizing capability.

Cytotoxic Effect of Isolated Protein-bound Polysaccharides from Hypsizigus marmoreus Extracts by Response Surface Methodology (반응표면분석에 의한 해송이버섯(Hypsizigus marmoreus) 추출물 중 단백다당체의 암세포 성장억제효과)

  • This study used response surface methodology (RSM) in an effort to optimize the water extraction conditions of Hypsizigus marmoreus in order to increase cytotoxicity activity of the extract. A central composite design was applied to investigate the effects of independent variables, which included the extraction temperature ($X_1$), extraction time ($X_2$), the ratio of solvent to sample ($X_3$) on dependent variables of the extracts, including extraction yield ($Y_1$) and protein content ($Y_2$). The estimated optimal conditions were as follows: $51.3^{\circ}C$ extraction temperature, 8.2 hrs extraction time, and 46.7 mL/g of solvent per sample. The extract (CE) was extracted at optimal condition and crude polysaccharides (CPS) were obtained from CE by ethanol precipitation, dialysis, and freeze drying. Neutral (NPS) and acidic (APS) fraction of polysaccharides were seperated from CPS by ion chromatography. The growth inhibitory effects of the APS (0.5 mg/mL) on AGS human cancer cells were 73.97%. CPS showed the highest growth inhibitory effects on HepG2 human cancer cell at 0.5 mg/mL. However all fraction polysaccharides from Hypsizigus marmoreus showed lower than 20% growth inhibition on SW480 human cancer cell.
