Studies on the Fermentative Production of Inosine-5′-monophosphate by Microorganisms (Part 1) Derivation of 5′-IMP Producing Mutants from Brevibacterium ammoniagenes (미생물에 의한 5′-이노신산의 생산에 관한 연구 (제1보) 5′-이노신산 생산균주의 분리)

  • As the first step of domestic developmint of the nucleic acid-related compounds, purine base required auxotrophs from Brevibacterium ammoniagenes ATCC 6872 were derived by the ultraviolet irradiation or the treatment of N-methyl-N'-nitro-N-nitroso guanidine (NTG), diethyl sulfate (DES), and ethylme-thyl sulfate (EMS). The optimum conditions of mutation by means of several mutagens were induced respectively. The yield of mutants was 0.083% by the ultraviolet irradiation, 0.67% by the NTG treatment, 1.1% by the DES treatment, and 0.45% by the EMS treatment. Six strains among 239 auxotrophs were screened out to accumulate 5'-lMP in the culture broth. Cry-stalline 5'-lMP was isolated from the culture broth of Brevifbacterium ammoniagenes adnine-guanine less mutant D-21530 by the use of anion exchange resin, Amberlite IRA-402, and it was identified physically and chemically as 5'-inosinic acid.

  • The water pollution by waste water is one of the important issue and the short of animal feed is too, in Korea. So, this experiment is accomplished to treat alcohol distillers' waste by micro-organisms and planning to produce yeasts, which can be used as animal feed, pharmacy and condiments. 1. The raw material, alcohol distillers' waste, of this experiment consists of insoluble solids (residue) and filterate (supernatant). The residue contains 33.08% of crude protein, 19.96% of total sugar. and 2.06% of ash, respectively. On the other hand the flterate through the Toyo filter paper No. 5C, contains 2.48% of crude protein, 1.54% of reducing sugar, and 0.43% of ash, respectively. 2. Optimum pH of the basal medium for the growth of Saccharomyces cerevisiae YF-1 is 4.0. Optimum culture condition of this is as follows : when 0.43g of urea, 0.43g of potassium phosphate monobasic, and 0.21g of magnesium sulfate are added to the 100m1 of basal medium. Optimum temperature and optimum incubation time are 30$^{\circ}C$ and 24-28 hrs. 3. Under these conditions, the maximum yield of dry yeast is 1.38% to the medium. 4. The composition of dry yeast, produced under these conditions, is as follows: crude protein, 56.96%, lipid, 1.30%. total sugar, 6.53%, and ash 9.62%.

FACTORS INVOLVED IN THAWING OF FROZEN ALASKA POLLACK AND REFREEZING OF THE FILLET (명태 FILLET 제조를 위한 냉동원료의 해동방법과 가공품의 재동결방법에 관한 연구)

  • Alaska pollack caught in the Northern Pacific Ocean and frozen aboard vessel are skipped to the plant and processed into frozen fillets. In the present paper quality changes during thwaing, refreezing and storage at $-20^{\circ}C$ are discussed. Natural, running-water, vacuum and steam thawing were employed as thawing methods. And contact plate, air blast, immersion in dry ice-alcohol solution freezing and storage at $-5^{\circ}C$ were applied to refreeze the thawed fillets. As quality factors content of drip released, salt-extractable protein, VBN, DNA in the drip and pH were determined. In addition, bacteriological tests were also carried out along with the whole process. In thawing of round material, the vacuum thawing was more effective than any other method, resulting in drip, salt-extractable protein $(N\%)$, VBN and DNA as $4.4\%,\;1.82\%,\;16.21mg\%$ and $13.70\;{\mu}g/ml$, respectively. Storage at $-5^{\circ}C$ as refreezing method yielded lower in drip and DNA content but similar to or slightly higher in both salt-extractable protein and VBN, which might postulate that the quality of the frozen fillet depends not largely on the secondary freezing but on the conditions of thawing and primary freezing. It seemed that most of the bacterial flora in thawed fillet came from skin and viscera of fish, worker's hands, utensils and other processing facilities, since sanitary indicative bacteria were not detected in the frozen flesh of round Alaska pollack. Bacterial quality of fillet varied with thawing methods, vacuum thawing appeared more sanitative compared with other methods as natural, running-water, and steam thawing. Bacterial colonies isolated from the thawed fillet were composed of $73.8\%$ Gram negative rod shape, $4.9\%$ Gram positive rod shape, $18.0\%$ cocci, and $3.3\%$ yeast. Decreasing rate of coliform group of the fillet during the storage at $-20^{\circ}C$ for 30 days was more than $70\%$ and that of plate count was less than of coliform group.

Assessment of VOCs Emission Characteristics from Building Materials such as Wall Paper, Paints, and Adhesives Using Small Chamber Method (소형챔버법을 이용한 건축자재 중 벽지, 페인트 및 접착제의 VOCs 방출특성 평가)

  • Building and furnishing materials and consumer product are important sources of volatile organic compounds(VOCs) and other aldehydes in the indoor environment. Some available evidence indicates that VOCs can cause adverse health effects to the building occupants and contribute to some of the symptoms of what we call, 'Sick House Syndrome' in Korea. The aims of this study were to evaluate the efficiency of emission system and to investigate comparison of the emission characteristics of different building materials such as wall-papers, paints, and adhesives. The emission of VOCs from building materials were determined in the small chambers defining the temperature, relative humidity, and ventilation rate in this study. VOCs were sampled for 20 minutes using Tenax-TA tubes and analysed by GC-MS with thermal desorption. The stability of conditions for temperature and relative humidity in this small chamber system showed that the fluctuation of temperature was between 25.4$\pm$0.3$^{\circ}C$ and that of relative humidity was 50.2$\pm$0.6$\%$ under the airflow rate of 167 mL/min. The emission tests from building materials resulted in TVOC emission rates of 0.011 $\~$ 3.108 mg/m$^{2}$h after 7 days. The general wall-papers emitted toluene abundantly and the natural wall-papers mainly emitted n-butanol and a minor amount of alkanes compound such as n -tetradecane. The remainder consisted of toluene, m,p -xylene, and styrene. The paints mainly emitted toluene and the adhesives mainly emitted chloroform as well as toluene. As a result, this study is expected to suggest meaningful data for future studies in exposure control through selecting healthy building materials and for the establishment of guidelines for various building materials in Korea.

Development of a Fennel (Foeniculum vulgare) Oil-based Anti-insect Sachet to Prevent the Indian Meal Moth (Plodia interpunctella) (화랑곡나방 유충 방제를 위한 회향오일 기반 방충향낭 개발)

  • The stored-product insects have been a serious problem during the entire process of distribution on the food industry. Especially, the Indian meal moth (Plodia interpunctella) is one of stored-product insects which causes harm through penetrating into the food packaging. The objective of this study was to develop the anti-insect packaging material with fennel (Foeniculum vulgare) oil (FO). The FO has been selected for insecticidal substance against P. interpunctella, which was tested by fumigant toxicity assay. An anti-insect sachet was prepared by FO and filterpaper placed in a small paper bag. Repellent test was performed to evaluate the repellent activity of anti-insect sachet. In addition, the controlled release of FO from the anti-insect sachet was determined at $28^{\circ}C$ by gas chromatography (GC). It was demonstrated that FO was an effective substance against P. interpunctella. The mortality of FO was 56% at 800 mg/0.5 L in 120 h. In repellent test, the FO sachet showed effective repellency against P. interpunctella. The developed anti-insect sachet could be a promising source for insect repellent materials in food packaging.

Hydrolysis of Agricultural Residues and Kraft Pulps by Xylanolytic Enzymes from Alkaliphilic Bacillus sp. Strain BK

  • An alkaliphilic bacterium, Bacillus sp. strain BK, was found to produce extracellular cellulase-free xylanolytic enzymes with xylan-binding activity. Since the pellet-bound xylanase is eluted with 2% TEA from the pellet of the culture, they contain a xylan-binding region that is stronger than the xylan-binding xylanase of the extracellular enzyme. The xylanases had a different molecular weight and xylan-binding ability. The enzyme activity of xylanase in the extracellular fraction was 6 times higher than in the pellet-bound enzyme. Among the enzymes, xylanase had the highest enzyme activity. When Bacillus sp. strain BK was grown in pH 10.5 alkaline medium containing xylan as the sole carbon source, the bacterium produced xylanase, arabinofuranosidase, acetyl esterase, and $\beta$-xylosidase with specific activities of 1.23, 0.11, 0.06, and 0.04 unit per mg of protein, respectively. However, there was no cellulase activity detected in the crude enzyme preparation. The hydrolysis of agricultural residues and kraft pulps by the xylanolytic enzymes was examined at 50$^{\circ}C$ and pH 7.0. The rate of xylan hydrolysis in com hull was higher than those of sugarcane bagasse, rice straw, com cop, rice husk, and rice bran. In contrast, the rate of xylan hydrolysis in sugarcane pulp was 2.01 and 3.52 times higher than those of eucalyptus and pine pulp, respectively. In conclusion, this enzyme can be used to hydrolyze xylan in agricultural residues and kraft pulps to breach and regenerate paper from recycled environmental resources.

Preparation of Functional Cosmetics Containing $\beta$-carotene Derived from Recombinant Escherichia coli and Evaluation of Anti-wrinkle Efficacy by Clinical Testing (재조합 대장균 유래 베타카로틴이 함유된 기능성 화장품 제조 및 이의 임상실험을 통한 주름개선 효능 평가)

  • This paper described the formulation of functional cosmetics and evaluation of anti-wrinkle efficacy in clinical test. Cosmetics were formulated with highly purified $\beta$-carotene obtained from the culture broth of recombinant E.coli cells. Edible oil for solubilizing $\beta$-carotene, vitamine E for long-term storage, detergent/stabilizer (2.0%) for the complete formation of oil/water emulsion, dimethicone (0.35%) for good skin care, and sorbitol for skin moisturizer were also added as ingredients. Physical or chemical degradation of formulated products stored at $0^{\circ}C$, $25^{\circ}C$, or $40^{\circ}C$ was not observed for 60-day testing period. In clinical test, 68% of applicants observed wrinkle decrease after 8-week treatment. This result indicates that newly formulated cosmetics have strong potential for improving wrinkle skin care.

  • In succession to the previous paper, the effect of the addition of food additives, such as glucose, glycine, sucrose and sorbic acid on the histamine formation and histidine decarboxylase activity in mackerel muscle during storage at $25^{\circ}C$ was studied. Additionally, the effect of different rasping condition of mackerel muscle on the histamine formation was also studied. It was estimated that the rasping condition of mackerel muscle also affected to the histamine formation, by showing that much histamine was detected in homogenized muscle than in ground muscle when glucose and sucrose were added. The addition of glycine was inhibitory upon the histamine formation and histidine decarboxylase activity, which, in the muscle added $10\%$ of glycine, the histamine content was below the critical concentration of poisoning for histamine during storage for 5 days at $25^{\circ}C$. The addition of sorbic acid was also inhibited the histamine formation and histidine decarboxylase activity, and the inhibitory effect of $0.2\%$ addition was greater than $0.1\%$ addition.

  • Nitrogen compounds are one of the major pollutants which cause eutrophication problems of the river or lake and red tides problems of the ocean. Currently available technologies for the removal of nitrogen compounds are mostly biological treatment. However, biological treatment is only effective for the wastewater which contains low concentration of nitrogen compounds. Leachate from solid waste landfill or industrial wastewater which contains high concentration of nitrogen can not be effectively treated by most of the currently available biological treatment technologies. With this connection. the objective of this study is to examine the applicability of ammonia stripping technology for the removal of high concentration of ammonia nitrogen compounds of the leachate from solid waste landfill. It can be concluded that ammonia stripping technology which was placed before the biological treatment process was very effective for the removal of high concentration of ammonium compounds. The chemical cost for the ammonia stripping was 16 percent higher than MLE process, so other methods like sludge recycling are needed for the reduction of operation cost. Further details are discussed in this paper.

Persistent Organic Pollution and Arsenic Contamination in Asia Pacific Water: Case Study of Emerging Environmental Problems in Vietnam

  • This paper provides a comprehensive overview of the present status of several environmental problems caused by emerging toxic substances such as persistent organic pollutants (POPs), endocrine disrupting chemicals (EDCs), and arsenic in various environmental media in Vietnam. Monitoring data reported during the 1990s demonstrated elevated contamination of DDTs in most of these compartments in Vietnam. Studies in frame of the Asia-Pacific Mussel Watch Program revealed that fish, mussels and resident birds from Vietnam contained higher concentrations of DDTs as compared to other countries in region, suggesting the role of Vietnamese environment as a significant emission source of DDT in the Southeast Asian region. The estimated dietary intakes of PCBs and DDTs for Vietnamese were relatively high among Asian developing countries, suggesting potential risk for humans posed by thesechemicals. Widespread contamination of some endocrine active compounds such as alkylphenols and phthalates was observed at various sites along the coasts of northern and middle Vietnam. The presence of significant source of bisphenol-A along Red River estuary was revealed with the concentrations comparable to those reported for developed nations. A case study on seasonal variation of alkylphenols and phthalates in surface water of river delta and estuary of north and middle Vietnam indicated the differences in distribution of these compounds between dry and rainy seasons. Higher concentrations of alkylphenols and phthalates were found in dry season in estuary; while the contrasting pattern was observed in the river delta, showing elevated residues in rainy season. This result suggests the different behavior of alkylphenols and phthalates in river delta and coastal environment. From ecotoxicological perspectives, concentrations of bis-phenol A and di(2-ethylhexyl)phthalates [DEHP] in surface water from some locations in Vietnam exceeded the guideline values for Ecotoxicological Effects and the Environmental Risk Limit, respectively, suggesting potential for toxic implications on aquatic wildlife. Widespread and elevated arsenic contamination was discovered inour recent surveys in groundwater in a large area of suburban areas of Hanoi city, the capital of Vietnam. The most recent investigation in 4 villages showed about more than 50 % of groundwater samples contained As concentrations exceeding 50 g/L (the WHO and Vietnamese standard). In particular, in Son Dong villages, 58 % of samples analyzed contained As concentrations higher than 200 g/L. Good correlations were found in As concentrations in water and hair and urine of peoples in corresponding families, suggesting the chronic exposure to As by people living in As-contaminated ground water areas. In Son Dong village, As levels in hair (mean: 1.7 mg/kg dry wt) and urine (g/g creatinine) exceeding the reference values recommended by WHO, suggesting potential for human risk posed by long term accumulation of As in human body. Future studies should be focused on the time trends of POPs and EDCs in biota in Vietnam in order to predict future trend of contamination and to reveal new clues for understanding possible toxic impacts on aquatic organisms. The issues of arsenic contamination in groundwater and their chronic toxic implications on human health should be systematically investigated in the future.
