The button mushroom (Agaricus bisporus) is one of the most widely cultivated important edible mushroom species. In the breeding of new button mushroom, 'Seolgang' was developed by crossing two monokaryons 'CM020913-27' and 'SSU423-31'. Because of the secondarily homothallism, only a small percentage of the basidia produce 3 or 4 spores, which are mostly haploid (n) and do not fruit. Single spore cultures derived from these types of spores produce a vegetative mycelium that also contain a variable number of genetically identical nuclei per cell called monokaryon. The lack of clamp connections between monokaryon and dikaryon required a series of mycelial culture and fruiting test. After crossing, hybrids were cultivated on a small scale and on a commercial scale at a farm. For this, the spawn was made by a commercial spawn producer and the spawned compost by a commercial compost producer. Mycelial growth of 'Seolgang' on CDA was better at $20^{\circ}C$ and $25^{\circ}C$ when it was compared with that of '505 Ho'. The mature cap shape of new strain 'Seolgang' is oblate spheroid and the immature cap shape is round to oblate spheroid. The cap diameter was 41.2 mm on average. In comparison with white strain '505 Ho', the strain had a yield that was 9% higher. It produced fruiting bodies which had a higher weight on average per fruiting body and were 19% firmer with a good shelf life. Days of fruiting body were 3-4 days later than those of '505 Ho'. The physical characteristics such as elasticity, chewiness, adhesiveness were better than that of '505 Ho'. Genetic analysis of the new strain 'Seolgang' showed different profiles compared to '505 Ho', CM02913-27, SSU413-31, when RAPD primers A02 and O04 were used.
'Gonji10ho', a new variety of Neolentinus lepideus, was bred by mating two monokaryotic strains isolated from 'GMNL66037' and 'GMNL66014' in Mushroom Research Institute, Gyonggi-Do ARES in 2012. The optimum temperature for the mycelial growth of 'Gonji10ho' was $29^{\circ}C$ on PDA medium and that for the primordia formation and the growth of fruiting body of 'Gonji10ho' was $20{\pm}1^{\circ}C$. In bottle cultivation of 'Gonji10ho', the period of spawn running was around 35 days at $22{\pm}1^{\circ}C$ and the period from scratching of inoculum to harvesting was 27 days. 'Gonji10ho' had thicker pileus and stipe than control variety('Solhyang'). The number of available stipe was fewer than that of control variety. During the cultivation of 'Conji10ho', the occurrence rate of Trichoderma spp. was lower than that in control variety. In bottle cultivation, the yield of fruiting bodies of 'Gonji10ho', which was 38% higher than that of control variety, was 69.6 g/900 ml bottle, and in bag cultivation, that of 'Gonji10ho', which was 10% higher than that of control variety, was 114.2 g/1 kg bag. In physical characteristics, the strongness and brittleness of the pileus of 'Gonji10ho' were higher than those of control variety. When considered in the period available for sale, the shelf life of 'Gonji10ho', which was 5 days longer than that of control variety, was 34 days.

A new variety of Pleurotus eryngii which is named 'Gonji No. 8' was bred by mating monokaryotic strain isolated from E085D2 and a monokaryotic strain 'aerini No.3' obtained from the Mushroom Research Institute, Gyeonggi-Do A.R.E.S. The characteristics of the new variety 'Gonji No.8' is as follows. The optimum temperature for mycelium growth was from $23^{\circ}C$ to $26^{\circ}C$ on potato dextrose agar (PDA) medium. For the primodia formation and the growth of fruit bodies, the optimum temperature was from $14^{\circ}C$ to $16^{\circ}C$. The period of spawn running was around 30 days at $22^{\circ}C$ and the days taken after the removal of the spawn layer to initiate primodia was seven days. The hardness value of fruit body was $8,432{\pm}2,193$$g/cm^2$, which was two times more than that of 'Keunneutari No.2'. The yield of 'Gonji No.8' was about 133 g per bottle(900cc) and it was same as 'Keunneutari No.2'.
Flammulina velutipes, known as winter mushroom or Enokitake, is an important edible mushroom in Korea. In 2017, approximately 28,000 tons of this mushroom were produced in Korea, putting its output at the third place, behind the oyster and king oyster mushrooms. Despite its high production, F. velutipes has the lowest distribution rate of domestic cultivars, estimated to be about 28 percent. To increase the income of farms and provide more options to consumers, we developed a new golden domestic cultivar 'Auram'. 'Auram' was bred by mating two monokaryotic strains isolated from ASI 4103 and ASI 4111. Auram has bright golden, hemispheric shaped caps, and light brown stipes, making it strikingly different from the control. The optimum mycelial growth temperature was 25℃, but Auram mostly displayed higher mycelial growth than the control in a wide range of temperatures. The yield of Auram (257.4 ± 13.5 g) was high, and the period of fruit body growth was also a couple of days ahead in bottle cultivation, compared with the control. Auram received a more favorable evaluation from farms and distributors than the control for its appearance, because the fruit body of Auram had an attractive golden color, and the spotted cap often seen in the control was not observed.
The inky cap, Coprinellus congregatus, produces several laccase isozymes during its life cycle: both hyphal tip laccase and sclerotial laccase are involved in the fungal development. When this fungus was transferred to an acid liquid medium (pH 4.0-4.5), a new laccase was synthesized and secreted into the culture supernatant. In order to examine its regulation by external pH, green fluorescent protein gene was ligated at the downstream of the promoters having different lengths. These expression vectors having different promoter lengths were inserted into the fungal transformation vector, pBARGEM7-1. These expression vectors were introduced to the mating type a1 and a2 monokaryons, and the transformants were selected by the phosphinothricin resistance. Transformant a1 (a1TF) and transformant a2 (a2TF) were mated with each other to generate homozygotic dikaryon transformants. All these transformants were grown in neutral liquid medium for 5 days, and then the whole cell homogenates were transferred to the acidic liquid medium (pH 4.1). After 36 h incubation at $25^{\circ}C$, cells were harvested for the analysis of GFP expression. GFP expression was detected in the transformant having full-length promoter (2.0 kb), but other transformants having shorter length promoter (shorter than 1.29 kb) failed to show the fluorescence. Therefore, the acid-responsive element in the laccase promoter should be localized between -2.0 kb ~ -1.29 kb region.
Four gilded bronze objects and a gilded silver object were examined for elemental diffusion between the gilding layer and the mating matrix. The gilded bronzes consist of three objects from three different historical periods, the Korean Three Kingdoms period, the Koryo and the Choseon periods and one from an unknown period. The gilded silver was from the Koryo period. The amalgam process seems to have been the major technique employed for all of them in gilding. The occurrence of substantial diffusion was observed in all but the anonymous object, particularly in the Choseon artifact where evidence was found that the diffusion phenomenon was intentionally utilized in gilding. The gold content in the gilded bronzes decreases gradually from surface to interior while the copper content increases to the interior, making it difficult to locate the boundary between the gilding layer and the matrix. This gradual change in composition must have resulted from elemental diffusion at elevated temperatures. The oxygen content negligible in the gilding layer precludes the possibility of corrosion being responsible for the varying composition. It is observed that non-uniform diffusion caused variation of colors in the surface of gilded bronzes. The change of colors induced by diffusion, which is always accompanied by the unique surface morphology and chemical compositions, is distinguished from the color change by corrosion. In the gilded silver object, diffusion of mercury was observed along with that of gold and silver.
This study was conducted to analyze the transgenic efficiency and sex ratio in ${\alpha}$-1,3-galactosyltransferase (GalT) knock-out (KO) transgenic pigs according to generation. GalT KO piglets were produced by artificial insemination or natural mating. The transgenic confirmation of GalT KO was evaluated by PCR amplification using specific primers. After electrophoresis, three types of bands were detected such as 2.3 kb single band (Wild), 2.3 and 3.6kb double bands (GalT KO -/+; heterozygote), and 3.6kb single band (GalT KO -/-; homozygote). Transgenic efficiency in F1 generation was 64.5% (23/35) of GalT KO (-/+). In F2 generation, GalT KO transgenic efficiency was 36.4% (21/57, Wild), 47.5% (28/57, GalT KO -/+), and 16.1% (8/57, GalT KO -/-), respectively. Interestingly, no homozygote piglets were born in 6 deliveries among total 11 deliveries, although they were pregnant between male (M) and female (F) $F_1$ heterozygote. In the 5 litters including at least one GalT KO -/- piglet, the transgenic efficiency was 13.3% (2/24, Wild), 51.3% (14/24, GalT KO -/+), and 35.3% (8/24, GalT KO -/-), respectively. The sex ratio of M and F was 40:60 in $F_1$ and 49:51 in $F_2$ generation, respectively. Based on these results, GalT KO transgenic pigs have had a reproductive ability with a normal range of transgenic efficiency and sex ratio.

Young Jong Grand Bridge is approach traffic road of New Inchon International Airport which covers hub airport function in northeast asia. The total span length of this bridge is $4,420{\cal}m$ and this main bridge type is, the first in the world, Double Deck Self Anchored Suspension Bridge, designed as double deck systems to be arranged by road and railroad. Approach bridges to be connected with main span also are composed double deck steel truss and steel box girder to consider a continuity with this span. Our company erected $1,375{\cal}m$(about 60,000tons) of double deck steel truss bridge type which is composed by 6 traffic lane on upper deck and 4 traffic lane and Double track railroad on lower deck. The original installation method of this bridge was planed to install about 75 meters bridge blocks to use floating crane, after temporary bent was constructed between permanent piers. But this method which had to construct many temporary bents in the sea had the matter that construction periods can become lengthen and construction cost can be risen. To overcome the uncertainty to ensure high qualify of bridge and economic project execution, our company developed new bridge erection method to assure both quality control and economic construction work. The new erection method which was developed by us was one that could transport and install long bridge block, $120{\cal}m$ unit at a time and that temporary bent was not required. We hope that this paper is used as technical data which will erect bridge in the western sea and others marine region.
Deutzia paniculata Nakai is a Korean endemic species that has a very restricted distribution in Gyeongsang-do, South Korea. The genetic diversity and structure of five populations of D. paniculata were investigated using 31 ISSR loci from six primers. The Shannon's index (0.429) and genetic diversity (0.271) were relatively higher than those of other rare plant species in Korea. The Miryang (MY) and Yangsan (YS) populations, which have higher flowering rates than the other populations, showed greater genetic diversity than the other populations. An analysis of the molecular variance (AMOVA) showed that 16% of the total variation could be attributed to differences among the populations, and 84% to the differences within populations, indicating moderate gene flow among adjacent populations. The high genetic diversity and low genetic differentiation in the Deutzia paniculata populations, which have a restricted distribution, is considered to be affected by outcrossing of the mating system and abundant individuals in the populations. These results suggest that ex situ conservation strategies are needed to sustain the current genetic diversity of D. paniculata.
This study was conducted to examine the effect of salinity on the early embryonic development stage of boreal digging frogs (Kaloula borealis). An experiment was carried out with the eggs of K. borealis mating couples laid along the coastal zone of Jeju Island in June 2013. The salinity was set to 0, 1, 3, 5, and 7 ‰, respectively. The survival and growth of the larvae at various stages from the egg phase to the closing of their external gills phase were observed. In another experiment, tadpoles in internal gill stage were placed in solutions with salinity of 0, 1, 3, 5, and 7 ‰, respectively, and their survival rate and growth rate were observed, and then the results of the two experiments above were compared. The results showed that in egg and external gill stage, there was no significant difference in survival rate in 3 ‰ solution, though there was low survival rate in 5 ‰ solution. Further, all the eggs were dead in 7 ‰ solution, so it appeared that solutions with salinity of 5 ‰ or higher affect the survival of K. borealis in the early embryonic development. Larvae in the internal gill stage showed no significant difference in survival rate from the control group up to a salinity of 5 ‰, but when placed in a solution of over 7 ‰ salinity, the survival rate decreased. The growth rate also slowed down with the higher salinity. It appeared that in both the external gill stage and the internal gill stage in 5 ‰ solution, the growth rate decreased significantly compared to the control group. From this study it is concluded that higher salinity values have a significant impact on the survival and growth of the K. borealis larvae and this finding can be used to conserve K. borealis species that are decreasing in number due to the side effects of development in coastal areas.
